Feedstock/Pretreatment Screening for Bioconversion of Sugars and Lignin Residues

This project will conduct biomass deconstruction (pretreatment and enzymatic saccharification) on two representative biomass feedstocks and four different pretreatment processes, including three high temperature steam/chemical pretreatments and one low temperature chemical/mechanical pretreatment. A third biomass feedstock will undergo biomass deconstruction with three different pretreatment processes, including two high temperature chemical pretreatments and one low temperature chemical/mechanical pretreatment. Several pretreatment conditions will be performed in a screening study using NLR pilot-scale pretreatment equipment to generate a range of pretreated biomass substrates. A selected number of these substrates will be chosen for enzymatic saccharification evaluation, based on standard compositional analysis of the pretreated substrates as a primary indicator of pretreatment efficacy. Resulting enzymatic hydrolysis slurries will be analyzed to determine overall biomass sugar yields. Additional compositional analysis will be performed to determine oligomeric sugar composition and structure, to analyze structural characteristics of solids fractions on native, pretreated, and enzymatically saccharified biomass residues for one of the biomass feedstocks, corn stover. The enzymatically saccharified materials will undergo 2,3-butanediol fermentation in a shaker flask as bench scale. Using relevant process performance data collected in these various conversion steps, technoeconomic analysis activities will be performed to compare the economic potential of the various biomass feedstock and pretreatment processes and to identify key economic drivers and sustainability metrics.

Pseudomonas aeruginosa gene PA4880 encodes a Dps-like protein with a Dps fold, bacterioferritin-type ferroxidase centers, and endonuclease activity

We report the biochemical, structural, and functional characterization of the protein coded by gene PA4880 in the P. aeruginosa PAO1 genome. The PA4880 gene had been annotated as coding a probable bacterioferritin. Our structural work shows that the product of gene PA4880 is a protein that adopts the Dps subunit fold, which oligomerizes into a 12-mer quaternary structure. Unlike Dps, however, the ferroxidase di-iron centers and iron coordinating ligands are buried within each subunit, in a manner identical to that observed in the ferroxidase center of P. aeruginosa bacterioferritin. Since these structural characteristics correspond to Dps-like proteins, we term the protein as P. aeruginosa Dps-like, or Pa DpsL. The ferroxidase centers in Pa DpsL catalyze the oxidation of Fe 2+ utilizing O 2 or H 2 O 2 as oxidant, and the resultant Fe 3+ is compartmentalized in the interior cavity. Interestingly, incubating Pa DpsL with plasmid DNA results in efficient nicking of the DNA and at higher concentrations of Pa DpsL the DNA is linearized and eventually degraded. The nickase and endonuclease activities suggest that Pa DpsL, in addition to participating in the defense of P. aeruginosa cells against iron-induced toxicity, may also participate in the innate immune mechanisms consisting of restriction endonucleases and cognate methyl transferases.

A Tension Shell Structure for Application to Entry Vehicles

The shape of a shell of revolution designed t o have only tensile stresses under axisymmetric aerodynamic loadings has been derived on the basis of linear membrane theory. values of circumferential tension is also given. The results of this paper indicate that the design of an entry vehicle based on the tension shell concept leads to desirable aerodynamic and structural characteristics, namely, high drag and low weight.

The reduction of a ""safety catastrophic'' potential hazard: A case history

A worst case analysis is reported on the safety of time watch movements for triggering explosive packages on the lunar surface in an experiment to investigate physical lunar structural characteristics through induced seismic energy waves. Considered are the combined effects of low pressure, low temperature, lunar gravity, gear train error, and position. Control measures constitute a seal control cavity and design requirements to prevent overbanking in the mainspring torque curve. Thus, the potential hazard is reduced to safety negligible.

Structural considerations in design of lightweight glass-fiber composite pressure vessels

The development of structurally efficient, metal-lined, glass-fiber composite pressure vessels. Both the current state-of-the-art and current problems are discussed along with fracture mechanics considerations for the metal liner. The design concepts used for metal-lined, glass-fiber, composite pressure vessels are described and the structural characteristics of the composite designs are compared with each other and with homogeneous metal pressure vessels. Specific design techniques and available design data are identified. Results of a current program to evaluate flaw growth and fracture characteristics of the metal liners are reviewed and the impact of these results on composite pressure vessel designs is discussed.

Development and application of vibroacoustic structural data banks in predicting vibration design and test criteria for rocket vehicle structures

A method of predicting broadband random vibration criteria for components on space vehicles is presented. Large amounts of vibration and acoustic data obtained from flights and static firing tests of space vehicle were formulated into vibroacoustic data banks for structural categories of ring frame, skin stringer, and honeycomb. The vibration spectra with their associated acoustic spectra are normalized to a reference acoustic spectrum. The individual normalized spectra are grouped according to definite structural characteristics and statistically analyzed to form the vibroacoustic data banks described in this report. These data banks represent the reference vibration criteria available for determining the new vehicle vibration criteria.

Small Interactive Image Processing System (SMIPS) system description

The Small Interactive Image Processing System (SMIPS) operates under control of the IBM-OS/MVT operating system and uses an IBM-2250 model 1 display unit as interactive graphic device. The input language in the form of character strings or attentions from keys and light pen is interpreted and causes processing of built-in image processing functions as well as execution of a variable number of application programs kept on a private disk file. A description of design considerations is given and characteristics, structure and logic flow of SMIPS are summarized. Data management and graphic programming techniques used for the interactive manipulation and display of digital pictures are also discussed.

Applied Superconductivity Conference, 5th, Annapolis, Md., May 1-3, 1972, Proceedings.

Recently developed practical applications of superconductivity are described in papers dealing with transportation and propulsion, power generation, power transmission, special-purpose magnets, superconducting materials, Josephson junctions, and microwave systems. Important topics considered include tracked magnetic-cushion vehicles, electrical machines, cryogenic underground power transmission systems, magnets for fusion reactors and particle accelerators, thin-film superconductors, effects of the structural characteristics of materials on transition temperatures, frequency measurement with Josephson devices, and superconducting scientific instruments. Individual items are announced in this issue.

Influence of acoustic parameters on the mechanical properties of low-alloy steels welded in a vibrated field

The advantages and disadvantages of slag welding are given. A suitable method is proposed for introducing ultrasonic oscillations in the process of depositing the metal in the slag bath. Some practical results are given of experimental studies of the mechanical and structural characteristics obtained with the welding of steels under vibrated and nonvibrated conditions, respectively. Certain physical phenomena that attend the moving metal particle in slag welding are analyzed.

Structural considerations in design of lightweight glass-fiber composite pressure vessels

The design concepts used for metal-lined glass-fiber composite pressure vessels are described, comparing the structural characteristics of the composite designs with each other and with homogeneous metal pressure vessels. Specific design techniques and available design data are identified. The discussion centers around two distinctly different design concepts, which provide the basis for defining metal lined composite vessels as either (1) thin-metal lined, or (2) glass fiber reinforced (GFR). Both concepts are described and associated development problems are identified and discussed. Relevant fabrication and testing experience from a series of NASA-Lewis Research Center development efforts is presented.

Infrared photometry of Titan

A set of observations of Titan at wavelengths from 1.6 through 34 microns shows that Titan has a remarkably low albedo near 1.6 micron. Thermal flux observations for 10 years at 10.6 microns and for 5 years at 21 microns suggest that the thermal properties of Titan remained constant over this period and are independent of phase. Narrowband photometry between 17 and 25 microns does not indicate the presence of a structure characteristic of a hydrogen-rich atmosphere. An atmospheric model consistent with the observations is proposed for Titan.-
