Gold Nanorod-Affibody Conjugates Mediate Cancer Photothermal Therapy under 808 nm LED Irradiation

Current HER2-targeted therapies including monoclonal antibodies, antibody-drug conjugates (ADCs) and small-molecule tyrosine kinase inhibitors face limitations such as hepatotoxicity, development of treatment resistance, and subsequent disease relapse. To overcome these challenges, this study combines the specificity of a HER2-targeting affibody molecule (Z HER2:2891 -Cys) with the photothermal properties of gold nanorods (AuNRs), forming bioconjugates (Affi–AuNRs) for selective treatment of HER2-positive cancer cells. Affi–AuNRs were fabricated via a two-step surface modification: (i) ligand exchange to displace cetyltrimethylammonium bromide (CTAB) with poly(ethylene glycol) methyl ether thiol (mPEG-SH), and (ii) covalent attachment of the affibody molecule via Au–S chemistry. Affi-AuNRs exhibited a photothermal conversion efficiency of 64 ± 5%, comparable to that of CTAB-stabilized AuNRs (59 ± 4%). Confocal microscopy confirmed that Affi-AuNRs are selectively internalized by HER2-positive SKOV-3 cells, with minimal uptake by HEK293T cells, which have low HER2 expression. Upon exposure to 808 nm near-infrared (NIR) LED irradiation (408 mW cm –2 , 15 min), Affi-AuNRs significantly reduced SKOV-3 cell viability by 85 ± 9% (p < 0.001) relative to untreated controls with no detectable cytotoxicity in HEK293T cells. These results demonstrate HER2-selective photothermal cytotoxicity mediated by Affi–AuNRs under defined 808 nm LED irradiation conditions, supporting their continued development as nanotheranostic tools that integrate affibody-mediated specificity with plasmonic heating.

From Obsolete to Optimal (ATR Demineralized Water System)

My project was migrating a Human-Machine Interface (HMI) and Programmable Logic Controller (PLC) program from legacy software to a modern platform, enhancing operational efficiency and eliminating unsupported, obsolete equipment. Initially, I used a migration tool to transfer the programs to the new software, then manually updated variables to align with the new PLC. I optimized the PLC code by leveraging new scaling features in the I/O cards, removing over 45 obsolete rungs, and enhancing clarity with functional tag aliases. Next, I modernized the HMI's visuals with an updated color scheme and 3D buttons. I developed multiple interface versions iteratively refining them based on operator feedback. I also designed and created a dedicated "Rounds" screen incorporating insights from a previous intern’s project to streamline daily rounds. To improve clarity, consistency, and efficiency I redesigned the entire interface. Adding visual indicators to enhance operational awareness: red is used for values outside normal specifications, green for normal operating modes, yellow for manual mode, and grey for offline units. Additionally, I added a dynamic visual representation of tank levels, creating PLC logic to show the level in inches as well as inches and feet. I then incorporated a navigation sidebar to facilitate efficient screen transitions. Risk of human error was mitigated by defining narrow input ranges for tank level shutoff and password protecting setpoint changes. I also fixed issues with datatypes that caused inaccuracies in the original code. Overall, this comprehensive upgrade significantly enhanced the HMI's functionality, user experience, and operational reliability.

Quantum Computing Strategy 2026

Quantum computing (QC) is a rapidly maturing technology with the potential for revolutionary impacts on stockpile stewardship science and national security. Recent developments in fault-tolerant architectures have compressed vendor roadmaps, and predictions of a production-ready quantum computer by the mid-2030s are becoming increasingly credible. This strategy provides a roadmap for integrating QC into the Advanced Simulation and Computing (ASC) program by investing in four strategic focus areas: 1. Develop Capabilities in Mission-Relevant Quantum Applications: ASC will prioritize developing quantum-ready applications in mission areas that have shown significant promise for quantum advantage, including simulations of materials in extreme environments, nuclear dynamics, solving linear and nonlinear partial differential equations, and uncertainty quantification. These applications directly support stockpile stewardship science and modernization objectives. 2. Conduct R&D in Algorithms, Software, and Hardware: Sustained research into quantum algorithms, robust software tools, and quantum hardware is essential. ASC will develop efficient quantum algorithms; invest in quantum compilers, debuggers, and performance tools; and explore specialized quantum hardware tailored to NNSA’s unique requirements. 3. Engage with Vendors and Partners: Early and active collaboration with commercial quantum hardware vendors and academic partners is critical. Through testbeds, co-design agreements, and quantum demonstration facilities, ASC will influence hardware design, gain early access to emerging technologies, and ensure that quantum platforms evolve to meet mission needs. 4. Build Knowledge, Experience, and Workforce: Expanding and upskilling the quantum-trained workforce is essential to long-term success. This includes hiring, internal training, university outreach, and postdoctoral support to ensure ASC maintains the expertise required to operate, program, and integrate quantum systems as they become available. While quantum computing will never replace classical computing, it has the potential to solve certain problems with speed and accuracy that would be unachievable using any conceivable classical high-performance computing (HPC) system. By investing strategically in QC, ASC will help propel the emergent QC industry, maintain U.S. technological leadership, ensure mission readiness, and position itself to rapidly adopt quantum technologies as they mature.

Numerical and experimental analysis of mechanically induced failure in electric vehicle battery modules

Mitigating thermal runaway and cell-to-cell propagation is essential for improving the safety of electric and hybrid vehicles. Enhancing digital twin capabilities to predict battery mechanical abuse is particularly critical for automotive and aerospace applications, where crashworthiness is a key concern. Understanding failure conditions and propagation in battery modules during mechanical abuse is complex due to interactions between structural deformation, heat transfer, electrochemical processes, exothermic reactions and mechanical fracture. While prior studies have focused on modeling cell-level behavior, extending these models to module or pack level is necessary for a system level understating of electric vehicle safety. This study develops coupled large deformation finite element models that simultaneously solve for electrochemistry, material failure, internal short circuit and thermal runaway propagation. The models account for mechanical and thermal interactions between lithium-ion cells and other battery components while the contact interfaces are evolving with time. Model-predicted voltage, temperature and force responses are compared with experimental data for validation. The results demonstrate that the approach captures key failure mechanisms, including thermal propagation through heat transfer, electrical propagation from short circuits in parallel-connected cells, and mechanical propagation via penetration and crack formation. These findings show that computational models are valuable tools for understanding battery module failure and providing insight that can reduce the need for extensive experimental testing.

Leveraging System Dynamics to Predict the Commercialization Success of Emerging Energy Technologies: Lessons from Wind Energy

The United States urgently needs to tackle the climate crisis while enhancing energy security and resiliency. The complexity of the U.S. energy system, with its interconnected elements, makes predicting future states challenging, especially with the introduction of novel energy systems like wind, solar, clean hydrogen, and advanced nuclear technologies. Modern systems engineering methods and tools can provide deeper insights into these dynamics and future behaviors. This research aims to develop a comprehensive model that captures the main elements and behaviors of new energy technologies within the existing energy system. We hypothesized that the market uptake of novel energy systems is influenced by multiple diverse factors, such as technological learning, availability of resources, and economic incentives; examined the history of electricity generation using land-based wind technologies; and developed a system dynamics model to investigate the relationships between capacity growth and influencing factors, both internal and external. The developed model yielded outcomes that confirmed the hypothesized dynamics of wind energy system diffusion through a quantitative comparison of installed capacity and highlighted the significant influence of resource availability, federal incentives (production tax credits), and technological learning on capacity growth and cost reduction. This research aims to support informed decision-making for investments in novel energy systems and aid in developing effective policies for technology deployment.

SwinCell: a 3D transformer and flow-based framework for improved cell segmentation

Segmentation of three-dimensional (3D) cellular images is fundamental for studying and understanding cell structure and function. However, 3D cellular segmentation is challenging, particularly for dense cells and tissues. This challenge arises mainly from the complex contextual information within 3D images, anisotropic properties, and the sensitivity to internal cellular structures, which often lead to incorrect segmentation. In this work, we introduce SwinCell, a 3D transformer-based framework that leverages Swin-transformer to predict flow and differentiate individual cell instances. We demonstrate SwinCell’s utility in the segmentation of nuclei, colon tissue cells, and densely cultured cells. SwinCell strikes a balance between maintaining detailed local feature recognition and understanding broader contextual information. Through extensive testing with both public and in-house 3D cell imaging datasets, SwinCell shows utility in segmenting dense cells, making it a valuable tool for 3D segmentation in cellular analysis that could expedite research in cell biology and tissue engineering.
