5' modification of duplex DNA with a ruthenium electron donor-acceptor pair using solid-phase DNA synthesis

Incorporation of metalated nucleosides into DNA through covalent modification is crucial to measurement of thermal electron-transfer rates and the dependence of these rates with structure, distance, and position. Here, we report the first synthesis of an electron donor-acceptor pair of 5' metallonucleosides and their subsequent incorporation into oligonucleotides using solid-phase DNA synthesis techniques. Large-scale syntheses of metal-containing oligonucleotides are achieved using 5' modified phosporamidites containing [Ru(acac)(2)(IMPy)](2+) (acac is acetylacetonato; IMPy is 2'-iminomethylpyridyl-2'-deoxyuridine) (3) and [Ru(bpy)(2)(IMPy)](2+) (bpy is 2,2'-bipyridine; IMPy is 2'-iminomethylpyridyl-2'-deoxyuridine) (4). Duplexes formed with the metal-containing oligonucleotides exhibit thermal stability comparable to the corresponding unmetalated duplexes (T(m) of modified duplex = 49 degrees C vs T(m) of unmodified duplex = 47 degrees C). Electrochemical (3, E(1/2) = -0.04 V vs NHE; 4, E(1/2) = 1.12 V vs NHE), absorption (3, lambda(max) = 568, 369 nm; 4, lambda(max) = 480 nm), and emission (4, lambda(max) = 720 nm, tau = 55 ns, Phi = 1.2 x 10(-)(4)) data for the ruthenium-modified nucleosides and oligonucleotides indicate that incorporation into an oligonucleotide does not perturb the electronic properties of the ruthenium complex or the DNA significantly. In addition, the absence of any change in the emission properties upon metalated duplex formation suggests that the [Ru(bpy)(2)(IMPy)](2+)[Ru(acac)(2)(IMPy)](2+) pair will provide a valuable probe for DNA-mediated electron-transfer studies.

Imaging photopolarimeter on Pioneer Saturn

Results of Pioneer 11 imaging photopolarimeter observations of Saturn, its rings, and Titan are presented. The imaging photopolarimeter is a pointable telescope with an aperture of 2.5 cm and passbands of 390 to 500 to 720 nm which uses the spin of the spacecraft to scan across an object. Images of the Saturn system and of the rings are presented, and the absence of a D ring, structures in the C, B and A rings and the Cassini division and the discoveries of the F ring and the provisionally named Pioneer division separating it from the A ring are reported. A mean particle size less than 15 meters is estimated from estimates of total ring mass and the optical depth of the B ring. The discovery of the satellite 1979 S 1 at 2.53 Saturn radii is also noted. Models of the vertical aerosol structure of Saturn's atmosphere are compared with the polarization data, and it is indicated that the density of cloud particles decreases with altitude with a scale height about one fourth that of the gas, and that an optical depth of one is to be found at 750 mbar.

Development and Utility of a Four-Channel Scanner for Wildland Fire Research and Applications

The Airborne Infrared Disaster Assessment System (AIRDAS) is a four-channel scanner designed and built at NASA-Ames for the specific task of supporting research and applications on fire impacts on terrestrial and atmospheric processes and also of serving as a vital instrument in the assessment of natural and man-induced disasters. The system has been flown on numerous airframes including the Navajo, King-Air, C0130, and Lear Jet 310 and a 206. The system includes a configuration composed of a 386 PC computer workstation, a non-linear detector amplifier, a sixteen-bit digitizer, dichroic filters, and Exabyte 8500 5Gb Tape output, VHS tape output, a Rockwell GPS and a 2-axis gyro. The AIRDAS system collects digital data in four wavelength regions, which can be filtered: band 1 (0.61-0.68 microns), band 2 (1.57-1.7 microns), band 3 (3.6-5.5 microns), and band 4 (5.5-13.0 microns), an FOV of 108 degrees, an IFOV of 2.62 mrads, and a digitized swath width of 720 pixels. The inclusion of the non-linear detector amplifier allows for the accurate measurement of emitted temperature from fires and hot spots. Lab testing of the scanner has indicated temperature assessments of 800 C without detector saturation. This has advantages over previous systems which were designed for thermal measurement of earth background temperatures, and were ill-equipped for accurate determination of high intensity conditions. The scanner has been flown successfully on data collection missions since 1992 in the western US as well as Brazil. These and other research and applications responses will be presented along with an assessment of future directions with the system.a

6.3-micron water-vapor-band derivatives

Measurement of the absorptance in the 6.3-micron band of a 1-m column of water vapor as a function of concentration c, pressure p, and temperature T. From the measured absorptance, the first derivatives with respect to c, p, and T, as well as the second derivatives with respect to cc, pp, cp, and pT have been obtained. These experimental results have been compared with values calculated from the line parameters of the 720 strongest rotational lines in the band.

Detection of the isotopes of heavy cosmic ray nuclei

A counter telescope designed to detect and resolve the isotopic composition of cosmic ray nuclei heavier than neon is being prepared. The telescope consists of a rather conventional charge measuring array using two scintillator elements and two solid Cerenkov radiators of differing refractive index. The mass measurement is obtained by combining the velocity information from one or both of the Cerenkov radiators operating near their threshold with residual range measured in a block of nuclear emulsion. Path length corrections and particle location in the emulsions is provided by a spark chamber fired in coincidence with potentially suitable particles. The telescope has a geometry factor of 530 sq cm sr roughly. It should be able to resolve the isotopes of iron over the energy range of 300 to 720 Mev/n and those of neon over 300 to 400 MeV/n. The expected response and characteristics of the telescope are described in detail and the sensitivity to rare isotopes discussed.

Al-rich Chondrules: Petrologic Basis for Their Diversity, and Relation to Type C CAIs

Al-rich chondrules share mineralogical and chemical properties with, and are intermediate in a volatility sense between, CAIs and ferromagnesian chondrules. In some way they must be petrogenetic links between the two. A recent upsurge of interest in Al-rich chondrules is due to their constituent plagioclase feldspar and Al-rich glass being amenable to successful ion microprobe searches for radiogenic Mg-26, the decay product of Al-26 (t(sub 1/2) = 720,000 y). This has allowed estimates to be made of the time duration between CAI formation and the onset of Al-rich (and possibly, by extension, ferromagnesian) chondrule formation, on the order of 1.5-2.5 million years.

Age alters the cardiovascular response to direct passive heating

During direct passive heating in young men, a dramatic increase in skin blood flow is achieved by a rise in cardiac output (Qc) and redistribution of flow from the splanchnic and renal vascular beds. To examine the effect of age on these responses, seven young (Y; 23 +/- 1 yr) and seven older (O; 70 +/- 3 yr) men were passively heated with water-perfused suits to their individual limit of thermal tolerance. Measurements included heart rate (HR), Qc (by acetylene rebreathing), central venous pressure (via peripherally inserted central catheter), blood pressures (by brachial auscultation), skin blood flow (from increases in forearm blood flow by venous occlusion plethysmography), splanchnic blood flow (by indocyanine green clearance), renal blood flow (by p-aminohippurate clearance), and esophageal and mean skin temperatures. Qc was significantly lower in the older than in the young men (11.1 +/- 0.7 and 7.4 +/- 0.2 l/min in Y and O, respectively, at the limit of thermal tolerance; P < 0. 05), despite similar increases in esophageal and mean skin temperatures and time to reach the limit of thermal tolerance. A lower stroke volume (99 +/- 7 and 68 +/- 4 ml/beat in Y and O, respectively, P < 0.05), most likely due to an attenuated increase in inotropic function during heating, was the primary factor for the lower Qc observed in the older men. Increases in HR were similar in the young and older men; however, when expressed as a percentage of maximal HR, the older men relied on a greater proportion of their chronotropic reserve to obtain the same HR response (62 +/- 3 and 75 +/- 4% maximal HR in Y and O, respectively, P < 0.05). Furthermore, the older men redistributed less blood flow from the combined splanchnic and renal circulations at the limit of thermal tolerance (960 +/- 80 and 720 +/- 100 ml/min in Y and O, respectively, P < 0. 05). As a result of these combined attenuated responses, the older men had a significantly lower increase in total blood flow directed to the skin.

Effects of High-Temperature Exposures on the Fatigue Life of Disk Superalloys Examined

Tests used to characterize the low-cycle-fatigue resistance of disk superalloys are usually performed at cyclic frequencies of 0.33 Hz or faster. However, service conditions for disks in some aerospace and land-based gas turbine engines can produce major cycle periods extending from minutes to hours and days. Over a service life, this can produce total service times near the maximum temperature that exceed 100 hr for aerospace applications and 100,000 hr for land-based applications. Such time-dependent effects of realistic mission cycles on fatigue resistance can be significant in superalloy disks, and need to be considered for accurate disk life prediction. The purpose of this study at the NASA Glenn Research Center was to examine the effects of extended exposures and extended cycle periods on the fatigue resistance of two disk superalloys. Current alloy Udimet 720 (Special Metals Corporation, Huntington, WV) disk material was provided by Solar Turbines/Caterpillar Co., and advanced alloy ME3 was provided by the NASA Ultra-Efficient Engine Technologies (UEET) Project, in powder-metallurgy-processed, supersolvus heat-treated form. Fatigue specimens were fully machined and exposed in air at temperatures of 650 to 704 C for extended times. Then, they were tested using conventional fatigue tests with a total strain range of 0.70 percent and a minimum-to-maximum strain ratio of zero to determine the effects of prior exposure on fatigue resistance. Subsequent tests with extended dwells at minimum strain in each fatigue cycle were performed to determine cyclic exposure effects.

Chemical Diversity along the Traverse of the Rover Spirit at Gusev Crater

The Alpha-Particle-X-ray Spectrometer (APXS) is part of the in situ payload of the Mars Exploration Rovers. It has determined the chemical composition of soils and rocks along the nearly 6 km long traverse of the rover Spirit. The measuring method a combination of PIXE and XRF using Cm244 sources - allowed the unambiguous identification of elemental compositions with high precision. Besides sample triage and quantification of saltforming elements as indicators for aqueous alteration, the APXS also delivered important constraints to mineralogy intruments (i.e., Mossbauer (MB), MiniTES, Pancam) on minerals and rock types. The mineralogy instruments on the other hand provided constraints on minerals used for APXS normative calculations and, e.g. allowed the attribution of S to sulfate, instead of sulfide or elemental sulfur. This abstract gives an updated overview of the data obtained up to our current rover position on sol 720 at the eastern base of the Columbia Hills. We will emphasize elemental correlations that imply the presence of certain minerals that can not be identified by the MER mineralogy instruments.

X-ray Reflected Spectra from Accretion Disk Models. III. A Complete Grid of Ionized Reflection Calculations

We present a new and complete library of synthetic spectra for modeling the component of emission that is reflected from an illuminated accretion disk. The spectra were computed using an updated version of our code xillver that incorporates new routines and a richer atomic data base. We offer in the form of a table model an extensive grid of reflection models that cover a wide range of parameters. Each individual model is characterized by the photon index Gamma of the illuminating radiation, the ionization parameter zeta at the surface of the disk (i.e., the ratio of the X-ray flux to the gas density), and the iron abundance A(sub Fe) relative to the solar value. The ranges of the parameters covered are: 1.2 <= Gamma <= 3.4, 1 <= zeta <= 104, and 0.5 <= A(sub Fe) <= 10. These ranges capture the physical conditions typically inferred from observations of active galactic nuclei, and also stellar-mass black holes in the hard state. This library is intended for use when the thermal disk flux is faint compared to the incident power-law flux. The models are expected to provide an accurate description of the Fe K emission line, which is the crucial spectral feature used to measure black hole spin. A total of 720 reflection spectra are provided in a single FITS file suitable for the analysis of X-ray observations via the atable model in xspec. Detailed comparisons with previous reflection models illustrate the improvements incorporated in this version of xillver.

Investigation of solidification in zero-gravity environment; M553 sphere forming experiment. Phase C: Evaluation of Skylab specimens

Results on specimen evaluation and discussion of solidification behavior in each case are reported in the following order: (1) specimen SL-1.6, (2) specimen SL-2.8, (3) specimen SL-2.4, (4) specimen SL-1.10 and (5) specimen SL-1.11. Comparison is made with ground-processed specimens of similar composition, whenever pertinent and meaningful. Among the nondestructive evaluation methods the measurement of sphericity was conducted by micrometric and shadowgraphic techniques. The intricate shape of specimens in some cases appeared difficult to define. In measuring the density, liquid penetration inside cavities that outcrop on the surface was avoided by sealing off these cavities. Among the destructive evaluation methods the use of the Quantimet 720 required particular attention, because of the small difference in contrast between second phases and micropores. With regard to microporosity microvoids in the core of some specimens were so fine that X-ray microradiography had to be used.

Circular-scan streak tube with solid-state readout

A circular-scan streak tube designed for use in a laser ranging system is described. Electrostatic focusing is used between the photocathode and a microchannel plate, and electrons from the microchannel plate are proximity focused onto an output phosphor screen. Electron beam deflection is achieved by driving two orthogonal sets of deflection plate assemblies in phase quadrature at a frequency of 200 MHz. The light intensity in the output beam trace is measured by using a circular Reticon array of 720 photodiodes, which is fiber-optically coupled to the output phosphor screen of the tube. Sample measurements of frequency-doubled Nd:YAG laser-pulse waveforms are given. Also the output beamwidth has been measured at the 200-MHz deflection frequency. The results suggest a limiting resolution of 33 psec for the circular-scan streak tube used for these tests.

Broadly tunable high-power operation of an all-solid-state titanium-doped sapphire laser system

Broadly tunable and high-power operation of a Ti-doped sapphire laser is obtained with a diode-laser-pumped frequency-doubled Nd:YAG laser as the pump source. A maximum broadband (FWHM = 25 nm) output pulse energy of 720 microJ at 795 nm in a TEM00 mode is obtained for 1850 microJ of energy of 532-nm pump light. A minimum pulse duration of 7 nsec is obtained from a 40-mm-long cavity. With the use of an intracavity prism, the Ti:sapphire laser is tunable continuously over the 696-1000-nm spectral range (with three different mirror sets).

How Do Various Maize Crop Models Vary in Their Responses to Climate Change Factors?

Potential consequences of climate change on crop production can be studied using mechanistic crop simulation models. While a broad variety of maize simulation models exist, it is not known whether different models diverge on grain yield responses to changes in climatic factors, or whether they agree in their general trends related to phenology, growth, and yield. With the goal of analyzing the sensitivity of simulated yields to changes in temperature and atmospheric carbon dioxide concentrations [CO2], we present the largest maize crop model intercomparison to date, including 23 different models. These models were evaluated for four locations representing a wide range of maize production conditions in the world: Lusignan (France), Ames (USA), Rio Verde (Brazil) and Morogoro (Tanzania). While individual models differed considerably in absolute yield simulation at the four sites, an ensemble of a minimum number of models was able to simulate absolute yields accurately at the four sites even with low data for calibration, thus suggesting that using an ensemble of models has merit. Temperature increase had strong negative influence on modeled yield response of roughly -0.5 Mg ha(sup 1) per degC. Doubling [CO2] from 360 to 720 lmol mol 1 increased grain yield by 7.5% on average across models and the sites. That would therefore make temperature the main factor altering maize yields at the end of this century. Furthermore, there was a large uncertainty in the yield response to [CO2] among models. Model responses to temperature and [CO2] did not differ whether models were simulated with low calibration information or, simulated with high level of calibration information.

Sampling system for wheat (Triticum aestivum L) area estimation using digital LANDSAT MSS data and aerial photographs

A procedure to estimate wheat (Triticum aestivum L) area using sampling technique based on aerial photographs and digital LANDSAT MSS data is developed. Aerial photographs covering 720 square km are visually analyzed. To estimate wheat area, a regression approach is applied using different sample sizes and various sampling units. As the size of sampling unit decreased, the percentage of sampled area required to obtain similar estimation performance also decreased. The lowest percentage of the area sampled for wheat estimation with relatively high precision and accuracy through regression estimation is 13.90% using 10 square km as the sampling unit. Wheat area estimation using only aerial photographs is less precise and accurate than those obtained by regression estimation.

Nuclear gamma rays from 720-MeV alpha-induced reactions on Al-27 and Si-28

Prompt gamma rays from the interaction of 720-MeV alpha particles with Al-27 and Si-28 were detected and analyzed to identify residual nuclei and to determine cross sections for production of specific levels. No gamma-ray transitions were detected from nuclei heavier than the target. From Doppler broadening, the momentum of the residual nuclei was estimated. The results are compared with previous results for 140- and 1600-MeV alphas on Al-27 and approximately 200-MeV positive or negative pions on Al-27 and Si-28 and fitted to a spallation-yield formula.

A new wideband, fully steerable, decametric array at Clark Lake

A new, fully steerable, decametric array for radio astronomy is under construction at the Clark Lake Radio Observatory near Borrego Springs, California. This array will be a T of 720 conical spiral antennas (teepee-shaped antennas, hence the array is called the TPT), 3.0 by 1.8 km capable of operating between 15 and 125 MHz. Both its operating frequency and beam position will be adjustable in less than one msec, and the TPT will provide a 49-element picture around the central beam position for extended-source observations. Considerable experience has been gained in the operation of completed portions of the array, and successful operation of the final array is assured. This paper describes the results of the tests which have been conducted with the conical spirals and outlines the planned electronics.
