Metabolic engineering of Caldicellulosiruptor bescii for 2,3-butanediol production from unpretreated lignocellulosic biomass and metabolic strategies for improving yields and titers

ABSTRACT The platform chemical 2,3-butanediol (2,3-BDO) is used to derive products, such as 1,3-butadiene and methyl ethyl ketone, for the chemical and fuel production industries. Efficient microbial 2,3-BDO production at industrial scales has not been achieved yet for various reasons, including product inhibition to host organisms, mixed stereospecificity in product formation, and dependence on expensive substrates (i.e., glucose). In this study, we explore engineering of a 2,3-BDO pathway inCaldicellulosiruptor bescii, an extremely thermophilic (optimal growth temperature = 78°C) and anaerobic bacterium that can break down crystalline cellulose and hemicellulose into fermentable C 5 and C 6 sugars. In addition, C. besciigrows on unpretreated plant biomass, such as switchgrass. Biosynthesis of 2,3-BDO involves three steps: two molecules of pyruvate are condensed into acetolactate; acetolactate is decarboxylated to acetoin, and finally, acetoin is reduced to 2,3-BDO.C. besciinatively produces acetoin; therefore, in order to complete the 2,3-BDO biosynthetic pathway,C. besciiwas engineered to produce a secondary alcohol dehydrogenase (sADH) to catalyze the final step. Two previously characterized, thermostable sADH enzymes with high affinity for acetoin, one from a bacterium and one from an archaeon, were tested independently. When either sADH was present inC. bescii, the recombinant strains were able to produce up to 2.5-mM 2,3-BDO from crystalline cellulose and xylan and 0.2-mM 2,3-BDO directly from unpretreated switchgrass. This serves as the basis for higher yields and productivities, and to this end, limiting factors and potential genetic targets for further optimization were assessed using the genome-scale metabolic model ofC. bescii. IMPORTANCE Lignocellulosic plant biomass as the substrate for microbial synthesis of 2,3-butanediol is one of the major keys toward cost-effective bio-based production of this chemical at an industrial scale. However, deconstruction of biomass to release the sugars for microbial growth currently requires expensive thermochemical and enzymatic pretreatments. In this study, the thermo-cellulolytic bacteriumCaldicellulosiruptor besciiwas successfully engineered to produce 2,3-butanediol from cellulose, xylan, and directly from unpretreated switchgrass. Genome-scale metabolic modeling ofC. besciiwas applied to adjust carbon and redox fluxes to maximize productivity of 2,3-butanediol, thereby revealing bottlenecks that require genetic modifications.

Calcium-mediated nitrogen reduction for electrochemical ammonia synthesis

Ammonia (NH 3 ) is a key commodity chemical for the agricultural, textile and pharmaceutical industries, but its production via the Haber–Bosch process is carbon-intensive and centralized. Alternatively, an electrochemical method could enable decentralized, ambient NH 3 production that can be paired with renewable energy. The first verified electrochemical method for NH 3 synthesis was a process mediated by lithium (Li) in organic electrolytes. So far, however, elements other than Li remain unexplored in this process for potential benefits in efficiency, reaction rates, device design, abundance and stability. In our demonstration of a Li-free system, we found that calcium can mediate the reduction of nitrogen for NH 3 synthesis. Here we verified the calcium-mediated process using a rigorous protocol and achieved an NH 3 Faradaic efficiency of 40 ± 2% using calcium tetrakis(hexafluoroisopropyloxy)borate (Ca[B(hfip) 4 ] 2 ) as the electrolyte. Our results offer the possibility of using abundant materials for the electrochemical production of NH 3 , a critical chemical precursor and promising energy vector

Synergistic effects of climate warming and atmospheric nutrient deposition on the alpine lake ecosystem in the south-eastern Tibetan Plateau during the Anthropocene

Alpine lakes on the Tibetan Plateau are highly sensitive to global change and have been recognized as the sentinel of climate warming. However, anthropogenic impacts in populated area are migrating to these remote areas via transporting particulate nutrients by atmospheric deposition. Whether warming and nutrient deposition would impose additive or synergistic effects on the lake ecosystem remains largely unknown. Here, we present multi-proxy (sediment pigment and geochemistry) records during the past two centuries at the Cuoqia Lake in the southeast Tibetan Plateau. We found that the lake exhibited rapid ecological changes since 1980 AD characterized by an increase in primary productivity due to algal proliferation, with more rapid growth of green algae and diatoms. These findings are in concert with many other lakes (e.g., Moon Lake and Shade Co) in the same area, suggesting a consistent pattern of ecosystem evolution at the region scale. Statistical analyses suggested that nutrient deposition and climate warming were strongly associated with the variations in primary productivity and algae composition, exerting both individual and interactive effects. In addition, scenario analyses with a well-established process-based ecosystem model further revealed that the two factors not only individually, but also synergistically promoted the algal proliferation and community succession. Such synergy is evident in that the effect of lake warming would be more pronounced under higher nutrient deposition scenario, which is potentially due to higher temperature-driven mineralization in warmer conditions, and higher efficiency of nutrient utilization under enhanced light availability attributing to declining ice thickness and duration in cold seasons. Overall, our study proposes the existence and quantifies the synergistic impacts of climate warming and anthropogenic activities in driving the ecological changes in remote alpine lakes on the Tibetan Plateau. The lake ecological consequences driven by individual factor would be worsen by such synergy, so that we cannot predict the lake ecosystem trajectory in the future based on each factor separately, and more efforts than previously expected would be needed for the lake restoration and management.

Biochemical and Regulatory Analyses of Xylanolytic Regulons in Caldicellulosiruptor bescii Reveal Genus-Wide Features of Hemicellulose Utilization

Microbial deconstruction of lignocellulose for the production of biofuels and chemicals requires the hydrolysis of heterogeneous hemicelluloses to access the microcrystalline cellulose portion. This work extends previous in vivo and in vitro efforts to characterize hemicellulose utilization by integrating genomic reconstruction, transcriptomic data, operon structures, and biochemical characteristics of key enzymes to understand the deployment and functionality of hemicellulases by the extreme thermophile Caldicellulosiruptor bescii .

Transcriptional Regulation of Plant Biomass Degradation and Carbohydrate Utilization Genes in the Extreme Thermophile Caldicellulosiruptor bescii

To develop functional metabolic engineering platforms for nonmodel microorganisms, a comprehensive understanding of the physiological and metabolic characteristics is critical. Caldicellulosiruptor bescii and other species in this genus have untapped potential for conversion of unpretreated plant biomass into industrial fuels and chemicals. The highly interactive and complex machinery used by C. bescii to acquire and process complex carbohydrates contained in lignocellulose was elucidated here to complement related efforts to develop a metabolic engineering platform with this bacterium.
