The recently developed technique of gas chromatography-mass spectrometry supported by computer has considerably improved the analysis of physiologic fluids. This study attempted to demonstrate the value of this system in the investigation of metabolite patterns in urine in two metabolic problems of prematurity, transient tyrosinemia and late metabolic acidosis. Serial 24-hr urine specimens were analyzed in 9 infants. Transient tyrosinemia, characterized by 5- 10-fold increases over basal excretion of tyrosine, p-hydroxyphenyllactate, and p-hydroxyphenylpyruvate in urine, was noted in five of the infants. Late metabolic acidosis was seen in four infants, but bore no relation to transient tyrosinemia.
A large suite of aliphatic dicarboxylic acids was found in extracts from the carbonaceous chondrite. This suite includes many of the branched and straight chained isomers with up to nine carbon atoms. The large number of compounds and the isomeric distribution suggest that the acids are the product of an abiotic synthesis. The dicarboxylic acids found have been identified by their gas chromatography retention values and by comparison of their mass spectra with those of authentic standards.
Description of a computerized method for the simultaneous quantitation of 12 amino acids in biological fluids using the technique of quadrupole mass in fragmentography. The time taken for one complete analysis, exclusive of derivatization, is 40 minutes for data collection with an additional 10 minutes before the computer presents the final analytical result.
A mass fragmentographic technique for a rapid, specific and sensitive determination of ethanol in blood and urine is described. A Varian gas chromatograph coupled through an all-glass membrane separator to a Finnigan quadripole mass spectrometer and interfaced to a computer system is used for ethanol determination in blood and urine samples. A procedure for plotting calibration curves for ethanol quantitation is also described. Quantitation is achieved by plotting the peak area ratios of undeuterated-to-deuterated ethanol fragment ions against the amount of ethanol added. Representative results obtained by this technique are included.

The mass spectra of six aurone epoxides have been rationalized with the aid of high resolution mass spectrometry and metastable ion evidence. These compounds fragment in a well defined manner and mechanisms are proposed for the formation of their characteristic ions. Some similarity was observed between the mass spectra of 6-methoxyaurone epoxide (II), 4-hydroxy-7-methoxy-3-phenylcoumarin (VII) and 7-methoxyflavonol (IX). The possibility that VII and IX are intermediates in the fragmentation of epoxide II is discussed. Thermal rearrangement of aurone epoxide II was shown to yield the corresponding flavonol IX and coumarin VII.
A specific and sensitive method for the identification and simultaneous quantitation by mass fragmentography of ten of the amino acids present in soil was developed. The technique uses a computer driven quadrupole mass spectrometer and a commercial preparation of deuterated amino acids is used as internal standards for purposes of quantitation. The results obtained are comparable with those from an amino acid analyzer. In the quadrupole mass spectrometer-computer system up to 25 pre-selected ions may be monitored sequentially. This allows a maximum of 12 different amino acids (one specific ion in each of the undeuterated and deuterated amino acid spectra) to be quantitated. The method is relatively rapid (analysis time of approximately one hour) and is capable of the quantitation of nanogram quantities of amino acids.

Mass spectra of alkyl carbamates derived from primary, secondary, and teriary alcohols by use of deuterium labeling and high resolution mass spectroscopy
A sensitive primary amine assay, capable of detecting 10 to the minus 11th g and utilizing the determination of the amine N-2,4-dinitrophenyl derivative by electron-capture gas chromatography is described. The method is exemplified by the determination of cyclohexylamine in sodium cyclamate.
The mass spectra of twenty alkyl carbamates derived from primary, secondary, and tertiary alcohols are investigated, using deuterium labeling and high-resolution mass spectrometry. Several of the primary and secondary alcohol derivatives are found to yield an ion formally equivalent to the product ion of a McLafferty rearrangement. Deuterium labeling, however, revealed a lack, in these carbamate derivatives, of the usual site specificity associated with the McLafferty rearrangement. A double hydrogen rearrangement process was observed in the mass spectra of several carbamates derived from tertiary alcohols.
Description of the DENDRAL (Dendritic Algorithm) project, the objectives of which were to base the computer program on an alogorithm that generates an exhaustive, nonredundant list of all the structural isomers of a given chemical composition, and to devise a computer program that would perform an organic structure determination, given a molecular formula and a mass spectrum. This program is called 'Heuristic DENDRAL' and it operates by using the known structure/spectrum correlations to constrain the DENDRAL isomer generator to produce a single isomer for that composition. The collaboration of chemists and computer scientists has produced a tool of some practical utility from the chemical viewpoint, and an interesting program from the viewpoint of artificial intelligence.
