Space biosynthesis systems

Model reactions based on chemical, enzymatic or cellular conversion of D glucose into d gluconic acid are designed to unequivocally define the advantages of microgravity on reaction mechanisms, mass-transfers and separation of organic chemicals and to serve as procedures to test the performance characteristics of space bioprocessing equipment.

Polyamines and plant stress - Activation of putrescine biosynthesis by osmotic shock

The putrescine content of oat leaf cells and protoplasts increases up to 60-fold within 6 hours of exposure to osmotic stress (0.4 to 0.6 molar sorbitol). Barley, corn, wheat, and wild oat leaves show a similar response. Increased arginine decarboxylase activity parallels the rise in putrescine, whereas ornithine decarboxylase remains unchanged. DL-alpha-Difluoromethylarginine, a specific irreversible inhibitor of arginine decarboxylase, prevents the stress-induced rise in increase in arginine decarboxylase activity and putrescine synthesis, indicating the preferential activation of this pathway.

Aromatic biosynthesis in pine tissues

Pinus elliotti is a woody plant species responsive to gravity and capable of synthesizing large quantities of lignin. Lignification begins very quickly after germination; lignin is detected in the vascular region within 4 days after germination and rapidly progresses up the hypocotyl. Young pine seedlings bend in response to geostimulation for about 10 days after germination, with the most rapid response time occurring in 4- to 5-day-old seedlings. Various chemicals were used to establish their effects on the geotropic response in this gymnosperm species. IAA completely arrests the geotropic response for 18 to 24 hr. Afterward the seedlings respond geostimulation as if they were not treated. The same pattern of response will occur with a second IAA treatment. If the synthetic auxin, 2-4,D, is used, the georesponse is permanently blocked. The method of application does not appear to be critical; addition of auxin to only one side of the seedling gave results similar to those obtained by treating the entire seedling.

Interactions of light and gravity on growth, orientation, and lignin biosynthesis in mung beans

Mung beans (Vigna radiata L.) seedlings grown on the third Space Transport Mission (STS-3) showed marked orientation problems (some of the stems elongated horizontally and many of the roots were growing upward) and had a lower lignin content than the ground based controls. This research was initiated to determine if the atypical growth characteristics of mung beans grown in microgravity could be simulated using horizontal clinostats. Most of the effort focused on the design, construction and testing of the clinostats. In order to closely approximate the growth conditions of the plants grown in the plant growth unit on STS-3, cylindrical lexan minichambers were constructed. Results showed that plants grown using these clinostats in the horizontal position exhibit similar growth characteristics to the plants grown on STS-3 (disorientation of both stems and roots), while the vertical stationary and vertical rotating controls exhibit normal growth.

Anaerobic biosynthesis of unsaturated fatty acids in the cyanobacterium, Oscillatoria limnetica

The mechanism for synthesis of monounsaturated fatty acids under aerobic and anaerobic conditions was studied in the facultative anaerobic cyanobacterium, Oscillatoria limnetica. The hexadecenoic acid (C16:1) of aerobically grown O. limnetica was shown to contain both the delta 7 (79%) and delta 9 (21%) isomers, while the octadecenoic (C18:1) acid was entirely the delta 9 acid. Incorporation of [2-14C] acetate into the fatty acids under aerobic conditions resulted in synthesis of the delta 7 and delta 9 C16:1 and the delta 9 C18:1. Synthesis of unsaturated fatty acids in the presence of DCMU required sulfide. Anaerobic incubations in the presence of DCMU and sulfide (less than 0.003% atmospheric oxygen) resulted in a two-fold increase in monounsaturated fatty acids of both delta 7 and delta 9 C16:1 and delta 9 and delta 11 C18:1. The synthesis of these is characteristic of a bacterial-type, anaerobic pathway.

Kinetic studies on the control of the bean rust fungus (Uromyces phaseoli L.) by an inhibitor of polyamine biosynthesis

alpha-Difluoromethylornithine (DFMO), a specific and irreversible inhibitor of the polyamine biosynthetic enzyme ornithine decarboxylase, effectively inhibits mycelial growth of several phytopathogenic fungi on defined media in vitro and provides systemic protection of bean plants against infection by Uromyces phaseoli L. race 0 (MV Rajam, AW Galston 1985 Plant Cell Physiol 26: 683-692; MV Rajam et al. 1985 Proc Natl Acad Sci USA 82: 6874-6878). We now find that application of 0.5 millimolar DFMO to unifoliolate leaves of Pinto beans up to 3 days after inoculation with uredospores of U. phaseoli completely inhibits the growth of the pathogen, while application 4 or 5 days after inoculation results in partial protection against the pathogen. Spores do not germinate on the surface of unifoliolate leaves treated with DFMO 1 day before infection, but addition of spermidine to the DFMO treatments partially reverses the inhibitory effect. The titer of polyamines in bean plants did not decline after DFMO treatment; rather, putrescine and spermidine contents actually rose, probably due to the known but paradoxical stimulation of arginine decarboxylase activity by DFMO.

Relief from glucose interference in microcin B17 biosynthesis by growth in a rotating-wall bioreactor

Glucose interference in production of microcin B17 by Escherichia coli ZK650 was decreased sevenfold by growth in a ground-based rotating-wall bioreactor operated in the simulated microgravity mode as compared with growth in flasks. When cells were grown in the bioreactor in the normal gravity mode, relief from glucose interference was even more dramatic, amounting to a decrease in glucose interference of over 100-fold.

Mutations in Arabidopsis thaliana genes involved in the tryptophan biosynthesis pathway affect root waving on tilted agar surfaces

Arabidopsis thaliana roots grow in a wavy pattern upon a slanted surface. A novel mutation in the anthranilate synthase alpha 1 (ASA1) gene, named trp5-2wvc1, and mutations in the tryptophan synthase alpha and beta 1 genes (trp3-1 and trp2-1, respectively) confer a compressed root wave phenotype on tilted agar surfaces. When trp5-2wvc1 seedlings are grown on media supplemented with anthranilate metabolites, their roots wave like wild type. Genetic and pharmacological experiments argue that the compressed root wave phenotypes of trp5-2wvc1, trp2-1 and trp3-1 seedlings are not due to reduced IAA biosynthetic potential, but rather to a deficiency in L-tryptophan (L-Trp), or in a L-Trp derivative. Although the roots of 7-day-old seedlings possess higher concentrations of free L-Trp than the shoot as a whole, trp5-2wvc1 mutants show no detectable alteration in L-Trp levels in either tissue type, suggesting that a very localized shortage of L-Trp, or of a L-Trp-derived compound, is responsible for the observed phenotype.

Lipid biosynthesis and abiotic stress resilience in photosynthetic organisms

This application describes consortium between fungi and algae, where the algae are incorporated within hyphae of the fungi. The fungi, the algae, or both can be modified to express heterologous lipid synthesizing enzymes. Incorporation of algae into fungi facilitates harvesting of the algae and products produced by the consortia. Such consortia are robust. For example, the fungi and algae can symbiotically provide nutrients to each other and are tolerant of environmental stresses.
