Roundup causes embryonic development failure and alters metabolic pathways and gut microbiota functionality in non-target species

Background: Research around the weedkiller Roundup is among the most contentious of the twenty-first century. Scientists have provided inconclusive evidence that the weedkiller causes cancer and other life-threatening diseases, while industry-paid research reports that the weedkiller has no adverse effect on humans or animals. Much of the controversial evidence on Roundup is rooted in the approach used to determine safe use of chemicals, defined by outdated toxicity tests. We apply a system biology approach to the biomedical and ecological model species Daphnia to quantify the impact of glyphosate and of its commercial formula, Roundup, on fitness, genome-wide transcription and gut microbiota, taking full advantage of clonal reproduction in Daphnia. We then apply machine learning-based statistical analysis to identify and prioritize correlations between genome-wide transcriptional and microbiota changes. Results: We demonstrate that chronic exposure to ecologically relevant concentrations of glyphosate and Roundup at the approved regulatory threshold for drinking water in the US induce embryonic developmental failure, induce significant DNA damage (genotoxicity), and interfere with signaling. Furthermore, chronic exposure to the weedkiller alters the gut microbiota functionality and composition interfering with carbon and fat metabolism, as well as homeostasis. Using the “Reactome,” we identify conserved pathways across the Tree of Life, which are potential targets for Roundup in other species, including liver metabolism, inflammation pathways, and collagen degradation, responsible for the repair of wounds and tissue remodeling. Conclusions: Our results show that chronic exposure to concentrations of Roundup and glyphosate at the approved regulatory threshold for drinking water causes embryonic development failure and alteration of key metabolic functions via direct effect on the host molecular processes and indirect effect on the gut microbiota. The ecological model species Daphnia occupies a central position in the food web of aquatic ecosystems, being the preferred food of small vertebrates and invertebrates as well as a grazer of algae and bacteria. The impact of the weedkiller on this keystone species has cascading effects on aquatic food webs, affecting their ability to deliver critical ecosystem services.

Evaluation of Multi-cell Failure Propagation

Failure propagation testing is of increasing interest to the designers and end users of battery systems. One of the chief difficulties, however, is choosing an appropriate initiation method to perform the test. Single cell abuse testing is typically used to initiate thermal runaway but this can involve a large amount of additional energy injected into the system. It is assumed that this will have some impact on the behavior of a propagating thermal runaway event, but there is little data available as to how significant this would be. Further, it is ultimately difficult to develop viable propagation tests for compliance and public safety activities without better knowledge of how test methods will impact the results. This work looks at propagating battery failure with a variety of chemistries, formats, configurations and initiation methods to determine the level of significance of the chosen initiation method on the test results. We have ultimately found while there is some impact on the detailed results of propagation testing, in most cases other factors, particularly the energy density of the system play a much greater role in the likelihood of a propagation event consuming an entire battery. We have also provided some guidelines for test design to support best practices in testing.

WhiteWind: White Etching Crack (WEC) Bearing Failures in Wind Turbine: Cooperative Research and Development, CRADA Number CRD-18-00758 (Final Report)

A WEC is a particularly aggressive, unpredictable and wide spread rolling element bearing failure mode that is common for large multi-megawatt (MW) wind turbines. WEC is considered the single most expensive failure mode for all wind turbine components, and there is currently no commercial solution. The Technical University of Denmark (DTU) is leading the WhiteWind project to investigate WECs through funding provided by the Innovation Fund Denmark. Other project partners include Vestas, SKF, Expanite, Rheinisch-Westfalische Technische Hochschule Aachen University (RWTH Aachen), and Argonne National Laboratory. The objective of the overall project is to develop a new surface engineered WEC-resistant bearing material using novel surface engineering techniques that shall provide a commercially competitive alternative to existing wind turbine bearings. NREL will support the project by providing existing measured bearing loads and validating models of drivetrain loads.

Laser Welder Startup Capacitor Failure Analysis & Suggested Potential Mitigation Factors [Slides]

On the morning of the failure (Mon 2/23/2023 5 AM), an unclassified Yttrium Aluminum Garnet Laser Welder system was working within operational parameters. However, when another part was positioned for drilling and it was activated, the coolant compressor was heard shutting off by everyone working on the laser. The coolant pump reportedly failed due to blown 5-amp fuses. Since the coolant pump was offline, the laser was automatically placed into an inoperable state. Failure of the coolant pump was due to a failed capacitor in the startup circuit for the pump.

Degradation and Failure Phenomena of Advanced Reactor Fuel Concepts: Sodium-Cooled Fast Reactor Metallic Fuel

The U.S. Nuclear Regulatory Commission (NRC) is anticipating licensing applications and commercial use of new fuel types in advanced nuclear power reactors that would be designed and built in the United States. Pacific Northwest National Laboratory (PNNL) is providing technical assistance to the NRC related to the newly proposed nuclear fuel and cladding designs that would be deployed in these reactors. This report focuses specifically on the metallic fuel that is being considered for sodium-cooled fast reactors and specifically on mechanisms that would cause damage or failure to the fuel under reactor operating conditions and design basis accident conditions. There is historic experience with both metallic and oxide fuels in experimental sodium-cooled fast reactors, but this report will focus solely on metallic fuel. Currently two U.S.-based reactor designers are engaged with the NRC in the application or pre-application review and considering a sodium-cooled fast reactor. TerraPower is engaged with NRC in pre-application review of its Natrium reactor. The current design for this fuel uses a sodium-bonded uranium-10wt% zirconium (U10Zr) fuel clad in HT9 stainless steel. ARC Clean Technology is engaged with NRC in pre-application review of its ARC-100 reactor. The current design for this fuel uses a sodium-bonded uranium-10wt% zirconium (U10Zr) fuel with steel cladding. This report will focus on this fuel system specifically, with broader information given regarding other metallic fuel systems with other stainless steel alloy claddings. To support the NRC’s readiness efforts, this report will identify and discuss degradation and failure modes of metallic fuel concepts for sodium-cooled fast reactors, including fuel performance characteristics that may not be addressed within existing regulatory documents.

Public values failure associated with Hurricane Ian power outages

Power outages from extreme weather events can diminish community resilience, making it difficult for the areas impacted to bounce back after such events. For socially vulnerable populations, the frequency and duration of power outages can be even more severe. Governments have an obligation to protect public values, or those values that are most fundamental to society, which includes equitable resilience. Using Jørgensen and Bozeman's inventory of public values, this manuscript explores how power outages from extreme weather events create public values failures. More specifically, the manuscript evaluates intraorganizational aspects of public administration during power outages in Florida during Hurricane Ian in 2022. Framing power outages as a public values failure may motivate greater time and effort toward improving equitable access to more resilient power systems.

Investigation of Failure Modes, Mechanisms and Driving Forces for Electrically Conductive Adhesives as Interconnects in PV Modules

In this work we describe our current progress into investigating the reliability of ECA interconnects for PV modules. Preliminary thermal cycling experiments demonstrated that both cohesive and adhesive failure of the ECA joint are relevant failure modes that result in an increase of the joints electrical resistance. A thorough viscoelastic characterization of the candidate ECA was conducted to enable accurate numerical modeling of the joint’s response to thermal cycling. Combined with critical and subcritical fracture measurements, the numerical model elucidates that an additional fatigue mechanism exists which can result in fracture well below subcritical debonding threshold values. It is also demonstrated that fracture of the ECA is more sensitive to humidity than temperature and subcritical debonding mechanisms do exist which are again more sensitive to humidity.

Investigating lab-scaled offshore wind aerodynamic testing failure and developing solutions for early anomaly detections

As offshore wind systems become more complex, the risk of human error or equipment malfunction increases during experimental testing. This study investigates a lab-scale incident involving a 1 : 50 scale 5 MW wind turbine, where a generator failure led to rotor overspeed and a blade–tower strike. To improve early fault detection, we propose a data-driven method based on multivariate long short-term memory (LSTM) models. High-frequency measurements are projected onto principal components, and anomalies are identified using reconstruction error and its time derivative. Two models are trained on different healthy datasets and tested using single- and multi-principal component (1PC and MPC) variations. Results show that combining both error and error derivative improves detection accuracy. The 1PC model detects faults faster, has a higher recall rate, and achieves a 43 % improvement in anomaly detection accuracy, while the MPC model yields higher precision. This approach provides a simple and effective tool for early anomaly detection in lab-scale experiments, helping to reduce the risk of future failures during the testing of new technologies.

Dimensionless parameters to categorize the failure modes of ductile plate perforation

In this work, two dimensionless parameters were derived organically from impact mechanics equations to remove the need for empirical constants. These two dimensionless parameters effectively collapse the ballistic performance of ductile metal plates along two distinct loci, depending on the projectile geometry and the corresponding failure mode. Projectiles that initiate ductile hole enlargement via cavity expansion fall along a linear curve, while shear-plugging failure modes tend to fall along a cubic curve. Results were demonstrated for various projectiles of different calibers impacting aluminum and Ti-6Al-4V alloys. The main advantage of the approach in this work is to drastically reduce the number of features for input into machine learning algorithms.

Tolerating memory stack failures in multi-stack systems

Memory management circuitry and processes operate to improve reliability of a group of memory stacks, providing that if a memory stack or a portion thereof fails during the product's lifetime, the system may still recover with no errors or data loss. A front-end controller receives a block of data requested to be written to memory, divides the block into sub-blocks, and creates a new redundant reliability sub-block. The sub-blocks are then written to different memory stacks. When reading data from the memory stacks, the front-end controller detects errors indicating a failure within one of the memory stacks, and recovers corrected data using the reliability sub-block. The front-end controller may monitor errors for signs of a stack failure and disable the failed stack.

Methods and systems for diagnosis of failure mechanisms and for prediction of lifetime of metal batteries

Methods for diagnosing failure mechanisms and for predicting lifetime of metal batteries include monitoring rest voltage and Coulombic Efficiency over relatively few cycles to provide profiles that indicate, by the trends thereof, a particular failure mechanism (e.g., electrolyte depletion, loss of metal inventory, increased cell impedance). The methods also include cycling over relatively few cycles an anode-free cell, having the same cathode and electrolyte as the metal battery, but with a current collector instead of the anode. Discharge capacity is monitored and profiled, and a discharge capacity curve is fitted to the discharge capacity profile to discern a capacity retention per cycle. The lifetime of the metal battery is determined using the capacity retention per cycle discerned from the anode-free cell. Related systems include a metal-based battery and an anode-free cell or a battery cell reconfigurable between a metal-based and an anode-free cell.

Hydrogen Equipment Enclosure Risk Reduction through Earlier Detection of Component Failures

Hydrogen component reliability and the hazard associated with failure rates is a critical area of research for the successful implementation and growth of hydrogen technology across the globe. The research team has partnered to quantify system risk reduction through earlier detection of hydrogen component failures. A model of hydrogen dispersion in a hydrogen equipment enclosure has been developed utilizing experimentally quantified hydrogen component leak rates as inputs. This model provides insight into the impact of hydrogen safety sensors and ventilation on the flammable mass within a hydrogen equipment enclosure. This model also demonstrates the change in safety sensor response time due to detector placement under various leak scenarios. The team looks to improve overall hydrogen system safety through an improved understanding of hydrogen component reliability and risk mitigation methods. This collaboration fits under the work program of IEA Hydrogen Task 43 Subtask E Hydrogen System Safety.

Development of mechanistic, microstructure-informed BISON models for fission product-induced failure mechanisms in advanced nuclear fuels

The US continues to prioritize commercial deployment of advanced reactors—particularly those utilizing U-Zr metallic and TRISO particle fuels. Fission product-induced failure mechanisms for these systems include fuel–cladding chemical interaction in metallic fuel rods, which threatens cladding integrity, and Pd penetration in TRISO particles, which degrades SiC layer properties. Empirical models can be applied within the bounds of existing irradiation databases, but their utility is limited when considering new designs or investigating fundamental material behaviors. The Nuclear Energy Advanced Modeling and Simulation Program is therefore developing mechanistic models for these behaviors, which are expected to aid in fuel design, assist in development of failure mitigation strategies, and provide support for qualification and licensing. These efforts leverage multiscale capabilities to capture the microstructural features and processes that govern these behaviors. This talk summarizes past and ongoing efforts to develop and validate these models for the BISON fuel performance code.
