Separation of granulocytes from whole blood by leukoadhesion, phase 1

Capillary glass tubes are investigated for the separation and retrieval of large quantities of viable granulocytes and monocytes from whole blood on a continuous basis from a single donor. This effort represented the feasibility demonstration of a three phase program for development of a capillary tube cell separation device. The activity included the analysis and parametric laboratory testing with subscale models required to design a prototype device. Capillary tubes 40 cm long with a nominal 0.030 cm internal diameter yielded the highest total process efficiency. Recovery efficiencies as high as 89% of the adhering cell population were obtained. Granulocyte phagocytosis of latex particles indicated approximately 90% viability. Monocytes recovered from the separation column retained their capability to stimulate human bone marrow colony growth, as demonstrated in an in vitro cell culture assay.

Mathematical synthesization of complex structures

A mathematical synthesization approach for rapidly modeling and analyzing complex structures comprised of hundreds or thousands of repeating individual structural members, interconnecting hardware, and other components was described. The techniques are applicable to any structure with repeating structural members. The capabilities are embodied within the Interactive Design and Evaluation of Advanced Spacecraft (IDEAS) computer-aided design system. IDEAS has a full range of integrated spacecraft modeling, design, analysis, performance, and cost estimating capabilities which include about 40 technical program and executive, data base, and file management systems. With IDEAS, a single user at an interactive terminal can create, design, analyze, and conduct parametric studies of Earth orbiting spacecraft in a timely, cost-efficient manner. The system is particularly useful in the conceptual design phases of advanced space missions when a multiplicity of concepts must be evaluated. The IDEAS interactive finite-element modeling capabilities are discussed. The mathematical synthesization programs were structured not only to provide a detailed finite-element model of the structure but also to automatically perform the many auxiliary computations needed to analyze the spacecraft in its orbital operational environment.

Physics-informed machine learning with differentiable programming for heterogeneous underground reservoir pressure management

Abstract Avoiding over-pressurization in subsurface reservoirs is critical for applications like CO $$_2$$ 2 sequestration and wastewater injection. Managing the pressures by controlling injection/extraction are challenging because of complex heterogeneity in the subsurface. The heterogeneity typically requires high-fidelity physics-based models to make predictions on CO $$_2$$ 2 fate. Furthermore, characterizing the heterogeneity accurately is fraught with parametric uncertainty. Accounting for both, heterogeneity and uncertainty, makes this a computationally-intensive problem challenging for current reservoir simulators. To tackle this, we use differentiable programming with a full-physics model and machine learning to determine the fluid extraction rates that prevent over-pressurization at critical reservoir locations. We use DPFEHM framework, which has trustworthy physics based on the standard two-point flux finite volume discretization and is also automatically differentiable like machine learning models. Our physics-informed machine learning framework uses convolutional neural networks to learn an appropriate extraction rate based on the permeability field. We also perform a hyperparameter search to improve the model’s accuracy. Training and testing scenarios are executed to evaluate the feasibility of using physics-informed machine learning to manage reservoir pressures. We constructed and tested a sufficiently accurate simulator that is 400 000 times faster than the underlying physics-based simulator, allowing for near real-time analysis and robust uncertainty quantification.

System maintenance manual for master modeling of aerodynamic surfaces by three-dimensional explicit representation

A system of computer programs has been developed to model general three-dimensional surfaces. Surfaces are modeled as sets of parametric bicubic patches. There are also capabilities to transform coordinate to compute mesh/surface intersection normals, and to format input data for a transonic potential flow analysis. A graphical display of surface models and intersection normals is available. There are additional capabilities to regulate point spacing on input curves and to compute surface intersection curves. Internal details of the implementation of this system are explained, and maintenance procedures are specified.

Sizing an emergency venting system for a cryogenic dewar

If the vacuum vessel that insulates a cryogenic dewar for a spaceborne experiment prior to launch is damaged, air will leak into the vacuum insulation space. As the sudden heat load causes the pressure to rise in the dewar, a safety disk in the emergency vent line will burst at the design pressure differential to allow vaporized cryogenic fluid to escape. The emergency vent line should be sized such that sufficient gaseous cryogen will be vented to keep the pressure inside the dewar below the design limit. On the other hand, the line should not be so large as to impose an unnecessary heat load on the dewar filled with cryogenic fluid. A vent-line computer program was generated to compute the maximum flow rate allowed for a proposed vent-line system. Parametric studies have been carried out for different burst disk pressure differentials, liquid cryogen ullage, and vent-line sizes.

Technology needs for lunar and Mars space transfer systems

The determination of appropriate space transportation technologies and operating modes is discussed with respect to both lunar and Mars missions. Three levels of activity are set forth to examine the sensitivity of transportation preferences including 'minimum,' 'full science,' and 'industrialization and settlement' categories. High-thrust-profile missions for lunar and Mars transportation are considered in terms of their relative advantages, and transportation options are defined in terms of propulsion and braking technologies. Costs and life-cycle cost estimates are prepared for the transportation preferences by using a parametric cost model, and a return-on-investment summary is given. Major technological needs for the programs are listed and include storable propulsion systems; cryogenic engines and fluids management; aerobraking; and nuclear thermal, nuclear electric, electric, and solar electric propulsion technologies.

Planar Inlet Design and Analysis Process (PINDAP)

The Planar Inlet Design and Analysis Process (PINDAP) is a collection of software tools that allow the efficient aerodynamic design and analysis of planar (two-dimensional and axisymmetric) inlets. The aerodynamic analysis is performed using the Wind-US computational fluid dynamics (CFD) program. A major element in PINDAP is a Fortran 90 code named PINDAP that can establish the parametric design of the inlet and efficiently model the geometry and generate the grid for CFD analysis with design changes to those parameters. The use of PINDAP is demonstrated for subsonic, supersonic, and hypersonic inlets.

6 um Pulsed OP-GaAs OPO Laser for Unambiguous Water Ice Detection on the Moon and Other Planetary Bodies

NASA demands a laser transmitter for planetary remote sensing to detect water-ice on the Moon and other planetary bodies. Based on the measurement of the Moon Mineralogy Mapper (M3) instrument, the discovery of water ice on the Moon was acclaimed, but the measurement is disputed because OH- and/or H2O-bearing materials share the absorption line around 3 μm wavelength. Lunar Flashlight, another mission project exploring the surface of Moon which will be launched later this year, enables to map the minerals on dark area of the Moon, but could not resolve the above-mentioned ambiguity. The absorption of 6.08 μm light is uniquely associated with the bending resonance of H2O since there is no comparable vibration in confounding OH-bearing materials. 6.08 μm laser between the atmospheric windows: middle-wave infrared (3-5 μm) and long-wave infrared (8-12 μm) has not been commercially available. Our approach is a Q-switched Ho:YLF laser pumped the orientation-pattern Gallium Arsenide optical parametric oscillator (OP-GaAs OPO) for generating high-energy laser pulses at the wavelength of 6.08 μm. In the current design, a 1.94 μm Tm:fiber is used as the pump source. In the compact design, a 1.94 μm laser diode will replace the Tm:fiber laser as the pump source. We will combine this proposed mid-infrared laser with the latest HgCdTe avalanche photodiode (APD) array to design a lidar for detecting water ice on the Moon and Mars from their respective orbits, enabling novel science and in-situ resource utilization. Our instrument is an enabling technology aboard the Artemis program and others.

Digital Elevation Map Parametric Error Analysis Using Corresponding NAC Images

Future lunar landing systems, particularly those used to land humans on the lunar surface aspart of the ARTEMIS program, will require precision navigation relative to the lunar surface. The most common way to meet these stringent navigation require-ments is through terrain relative navigation (TRN), which localizes a spacecraft by comparing descent im-agery with a predefined map of the surface. The accu-racy achievable using TRN is limited by the accuracy of the reference Digital Elevation Map (DEM). It is therefore critical for future lunar missions that potential errors in DEMs be quantified. This paper describes one of NASA’s current efforts to develop a process for evaluating lunar DEM quality.

Development of a hydrogen external burning flight test experiment on the NASA Dryden SR-71A aircraft

A captive-carry flight test experiment of the transonic external burning concept using gaseous hydrogen fuel has ben proposed for the SR-71A flight research test bed aircraft at the NASA Dryden Flight Research Facility, Edwards, California. This program will be the first large-scale, near-field flight test investigation of this base drag reduction technique at Mach numbers up to 3. Low-speed base drag reduction for supersonic and hypersonic flight vehicles is important in reducing vehicle drag and improving nozzle efficiency. Flight data will be correlated with a concurrent wind tunnel ground test of a similar test apparatus to confirm geometry and component performance scalability predictions and to investigate design parametrics under actual flight conditions. This paper describes the problem of transonic base drag for supersonic and hypersonic aircraft, a planned flight research program to address this problem, the test bed aircraft, and the flight experiment apparatus.

A cross-sectional aeroelastic analysis and structural optimization tool for slender composite structures

A fully open-source available framework for the parametric cross-sectional analysis and design optimization of slender composite structures, such as helicopter or wind turbine blades, is presented. The framework—Structural Optimization and Aeroelastic Analysis (SONATA)—incorporates two structural solvers, the commercial tool VABS, and the novel open-source code ANBA4. SONATA also parameterizes the design inputs, postprocesses and visualizes the results, and generates the structural inputs to a variety of aeroelastic analysis tools. It is linked to the optimization library OpenMDAO. This work presents the methodology and explains the fundamental approaches of SONATA. Structural characteristics were successfully verified for both VABS and ANBA4 using box beam examples from literature, thereby verifying the parametric approach to generating the topology and mesh in a cross section as well as the solver integration. The framework was furthermore exercised by analyzing and evaluating a fully resolved highly flexible wind turbine blade. Computed structural characteristics correlated between VABS and ANBA4, including off-diagonal terms. Stresses, strains, and deformations were recovered from loads derived through coupling with aeroelastic analysis. The framework, therefore, proves effective in accurately analyzing and optimizing slender composite structures on a high-fidelity level that is close to a three-dimensional finite element model.

stor4build

The EnergyPlus simulation engine supports modeling and simulation of thermal energy storage (TES) systems in several ways, including using the Python-EMS feature, which extends the operation of the engine with custom code written in Python. Creation of models using this feature can be tedious and error prone, with the connection of the model components to the Python code a particularly troublesome area. The stor4build Python package simplifies this process by modifying an input model to add a selected TES technology (implemented with the Python-EMS feature) and runs the simulation. The package leverages the OpenStudio middleware software development kit to automate this process as much as possible, eliminating potential errors and simplifying usage of EnergyPlus. The package provides objects, functions, and OpenStudio measures that implement the necessary operations to automate the creation of EnergyPlus models that integrate TES technologies with building systems. In addition, two user interfaces are provided: a command line interface and a web application programming interface. The automated process implemented by the package greatly simplifies the modeling and simulation process, allowing for parametric studies to be executed much more efficiently and effectively. The OpenStudio-based workflow is also very flexible and will allow for future additions of new technologies.
