Advances in Regioselective Functionalization of Endohedral Metallofullerenes †

Comprehensive Summary Endohedral metallofullerenes (EMFs) are a family of molecules with metal clusters or ions encapsulated inside a fullerene cage. Their unique structures provide a powerful interface for organic chemists to manipulate metal elements. Over a decade ago, many reactions on empty fullerenes were adopted for EMFs, resulting in a series of reactions that are well‐established and reviewed in the literature. In recent years, new topics have emerged in EMF chemistry. First, new addition reactions were developed, notably some that are not based on known empty‐fullerene reactions. Second, considerable progress has been made on regioselective multi‐additions on EMFs. Finally, EMFs have been covalently conjugated with fullerene C 60 to form functional materials. This Recent Advances article will focus on these new developments and discuss their implication for future directions of EMFs.

Interactions between fullerene derivatives and biological systems

Attention towards nanoparticles from the pharmaceutical and biomedical fields has significantly increased due to their attractive surface modification, high drug-loading, and improved pharmacokinetics. Fullerenes, an allotrope of carbon, stand out for their molecularly precise structure, potent radical-scavenging activity, photoactivatable reactive-oxygen species generation, and ability to definitively confine metal atoms and clusters. Accordingly, fullerene systems have been applied in various biological contexts, including increased and controlled drug delivery, antioxidative, anti-inflammatory, and photodynamic therapy, and magnetic resonance imaging. Ultimately, the pleiotropic activity of fullerenes, coupled with its precise structure and functionalization, can realize precise and tailorable medicines. Here, different from some excellent reviews focusing on the structure and chemistry of fullerene derivatives and their biomedical applications, this review highlights the interaction of fullerene materials with biological systems, with insights into their structural influence on their interactions with the cellular environment.

A series of cation-modified robust zirconium-based metal–organic frameworks for carbon dioxide capture

Metal–organic frameworks (MOFs) represent one of the most promising porous solids for possible use as sorbents to control and reduce the greenhouse gas emission. Studies have shown that open metal sites (OMS) interact strongly with carbon dioxide and thus, serve as efficient binding sites for CO 2 capture. However, many OMS-bearing MOFs are lack of framework stability and often have high regeneration temperature. To seek ways to solve the stability issue, we designed a series of isoreticular MOFs, Zr-tcpb-COOM (M = alkali/alkaline earth metal), by exchange of protons with metal ions on Zr-tcpb-COOH via post-synthetic modification (PSM). The pristine MOF (Zr-tcpb-COOH) has a very robust framework. The PSM process does not deteriorate the framework stability but creates metal binding sites that form strong bonds with carbon dioxide. The results show that at low CO 2 pressure, the uptake amount is enhanced considerably using Zr-tcpb-COOM and is in trend of increasing atomic number (Li + < Na + < K + < Ca 2+ ). High adsorption selectivity (CO 2 /N 2 IAST selectivity (15 : 85) = 539.5) is also achieved for CO 2 over N 2 at room temperature. This approach offers a feasible method to improve CO 2 capture capacity, especially at low concentrations.

Structurally Defined Water‐Soluble Metallofullerene Derivatives towards Biomedical Applications

Abstract Endohedral metallofullerenes (EMFs) are excellent carriers of rare‐earth element (REE) ions in biomedical applications because they preclude the release of toxic metal ions. However, existing approaches to synthesize water‐soluble EMF derivatives yield mixtures that inhibit precise drug design. Here we report the synthesis of metallobuckytrio (MBT), a three‐buckyball system, as a modular platform to develop structurally defined water‐soluble EMF derivatives with ligands by choice. Demonstrated with PEG ligands, the resulting water‐soluble MBTs show superb biocompatibility. The Gd MBTs exhibit superior T 1 relaxivity than typical Gd complexes, potentially superseding current clinical MRI contrast agents in both safety and efficiency. The Lu MBTs generated reactive oxygen species upon light irradiation, showing promise as photosensitizers. With their modular nature to incorporate other ligands, we anticipate the MBT platform to open new paths towards bio‐specific REE drugs.

Synthesis of [60]- and [70]Fullerene-Fused Tetrahydroquinoxaline Derivatives by Oxidative [4 + 2] Cycloaddition with Unusual Reactivity and Regioselectivity

The oxidative [4 + 2] reaction of o-phenylenediamine-derived disulfonamides with fullerene C 60 and C 70 is reported, in which electron-deficient reactants showed high reactivity. The reaction of C 70 exhibited unusual regioselectivity, yielding a [5,6]-adduct as the major product, which was characterized by 1 H, 13 C NMR and single-crystal X-ray diffraction. Here, DFT calculations revealed the reaction is an inverse-electron-demand Diels–Alder (IEDDA) reaction, and the [5,6]-adduct of C 70 is a kinetic product.

Multicomponent Reactions Among Alkyl Isocyanides, sp Reactants, and sp 2 Carbon Cages

We explored the reactivity and substrate scope of the reactions among an alkyl isocyanide, an sp-hybridized reactant (i.e. alkyne or allene), and a carbon cage, as a new approach to functionalize fullerenes and metallofullerenes. Furthermore, this account summarizes the key findings in our recent published work, and some original data for the reaction involving an isocyanide, allenes, and metallofullerene Lu 3 N@C 80 .

Isocyanide-Induced Annulation Leading to Cyclopento-, Methano-, and Cyclopentano-[60]Fullerene Derivatives

The three-component annulation reactions of C 60 , alkyl isocyanide, and dimethyl acetylenedicarboxylate (DMAD) or unsymmetric alkynes are investigated to afford cyclopent-2-en-1-imino- and ketenimine methano-[60]fullerene derivatives, which, upon hydration in the presence of acid, yield the corresponding fullerene amides. Dimethyl 2,3-pentadienedioate, the allene counterpart of DMAD, and ethyl buta-2,3-dienoate undergo four-component annulation with C 60 , alkyl isocyanide, and water under similar conditions to yield cyclopentano-[60]fullerene derivatives with similar amide groups.

Unexpected Formation of Metallofulleroids from Multicomponent Reactions, with Crystallographic and Computational Studies of the Cluster Motion

Abstract New multicomponent reactions involving an isocyanide, terminal or internal alkynes, and endohedral metallofullerene (EMF) Lu 3 N@C 80 yield metallofulleroids which are characterized by mass‐spectrometry, HPLC, and multiple 1D and 2D NMR techniques. Single crystal studies revealed one ketenimine metallofulleroid has ordered Lu 3 N cluster which is unusual for EMF monoadducts. Computational analysis, based on crystallographic data, confirm that the endohedral cluster motion is controlled by the position of the exohedral organic appendants. Our findings provide a new functionalization reaction for EMFs, and a potential facile approach to freeze the endohedral cluster motion at relatively high temperatures.
