SARS-CoV-2 Infection Severity Is Linked to Superior Humoral Immunity against the Spike

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is currently causing a global pandemic. The antigen specificity of the antibody response mounted against this novel virus is not understood in detail. Here, we report that subjects with a more severe SARS-CoV-2 infection exhibit a larger antibody response against the spike and nucleocapsid protein and epitope spreading to subdominant viral antigens, such as open reading frame 8 and nonstructural proteins. Subjects with a greater antibody response mounted a larger memory B cell response against the spike, but not the nucleocapsid protein. Additionally, we revealed that antibodies against the spike are still capable of binding the D614G spike mutant and cross-react with the SARS-CoV-1 receptor binding domain. Together, this study reveals that subjects with a more severe SARS-CoV-2 infection exhibit a greater overall antibody response to the spike and nucleocapsid protein and a larger memory B cell response against the spike.

Nonmonotonic crossover in electronic phase separated manganite superlattices driven by the superlattice period

Studying manganite superlattices [(LCMO) 2n / (PCMO) n ] t made of La 0.625 Ca 0.375 MnO 3 (LCMO) and Pr 0.625 Ca 0.375 MnO 3 (PCMO), we found an unexpected behavior varying the period n. At small n, the ensemble is a three-dimensional ferromagnetic metal due to interfacial charge transfer. At large n , the LCMO layers dominate transport. However, rather than a smooth interpolation between these limits a sharp transport and magnetic anomaly is found at an intermediate critical PCMO thickness n*. Magnetic force microscopy reveals that the phase-separation length scale also maximizes at n* where, unexpectedly, it becomes comparable to that of the (La 1-y Pr y ) 0.625 Ca 0.375 MnO 3 (LPCMO) alloy. Finally, we conjecture the phenomenon originates in a disorder-related length scale: Large charge-ordered clusters as in LPCMO can only nucleate when Pr-rich regions reach a critical size related to n*.

3D-Printed Membranes with a Zwitterionic Hydrogel Coating for More Robust Oil–Water Separation

Three-dimensional (3D)-printed membranes via stereolithography (SLA) are promising in oil–water separation, which is the key in the purification of industrial oily wastewater. To achieve gravity-driven oil–water separation, the membrane material needs to be simultaneously hydrophilic/oleophobic. However, most of the state-of-the-art materials for SLA do not meet the requirement. While water-adsorbing hydrogel is simultaneously hydrophilic/oleophobic and there have been reports on 3D printing of hydrogels in biomedical applications, the hydrogel is too soft for membrane application. Here, we report a simple approach to tackle the issue: a hydrogel coating on SLA-based plastic membranes. The coating is fabricated, using [2-(methacryloyloxy) ethyl] dimethyl-(3-sulfopropyl) ammonium hydroxide as the zwitterionic monomer and acrylamide as the comonomer, via in situ polymerization on SLA-based plastic membranes. The contact angle tests and Fourier transform infrared spectrum show that such a membrane readily adsorbs water and becomes simultaneously hydrophilic/oleophobic. The oil–water separation tests indicate that the water-adsorbed membrane is highly efficient in gravity-driven oil–water separation in 31 repeating cycles. Our results indicate the great potential of 3D-printed membranes in oil–water separation.

Structural basis for substrate recognition and chemical inhibition of oncogenic MAGE ubiquitin ligases

Testis-restricted melanoma antigen (MAGE) proteins are frequently hijacked in cancer and play a critical role in tumorigenesis. MAGEs assemble with E3 ubiquitin ligases and function as substrate adaptors that direct the ubiquitination of novel targets, including key tumor suppressors. However, how MAGEs recognize their targets is unknown and has impeded the development of MAGE-directed therapeutics. Here, we report the structural basis for substrate recognition by MAGE ubiquitin ligases. Biochemical analysis of the degron motif recognized by MAGE-A11 and the crystal structure of MAGE-A11 bound to the PCF11 substrate uncovered a conserved substrate binding cleft (SBC) in MAGEs. Mutation of the SBC disrupted substrate recognition by MAGEs and blocked MAGE-A11 oncogenic activity. A chemical screen for inhibitors of MAGE-A11:substrate interaction identified 4-Aminoquinolines as potent inhibitors of MAGE-A11 that show selective cytotoxicity. These findings provide important insights into the large family of MAGE ubiquitin ligases and identify approaches for developing cancer-specific therapeutics.

Nucleon pair shell model in $\textit{M}$ scheme

The nucleon pair shell model (NPSM) is cast into the so-called $\textit{M}$ scheme for the cases with isospin symmetry and without isospin symmetry. The odd system and even system are treated on the same footing. The uncoupled commutators for nucleon pairs, which are suitable for the $\textit{M}$ scheme, are given. Explicit formula of matrix elements in $\textit{M}$ scheme for overlap, one-body operators, and two-body operators are obtained. In this work, it is found that the CPU time used in calculating the matrix elements in $\textit{M}$ scheme is much shorter than that in the $\textit{J}$ scheme of NPSM.

3D-Printed Repeating Re-Entrant Topography to Achieve On-Demand Wettability and Separation

While it is difficult and expensive to fabricate a complicated surface structure via conventional techniques, three-dimensional (3D) printing serves as a time-efficient and cost-efficient alternative. In the current study, a novel repeating re-entrant topography is fabricated by two-photon polymerization 3D printing. The experimental results show that the repeating re-entrant surface enhances the desired on-demand surface wettability. Furthermore, the 3D-printed membranes with the repeating re-entrant structures enable the efficient on-demand separation of liquid mixtures with high flux, which is critical for the wastewater treatment in the chemical industry.

Joining of Thermoset Carbon Fiber Reinforced Polymer and AZ31 Magnesium Alloy Sheet Via Friction Stir Interlocking

A new technique called friction stir interlocking (FSI) was applied for lap-joining of AZ31 magnesium sheet and thermoset carbon fiber reinforced polymer (TS-CFRP) sheet. Instead of directly joining AZ31 and TS-CFRP, a series of magnesium interlocks were used to friction stir weld with AZ31 sheet to enable joining with TS-CFRP. Microstructural characterization of joint cross sections showed that a tool rotation rate of 800 rpm produced incomplete mixing of magnesium interlocks and the top AZ31 sheet, while 1200 rpm achieved sufficient material mixing to avoid unwelded regions. Mechanical property characterization showed that AZ31/TS-CFRP dissimilar joints failed through the magnesium interlock shanks during lap shear tensile testing, with a maximum normalized joint strength reaching ~100 MPa.

Bioinformatic and experimental evidence for suicidal and catalytic plant THI4s

Like fungi and some prokaryotes, plants use a thiazole synthase (THI4) to make the thiazole precursor of thiamin. Fungal THI4s are suicide enzymes that destroy an essential active-site Cys residue to obtain the sulfur atom needed for thiazole formation. In contrast, certain prokaryotic THI4s have no active-site Cys, use sulfide as sulfur donor, and are truly catalytic. The presence of a conserved active-site Cys in plant THI4s and other indirect evidence implies that they are suicidal. To confirm this, we complemented the Arabidopsistz-1 mutant, which lacks THI4 activity, with a His-tagged Arabidopsis THI4 construct. LC–MS analysis of tryptic peptides of the THI4 extracted from leaves showed that the active-site Cys was predominantly in desulfurated form, consistent with THI4 having a suicide mechanism in planta. Unexpectedly, transcriptome data mining and deep proteome profiling showed that barley, wheat, and oat have both a widely expressed canonical THI4 with an active-site Cys, and a THI4-like paralog (non-Cys THI4) that has no active-site Cys and is the major type of THI4 in developing grains. Transcriptomic evidence also indicated that barley, wheat, and oat grains synthesize thiamin de novo, implying that their non-Cys THI4s synthesize thiazole. Structure modeling supported this inference, as did demonstration that non-Cys THI4s have significant capacity to complement thiazole auxotrophy in Escherichia coli. There is thus a prima facie case that non-Cys cereal THI4s, like their prokaryotic counterparts, are catalytic thiazole synthases. Bioenergetic calculations show that, relative to suicide THI4s, such enzymes could save substantial energy during the grain-filling period.
