Structural and Spectroscopic Comparison of Soft‐Se vs. Hard‐O Donor Bonding in Trivalent Americium/Neodymium Molecules

Covalency is often considered to be an influential factor in driving An 3+ vs. Ln 3+ selectivity invoked by soft donor ligands. This is intensely debated, particularly the extent to which An 3+ /Ln 3+ covalency differences prevail and manifest as the f-block is traversed, and the effects of periodic breaks beyond Pu. Herein, two Am complexes, [Am{N(E=PPh 2 ) 2 } 3 ] (1-Am, E=Se; 2-Am, E=O) are compared to isoradial [Nd{N(E=PPh 2 ) 2 } 3 ] (1-Nd, 2-Nd) complexes. Covalent contributions are assessed and compared to U/La and Pu/Ce analogues. In conclusion, through ab initio calculations grounded in UV-vis-NIR spectroscopy and single-crystal X-ray structures, we observe differences in f orbital involvement between Am–Se and Nd–Se bonds, which are not present in O-donor congeners.

Deep Learning‐Assisted Automated Single Cell Electroporation Platform for Effective Genetic Manipulation of Hard‐to‐Transfect Cells

Abstract Genome engineering of cells using CRISPR/Cas systems has opened new avenues for pharmacological screening and investigating the molecular mechanisms of disease. A critical step in many such studies is the intracellular delivery of the gene editing machinery and the subsequent manipulation of cells. However, these workflows often involve processes such as bulk electroporation for intracellular delivery and fluorescence activated cell sorting for cell isolation that can be harsh to sensitive cell types such as human‐induced pluripotent stem cells (hiPSCs). This often leads to poor viability and low overall efficacy, requiring the use of large starting samples. In this work, a fully automated version of the nanofountain probe electroporation (NFP‐E) system, a nanopipette‐based single‐cell electroporation method is presented that provides superior cell viability and efficiency compared to traditional methods. The automated system utilizes a deep convolutional network to identify cell locations and a cell‐nanopipette contact algorithm to position the nanopipette over each cell for the application of electroporation pulses. The automated NFP‐E is combined with microconfinement arrays for cell isolation to demonstrate a workflow that can be used for CRISPR/Cas9 gene editing and cell tracking with potential applications in screening studies and isogenic cell line generation.

Identification and mapping of quantitative trait loci for Fusarium head blight resistance in a synthetic hexaploid × hard red spring wheat population

Abstract Fusarium head blight (FHB), caused byFusarium graminearumSchwabe, is one of the most devastating diseases in wheat (Triticum aestivumL.). The synthetic hexaploid wheat line Largo was developed from a cross between the durum wheat [T. turgidumssp.durum(Desf.) Husn.] variety Langdon and theAegilops tauschiiCosson accession PI 268210, and it was previously found to have a moderate level of FHB resistance. This study was conducted to identify quantitative trait loci (QTL) associated with FHB resistance using a population of 188 recombinant inbred lines (RILs) from a cross between Largo and the susceptible wheat line ND495. The RILs were evaluated for Type II resistance in two greenhouse and two field environments. The disease severity and 90K single‐nucleotide polymorphism marker data were used for QTL analysis, which revealed six QTL on chromosomes 1D, 2D, 5B, and 7D. Four QTL (QFhb.rwg‐1D,QFhb.rwg‐5B,QFhb.rwg‐7D.1, andQFhb.rwg‐7D.3) from Largo had minor effects, whereas two QTL (QFhb.rwg‐2DandQFhb.rwg‐7D.2) from ND495 showed large effects on FHB resistance. The result suggested that ND495 may possess suppressor or susceptibility gene(s) suppressing or masking FHB resistance controlled by the resistance QTL. Among these QTL, four coincided with previously reported QTL, includingFhb9, and two (QFhb.rwg‐1DandQFhb.rwg‐7D.1) are likely novel QTL. From the six QTL regions, 10 Kompetitive allele‐specific PCR markers were developed and validated for marker‐assisted selection. The QTL detected from the resistant and susceptible parents enhance our understanding of FHB resistance expression and provide new resources for improving FHB resistance in wheat.

Rethinking the ill-posedness of the spectral function reconstruction — Why is it fundamentally hard and how Artificial Neural Networks can help

Reconstructing hadron spectral functions through Euclidean correlation functions are of the important missions in lattice QCD calculations. However, in a Källen–Lehmann (KL) spectral representation, the reconstruction is observed to be ill-posed in practice. It is usually ascribed to the fewer observation points compared to the number of points in the spectral function. In this paper, by solving the eigenvalue problem of continuous KL convolution, we show analytically that the ill-posedness of the inversion is fundamental and it exists even for continuous correlation functions. We discussed how to introduce regulators to alleviate the predicament, in which include the Artificial Neural Networks (ANNs) representations recently proposed by the Authors in another study. The uniqueness of solutions using ANNs representations is manifested analytically and validated numerically. Reconstructed spectral functions using different regularization schemes are also demonstrated, together with their eigen-mode decomposition. We observe that components with large eigenvalues can be reliably reconstructed by all methods, whereas those with low eigenvalues need to be constrained by regulators.

Mineral Recovery Enhanced Desalination (MRED) process: An innovative technology for desalinating hard brackish water

Desalination of water containing high concentrations of calcium, dissolved carbon dioxide, sulfate, silica and other sparingly soluble salts is difficult because of the scaling potential. An innovative pre-treatment scheme was investigated at bench scale that selectively removes these constituents and produces a soft water to enable desalination with high feed water recovery. The process first removes dissolved carbon dioxide by air stripping at low pH. Magnesium is removed by precipitation at high pH. Calcium is removed by ion exchange (IX), and sulfate is removed by nanofiltration (NF). Cation regenerant from IX, containing calcium, and concentrate from NF, containing sulfate, is combined to precipitate gypsum. Concentrate from the desalination process consisting of a concentrated NaCl solution is used to regenerate the IX resins. The selective precipitation, IX, and NF processes were tested in laboratory experiments and produced magnesium hydroxide and gypsum at greater than 90% and 95% purity respectively. A process model was developed to calculate process performance, mass and liquid flow rates. Finally, the MRED process offers the following benefits: 1) greater recovery of brackish feedwater by a desalination process, 2) recovery of marketable commodities and 3) reduction in the volume and mass of waste products from the treatment process.

Experimental Investigation of Drillstring Torsional Vibration Effect on Rate of Penetration with PDC Bits in Hard Rock

Polycrystalline diamond compact (PDC) bits have shown tremendous performance compared to roller cone bits and have gained attraction in deep geothermal drilling applications. While PDC bits can reach a higher rate of penetration compared to other bits, they are more prone to drilling vibrations. The scope of this paper is to quantify the effects of drillstring torsional vibrations on PDC bit performance using a laboratory testing facility. Here, the tests were performed with three drillstring configurations to evaluate the effect of drillstring vibrations. The results showed that rigid testing overestimates the bit rate of penetration by 35% due to torsional vibrations.

Structural evolution and magnetic hardness of (Sm,Zr)(Fe,Co,Ti) 12 alloy particles via reduction-diffusion

The quest for achieving high coercivity in Sm(Fe,Co,Ti) 12 alloys, despite their inherent strong magnetocrystalline anisotropy, has posed significant challenges. Recently, (Sm,Zr)(Fe,Co,Ti) 12 monocrystalline particles have exhibited coercivity μ 0 H c > 1.2 T, showcasing promising prospects and significant potential for both manufacturing and research endeavors. This study delves into the structural evolution of (Sm,Zr)(Fe,Co,Ti) 12 (1:12) alloy particles made via the calciothermic reduction-diffusion synthesis process as influenced by the molar ratios of Ca atoms to O 2- ions (Ca/O), annealing time and annealing temperature. Critical insight that informs conditions to optimize the magnetic response is gained via systematic experimentation and advanced electron microscopy. Complex structural features, including core-shell morphologies and intricate multiphase compositions within individual particles, are unveiled. An optimal Ca/O ratio of 1.30 produces particles with a coercivity up to μ 0 H c = 1.63 T, while higher Ca/O ratios induce the formation of a Sm-rich TbCu 7 -type (1: 7 ) phase, which only partially transforms into the desired 1:12 phase during annealing. Persistent remnants of the 1:7 phase locally impact atomic structure, particle morphology, and coercivity. Furthermore, these findings underscore the complex interplay between synthesis parameters, resulting structures, and magnetic properties, informing the design and optimization of high-performance permanent magnets comprised of the (1:12) compound.
