Foliar Application of Nettle and Japanese Knotweed Extracts on Vitis vinifera: Consequences for Plant Physiology, Biochemical Parameters, and Yield

Climate change is expected to affect grapevine physiology, berry quality, and yield in the Douro Demarcated Region (DDR). In this study, nettle (NE) and Japanese knotweed (JKE) extracts were tested to verify their biostimulant effect on the physiological and biochemical parameters of grapevine leaves and in vine yields. In fact, some parameters were improved after foliar application of the plant extracts, namely the photosynthetic activity and consequently, the levels of photosynthetic pigments (Clb), starch, and total soluble sugars. We also observed a reduction in lipid peroxidation, which could play a crucial role in protecting cell membranes from oxidative damage induced by the climatic conditions prevalent in this region. Therefore, we confirmed that the foliar application of plant extracts, along with the enhancement of secondary metabolites and the upregulation of plant defense genes, as previously reported, resulted in the enhancement of grapevine physiology, while also increasing the yield at harvest. In the future, these plant extracts could serve as a vital tool for winegrowers in mitigating the effects of expected changes in climatic conditions.

Field Testing of an Affordable Zero-Liquid-Discharge Arsenic-Removal Technology for a Small-Community Drinking Water System in Rural California

Arsenic contamination in groundwater threatens public health, particularly in small, low-income communities lacking affordable treatment solutions. This study investigated the field implementation of novel air cathode assisted iron electrocoagulation (ACAIE) technology for arsenic removal in Allensworth, California, where groundwater arsenic concentrations exceeded 250 µg/L. Over four months, a pilot-scale ACAIE system, operating at 600 L/h, consistently reduced arsenic levels to below the EPA’s maximum contaminant level of 10 µg/L. Laboratory experiments informed the optimization of charge dosage and flow rates, which were validated during field testing of the ACAIE 600 L/h system. The in-situ generation of hydrogen peroxide at the cathode speeded up the reaction kinetics, ensuring high arsenic removal efficiency while allowing high throughput, even with a compact reactor size. An economic analysis demonstrated a treatment cost of USD 0.02/L excluding labor, highlighting the system’s affordability compared to conventional methods. Adding labor costs increased the treatment cost to USD 0.09/L. The regeneration of air cathodes extended their operational life, addressing a key maintenance challenge, thus reducing the costs slightly. Intermittent challenges were encountered with filtration and secondary contaminant removal; these issues highlight opportunities for further operational improvements. Despite these challenges, ACAIE’s low operational complexity, scalability, and cost-effectiveness make it a promising solution for underserved small communities. These findings provide critical insights into deploying sustainable arsenic remediation technologies that are tailored to the needs of rural, low-resource communities.

Zymomonas mobilis oxidative stress transcriptomics

Zymomonas mobilis is an important bioenergy organism that has potential to produce biofuels, including ethanol, in high volumes. Here we examined the response of Zymomonas mobilis to various oxidative stresses using genome-scale transcriptomics data. We first examined the transcrpit abundance in WT aerobic growth compared to aerobic grown in paraquat, which forms superoxide. Under anaerobic growth conditions we compared WT Zymomonas mobilis with strains grown in media lacking iron as well as strains lacking iron that were treated with the iron chelator DIP before collection. Finally we examined transcript abundance in cells lacking ZMO_0422 (Rrf2 family transcription factor homolog) and ZMO_1411 (Fur homolog) grown under anaerobic conditions. Overall design: Transcriptomic analysis of WT, a deletion of ZMO_0422, and a deletion of ZMO_1411 in Zymomonas mobilis ZM4 under aerobic and anaerobic growth conditions along with various oxidative stresses: Paraquate addition, No Iron, and hydrogen peroxide addition.

Microorganisms detected by enzyme-catalyzed reaction

Enzymes detect the presence of microorganisms in soils. The enzyme lysozymi is used to release the enzyme catalase from the microorganisms in a soil sample. The catalase catalyzes the decomposition of added hydrogen peroxide to produce oxygen which is detected manometrically. The partial pressure of the oxygen serves as an index of the samples bacteria content.

Improved chlorate candle provides concentrated oxygen source

Improved chlorate candle is used as a solid, portable source of oxygen in emergency situations. It contains sodium chlorate, iron, barium peroxide, and glass mixed in powdered form. The oxygen evolves from the decomposition of the sodium chlorate when an ignition pellet is electrically initiated.
