Comparative transcriptomics provides insights into molecular mechanisms of zinc tolerance in the ectomycorrhizal fungus Suillus luteus

Zinc (Zn) is a major soil contaminant and high Zn levels can disrupt growth, survival, and reproduction of fungi. Some fungal species evolved Zn tolerance through cell processes mitigating Zn toxicity, although the genes and detailed mechanisms underlying mycorrhizal fungal Zn tolerance remain unexplored. To fill this gap in knowledge, we investigated the gene expression of Zn tolerance in the ectomycorrhizal fungus Suillus luteus. We found that Zn tolerance in this species is mainly a constitutive trait that can also be environmentally dependent. Zinc tolerance in S. luteus is associated with differences in the expression of genes involved in metal exclusion and immobilization, as well as recognition and mitigation of metal-induced oxidative stress. Differentially expressed genes were predicted to be involved in transmembrane transport, metal chelation, oxidoreductase activity, and signal transduction. Some of these genes were previously reported as candidates for S. luteus Zn tolerance, while others are reported here for the first time. Our results contribute to understanding the mechanisms of fungal metal tolerance and pave the way for further research on the role of fungal metal tolerance in mycorrhizal associations.

A Fully Explicit Integrator for Modeling Astrophysical Reactive Flows

Simulating complex astrophysical reacting flows is computationally expensive—reactions are stiff and typically require implicit integration methods. The reaction update is often the most expensive part of a simulation, which motivates the exploration of more economical methods. In this research note, we investigate how the explicit Runge–Kutta–Chebyshev (RKC) method performs compared to an implicit method when applied to astrophysical reactive flows. These integrators are applied to simulations of X-ray bursts arising from unstable thermonuclear burning of accreted fuel on the surface of neutron stars. We show that the RKC method performs with similar accuracy to our traditional implicit integrator, but is more computationally efficient when run on CPUs.

Biogeochemistry of upland to wetland soils, sediments, and surface waters across Mid-Atlantic and Great Lakes coastal interfaces

Transferable and mechanistic understanding of cross-scale interactions is necessary to predict how coastal systems respond to global change. Cohesive datasets across geographically distributed sites can be used to examine how transferable a mechanistic understanding of coastal ecosystem control points is. To address the above research objectives, data were collected by the EXploration of Coastal Hydrobiogeochemistry Across a Network of Gradients and Experiments (EXCHANGE) Consortium – a regionally distributed network of researchers that collaborated on experimental design, methodology, collection, analysis, and publication. The EXCHANGE Consortium collected samples from 52 coastal terrestrial-aquatic interfaces (TAIs) during Fall of 2021. At each TAI, samples collected include soils from across a transverse elevation gradient (i.e., coastal upland forest, transitional forest, and wetland soils), surface waters, and nearshore sediments across research sites in the Great Lakes and Mid-Atlantic regions (Chesapeake and Delaware Bays) of the continental USA. The first campaign measures surface water quality parameters, bulk geochemical parameters on water, soil, and sediment samples, and physicochemical parameters of sediment and soil.

Assessment of TLE-Based Orbit Determination and Prediction for Cubesats

The IceCube satellite is a 3U cubesat which was deployed from the International Space Station Nanoracks in May 2017. IceCube uses onboard Global Positioning System (GPS) measurements for both science data processing and orbit determination; additionally, IceCube uses Two Line Element (TLE) sets provided by USSTRATCOM through Space Track for ground station acquisition. Once acquired, GPS measurements are downlinked and can be verified against nearby TLE states.TLE sets are typically used for cubesat acquisition data, however the accuracy of their propagation has a limited lifespan which can cause issues when using an older TLE to attempt spacecraft acquisition. This presentation details the merits of TLE-based orbit determination and prediction and discusses techniques for improving both. These refinements allow for improved cubesat acquisition as well as calibration and on-orbit checkout of the GPS. Using IceCube data as representative of a typical cubesat, TLE states are simulated as tracking data and processed through an Extended Kalman Filter. This filtered solution provides not only an ongoing estimate of spacecraft drag state parameters, but also improves the definitive orbit determination estimation as well as the propagation accuracy and therefore spacecraft acquisition. Next, using the definitive TLE-based solution, raw GPS measurements are introduced into the filter where the combined measurement types allow the GPS measurements to be subsequently calibrated with respect to an unknown timing bias. These analyses provide a more accurate state estimation and prediction of cubesats during early orbit operations. Additionally, these techniques showcase methods for TLE-based state estimation and GPS calibration when other methods of tracking are unavailable.

A Web-Based Search Service to Support Imaging Spectrometer Instrument Operations

Imaging spectrometers yield rich and informative data products, but interpreting them demands time and expertise. There is a continual need for new algorithms and methods for rapid first-draft analyses to assist analysts during instrument opera-tions. Intelligent data analyses can summarize scenes to draft geologic maps, searching images to direct op-erator attention to key features. This validates data quality while facilitating rapid tactical decision making to select followup targets. Ideally these algorithms would operate in seconds, never grow bored, and be free from observation bias about the kinds of mineral-ogy that will be found.
