Automated Particle Analysis of Hanford Tank Wastes 241-AN-106, 241-AN-101, and 241-AW-105

This effort is involved in developing particle size and density distribution (PSDD) for the major phases within Hanford tank waste sludge. The object of this work has been to use automated particle analysis (APA) methods on the Scanning Electron Microscope (SEM) to provide PSDDs. The PSDD can be used to quantify the proportion of gibbsite particles likely to settle quickly and enable blending of wastes that may be prone to pipeline plugging such as the uranium phase, clarkeite with other wastes. The Hanford tank waste solids were analyzed with SEM combined with x-ray Energy Dispersive Spectroscopy (EDS) using APA. This method can allow thousands of individual particles to be characterized and so may provide a more representative view of the samples and information PSDDs. We characterized several as-received sludge samples and developed PSDD with APA. Sample preparation methods were found to impact the collected results, as it was more difficult to collect representative samples of the larger particles. Much of the material was also dried and cemented together that further impacted results.

Joint Airport Weather Studies (AWS) project

The joint airport weather studies (JAWS) project is discussed. The major objectives of the JAWS Project are a fundamental description of the phenomenon, a determination of the hazard potential and a definition of a protection and warning system, all of which are relative to low level wind shear. Aspects of the low level wind shear phenomenon. The principal focus, however, is the microburst. The microburst is fundamentally a rather simple atmospheric flow. It is a downdraft that, upon approaching the surface, spreads out horizontally, producing a diverging radial flow in all directions. For any direction that an aircraft flies through the microburst, it will first encounter increasing head winds; then the remnants of the downdraft; and then, increasing tail wind.

Automatic Weather Station (AWS) Lidar

A ground based, autonomous, low power atmospheric lidar instrument is being developed at NASA Goddard Space Flight Center. We report on the design and anticipated performance of the proposed instrument and show data from two prototype lidar instruments previously deployed to Antarctica.

The Awful Truth About Zero-Gravity: Space Acceleration Measurement System; Orbital Acceleration Research Experiment

Earth's gravity holds the Shuttle in orbit, as it does satellites and the Moon. The apparent weightlessness experienced by astronauts and experiments on the Shuttle is a balancing act, the result of free-fall, or continuously falling around Earth. An easy way to visualize what is happening is with a thought experiment that Sir Isaac Newton did in 1686. Newton envisioned a mountain extending above Earth's atmosphere so that friction with the air would be eliminated. He imagined a cannon atop the mountain and aimed parallel to the ground. Firing the cannon propels the cannonball forward. At the same time, Earth's gravity pulls the cannonball down to the surface and eventual impact. Newton visualized using enough powder to just balance gravity so the cannonball would circle the Earth. Like the cannonball, objects orbiting Earth are in continuous free-fall, and it appears that gravity has been eliminated. Yet, that appearance is deceiving. Activities aboard the Shuttle generate a range of accelerations that have effects similar to those of gravity. The crew works and exercises. The main data relay antenna quivers 17 times per second to prevent 'stiction,' where parts stick then release with a jerk. Cooling pumps, air fans, and other systems add vibration. And traces of Earth's atmosphere, even 200 miles up, drag on the Shuttle. While imperceptible to us, these vibrations can have a profound impact on the commercial research and scientific experiments aboard the Shuttle. Measuring these forces is necessary so that researchers and scientists can see what may have affected their experiments when analyzing data. On STS-107 this service is provided by the Space Acceleration Measurement System for Free Flyers (SAMS-FF) and the Orbital Acceleration Research Experiment (OARE). Precision data from these two instruments will help scientists analyze data from their experiments and eliminate outside influences from the phenomena they are studying during the mission.

An expanded role for the transcription factor WRINKLED1 in the biosynthesis of triacylglycerols during seed development

The transcription factor WRINKLED1 ( WRI1 ) is known as a master regulator of fatty acid synthesis in developing oilseeds of Arabidopsis thaliana and other species. WRI1 is known to directly stimulate the expression of many fatty acid biosynthetic enzymes and a few targets in the lower part of the glycolytic pathway. However, it remains unclear to what extent and how the conversion of sugars into fatty acid biosynthetic precursors is controlled by WRI 1. To shortlist possible gene targets for future in-planta experimental validation, here we present a strategy that combines phylogenetic foot printing of cis-regulatory elements with additional layers of evidence. Upstream regions of protein-encoding genes in A. thaliana were searched for the previously described DNA-binding consensus for WRI1, the ASML1/WRI1 (AW)-box. For about 900 genes, AW-box sites were found to be conserved across orthologous upstream regions in 11 related species of the crucifer family. For 145 select potential target genes identified this way, affinity of upstream AW-box sequences to WRI1 was assayed by Microscale Thermophoresis. This allowed definition of a refined WRI1 DNA-binding consensus. We find that known WRI1 gene targets are predictable with good confidence when upstream AW-sites are phylogenetically conserved, specifically binding WRI1 in the in vitro assay, positioned in proximity to the transcriptional start site, and if the gene is co-expressed with WRI1 during seed development. When targets predicted in this way are mapped to central metabolism, a conserved regulatory blueprint emerges that infers concerted control of contiguous pathway sections in glycolysis and fatty acid biosynthesis by WRI1. Several of the newly predicted targets are in the upper glycolysis pathway and the pentose phosphate pathway. Of these, plastidic isoforms of fructokinase ( FRK 3) and of phosphoglucose isomerase ( PGI 1) are particularly corroborated by previously reported seed phenotypes of respective null mutations.
