T-Cell Shenanigans: The Impact of MHC Pathway & Lipid Metabolism Genes on T-Cell Differentiation in the Thymus

Astronaut health and proper immune function are key to the success and sustainability of long term missions in space. While previous studies have observed adaptive immune dysfunction such as diminished WBC counts and cytokine production during spaceflight, the underlying mechanisms behind why immune function worsens are poorly understood. Using transcriptomic data analyzed from mouse thymus tissues available from the GLDS-421 experiment on the Rodent Research-9 mission, we used the GeneLab standardized RNA-Seq pipeline and identified four genes that were largely dysregulated (p<0.05): lipid metabolic genes Hpgd and Pgr and cell cycle regulator genes Cenpe and Kif11. We analyzed how these genes regulate the MHC-TCR interaction, and used this understanding to propose a novel mechanism for alterations to T-cell differentiation. In our experimental methodology, we propose to use 36 transgenic CD4-CD8 mice and 12 wild-type mice from the Jackson Lab to create a microgravity-simulating hindlimb unloading model and test our hypothesis. By silencing the aforementioned genes using custom siRNA based primers, we will use flow cytometry and RNA-Seq to quantify Treg counts in comparison to naive CD4+ cells, Alamar Blue assay for observing the metabolic activity in T-cells, and Enzyme-Linked Immunosorbent Assay (ELISA) to observe the general immune response in terms of T-cell differentiation. We hope this methodology will improve our knowledge of MHC-TCR interaction, contribute to a better understanding of the mechanisms of autoimmune diseases such as lupus, and improve the success rate of newer cancer treatments such as CAR-T cell therapy.

Anthropogenic Aerosols Delay the Emergence of GHGs-Forced Wetting of South Asian Rainy Seasons Under a Fossil-Fuel Intensive Pathway

With continued fossil-fuel dependence, anthropogenic aerosols over South Asia are projected to increase until the mid-21st century along with greenhouse gases (GHGs). Using the Community Earth System Model (CESM1) Large Ensemble, we quantify the influence of aerosols and GHGs on South Asian seasonal precipitation patterns over the 21st century under a very high-emissions (RCP 8.5) trajectory. We find that increasing local aerosol concentrations could continue to suppress precipitation over South Asia in the near-term, delaying the emergence of precipitation increases in response to GHGs by several decades in the monsoon season and a decade in the post-monsoon season. Emergence of this wetting signal is expected in both seasons by the mid-21st century. Our results demonstrate that the trajectory of local aerosols together with GHGs will shape near-future precipitation patterns over South Asia. Therefore, constraining precipitation response to different trajectories of both forcers is critical for informing near-term adaptation efforts.

Establishing a Pathway to Greener Aviation

The burning and production of jet fuel in both commercial and regional planes at a high volume releases extreme levels of carbon emission as of its current state. This paper analyzes the implementation and benefits of three sustainable alternatives to traditional jet fuel: sustainable aviation fuel (SAF), hydrogen fuel cells, and electric batteries. A roadmap to show progress in reducing the carbon footprint of aviation is then proposed. The first phase of the roadmap involves the evolution of technology, validation, and testing of SAF-powered planes and the preliminary development of hydrogen production and propulsion and battery technology and the development of electric propulsion technologies. The second phase illustrates the widespread implementation of all three fuel sources where electric aviation will be enforced in short-range travel (30 min-3 hrs), medium-range aircraft (3-6 hrs) will use hydrogen fuel cells, and long-range (6+ hrs) airliners will utilize SAF. The third and final phase will enact the use of SAF and electric batteries for long-range applications while hydrogen fuel cells will dominate short- and medium-range aircraft. This roadmap, in combination with proper legislative action and corporate cooperation paired with public awareness, can help eliminate the usage of jet fuel in the aviation industry and replace it with three more sustainable and clean fuel sources.

2023 Astrophotonics Roadmap: Pathways to Realizing Multi-Functional Integrated Astrophotonic Instruments

Photonic technologies offer numerous functionalities that can be used to realize astrophotonic instruments. The most spectacular example to date is the ESO Gravity instrument at the Very Large Telescope in Chile that combines the light-gathering power of four 8 m telescopes through a complex photonic interferometer. Fully integrated astrophotonic devices stand to offer critical advantages for instrument development, including extreme miniaturization when operating at the diffraction-limit, as well as integration, superior thermal and mechanical stabilization owing to the small footprint, and high replicability offering significant cost savings. Numerous astrophotonic technologies have been developed to address shortcomings of conventional instruments to date, including for example the development of photonic lanterns to convert from multimode inputs to single mode outputs, complex aperiodic fiber Bragg gratings to filter OH emission from the atmosphere, complex beam combiners to enable long baseline interferometry with for example, ESO Gravity, and laser frequency combs for high precision spectral calibration of spectrometers. Despite these successes, the facility implementation of photonic solutions in astronomical instrumentation is currently limited because of (1) low throughputs from coupling to fibers, coupling fibers to chips, propagation and bend losses, device losses, etc, (2) difficulties with scaling to large channel count devices needed for large bandwidths and high resolutions, and (3) efficient integration of photonics with detectors, to name a few. In this roadmap, we identify 24 key areas that need further development. We outline the challenges and advances needed across those areas covering design tools, simulation capabilities, fabrication processes, the need for entirely new components, integration and hybridization and the characterization of devices. To realize these advances the astrophotonics community will have to work cooperatively with industrial partners who have more advanced manufacturing capabilities. With the advances described herein, multi-functional integrated instruments will be realized leading to novel observing capabilities for both ground and space based platforms, enabling new scientific studies and discoveries.

Enhancing Metal Additive Manufacturing Training with the Advanced Vision Language Model: A Pathway to Immersive Augmented Reality Training for Non-Experts

This paper introduces an innovative training system for the Renishaw AM400 metal printer, leveraging the synergy of the advanced Vision Language Model (VLM) with Augmented Reality (AR) within the Digital Twins (DT) framework. Aimed at overcoming the limitations of conventional training methods in metal additive manufacturing (AM), our system integrates AR to provide an immersive learning environment, enhancing the real-world experience with interactive digital overlays. The core of the system lies in its use of VLM, which, pre-trained on diverse datasets, excels in processing multi-modal data, thereby offering nuanced and contextually relevant guidance for trainees. Key experiments demonstrate the system’s effectiveness, particularly highlighting the usage of VLM as an Artificial Intelligence (AI) agent to integrate external tools like YOLO-v7 for valve state classification and CRAFT for control panel text recognition. This approach significantly improves recognition accuracy, operational understanding, and human–machine interaction, especially for non-expert users, making complex metal AM operations more accessible. The research not only showcases the potential of AR and VLM in industrial training but also sets a new standard for smart manufacturing practices, indicating broader applications in various industrial domains.
