The voltage-dependence of MscL has dipolar and dielectric contributions and is governed by local intramembrane electric field

Channels without canonical voltage sensors can be modulated by voltage acting on other domains. Here we show that besides protein dipoles, pore hydration can be affected by electric fields. In patches, both WT MscL and its V23T mutant show a decrease in the tension midpoint with hyperpolarization. The mutant exhibits a stronger parabolic dependence of transition energy on voltage, highly consistent with the favourable dielectric contribution from water filling the expanding pore. Purified V23T MscL in DPhPC droplet interface bilayers shows a similar voltage dependence. When reconstituted in an asymmetric DOPhPC/DPhPC bilayer carrying a permanent bias of ~130mV due to a dipole potential difference between the interfaces, the channel behaved as if the local intramembrane electric field sets the tension threshold for gating rather than just the externally applied voltage. The data emphasize the roles of polarized water in the pore and interfacial lipid dipoles in channel gating thermodynamics.

Dielectric relaxation behavior of poly(acrylonitri1e-co-methacrylonitrile) microcapsules dispersed in a silicone matrix

The dielectric relaxation behavior of poly(acry1onitrile-co-methacrylonitrile) dispersed in a cured polydimethyl siloxane (PDMS) matrix as microcapsules was investigated over multiple thermal cycles and at varying concentrations. The copolymer microcapsules contained an isopentane core. In the PDMS matrix this copolymer displayed a pronounced relaxation signal at temperatures above the glass transition of the copolymers due to Maxwell-Wagner-Sillars (MWS) relaxation. The mechanism of MWS relaxation interpreted by the Havriliak-Negami and Kohlrausch-Williams-Watts relaxation functions was found to be very similar to previous studies of neat polyacrylonitrile and its copolymer. The activation energy of the relaxation decreased over successive thermal cycling coincident with a decreasing strength of the relaxation. These observations were attributed to the decreasing concentration of nitrile groups due to intramolecular cyclizations.

Efficient Hybrid Actuation Using Solid-State Actuators

Piezohydraulic actuation is the use of fluid to rectify the motion of a piezoelectric actuator for the purpose of overcoming the small stroke limitations of the material. In this work we study a closed piezohydraulic circuit that utilizes active valves to rectify the motion of a hydraulic end affector. A linear, lumped parameter model of the system is developed and correlated with experiments. Results demonstrate that the model accurately predicts the filtering of the piezoelectric motion caused by hydraulic compliance. Accurate results are also obtained for predicting the unidirectional motion of the cylinder when the active valves are phased with respect to the piezoelectric actuator. A time delay associated with the mechanical response of the valves is incorporated into the model to reflect the finite time required to open or close the valves. This time delay is found to be the primary limiting factor in achieving higher speed and greater power from the piezohydraulic unit. Experiments on the piezohydraulic unit demonstrate that blocked forces on the order of 100 N and unloaded velocities of 180 micrometers/sec are achieved.

Project Cerberus: Flyby Mission to Pluto

The goal of the Cerberus Project was to design a feasible and cost-effective unmanned flyby mission to Pluto. The requirements in the request for proposal for an unmanned probe to Pluto are presented and were met. The design stresses proven technology that will avoid show stoppers which could halt mission progress. Cerberus also utilizes the latest advances in the spacecraft industry to meet the stringent demands of the mission. The topics covered include: (1) mission management, planning, and costing; (2) structures; (3) power and propulsion; (4) attitude, articulation, and control; (5) command, control, and communication; and (6) scientific instrumentation.
