Modular adjuvant-free pan-HLA-DR-immunotargeting subunit vaccine against SARS-CoV-2 elicits broad sarbecovirus-neutralizing antibody responses

Subunit vaccines typically require co-administration with an adjuvant to elicit protective immunity, adding development hurdles that can impede rapid pandemic responses. To circumvent the need for adjuvant in a severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) subunit vaccine, we engineer a thermostable immunotargeting vaccine (ITV) that leverages the pan-HLA-DR monoclonal antibody 44H10 to deliver the viral spike protein receptor-binding domain (RBD) to antigen-presenting cells. X-ray crystallography shows that 44H10 binds to a conserved epitope on HLA-DR, providing the basis for its broad HLA-DR reactivity. Adjuvant-free ITV immunization in rabbits and ferrets induces robust anti-RBD antibody responses that neutralize SARS-CoV-2 variants of concern and protect recipients from SARS-CoV-2 challenge. We demonstrate that the modular nature of the ITV scaffold with respect to helper T cell epitopes and diverse RBD antigens facilitates broad sarbecovirus neutralization. Our findings support anti-HLA-DR immunotargeting as an effective means to induce strong antibody responses to subunit antigens without requiring an adjuvant.

Comparison of Multi-Scale Nonlinear and Conventional Linear Methods for Stress Analysis of Nb 3 Sn Superconducting Magnets

Here this is an extension of the work on a multi-scale nonlinear procedure that demonstrated how to conduct nonlinear stress analysis of Nb 3 Sn superconducting accelerator magnets. The nonlinear procedure uses measured stress-strain curves of annealed copper, Nb 3 Sn strands and coil samples as inputs, which reduces the number of assumptions made for material properties—key uncertainties of any engineering analysis. The results from nonlinear analysis, semi-nonlinear analysis, and linear analysis of the same QFFB2 Nb 3 Sn quadrupole magnet under the same temperature and loads, were compared and discussed. The comparison illustrates that nonlinear stress analysis is significantly more accurate than other methods. As the superconducting coil block is a complex, composite material, it would be inadequate to assume the whole coil block to be linear, either isotropic or orthotropic. It is imperative to conduct nonlinear analysis by simulating the superconducting coil to the level of detail of the individual cables and individual strands. The nonlinear stress analysis procedure can be used to simulate not only Nb 3 Sn magnets, but also NbTi and HTS magnets.

Test Results of Fast Decaying Current-Induced AC Losses in SHMS Superconducting Magnets at Jefferson Lab

The 11 GeV Super High Momentum Spectrometer (SHMS) of Hall C, part of the recent 12 GeV accelerator Upgrade at Jefferson Lab, was successfully commissioned in 2017. Fast current discharges of the SHMS Q2, Q3 quadrupoles and dipole superconducting magnets experienced some level of operational difficulty, during commissioning and normal operation. Measurements and analyses demonstrate that the fast current discharge leads to substantial AC losses in the conductor and subsequently triggers a quench-back effect. The details of the measurements and analyses have been reported previously. This paper focuses on the test results of the magnets using a reduced value of the protection dump resistor for each magnet to lower the current decay rate. Finally, the test results confirmed that the reduced dump resistances eliminated the quench-back effect for the SHMS Q2, Q3 and Dipole magnets, thus improving their cryogenic operability.

Quench-Back Management for Fast Decaying Currents in SHMS Superconducting Magnets at Jefferson Lab

The Super High Momentum Spectrometer (SHMS) of Hall C, part of the 12-GeV upgrade at Jefferson Lab, was successfully commissioned in 2017. Early operation demonstrated that fast current dumps decays of the SHMS Q2/Q3 and dipole superconducting magnets triggered quenches, causing some level of operational difficulty. Tests and detailed assessments suggest that nonquench induced fast current decay could result in substantial ac loss in the conductor and subsequently trigger a quench-back effect. The OPERA/ELECTRA software package was used to calculate the amount of heat deposited in the copper stabilizer from a fast current decay. The resistances of external energy dump resistors were lowered to slow the fast dumping of the magnet's current to reduce or eliminate the quench-back effect. A worst-case adiabatic quench scenario was analyzed, assuming no external dump resistor and no liquid helium surrounding the coil, to ensure the safety of the magnets. The stress levels in the coil imposed by winding, collaring preload, Lorentz force, and temperature gradient during a quench were also examined. The Tsai-Wu material failure criterion was used to determine the acceptable combined stress level. Linear orthotropic analysis of the coil indicates that the magnets can be operated safely with appropriately sized dump resistors. Fast dump tests with the modified dump resistors have been planned to verify the performance and suitability.

Safe assembly and installation of a flywheel

A flywheel device includes structures allowing the flywheel system to be assembled offsite, transported safely, and installed with relatively few steps. The flywheel includes a rotor and a housing enclosing the rotor, where the housing includes a bottom plate, a top plate and side walls. The bottom plate and the top plate each includes a hole aligned with the center axis of the rotor. The flywheel also includes multiple bearing housings substantially covering the holes of the bottom plate and the top plate that are aligned to the center axis of the rotor. The flywheel also includes posts that physically contact the primary rotational mass of the rotor to prevent motion of the rotor during transport of the flywheel system. Some or all of these posts may be repositioned or removed during installation so that the rotor can spin freely.
