Hsa_circ_0000515 is a novel circular RNA implicated in the development of breast cancer through its regulation of the microRNA‐296‐5p/CXCL10 axis

Cancer metastasis is a major cause of death among women afflicted with breast cancer (BC) and understanding the molecular processes involved is a major focus in BC research. Circular RNAs (circRNAs) have emerged as genomic regulatory molecules in carcinogenesis and metastasis; however, their role in BC is unclear. We characterized a novel circRNA, hsa_circ_0000515, in context of BC. We collected 340 cancerous tissues surgically resected from BC patients and found hsa_circ_0000515 was upregulated in BC tissues and associated with poor prognosis of BC. Silencing of hsa_circ_0000515 impaired cell cycle progression, cell proliferation, and invasion, attenuated inflammatory response, and reduced the proangiogenetic potential of BC cells. RNA pull‐down and dual‐luciferase reporter gene assays showed that hsa_circ_0000515 binds miR‐296‐5p, preventing it from repressing CXCL10 expression. We also observed that miR‐296‐5p inhibition or CXCL10 overexpression promoted cell cycle progression, restored proliferative, invasive and proangiogenetic abilities, and increased inflammatory response in MCF‐7 cells in the absence of hsa_circ_0000515. In vivo analyses showed that partial loss of hsa_circ_0000515 reduced the tumor growth of MCF‐7 cells in nude mice. The key findings from this study revealed that targeting hsa_circ_0000515 might be an effective strategy to combat BC.

Open Water Blade Strain Measurements on a Vertical-Axis Tidal Turbine

Open-water testing of marine renewable energy devices represents a significant milestone and hurdle for research teams and companies that seek to reduce the levelized cost of energy to allow these devices to compete in the open electrical generation market. For open-water testing of tidal energy converters (TECs), accurate measurements of loading characteristics on the blades and other structural components correlated with power performance metrics can be invaluable to further refine design principles and models, allowing continued improvement and cost reductions of new turbine designs. This paper will describe the modifications and additions made to a TEC system to achieve time-correlated blade strain measurements during full turbine operation along with a discussion on the overall impacts the required modifications had on the unmodified turbine.

Coevolution of the Ess1-CTD axis in polar fungi suggests a role for phase separation in cold tolerance

Most of the world’s biodiversity lives in cold (-2° to 4°C) and hypersaline environments. To understand how cells adapt to such conditions, we isolated two key components of the transcription machinery from fungal species that live in extreme polar environments: the Ess1 prolyl isomerase and its target, the carboxy-terminal domain (CTD) of RNA polymerase II. Polar Ess1 enzymes are conserved and functional in the model yeast, Saccharomyces cerevisiae. By contrast, polar CTDs diverge from the consensus (YSPTSPS) 26 and are not fully functional in S. cerevisiae. These CTDs retain the critical Ess1 Ser-Pro target motifs, but substitutions at Y1, T4, and S7 profoundly affected their ability to undergo phase separation in vitro and localize in vivo. We propose that environmentally tuned phase separation by the CTD and other intrinsically disordered regions plays an adaptive role in cold tolerance by concentrating enzymes and substrates to overcome energetic barriers to metabolic activity.

The SEMA3F-NRP1/NRP2 axis is a key factor in the acquisition of invasive traits in in situ breast ductal carcinoma

A better understanding of ductal carcinoma in situ (DCIS) is urgently needed to identify these preinvasive lesions as distinct clinical entities. Semaphorin 3F (SEMA3F) is a soluble axonal guidance molecule, and its coreceptors Neuropilin 1 (NRP1) and NRP2 are strongly expressed in invasive epithelial BC cells. We utilized two cell line models to represent the progression from a healthy state to the mild-aggressive or ductal carcinoma in situ (DCIS) stage and, ultimately, to invasive cell lines. Additionally, we employed in vivo models and conducted analyses on patient databases to ensure the translational relevance of our results. We revealed SEMA3F as a promoter of invasion during the DCIS-to-invasive ductal carcinoma transition in breast cancer (BC) through the action of NRP1 and NRP2. In epithelial cells, SEMA3F activates epithelialmesenchymal transition, whereas it promotes extracellular matrix degradation and basal membrane and myoepithelial cell layer breakdown. Together with our patient database data, these proof-of-concept results reveal new SEMA3F-mediated mechanisms occurring in the most common preinvasive BC lesion, DCIS, and represent potent and direct activation of its transition to invasion. Moreover, and of clinical and therapeutic relevance, the effects of SEMA3F can be blocked directly through its coreceptors, thus preventing invasion and keeping DCIS lesions in the preinvasive state.

Unique Single-Axis Tracking Planar Waveguide Optical Collector for CSP Modules

During the project Agira was pursued: (a) R&D efforts leading to the development of an innovative and revolutionary future-generation waveguide concentrating solar collector that will have improved value or productivity when compared to best-in-class, near-commercial concentrating solar power (CSP) solar collector technologies; (b) demonstrate results supporting that the proposed innovation meets or goes beyond the 6 cents per kWh levelized cost of electricity (LCOE) goals set forth by the SunShot Initiative (c) pursue clear market- and application-driven objectives and partnerships as part of a credible pathway to commercial development and eventual deployment. The novel optical approach consisted of a primary light bending optics converting normal incidence light to shallow angles which is then input onto a shallow secondary reflector which redirects and concenrates the light onto the evacuated solar tube.

2020 Review of Axis 2 rf

An intense relativistic electron beam can produce radiofrequency (rf) radiation in many ways. The DARHT II linear induction accelerator produces a 17 MeV, 1.65 kA beam. The dominant sources of rf at DARHT-II and the methods used to suppress this rf are well documented. This note will examine the observed rf over the past six years and its impact on the beam. The recorded waveforms for each hydro starting with 4011 from 10/29/14 are examined to identify the presence of rf. The hydros are chosen because they represent the optimal accelerator performance at any given time rather than tuning or beam studies.
