Identifying the gene responsible for non‐photochemical quenching reversal in Phaeodactylum tricornutum

SUMMARY Algae such as diatoms and haptophytes have distinct photosynthetic pigments from plants, including a novel set of carotenoids. This includes a primary xanthophyll cycle comprised of diadinoxanthin and its de‐epoxidation product diatoxanthin that enables the switch between light harvesting and non‐photochemical quenching (NPQ)‐mediated dissipation of light energy. The enzyme responsible for the reversal of this cycle was previously unknown. Here, we identified zeaxanthin epoxidase 3 (ZEP3) from Phaeodactylum tricornutum as the candidate diatoxanthin epoxidase. Knocking out the ZEP3 gene caused a loss of rapidly reversible NPQ following saturating light exposure. This correlated with the maintenance of high concentrations of diatoxanthin during recovery in low light. Xanthophyll cycling and NPQ relaxation were restored via complementation of the wild‐type ZEP3 gene. The zep3 knockout strains showed reduced photosynthetic rates at higher light fluxes and reduced specific growth rate in variable light regimes, likely due to the mutant strains becoming locked in a light energy dissipation state. We were able to toggle the level of NPQ capacity in a time and dose dependent manner by placing the ZEP3 gene under the control of a β‐estradiol inducible promoter. Identification of this gene provides a deeper understanding of the diversification of photosynthetic control in algae compared to plants and suggests a potential target to improve the productivity of industrial‐scale cultures.

Green diatom mutants reveal an intricate biosynthetic pathway of fucoxanthin

Fucoxanthin is a major light-harvesting pigment in ecologically important algae such as diatoms, haptophytes, and brown algae (Phaeophyceae). Therefore, it is a major driver of global primary productivity. Species of these algal groups are brown colored because the high amounts of fucoxanthin bound to the proteins of their photosynthetic machineries enable efficient absorption of green light. While the structure of these fucoxanthin-chlorophyll proteins has recently been resolved, the biosynthetic pathway of fucoxanthin is still unknown. Here, we identified two enzymes central to this pathway by generating corresponding knockout mutants of the diatom Phaeodactylum tricornutum that are green due to the lack of fucoxanthin. Complementation of the mutants with the native genes or orthologs from haptophytes restored fucoxanthin biosynthesis. We propose a complete biosynthetic path to fucoxanthin in diatoms and haptophytes based on the carotenoid intermediates identified in the mutants and in vitro biochemical assays. It is substantially more complex than anticipated and reveals diadinoxanthin metabolism as the central regulatory hub connecting the photoprotective xanthophyll cycle and the formation of fucoxanthin. Moreover, our data show that the pathway evolved by repeated duplication and neofunctionalization of genes for the xanthophyll cycle enzymes violaxanthin de-epoxidase and zeaxanthin epoxidase. Brown algae lack diadinoxanthin and the genes described here and instead use an alternative pathway predicted to involve fewer enzymes. Our work represents a major step forward in elucidating the biosynthesis of fucoxanthin and understanding the evolution, biogenesis, and regulation of the photosynthetic machinery in algae.

Investigating the dislocation reactions on Σ3{111} twin boundary during deformation twin nucleation process in an ultrafine-grained high-manganese steel

Abstract Some of ultrafine-grained (UFG) metals including UFG twinning induced plasticity (TWIP) steels have been found to overcome the paradox of strength and ductility in metals benefiting from their unique deformation modes. Here, this study provides insights into the atomistic process of deformation twin nucleation at Σ3{111} twin boundaries, the dominant type of grain boundary in this UFG high manganese TWIP steel. In response to the applied tensile stresses, grain boundary sliding takes place which changes the structure of coherent Σ3{111} twin boundary from atomistically smooth to partly defective. High resolution transmission electron microscopy demonstrates that the formation of disconnection on Σ3{111} twin boundaries is associated with the motion of Shockley partial dislocations on the boundaries. The twin boundary disconnections act as preferential nucleation sites for deformation twin that is a characteristic difference from the coarse-grained counterpart, and is likely correlated with the lethargy of grain interior dislocation activities, frequently seen in UFG metals. The deformation twin nucleation behavior will be discussed based on in-situ TEM deformation experiments and nanoscale strain distribution analyses results.

A correlation between grain boundary character and deformation twin nucleation mechanism in coarse-grained high-Mn austenitic steel

Abstract In polycrystalline materials, grain boundaries are known to be a critical microstructural component controlling material’s mechanical properties, and their characters such as misorientation and crystallographic boundary planes would also influence the dislocation dynamics. Nevertheless, many of generally used mechanistic models for deformation twin nucleation in fcc metal do not take considerable care of the role of grain boundary characters. Here, we experimentally reveal that deformation twin nucleation occurs at an annealing twin (Σ3{111}) boundary in a high-Mn austenitic steel when dislocation pile-up at Σ3{111} boundary produced a local stress exceeding the twining stress, while no obvious local stress concentration was required at relatively high-energy grain boundaries such as Σ21 or Σ31. A periodic contrast reversal associated with a sequential stacking faults emission from Σ3{111} boundary was observed by in-situ transmission electron microscopy (TEM) deformation experiments, proving the successive layer-by-layer stacking fault emission was the deformation twin nucleation mechanism, different from the previously reported observations in the high-Mn steels. Since this is also true for the observed high Σ-value boundaries in this study, our observation demonstrates the practical importance of taking grain boundary characters into account to understand the deformation twin nucleation mechanism besides well-known factors such as stacking fault energy and grain size.

Nonmonotonic crossover in electronic phase separated manganite superlattices driven by the superlattice period

Studying manganite superlattices [(LCMO) 2n / (PCMO) n ] t made of La 0.625 Ca 0.375 MnO 3 (LCMO) and Pr 0.625 Ca 0.375 MnO 3 (PCMO), we found an unexpected behavior varying the period n. At small n, the ensemble is a three-dimensional ferromagnetic metal due to interfacial charge transfer. At large n , the LCMO layers dominate transport. However, rather than a smooth interpolation between these limits a sharp transport and magnetic anomaly is found at an intermediate critical PCMO thickness n*. Magnetic force microscopy reveals that the phase-separation length scale also maximizes at n* where, unexpectedly, it becomes comparable to that of the (La 1-y Pr y ) 0.625 Ca 0.375 MnO 3 (LPCMO) alloy. Finally, we conjecture the phenomenon originates in a disorder-related length scale: Large charge-ordered clusters as in LPCMO can only nucleate when Pr-rich regions reach a critical size related to n*.

Effect of dietary fiber fermentation on short‐chain fatty acid production and microbial composition in vitro

Abstract BACKGROUND The efficient utilization of fiber‐rich co‐products is important for optimizing feed resource utilization and animal health. This study was conducted to evaluate the fermentation characteristics of fiber‐rich co‐products, which had equal quantities of total dietary fiber (TDF), at different time points using batch in vitro methods. It considered their gas production, short‐chain fatty acid (SCFA) production, and microbial composition. RESULTS The fermentation of wheat bran (WB) and oat bran (OB) showed higher and faster ( P < 0.05) gas and SCFA production than corn bran (CB), sugar beet pulp (SBP), and soybean hulls (SH). The α ‐diversity was higher in the CB, SBP, and SH groups than in the WB and OB groups ( P < 0.05). At the phylum level, OB and WB fermentation showed lower ( P < 0.05) relative abundance of Actinobacteria than the CB, SBP, and SH groups. At the genus level, OB and WB fermentation increased the Enterococcus population in comparison with the CB, SBP, and SH groups, whereas CB and SBP fermentation improved the relative abundance of the Christensenellaceae R‐7 group more than the WB, OB, and SH groups ( P < 0.05). CONCLUSION Overall, WB and OB were rapidly fermented by fecal microbiota, in contrast with SBP, SH, and CB. Fermentation of different fiber‐rich co‐products with an equal TDF content gives different responses in terms of microbial composition and SCFA production due to variations in their physicochemical properties and molecular structure. © 2020 Society of Chemical Industry

Modeling the effect of temperature gradient on moisture and ionic transport in concrete

Most reinforced concrete structures are under the influence of environmental conditions, such as temperature fluctuations, moisture and chloride variations simultaneously. Thus, the moisture transfer in concrete is driven by the moisture gradient as well as the temperature gradient. Similarly, the ionic transfer in concrete is driven by the concentration gradient of the ion as well as the temperature gradient. This paper developed an analytical solution that can be used for the moisture or the chloride transport in concrete considering the effect of temperature gradient. The effect of mass transfer on the thermal conduction was not considered, so the problem is simplified as a one-way coupled problem. The analytical model was validated by using two CASE studies, in which realistic material parameters of concrete were used, the parameters were obtained from experiments in the literature. The comparisons between analytical results and experimental results showed good agreement. It indicated that the present model can successfully characterize the effect of temperature gradient on moisture transfer and the effect of temperature gradient on chloride transport in concrete.

Integrated Approach to Density-Based Spatial Clustering of Applications with Noise and Dynamic Time Warping for Breakout Prediction in Slab Continuous Casting

Mold breakout is a catastrophic accident that has serious impacts on smooth production, slab quality, and caster equipment. Accurate identification and prediction of an impending breakout are always top priorities in continuous casting operations. In view of crucial common features of mold copper plate temperatures during a breakout, such as time lag and space inversion, the concepts of density-based spatial clustering of applications with noise and dynamic time warping are introduced, and an integrated novel method for breakout prediction is developed. Through extracting and fusing the representative singularity and approximation of temperature variation, the typical temporal and spatial temperature characteristics during breakout can be distinguished and predicted accurately. Compared with traditional methods of logical judgment and artificial neural network, the method based on clustering does not need to modify forecast thresholds or parameters artificially, which overcomes the limitation of model dependence on human beings, and demonstrates excellent adaptability and robustness for online abnormality prevention.

Precision Higgs physics at the CEPC

The discovery of the Higgs boson with its mass around 125 GeV by the ATLAS and CMS Collaborations marked the beginning of a new era in high energy physics. The Higgs boson will be the subject of extensive studies of the ongoing LHC program. At the same time, lepton collider based Higgs factories have been proposed as a possible next step beyond the LHC, with its main goal to precisely measure the properties of the Higgs boson and probe potential new physics associated with the Higgs boson. The Circular Electron Positron Collider~(CEPC) is one of such proposed Higgs factories. The CEPC is an $e^+e^-$ circular collider proposed by and to be hosted in China. Located in a tunnel of approximately 100~km in circumference, it will operate at a center-of-mass energy of 240~GeV as the Higgs factory. In this paper, we present the first estimates on the precision of the Higgs boson property measurements achievable at the CEPC and discuss implications of these measurements.

Observing a previously hidden structural-phase transition onset through heteroepitaxial cap response

Significance We provide an approach to study structural transitions by using appropriate heteroepitaxial caps. Previously unobservable phase transitions that involve small changes in lattice constants, but may have important impact on the physical properties of the material, can now be detected in real space. Using the coherent nanotwinning waves of epitaxial La 0.7 Sr 0.3 MnO 3 /SrTiO 3 thin films as a sensitive detector, we are able to uncover the onset stage of the cubic-to-tetragonal structural-phase transition of SrTiO 3 in real space. Researchers involved in the study of material properties as well as technologists in a variety of industries, such as electronics, will greatly benefit from this approach.

Machine Learning Applied to Star–Galaxy–QSO Classification and Stellar Effective Temperature Regression

In modern astrophysics, machine learning has increasingly gained popularity with its incredibly powerful ability to make predictions or calculated suggestions for large amounts of data. We describe an application of the supervised machine-learning algorithm, random forests (RF), to the star/galaxy/QSO classification and the stellar effective temperature regression based on the combination of Large Sky Area Multi-Object Fiber Spectroscopic Telescope and Sloan Digital Sky Survey spectroscopic data. This combination enables us to obtain reliable predictions with one of the largest training samples ever used. The training samples are built with a nine-color data set of about three million objects for the classification and a seven-color data set of over one million stars for the regression. The performance of the classification and regression is examined with validation and blind tests on the objects in the RAdial Velocity Extension, 6dFGS, UV-bright Quasar Survey and Apache Point Observatory Galactic Evolution Experiment surveys. We demonstrate that RF is an effective algorithm, with classification accuracies higher than 99% for stars and galaxies, and higher than 94% for QSOs. These accuracies are higher than machine-learning results in former studies. The total standard deviations of the regression are smaller than 200 K, which is similar to those of some spectrum-based methods. The machine-learning algorithm with the broad-band photometry provides us with a more efficient approach for dealing with massive amounts of astrophysical data than do traditional color cuts and spectral energy distribution fits.

A Versatile Integrated Ambient Ionization Source Platform

The pursuit of high-throughput sample analysis from complex matrix demands development of multiple ionization techniques with complementary specialties. A versatile integrated ambient ionization source (iAmIS) platform is proposed in this work, based on the idea of integrating multiple functions, enhancing the efficiency of current ionization techniques, extending the applications, and decreasing the cost of the instrument. The design of the iAmIS platform combines flowing atmospheric pressure afterglow (FAPA) source/direct analysis in real time (DART), dielectric barrier discharge ionization (DBDI)/low-temperature plasma (LTP), desorption electrospray ionization (DESI), and laser desorption (LD) technique. All individual and combined ionization modes can be easily attained by modulating parameters. In particular, the FAPA/DART&DESI mode can realize the detection of polar and nonpolar compounds at the same time with two different ionization mechanisms: proton transfer and charge transfer. The introduction of LD contributes to the mass spectrometry imaging and the surface-assisted laser desorption (SALDI) under ambient condition. Compared with other individual or multi-mode ion source, the iAmIS platform provides the flexibility of choosing different ionization modes, broadens the scope of the analyte detection, and facilitates the analysis of complex samples. .

Unexpected Intermediate State Photoinduced in the Metal-Insulator Transition of Submicrometer Phase-Separated Manganites

At ultrafast timescales, the initial and final states of a first-order metal-insulator transition often coexist forming clusters of the two phases. In this work, we report an unexpected third long-lived intermediate state emerging at the photoinduced first-order metal-insulator transition of La 0.325 Pr 0.3 Ca 0.375 MnO 3 , known to display submicrometer length-scale phase separation. Using magnetic force microscopy and time-dependent magneto-optical Kerr effect, we determined that the third state is a nanoscale mixture of the competing ferromagnetic metallic and charge-ordered insulating phases, with its own physical properties. This discovery bridges the two different families of colossal magnetoresistant manganites known experimentally and shows for the first time that the associated states predicted by theory can coexist in a single sample.
