De Novo Design of High‐Affinity Miniprotein Binders Targeting Francisella Tularensis Virulence Factor

Abstract Francisella tularensis poses considerable public health risk due to its high infectivity and potential for bioterrorism. Francisella‐like lipoprotein (Flpp3), a key virulence factor unique to Francisella, plays critical roles in infection and immune evasion, making it a promising target for therapeutic development. However, the lack of well‐defined binding pockets and structural information on native interactions has hindered structure‐guided ligand discovery against Flpp3. Here, we used a combination of physics‐based and deep‐learning methods to design high‐affinity miniprotein binders targeting two distinct sites on Flpp3. We identified four binders for site I with binding affinities ranging between 24–110 nM. For the second site, an initial binder showed a dissociation constant ( K D ) of 81 nM, and subsequent site saturation mutagenesis yielded variants with sub‐nanomolar affinities. Circular dichroism confirmed the topology of designed miniproteins. The X‐ray crystal structure of Flpp3 in complex with a site I binder is nearly identical to the design model (Cα root‐mean‐square deviation (RMSD): 0.9 Å). These designed miniproteins provide research tools to explore the roles of Flpp3 in tularemia and should enable the development of new therapeutic candidates.

Turbomachinery Simulation Impact on Design, Understanding, and Optimization

This presentation shows the impact of Turbomachinery Simulation from simple analytical simulation to high fidelity CFD and Finite Element Analysis on the design of turbomachinery and the understanding of flow physics that is then used to improve design approaches. The impact of Optimization is also presented. The best approach for the tool development is to work with a compressor, fan, or turbine designer or to work on the design process directly. The presentation represents the work and impact of the author over his 45-year career and provides insight for both new and experienced engineers. The presentation explores applications of distortion from a downstream fan frame, the first uses of 3D CFD for fan, compressor and turbine design, and approaches to optimization for performance and structures.

Understanding Photovoltage Deficits in Electrochemically Grown Tin Sulfide (SnS) Thin-Film Photovoltaic Devices

Tin­(II) sulfide (SnS) is an earth-abundant semiconductor with a direct optical bandgap of ca. 1.1 eV, which makes it a promising absorber material for thin-film photovoltaic (PV) devices. However, existing devices have significant photovoltage deficits, which may be related to the anisotropic structure of the layered Herzenbergite SnS crystal structure. Here, we explore electrochemical deposition as a near room temperature path to oriented SnS crystal films on Mo and FTO substrates and employ vibrating Kelvin probe surface photovoltage (SPV) spectroscopy and J–V measurements to identify conversion losses in them. According to grazing-incidence X-ray diffraction and SEM, the SnS films consist of crystalline microplates with preferred orientation in the [111] and [001] directions. The bare SnS films produce only small and irreversible surface photovoltage signals, due charge trapping and recombination at the SnS surfaces, but addition of a CdS buffer layer lowers the charge recombination rate by 2 orders of magnitude and increases both the photovoltage and its reversibility due to the formation of a p-SnS/n-CdS junction. According to SPV, the FTO/SnS back interface (but not the Mo/SnS interface) forms a detrimental p–n junction that hinders hole transfer. Additional shunting through the relatively open microcrystal SnS layers and a lower conductivity of the FTO substrate explain the low power conversion efficiencies of the final devices (0.18 and 0.10% for the Mo and FTO substrates). Altogether, this work establishes a low-temperature path for the fabrication of crystalline SnS film-based solar cells and identifies the bottlenecks that limit high photoconversion efficiency.

Optimal experimental design using eigenvalue-based criteria with Pyomo.DoE

New developments in automated optimal experimental design within the PSE+ software ecosystem. Advancements in user experience (to reduce the time taken to perform optimal experiment design) and computational capabilities (allowing more diverse experimental design) are shown with an example relevant to critical minerals and materials. Also, a small tutorial on science-based optimal experimental design and novel contributions therein are presented.

Transit Thermal Control Design for Galileo Entry Probe for Planet Jupiter

A totally passive design was completed for the thermal control of the Galileo entry probe during its transit to the planet Jupiter. The design utilizes radio isotope heater units, multilayer insulation blankets and a thermal radiator in conjunction with a design conductance support structure to achieve both the required storage and critical initial planet atmosphere entry temperatures. The probe transit thermal design was completed and verified based on thermal vacuum testing of a prototype probe thermal test model.

NASA battery testbed: A designed experiment for the optimization of LEO battery operational parameters

Simulation of spacecraft battery operation is implemented. Robust design experiment to obtain optimum battery operational parameters is performed. It is found that short term tests using robust design of experiments can provide guidelines for optimum battery operation. It is decided to use robust design approach to provide guidelines for battery operation on current spacecraft in orbit as batteries age (GRO, UARS, EUVE, TOPEX).

Recent Developments in Safe Lithium Ion Battery Design for Human Space Flight

This presentation provides an overview of EVA batteries and introduces the concept of designing lithium ion batteries that resist the propagation of a single cell catastrophic failure. Thermal runaway initiation methods are briefly discussed, and the safe performance of the resulting designs is summarized. Approached in an incremental fashion, each subsequent battery design is introduced, ending with a current development for the Exploration EVA spacesuit. Challenges in achieving safe design performance with limited internal volume are briefly discussed and forward work is identified. Video examples of both propagating and non-propagating designs are included.
