The ethylene response pathway in Arabidopsis

The simple gas ethylene influences a diverse array of plant growth and developmental processes including germination, senescence, cell elongation, and fruit ripening. This review focuses on recent molecular genetic studies, principally in Arabidopsis, in which components of the ethylene response pathway have been identified. The isolation and characterization of two of these genes has revealed that ethylene sensing involves a protein kinase cascade. One of these genes encodes a protein with similarity to the ubiquitous Raf family of Ser/Thr protein kinases. A second gene shows similarity to the prokaryotic two-component histidine kinases and most likely encodes an ethylene receptor. Additional elements involved in ethylene signaling have only been identified genetically. The characterization of these genes and mutants will be discussed.

LDPC-PPM Coding Scheme for Optical Communication

In a proposed coding-and-modulation/demodulation-and-decoding scheme for a free-space optical communication system, an error-correcting code of the low-density parity-check (LDPC) type would be concatenated with a modulation code that consists of a mapping of bits to pulse-position-modulation (PPM) symbols. Hence, the scheme is denoted LDPC-PPM. This scheme could be considered a competitor of a related prior scheme in which an outer convolutional error-correcting code is concatenated with an interleaving operation, a bit-accumulation operation, and a PPM inner code. Both the prior and present schemes can be characterized as serially concatenated pulse-position modulation (SCPPM) coding schemes. Figure 1 represents a free-space optical communication system based on either the present LDPC-PPM scheme or the prior SCPPM scheme. At the transmitting terminal, the original data (u) are processed by an encoder into blocks of bits (a), and the encoded data are mapped to PPM of an optical signal (c). For the purpose of design and analysis, the optical channel in which the PPM signal propagates is modeled as a Poisson point process. At the receiving terminal, the arriving optical signal (y) is demodulated to obtain an estimate (a^) of the coded data, which is then processed by a decoder to obtain an estimate (u^) of the original data.

Comparing bandwidth requirements for digital baseband signals.

This paper describes the relative bandwidth requirements of the common digital baseband signaling techniques used for data transmission. Bandwidth considerations include the percentage of total power in a properly encoded PN sequence passed at bandwidths of 0.5, 1, 2 and 3 times the reciprocal of the bit interval. The signals considered in this study are limited to the binary class. The study compares such signaling techniques as delay modulation, bipolar, biternary, duobinary, pair selected ternary and time polarity control in addition to the conventional NRZ, RZ and BI-phi schemes.

VPS26 Moonlights as a β-Arrestin-like Adapter for a 7-Transmembrane RGS Protein in Arabidopsis thaliana

Extracellular signals perceived by 7-transmembrane (7TM)-spanning receptors initiate desensitization that involves the removal of these receptors from the plasma membrane. Agonist binding often evokes phosphorylation in the flexible C-terminal region and/or intracellular loop 3 of many 7TM G-protein-coupled receptors in animal cells, which consequently recruits a cytoplasmic intermediate adaptor, β-arrestin, resulting in clathrin-mediated endocytosis (CME) and downstream signaling such as transcriptional changes. Some 7TM receptors undergo CME without recruiting β-arrestin, but it is not clear how. Arrestins are not encoded in the Arabidopsis thaliana genome, yet Arabidopsis cells have a well-characterized signal-induced CME of a 7TM protein, designated Regulator of G Signaling 1 (AtRGS1). Here we show that a component of the retromer complex, Vacuolar Protein Sorting-Associated 26 (VPS26), binds the phosphorylated C-terminal region of AtRGS1 as a VPS26A/B heterodimer to form a complex that is required for downstream signaling. We propose that VPS26 moonlights as an arrestin-like adaptor in the CME of AtRGS1.

Bandwidth compression of the digitized HDTV images for transmission via satellites

This paper investigates a subband coding scheme to reduce the transmission bandwidth of the digitized HDTV images. The HDTV signals are decomposed into seven bands. Each band is then independently encoded. The based band is DPCM encoded and the high bands are encoded by using nonuniform Laplacian quantizers with a dead zone. By selecting the dead zone on the basis of energy in the high bands an acceptable image quality is achieved at an average of 45 Mbits/sec (Mbps) rate. This rate is comparable to some very hardware intensive schemes of transform compression or vector quantization proposed in the literature. The subband coding scheme used in this study is considered to be of medium complexity. The 45 Mbps rate is suitable for transmission of HDTV signals via satellites.

Method and apparatus for optical encoding with compressible imaging

The present invention presents an optical encoder with increased conversion rates. Improvement in the conversion rate is a result of combining changes in the pattern recognition encoder's scale pattern with an image sensor readout technique which takes full advantage of those changes, and lends itself to operation by modern, high-speed, ultra-compact microprocessors and digital signal processors (DSP) or field programmable gate array (FPGA) logic elements which can process encoder scale images at the highest speeds. Through these improvements, all three components of conversion time (reciprocal conversion rate)--namely exposure time, image readout time, and image processing time--are minimized.

System for controller area network payload decoding

A system for decoding an unknown automotive controller area network (“CAN”) message definitions. CAN data vehicle signal mappings are typically held in secret and varied by automotive model and year. Without knowledge of the mappings, the wealth of real-time vehicle data hidden in the automotive CAN packets is uninterpretable—impeding research, after-market tuning, efficiency and performance monitoring, fault diagnosis, and privacy-related technologies. This system can ascertain the CAN signals' boundaries (start bit and length), endianness (byte ordering), signedness (binary-to-integer encoding) from raw CAN data. This allows conversion of CAN data to time series. Interpreting the translated CAN data's physical meaning and finding a linear mapping to standard units (e.g., knowing the signal is speed and scaling values to represent units of miles per hour) can be achieved for many signals by leveraging diagnostic standards to obtain real-time measurements of in-vehicle systems. The system can be integrated into lightweight hardware enabling an OBD-II plugin for real-time in-vehicle CAN decoding or run on standard computers. The system can output a standard DBC file with the signal definition information.

Conformation-specific synthetic intrabodies modulate mTOR signaling with subcellular spatial resolution

Subcellular compartmentalization is integral to the spatial regulation of mechanistic target of rapamycin (mTOR) signaling. However, the biological outputs associated with location-specific mTOR signaling events are poorly understood and challenging to decouple. Here, we engineered synthetic intracellular antibodies (intrabodies) that are capable of modulating mTOR signaling with genetically programmable spatial resolution. Epitope-directed phage display was exploited to generate high affinity synthetic antibody fragments (Fabs) against the FKBP12–Rapamycin binding site of mTOR (mTOR FRB ). We determined high-resolution crystal structures of two unique Fabs that discriminate distinct conformational states of mTOR FRB through recognition of its substrate recruitment interface. By leveraging these conformation-specific binders as intracellular probes, we uncovered the structural basis for an allosteric mechanism governing mTOR complex 1 (mTORC1) stability mediated by subtle structural adjustments within mTOR FRB . Furthermore, our results demonstrated that synthetic binders emulate natural substrates by employing divergent yet complementary hydrophobic residues at defined positions, underscoring the broad molecular recognition capability of mTOR FRB . Intracellular signaling studies showed differential time-dependent inhibition of S6 kinase 1 and Akt phosphorylation by genetically encoded intrabodies, thus supporting a mechanism of inhibition analogous to the natural product rapamycin. Finally, we implemented a feasible approach to selectively modulate mTOR signaling in the nucleus through spatially programmed intrabody expression. These findings establish intrabodies as versatile tools for dissecting the conformational regulation of mTORC1 and should be useful to explore how location-specific mTOR signaling influences disease progression.

An orphan gene BOOSTER enhances photosynthetic efficiency and plant productivity

Organelle-to-nucleus DNA transfer is an ongoing process playing an important role in the evolution of eukaryotic life. Here, genome-wide association studies (GWAS) of non-photochemical quenching parameters in 743 Populus trichocarpa accessions identified a nuclear-encoded genomic region associated with variation in photosynthesis under fluctuating light. The identified gene, BOOSTER (BSTR), comprises three exons, two with apparent endophytic origin and the third containing a large fragment of plastid-encoded Rubisco large subunit. Higher expression of BSTR facilitated anterograde signaling between nucleus and plastid, which corresponded to enhanced expression of Rubisco, increased photosynthesis, and up to 35% greater plant height and 88% biomass in poplar accessions under field conditions. Overexpression of BSTR in Populus tremula × P. alba achieved up to a 200% in plant height. Similarly, Arabidopsis plants heterologously expressing BSTR gained up to 200% in biomass and up to 50% increase in seed.

Quantum frequency resampling

In signal processing, resampling algorithms can modify the number of resources encoding a collection of data points. Downsampling reduces the cost of storage and communication, while upsampling interpolates new data from limited one, e.g., when resizing a digital image. We present a toolset of quantum algorithms to resample data encoded in the probabilities of a quantum register, using the quantum Fourier transform to adjust the number of high-frequency encoding qubits. We discuss advantage over classical resampling algorithms.

Progress in developing a superconducting accelerator for efficient eBeam and X-ray production

The PIP-II Accelerator is an 800 MeV superconducting Linac in the injection chain of the Fermilab accelerator complex. The LLRF systems for the 125 cavities which include a few normal conducting cavities in the warm front-end section, use a variety of LLRF hardware components and sub-systems that are part of the timing and synchronization system. This includes a master oscillator and phase averaging reference line, a beam pattern generator to enable beam transfer between non-harmonic RF systems and beam loading compensation systems synchronized to a start of beam trigger. The timing signals are based on a 650 MHz clock with manchester encoded event information and data provided over optical fiber. The timing and synchronization systems and their performance parameters will be described here.

This includes a master oscillator and phase averaging reference line, a beam pattern generator to enable beam transfer between non-harmonic RF systems and beam loading compensation systems synchronized to a start of beam trigger. The timing signals are based on a 650 MHz clock with manchester encoded event information and data provided over optical fiber. The timing and synchronization systems and their performance parameters will be described here.

Variable word length encoder reduces TV bandwith requirements

Adaptive variable resolution encoding technique provides an adaptive compression pseudo-random noise signal processor for reducing television bandwidth requirements. Complementary processors are required in both the transmitting and receiving systems. The pretransmission processor is analog-to-digital, while the postreception processor is digital-to-analog.

Digital plus analog output encoder

The disclosed encoder is adapted to produce both digital and analog output signals corresponding to the angular position of a rotary shaft, or the position of any other movable member. The digital signals comprise a series of binary signals constituting a multidigit code word which defines the angular position of the shaft with a degree of resolution which depends upon the number of digits in the code word. The basic binary signals are produced by photocells actuated by a series of binary tracks on a code disc or member. The analog signals are in the form of a series of ramp signals which are related in length to the least significant bit of the digital code word. The analog signals are derived from sine and cosine tracks on the code disc.

Telephone multiline signaling using common signal pair

An operator can rapidly and automatically produce coded electrical signals by manipulating mechanical thumb wheel switches so as to instruct a service center to connect any number of telephone lines to the console thus enabling the operator to listen and/or talk over several lines simultaneously. The system includes an on-site console having several mechanically operated thumb wheel switches to which the desired lines to be connected can be dialed in. Electrical coded signals are fed to a number of banks of line AND gates representing units, tens and hundreds, a group of channel gates, and a command gate. These signals are gated out in a controlled manner to an encoder which generates tones that are transmitted over a single line to a communication service center.

Improved Speech Coding Based on Open-Loop Parameter Estimation

A nonlinear optimization algorithm for linear predictive speech coding was developed early that not only optimizes the linear model coefficients for the open loop predictor, but does the optimization including the effects of quantization of the transmitted residual. It also simultaneously optimizes the quantization levels used for each speech segment. In this paper, we present an improved method for initialization of this nonlinear algorithm, and demonstrate substantial improvements in performance. In addition, the new procedure produces monotonically improving speech quality with increasing numbers of bits used in the transmitted error residual. Examples of speech encoding and decoding are given for 8 speech segments and signal to noise levels as high as 47 dB are produced. As in typical linear predictive coding, the optimization is done on the open loop speech analysis model. Here we demonstrate that minimizing the error of the closed loop speech reconstruction, instead of the simpler open loop optimization, is likely to produce negligible improvement in speech quality. The examples suggest that the algorithm here is close to giving the best performance obtainable from a linear model, for the chosen order with the chosen number of bits for the codebook.

LISA Framework for Enhancing Gravitational Wave Signal Extraction Techniques

This paper describes the development of a Framework for benchmarking and comparing signal-extraction and noise-interference-removal methods that are applicable to interferometric Gravitational Wave detector systems. The primary use is towards comparing signal and noise extraction techniques at LISA frequencies from multiple (possibly confused) ,gravitational wave sources. The Framework includes extensive hybrid learning/classification algorithms, as well as post-processing regularization methods, and is based on a unique plug-and-play (component) architecture. Published methods for signal extraction and interference removal at LISA Frequencies are being encoded, as well as multiple source noise models, so that the stiffness of GW Sensitivity Space can be explored under each combination of methods. Furthermore, synthetic datasets and source models can be created and imported into the Framework, and specific degraded numerical experiments can be run to test the flexibility of the analysis methods. The Framework also supports use of full current LISA Testbeds, Synthetic data systems, and Simulators already in existence through plug-ins and wrappers, thus preserving those legacy codes and systems in tact. Because of the component-based architecture, all selected procedures can be registered or de-registered at run-time, and are completely reusable, reconfigurable, and modular.

DOT Transmit Module

The Deep Space Optical Terminal (DOT) transmit module demonstrates the DOT downlink signaling in a flight electronics assembly that can be qualified for deep space. The assembly has the capability to generate an electronic pulse-position modulation (PPM) waveform suitable for driving a laser assembly to produce the optical downlink signal. The downlink data enters the assembly through a serializer/ deserializer (SERDES) interface, and is encoded using a serially concatenated PPM (SCPPM) forward error correction code. The encoded data is modulated using PPM with an inter-symbol guard time to aid in receiver synchronization. Monitor and control of the assembly is via a low-voltage differential signal (LVDS) interface
