SARS-CoV-2 variant nanobodies and constructs comprising such nanobodies

A large and highly diverse nanobody library was constructed and screened against multiple variants of SARS-COV-2 to find nanobodies with high sensitivity and specificity for the variants. Four rounds of positive selection against a panel of six diverse SARS-COV-2 variant RBDs was performed with our high-diversity. At least 59 of these nanobodies were found to work well against Alpha, Beta, Gamma, Delta, Kappa, Lambda and Mu with some overlap efficacy against other variants. These nanobodies have efficacy as stand-alone nanobodies and as a construct comprising nanobodies linked to the human IgG1 constant fragment (Fc) (nanobody-hFc constructions or nb-hFcs) to make enhanced humanized sdAbs with all the attributes of nanobodies with improved half-life and optimized effector functions. Several promising nanobodies that neutralize the original SARS-COV-2 and several of its variants have been identified, including Delta, with high efficacy. In particular, a subset of these nanobodies bind to the Omicron RBD.

Techno-Economic Analysis of Sorbent-Based Direct Air Capture Informed by EPC Input and Recent Technological Advancements

Direct air capture (DAC) technology has been developing at an accelerated rate. Independent, comprehensive, and transparent techno-economic analyses (TEAs) of DAC technologies are required to inform future development. NETL is publishing an update to the 2022 sorbent-based DAC case study report. This manuscript highlights select TEA results that will be presented in the upcoming NETL report. The analysis considers three sorbent-based DAC system configurations (direct steam temperature swing, indirect steam temperature swing, and direct steam temperature/vacuum swing) and two scenarios for providing the required electricity and steam (stand-alone system with a combined heat and power plant generating the required steam and electricity, and a purchased power case utilizing an electric boiler). Detailed sensitivity analyses are included and highlight the impact of external market factors, sorbent performance, and economies of scale. The potential for cost reductions by opting for temperature vacuum swing adsorption is highlighted. More detailed background and TEA results will be presented in the upcoming NETL report titled “Direct Air Capture Case Studies: Sorbent System (Revision 1)”.

Grid-Forming PV Inverter: Technology Development and Microgrid Applications

Presently, excluding residential backup power applications, grid-forming (GFM) inverters are commercially available for battery energy storage system (BESS) and some PV plus battery systems but not yet for stand-alone PV. This tech update investigates the control design for GFM PV inverter, especially the control required to provide active power reserve and to stabilize the dc link voltage. Subsequently, the use cases of GFM PV plants in utility-level microgrids are discussed considering fully inverter-based microgrids (i.e., with PV and BESS) as well as mixed-source microgrids (i.e., with PV and diesel generator). Scenarios where active power up-reserve from GFM PV plants is beneficial are illustrated. Moreover, requirements and specifications for GFM PV plant, especially for its active power reserve function are developed. In addition, stability of an example utility-level microgrid with two GFM PV plants and a diesel generator is analyzed to shed light on the potential unstable interaction between multiple GFM resources and the countermeasures. This research aims to advance GFM PV technology to offer more flexibility in microgrid design. With GFM PV available, depending on the level of reliability required, cost-benefit analysis, etc., the GFM BESS or diesel generator may be sized smaller while leveraging the full potential of PV generation inside the microgrid.

Current status of anaerobic digestion of food waste in the United States

Utilization of food waste (FW) and food processing waste streams in waste-to-energy technologies has increased over the years in United States (US). This review paper compiled information from published literature and open data sources to obtain a holistic view of the current state of FW availability and its management using anaerobic digestion (AD). The review examined the usage of FW as a feedstock for AD by considering the feedstock composition and availability of FW in the US as well as challenges and opportunities with implementing FW in mono- and co-digestion. Also presented are global trends in FW digestion and the current status of FW codigestion in the US. FW in the US was characterized with regard to volume and composition to show a need for waste management and an opportunity for valuable resource recovery. Challenges and opportunities of processing via AcoD were presented to review best practices for successful operations. Environmental standards, economic incentives, and governmental policies in the US and around the world regarding waste management and utilization were discussed to identify barriers and sources of opportunity for large-scale implementation of AcoD. Additionally, a range of facilities throughout the US was examined for information regarding digestion type, feedstock (s), biogas production and utilization, and energy self-sufficiency to better understand the current status of FW digestion. Several US facilities were outlined as case studies that co-digest FW and produce and utilize biogas for various applications. Future strategies, perspectives, and roadmaps were discussed to elucidate the shaping of future FW management practices.

Nb$_{3}$Sn SRF Photogun High Power Test at Cryogenic Temperatures

Superconducting RF (SRF) photoguns are emerging as promising candidates to produce highly stable electrons for Ultra fast Electron Diffraction/Microscopy (EUD/UEM) applications due to the ultrahigh shot-to-shot stability compared to room temperature RF photoguns. SRF technology was prohibitively expensive for industrial use until two recent advancements: Nb 3 Sn and conduction cooling. SRF gun can provide a CW operation capability while consuming only several Watts of RF power which eliminates the need of an expensive high power RF system and saves a facility footprint. Euclid is developing a continuous wave (CW), 1.5-cell, MeV-scale SRF conduction cooled photogun operating at 1.3 GHz. Here, in this paper, we present first high power results of the gun covered with Nb 3 Sn.

Use of Operating Agreements and Energy Storage to Reduce Photovoltaic Interconnection Costs: Conceptual Framework

This report explores one integrated technical and process concept designed to manage interconnection costs and streamline interconnection timelines to support near-term renewable energy deployment. We describe a new agreement between renewable energy developers and utilities, informed by the technical analysis. The agreement defines the operational parameters for a renewable energy system, with the goal of reducing risk and cost to all parties. This work provides a foundation upon which other states and utilities may build proof of concept. This report is supported by a technical analysis that is detailed in a companion report, "Use of Operating Agreements and Energy Storage to Reduce Photovoltaic Interconnection Costs: Technical and Economic Analysis" (McLaren et al. 2022).
