Evolving a New Electron Transfer Pathway for Nitrogen Fixation Uncovers an Electron Bifurcating-Like Enzyme Involved in Anaerobic Aromatic Compound Degradation

There is increasing evidence that protein electron carriers like Fd evolved to form specific partnerships with select electron donors and acceptors to keep native electron transfer pathways insulated from one another. This makes it challenging to integrate a Fd-dependent pathway such as biological nitrogen fixation into non-nitrogen-fixing organisms and provide the high-energy reducing power needed to fix nitrogen.

Isomer-specific spectroscopy of aromatics and their resonance-stabilized radical intermediates

There are many circumstances in which complex gas-phase reaction mixtures must be interrogated and understood, including combustion, plasmas, atmospheres, and molecular clouds. Spectroscopic tools are essential to selective detection and characterization of the widening array of precursors and the reactive intermediates they generate. This research program has focused on the development and implementation of sensitive spectroscopic detection schemes that are isomer-specific and even conformation-specific. In particular, we used an array of laser-based and broadband microwave methods to carry out isomer-specific and conformation-specific spectroscopy on large flexible molecules and reactive intermediates formed by photolysis, pyrolysis, combustion, or electron-driven chemistry.

Energetics and Kinetics of Syntrophic Aromatic Degradation (Final Technical Report)

This DOE Basic Energy Physical Biosciences project spanned a 28-year period and two project investigators. Numerous significant research discoveries have occurred over this time frame. Much of this work has been reported in peer-reviewed literature, with the publication of results from the last four years forthcoming. The overarching project themes have focused on probing the metabolism of syntrophic bacteria and their environments. Research supported by this project has been central to ten doctoral dissertations and one master’s thesis at the University of Oklahoma. The project has also supported numerous undergraduate research projects throughout the years. Additionally, several microbial genomes were sequenced and annotated in association with this work 12-16. Key findings and publications from this work are highlighted. More detailed results are provided for unpublished and embargoed work. A complete list of publications and research products associated with this project is at the report's end.

Process for enhanced production of desired hydrocarbons from biologically-derived compounds and bio-oils containing cyclic compounds by opening of aromatics and naphthenic ring-containing compounds

A system and process for processing biologically-derived compounds or a complex bio-oil by converting cyclic compounds in a complex bio-oil or biologically-derived compounds to desired materials such as high molecular weight paraffins with minimal carbon loss by using a ring-contraction catalyst to selectively produce C5 ring containing compounds; and then reacting the C5 ring containing compounds with a C5 ring opening catalyst in a second reactor to minimize carbon loss via cracking reactions.
