A simple model of fluid flow and electrolyte balance in the body

The model is basically a three-compartment model, the three compartments being the plasma, interstitial fluid and cellular fluid. Sodium, potassium, chloride and urea are the only major solutes considered explicitly. The control of body water and electrolyte distribution is affected via drinking and hormone levels. Basically, the model follows the effect of various oral input water loads on solute and water distribution throughout the body.

Effects of gravity on contractile proteins

A method was established for the isolation and purification of nuclei in high yield from the microplasmodia of Physarum flavicomum. Purified nuclei were resistant to breakage by methods commonly employed for isolated plant and animal nuclei. Several methods for the extraction of nuclear protein were compared. Incubation of nuclear lysates with either 2 M NaCl, with or without 5 M urea, or 1 M CaCl2 resulted in the extraction of nuclear action together with histones. The histones were chemically fractionated into the 5 basic groups common to other eucaryotic tissue. Amino acid analyses of the total histone were also performed. Nuclear actin was found to have a molecular weight of 41,000 ? 4,000 daltons as determined by SDS polyacrylamide gel electrophoresis. The amino acid composition of the nuclear action was established.

Cyanamide mediated synthesis under plausible primitive earth conditions. VI - The synthesis of glycerol and glycerophosphates

The formation of glycerol occurs when a solution of DL-glyceraldehyde is heated in the presence of hydrogen sulfide at room temperature. DL-glyceraldehyde and dihydroxyacetone treated with hydrazine, as well as DL-glyceraldehyde incubated with formaldehyde are also partially converted to glycerol. The yields of the above reactions are from approximately 1% to about 3%. The formation of glycerophosphates occurs when glycerol is heated with ammonium dihydrogen phosphate and either urea or cyanamide. The yield of glycerophosphates is about 30%, most of which is sn-glycero-1(3)-phosphate. These findings indicate that glycerol and sn-glycero-3-phosphate, which are moieties of glycerolipids, could have been formed under conditions which may have prevailed on the primitive earth.

Neutralizing Amine-Cured Epoxy Surfaces

New surface treatment is a rapid, convenient, and effective method for converting unused amines and amine functional groups into neutral, noncorrosive substituted urea. Reaction proceeds at room temperature, takes only a few minutes, and leaves no corrosive residue. Surface should first be washed with alcohol to remove as much as possible of unreacted amine. Then it should be dried, since residual moisture or alcohol may consume some of treatment reagent and neutralization may then be incomplete.

Studies on chemical evolution using carbon suboxide

It was learned that when carbon suboxide polymers react with hydroxylamine in modified sea water the amino acids glycine and lysine are produced and that they react with urea to produce nucleic acid-like substances. These results lead to the supposition that it is possible that carbon suboxide polymers may have accumulated in the primeval sea some three or more billion years ago and were transformed into the procursors of living molecules.

Structural studies on HCN oligomers

NMR spectral studies on the HCN oligomers suggest the presence of carboxamide and urea groupings. The release of CO2, H2O, HCN, CH3CN, HCONH2 and pyridine on pyrolysis is consistent with the presence of these groupings as well as carboxylic acid groups. No basic primary amine groupings could be detected with fluorescamine. Hydrazinolysis of the HCN oligomers releases 10% of the amino acids normally released by acid hydrolysis. The oligomers give a positive biuret test but this is not due to the presence of peptide bonds. There is no conclusive evidence for the presence of peptide bonds in the HCN oligomers. No diglycine was detected on partial hydrolysis of the HCN oligomers at pH 8.5 suggesting that HCN oligomers were not a source of prebiotic peptides.

Suspension restraint - Induced hypokinesia and antiorthostasis as a simulation of weightlessness

Muscle, renal, fluid and electrolyte responses were measured in suspended rats; the hind limbs are non-load bearing and the front limbs can be used for feeding and grooming. Hind limb hypokinesia reverses after removal from the suspension harness. This suspension system is adjustable for a head-down tilt to produce antiorthostatic responses which are also reversible. Responses to hypokinesia or antiorthostatic hypokinesia for up to 14 days were measured, e.g., muscle atrophy: soleus greater than gastrocnemius equals plantaris greater than extensor digitorum longus, kaliuresis, and increased excretion of urea, NH3, and 3 methylhistidine. Muscle protein loss, a response to a reduction in RNA, is also reversible. A head-down tilt for 7-14 days results in diuresis and natriuresis. These changes are reversed within 24 hours after removal from the restraint harness. Physiological effects of suspension restraint can be used to simulate and predict responses to microgravity exposure.

Method of neutralizing the corrosive surface of amine-cured epoxy resins

The corrosive alkaline surface layer of an epoxy resin product formed by the curing of the epoxy with an aliphatic amine is eliminated by first applying a non-solvent to remove most or all of the free unreacted amine and then applying a layer of a chemical reagent to neutralize the unused amine or amine functional groups by forming a substituted urea. The surface then may be rinsed with acetone and then with alcohol. The non-solvent may be an alcohol. The neutralizing chemical reagent is a mono-isocyanate or a mono-isothiocyanate. Preferred is an aromatic mono-isocyanate such as phenyl isocyanate, nitrophenyl isocyanate and naplthyl isocyanate.

Prebiotic synthesis in atmospheres containing CH4, CO, and CO2. I - Amino acids

The prebiotic synthesis of amino acids, HCN, H2CO, and NH3 using a spark discharge on various simulated primitive earth atmospheres at 25 C is investigated. Various mixtures of CH4, CO, CO2, N2, NH3, H2O, and H2 were utilized in different experiments. The yields of amino acids (1.2-4.7 percent based on the carbon) are found to be approximately independent of the H2/CH4 ratio and the presence of NH3, and a wide variety of amino acids are obtained. Glycine is found to be almost the only amino acid produced from CO and CO2 model atmospheres, with the maximum yield being about the same for the three carbon sources at high H2/carbon ratios,whereas CH4 is superior at low H2/carbon ratios. In addition, it is found that the directly synthesized NH3 together with the NH3 obtained from the hydrolysis of HCN, nitriles, and urea could have been a major source of ammonia in the atmosphere and oceans of the primitive earth. It is determined that prebiotic syntheses from HCN and H2CO to give products such as purines and sugars and some amino acids could have occurred in primitive atmospheres containing CO and CO2 provided the H2/CO and H2/CO2 ratios were greater than about 1.0.

Extensions of suspension systems to measure effects of hypokinesia/hypodynamia and antiorthostasis in rats

Suspension systems are used to simulate hypokinetic/hypodynamic (H/H) and anitorthostatic (AO) responses seen under conditions of weightlessness. Growing rats in H/H suspension with unloaded hindlimbs for one and two weeks respond with muscle atrophy and increased excretion of nitrogenous end products such as urea, NH3 and 3 methyl histidine. Since muscle is in a dynamic state of synthesis and breakdown of protein, relationships between protein, RNA and DNA contents in the four muscles which reflect weight bearing and non-weight bearing functions were assessed. Protein and RNA progressively decreased over a one and two week period of H/H suspension: soleus gastrocnemius=plantaris EDL. Concommitant analysis of DNA contents showed there were no changes. The interpretation was that protein synthesis was slowed during H/H. As with muscle mass, protein and RNA levels recovered rapidly after removal from H/H. The AO rats (which are also H/H) respond with diuresis, natriuresis and kaliuresis in a manner comparable to responses seen when thoracic blood vessels are volume loaded.

Metal phthalocyanine polymers

Metal 4, 4', 4", 4"'=tetracarboxylic phthalocyanines (MPTC) are prepared by reaction of trimellitic anhydride, a salt or hydroxide of the desired metal (or the metal in powdered form), urea and a catalyst. A purer form of MPTC is prepared than heretofore. These tetracarboxylic acids are then polymerized by heat to sheet polymers which have superior heat and oxidation resistance. The metal is preferably a divalent metal having an atomic radius close to 1.35A.

Rat hindlimb muscle responses to suspension hypokinesia/hypodynamia

Hypokinetic/hyupodynamic (H/H) whole body suspension of rats eliminates hindlimb load bearing functions while permitting continued use of the forelimbs. Responses of hindlimb muscles were assessed in terms of absolute and relative weights during 1 and 2 weeks of H/H suspension. Muscle mass loss was in the order soleus greater than gastrocnemius equal to plantaris greater than extensor digitorum longus (EDL). The soleus, a postural antigravity muscle composed mainly of slow twitch fibers, was most sensitive, losing 35 and 45 percent of its weight during the first and second weeks, respectively. The gastrocnemius and plantaris showed losses during the first week but no significant loss during the second wee. The EDL showed little or no weight loss. During post suspension recovery all muscles showed a weight gain. H/H suspended rats failed to grow; following removal from suspension they gained weight linearly, comparable to controls. Products of muscle metabolism including urea, ammonia, and 3-methylhistidine increased in the urine during H/H suspension and were significantly reduced approaching control levels during recovery. This suspension model offers considerable promise for comparison with H/H responses during weightlessness.

Laser-induced fluorescence of green plants. I - A technique for the remote detection of plant stress and species differentiation

The laser-induced fluorescence (LIF) of green plants was evaluated as a means of remotely detecting plant stress and determining plant type. Corn and soybeans were used as representatives of monocots and dicots, respectively, in these studies. The fluorescence spectra of several plant pigments was excited with a nitrogen laser emitting at 337 nm. Intact leaves from corn and soybeans also fluoresced using the nitrogen laser. The two plant species exhibited fluorescence spectra which had three maxima in common at 440, 690, and 740 nm. However, the relative intensities of these maxima were distinctly different for the two species. Soybeans had an additional slight maxima at 525 nm. Potassium deficiency in corn caused an increase in fluorescence at 690 and 740 nm. Simulated water stress in soybeans resulted in increased fluorescence at 440, 525, 690, and 740 nm. The inhibition of photosynthesis in soybeans by 3-(3-4-dichlorophenyl)-1-1-dimethyl urea (DCMU) gave incresed fluorescence primarily at 690 and 740 nm. Chlorosis as occurring in senescent soybean leaves caused a decrease in fluorescence at 690 and 740 nm. These studies indicate that LIF measurements of plants offer the potential for remotely detecting certain types of stress condition and also for differentiating plant species.

Hyperfiltration wash water recovery subsystem - Design and test results

The Hyperfiltration Wash Water Recovery (HWWR) subsystem, designed to offer low-power high-volume wash water purification for extended mission spacecraft, is discussed in terms of preprototype design and configuration. Heated wash water collected from the shower, hand wash, and laundry flows into a temperature-controlled (374 K) waste storage tank. Two parallel 25 micron absolute filters at the tank outlet remove large particles from the feed stream. A positive displacement feed pump delivers wash water to the hyperfiltration module at a constant flow rate of 0.20 lpm with discharge pressure variations from 4181-7239 Kpa. The hyperfiltration membrane module is a single-pass design including 36 porous stainless steel tubes, and is designed to provide an approximate water recovery rate of 90 percent. Permeate and brine water flows are monitored by flow meters, and removal of urea and ammonia is achieved by adding 15 percent NaOCl solution to the permeate fluid stream. An alternate module design using two diameters of tubing (allowing a smaller pressure drop and a larger membrane area) gave a superior predicted performance over the first module with larger tubing throughout.

A suspension model for hypokinetic/hypodynamic and antiorthostatic responses in the mouse

Hypokinetic/hypodynamic and antiorthostatic responses to weightlessness and bedrest were simulated in mice using a suspension technique. Animals were suspended for 1 or 2 weeks in an antiorthostatic posture and positioned to permit freedom of movement but eliminate load bearing by the hindlimbs. Suspended mice exhibited reduced food and water intakes and a rapid 10 percent decrease in body weight to a level which was maintained for the remainder of the suspension period. Diuresis was evident in suspended mice, but the natriuresis and kaliuresis previously observed in the suspended rat were not evident. Differential hindlimb muscle atrophy and increased excretion of urea and ammonia were also noted in suspended mice. Postsuspension recovery studies indicated that the recovery process was highly effective. These results document specific responses in the mouse and similarities in the responses of mice and rats to suspension. These studies expand the utility of the suspension model and suggest that the mouse may be useful in future studies simulating both weightlessness and bedrest.

Progress in optical parametric oscillators

It is pointed out that tunable coherent sources are very useful for many applications, including spectroscopy, chemistry, combustion diagnostics, and remote sensing. Compared with other tunable sources, optical parametric oscillators (OPO) offer the potential advantage of a wide wavelength operating range, which extends from 0.2 micron to 25 microns. The current status of OPO is examined, taking into account mainly advances made during the last decade. Attention is given to early LiNbO3 parametric oscillators, problems which have prevented wide use of parametric oscillators, the demonstration of OPO's using urea and AgGaS2, progress related to picosecond OPO's, a breakthrough in nanosecond parametric oscillators, the first demonstration of a waveguide and fiber parametric amplification and generation, the importance of chalcopyrite crystals, and theoretical work performed with the aim to understand the factors affecting the parametric oscillator performance.

Atrophy and growth failure of rat hindlimb muscles in tail-cast suspension

The primary objective of the present study is related to an evaluation of a modified tail-cast suspension model as a means of identifying metabolic factors which control or are associated with muscle atrophy and growth failure. Two different control conditions (normal and tail-casted weight bearing) were studied to determine the appropriate control for tail-cast suspension. A description is presented of a model which is most useful for studying atrophy of hindlimb muscles under certain conditions. Female Sprague-Dawley rats were employed in the experiments. Attention is given to growth rate and urinary excretion of urea and ammonia in different types of rats, the relationship between body weight and skeletal muscle weight, and the relationship between animal body weight and rates of protein synthesis and protein degradation.

Growth of Organic Crystals by Ostwald Ripening

The objective of this investigation is to evaluate the growth of various organic crystals by chemical precipitation and Ostwald ripening. Six precipitation reactors were flown on STS-51A. Five of the reactors contained proprietary materials. The sixth contained urea dissolved in ethanol with toluene as the precipitating agent. The size distribution will be analyzed and compared with a similar model being developed.
