A dynamic interaction between CD19 and the tetraspanin CD81 controls B cell co-receptor trafficking

CD81 and its binding partner CD19 are core subunits of the B cell co-receptor complex. While CD19 belongs to the extensively studied Ig superfamily, CD81 belongs to a poorly understood family of four-pass transmembrane proteins called tetraspanins. Tetraspanins play important physiological roles by controlling protein trafficking and other processes. Here, we show that CD81 relies on its ectodomain to traffic CD19 to the cell surface. Moreover, the anti-CD81 antibody 5A6, which binds selectively to activated B cells, recognizes a conformational epitope on CD81 that is masked when CD81 is bound to CD19. Mutations of CD81 in this interface suppress its CD19 export activity. These data indicate that the CD81 - CD19 interaction is dynamically regulated upon B cell activation and this dynamism can be exploited to regulate B cell function. These results are not only valuable for understanding B cell biology, but also have important implications for understanding tetraspanin function generally.

Low pressure radon diffusion - A laboratory study and its implications for lunar venting

Results of a study of radon migration through columns of fine particulate materials, at total pressures of 0.02-0.2 torr, are reported. Materials studied were: NBS Glass Spheres (SRM 1003), Emerson & Cuming Eccospheres (IG-101), activated coconut charcoal, Lipaci obsidian, and W-1 Standard Diabase. Rates of diffusion were used to derive heats of adsorption for radon on the materials tested. The most reliable values found clustered around 8-9 kcal/mole. These high heats of adsorption, if typical for most materials, combined with low percentages of radon emanation by lunar soils found by other researchers, imply that random walk diffusion will not be an important mechanism for redistributing the radon and the radon daughters produced in the lunar regolith. In particular, since random walk migration is not a sufficient mechanism to account for localized high concentrations of radon-222 and its daughter polonium-210 observed by the Apollo 15 and 16 command modules, an alternative mechanism is proposed, in which radon would be swept to the surface by other gases during intermittent venting events.

Gravity and magnetic anomaly modeling and correlation using the SPHERE program and Magsat data

The spherical Earth inversion, modeling, and contouring software were tested and modified for processing data in the Southern Hemisphere. Preliminary geologic/tectonic maps and selected cross sections for South and Central America and the Caribbean region are being compiled and as well as gravity and magnetic models for the major geological features of the area. A preliminary gravity model of the Andeas Beniff Zone was constructed so that the density columns east and west of the subducted plates are in approximate isostatic equilibrium. The magnetic anomaly for the corresponding magnetic model of the zone is being computed with the SPHERE program. A test tape containing global magnetic measurements was converted to a tape compatible with Purdue's CDC system. NOO data were screened for periods of high diurnal activity and reduced to anomaly form using the IGS-75 model. Magnetic intensity anomaly profiles were plotted on the conterminous U.S. map using the track lines as the anomaly base level. The transcontinental magnetic high seen in POGO and MAGSAT data is also represented in the NOO data.

Parallelism and pipelining in high-speed digital simulators

The attainment of high computing speed as measured by the computational throughput is seen as one of the most challenging requirements. It is noted that high speed is cardinal in several distinct classes of applications. These classes are then discussed; they comprise (1) the real-time simulation of dynamic systems , (2) distributed parameter systems, and (3) mixed lumped and distributed systems. From the 1950s on, the quest for high speed in digital simulators concentrated on overcoming the limitations imposed by the so-called von Neumann bottleneck. Two major architectural approaches have made ig possible to circumvent this bottleneck and attain high speeds. These are pipelining and parallelism. Supercomputers, peripheral array processors, and microcomputer networks are then discussed.

Oxygen and carbon impurities and related defects in silicon

Oxygen and carbon are the predominant impurities in Czochralski-grown silicon. The incorporation of oxygen and carbon during crystal growth is reviewed and device effects are discussed. Methods for controlling oxygen and carbon incorporation during crystal growth are discussed and results supporting a segregation coefficient of k=0.5 for oxygen are presented. The nucleation and precipitation behavior of oxygen is complex. Temperature and doping level effects which add insight into the role of point defects in the nucleation process are highlighted. In general, precipitation is found to be retarded in N+ and P+ silicon. The types and quantities of defects resulting from the oxygen precipitates is of interest as they are technologically useful in the process called intrinsic gettering. A comparison is made between the available defect sites and the quantities of metallic impurities present in a typical wafer which need to be gettered. Finally, a discussion of the denuded-zone, intrinsic-gettered (DZ-IG) structure on device properties is presented.

Rat maintenance in the Research Animal Holding Facility during the flight of Space Lab 3

To test the husbandry capabilities of the Research Animal Holding Facility (RAHF) during space flight, 24 male rats were flown on Spacelab 3 for 7 days. Twelve large rats (400 g, LF), 5 of which had telemetry devices implanted (IF), and 12 small rats (200 g, SF) were housed in the RAHF. Examination 3 hr after landing (R + 3) revealed the rats to be free of injury, well nourished, and stained with urine. At R + 10 the rats were lethargic and atonic with hyperemia of the extremities and well groomed except for a middorsal area stained with urine and food. Both LF and SF rats showed weight gains comparable to their IG controls; IF rats grew less than controls. Food and water consumption were similar for flight and control groups. Plasma concentrations of total protein, sodium, albumin and creatinine did not differ between flight and control groups. LF and SF rats had elevated plasma glucose, and SF rats had increased blood urea nitrogen, potassium and glutamic pyruvic transaminase. These observations indicate that rats maintained in the RAHF were healthy, well nourished and experienced minimal stress; physiological changes in the rats can thus be attributed to the effects of space flight.

Affinity immunoblotting - High resolution isoelectric focusing analysis of antibody clonotype distribution

A sensitive and specific method is proposed for the analysis of specific antibody clonotype changes occurring during an immune response and for comparing multiple sera for antibody clonotype similarities. Polyclonal serum antibodies separated by isoelectric focusing (IEF) were analyzed by an affinity immunoblotting method using antigen-coated nitrocellulose membranes. Antibodies present on the surface of the acrylamide gels following IEF bind the antigen on the nitrocellulose when the coated nitrocellulose is laid over the gels. The technique has been used to analyze Ig clonotypes specific for five protein antigens and two carbohydrate antigens. Optimal antigen concentrations for coating the nitrocellulose membranes were found to range from 10-100 microgram/ml.

Effect of sudden solar wind dynamic pressure changes at subauroral latitudes - Change in magnetic field

The observations obtained during the International Magnetospheric Study (IMS) from the magnetometers of the IGS network extending from Cambridge, England, to Tromso, Norway, are used to study the response of subauroral current systems to sudden changes in solar wind dynamic pressure. Observations show that the response is very strong at subauroral latitudes. The preliminary response in the H component is a brief, small increase in the dayside morning sector and a decrease in the aftenoon and night sectors. The main response in the horizontal field (the H and D components) is toward the pole except in the dayside morning sector. The inferred ionospheric current is mainly a circulatory system flowing counterclockwise when viewed from the north pole everywhere at subauroral latitudes except the dayside morning sector.

Combustion synthesis of ceramic-metal composite materials in microgravity

Combustion synthesis, self-propagating high temperature synthesis (SHS) or reactive synthesis provides an attractive alternative to conventional methods of producing advanced materials since this technology is based on the ability of highly exothermic reactions to be self sustaining and, therefore, energetically efficient. The exothermic SHS reaction is initiated at the ignition temperature, T(sub ig), and generates heat which is manifested in a maximum or combustion temperature, T(sub c), which can exceed 3000 K . Such high combustion temperatures are capable of melting and/or volatilizing reactant and product species and, therefore, present an opportunity for producing structure and property modification and control through liquid-solid, vapor-liquid-solid, and vapor-solid transformations.

Occurrency frequency of substorm field and plasma signatures observed near-earth by ISEE-1/2

The onset of the majority of substorms occurs when the tail field stops growing more tail-like and begins to become more dipolar. This corresponds to the onset signatures on the ground and in geosynchronous orbit. The AE indices and the IGS Pi 2 data were used to determine the major substorm onsets of 1978 and 1979. The time delay between successive substorms, the distribution of the substorm growth phase duration and the probability of tailward flows were determined as a function of spacecraft location. About a half of the substorms exhibit a plasma signature including earthward or tailward flows or plasma sheet drop out and recovery. Earthward flows are often seen at substorm onset, and almost always during substorm recovery. Tailward flows are occasionally seen at onset as the spacecraft is close enough to the neutral sheet. The experimental results are compared to predictions based on the neutral line and current sheet disruption models.

Performance Assessment of Two GPS Receivers on Space Shuttle

Space Shuttle STS-69 was launched on September 7, 1995, carrying the Wake Shield Facility (WSF-02) among its payloads. The mission included two GPS receivers: a Collins 3M receiver onboard the Endeavour and an Osborne flight TurboRogue, known as the TurboStar, onboard the WSF-02. Two of the WSF-02 GPS Experiment objectives were to: (1) assess the ability to use GPS in a relative satellite positioning mode using the receivers on Endeavour and WSF-02; and (2) assess the performance of the receivers to support high precision orbit determination at the 400 km altitude. Three ground tests of the receivers were conducted in order to characterize the respective receivers. The analysis of the tests utilized the Double Differencing technique. A similar test in orbit was conducted during STS-69 while the WSF-02 was held by the Endeavour robot arm for a one hour period. In these tests, biases were observed in the double difference pseudorange measurements, implying that biases up to 140 m exist which do not cancel in double differencing. These biases appear to exist in the Collins receiver, but their effect can be mitigated by including measurement bias parameters to accommodate them in an estimation process. An additional test was conducted in which the orbit of the combined Endeavour/WSF-02 was determined independently with each receiver. These one hour arcs were based on forming double differences with 13 TurboRogue receivers in the global IGS network and estimating pseudorange biases for the Collins. Various analyses suggest the TurboStar overall orbit accuracy is about one to two meters for this period, based on double differenced phase residuals of 34 cm. These residuals indicate the level of unmodeled forces on Endeavour produced by gravitational and nongravitational effects. The rms differences between the two independently determined orbits are better than 10 meters, thereby demonstrating the accuracy of the Collins-determined orbit at this level as well as the accuracy of the relative positioning using these two receivers.

Helicopter Rotor Blade Computation in Unsteady Flows Using Moving Overset Grids

An overset grid thin-layer Navier-Stokes code has been extended to include dynamic motion of helicopter rotor blades through relative grid motion. The unsteady flowfield and airloads on an AH-IG rotor in forward flight were computed to verify the methodology and to demonstrate the method's potential usefulness towards comprehensive helicopter codes. In addition, the method uses the blade's first harmonics measured in the flight test to prescribe the blade motion. The solution was impulsively started and became periodic in less than three rotor revolutions. Detailed unsteady numerical flow visualization techniques were applied to the entire unsteady data set of five rotor revolutions and exhibited flowfield features such as blade vortex interaction and wake roll-up. The unsteady blade loads and surface pressures compare well against those from flight measurements. Details of the method, a discussion of the resulting predicted flowfield, and requirements for future work are presented. Overall, given the proper blade dynamics, this method can compute the unsteady flowfield of a general helicopter rotor in forward flight.

Premixed Turbulent Flame Propagation in Microgravity

A combined numerical-experimental study has been carried out to investigate the structure and propagation characteristics of turbulent premixed flames with and without the influence of buoyancy. Experimentally, the premixed flame characteristics are studied in the wrinkled regime using a Couette flow facility and an isotropic flow facility in order to resolve the scale of flame wrinkling. Both facilities were chosen for their ability to achieve sustained turbulence at low Reynolds number. This implies that conventional diagnostics can be employed to resolve the smallest scales of wrinkling. The Couette facility was also built keeping in mind the constraints imposed by the drop tower requirements. Results showed that the flow in this Couette flow facility achieves full-developed turbulence at low Re and all turbulence statistics are in good agreement with past measurements on large-scale facilities. Premixed flame propagation studies were then carried out both using the isotropic box and the Couette facility. Flame imaging showed that fine scales of wrinkling occurs during flame propagation. Both cases in Ig showed significant buoyancy effect. To demonstrate that micro-g can remove this buoyancy effect, a small drop tower was built and drop experiments were conducted using the isotropic box. Results using the Couette facility confirmed the ability to carry out these unique reacting flow experiments at least in 1g. Drop experiments at NASA GRC were planned but were not completed due to termination of this project.
