Lawrence Livermore National Laboratory Experimental Test Site, Site 300: Soil Study Institutional Review Board #1081 Soil Sampling and Analysis Plan

This Soil Sampling and Analysis Plan (SAP) was prepared by the Environmental Functional Area/Technical Services Department (TSD) of the Environment, Safety & Health (ES&H) Directorate for the Project Management Office (PMO) to collect soil quality data at an area located at Lawrence Livermore National Laboratory’s (LLNL’s) Experimental Test Site, Site 300 (Site 300) (project) (Figures 1 and 2). The purpose of the project is to determine the chemical and radiological quality of soil in the project area to support feasibility and design of future improvements. This SAP identifies chemicals of concern and describes the procedures for collection and analysis of environmental samples, and evaluation of analytical data (chemical and radiological) to determine the reuse potential of shallow soil at the project area. This SAP follows the criteria established in LLNL’s Soils Screening and Management Plan (SSMP) (LLNL 2022), which is consistent with U.S. Environmental Protection Agency (EPA) guidance for developing Data Quality Objectives for environmental data (EPA 2006) and the Multi-Agency Radiation Survey and Site Investigation Manual (MARSSIM) guidance (U.S. NRC, U.S. EPA, U.S. DOE, U.S. DOD 2000). The scope of this SAP is based on preliminary design information provided by PMO.

Impacts of divalent cations (Mg 2+ and Ca 2+ ) on PFAS bioaccumulation in freshwater macroinvertebrates representing different foraging modes

Per- and polyfluoroalkyl substances (PFAS) have extensively contaminated freshwater aquatic ecosystems where they can be transported in water and partition to sediment and biota. In this paper, three freshwater benthic macroinvertebrates with different foraging modes were exposed to environmentally relevant concentrations of eight perfluoroalkyl carboxylates (PFCA), three perfluoroalkyl sulfonates (PFSA), and three fluorotelomer sulfonates (FTS) at varying divalent cation concentrations of magnesium (Mg 2+ ) and calcium (Ca 2+ ). Divalent cations can impact PFAS partitioning to solids, especially to sediments, at higher concentrations. Sediment dwelling worms (Lumbriculus variegatus), epibenthic grazing snails (Physella acuta), and sediment-dwelling filter-feeding bivalves (Elliptio complanata) were selected due to their unique foraging modes. Microcosms were composed of synthetic sediment, culture water, macroinvertebrates, and PFAS and consisted of a 28-day exposure period. L. variegatus had significantly higher PFAS bioaccumulation than P. acuta and E. complanata, likely due to higher levels of interactions with and ingestion of the contaminated sediment. “High Mg 2+ ” (7.5 mM Mg 2+ ) and “High Ca 2+ ” (7.5 mM Ca 2+ ) conditions generally had statistically higher bioaccumulation factors (BAF) than the “Reference Condition” (0.2 mM Ca 2+ and 0.2 mM Mg 2+ ) for PFAS with perfluorinated chain lengths greater than eight carbons. Long-chain PFAS dominated the PFAS profiles of the macroinvertebrates for all groups of compounds studied (PFCA, PFSA, and FTS). Furthermore, these results indicate that the study organism has the greatest impact on bioaccumulation, although divalent cation concentration had observable impacts between organisms depending on the environmental conditions. Elevated cation concentrations in the microcosms led to significantly greater bioaccumulation in the test organisms compared to the experimental reference conditions for long-chain PFAS.

Application of printed strain gauges in prototypical nuclear reactor conditions

This report discusses the current development and testing of additively manufactured temperature sensors and resistive/ capacitive strain gauges. This has potential to improve the sensor design and manufacturing techniques to meet the requirements of in-pile monitoring of mechanical properties and structural health of materials and specimens in current and advanced nuclear test reactors (i.e., in terms of environment conditions, sample geometry, and materials compatibility). The developmental additively manufactured strain gauges are exposed to separate effects testing (i.e., mechanical strain (up to 1000 µe), high temperature (up to 700 °C)) to determine environmental factors that affect the performance of the strain gauge. The robustness and integrity of a printed strain gauge layer exposed to a molten salt environment for up to 500 hours was evaluated, with the goal of assessing material compatibility of printed strain sensors for instrumented surveillance test articles in molten salt reactors. In addition, sensor qualification methodologies are further developed for determining the reliability and robustness at the interface of the additively manufactured strain gauge materials.

Multi‐Omics Analyses Reveal Divergent Molecular Mechanisms Underlying Plant Biomass Conversion by Five Fungi

Fungal plant biomass conversion (FPBC) is of great importance to the global carbon cycle and has been increasingly applied for the production of biofuel and biochemicals from lignocellulose. However, the comprehensive understanding of relevant molecular mechanisms in different fungi remains challenging. Here, we comparatively analyzed the transcriptome, proteome and metabolome profile of four ascomycetes and one basidiomycete fungi during their growth on two common agricultural feedstocks (soybean hulls and corn stover). We revealed strong time‐, substrate‐ and species‐specific responses at multi‐omics levels for the tested fungi, highlighting species‐specific carbon utilization approaches and evolutionary adaptation to environmental niches. Notably, a remarkable expressional diversity of lignocellulose degrading enzymes, sugar transporter and metabolic genes, as well as industrially relevant metabolites were identified across different fungi and cultivation conditions. The findings improves our understanding of complex molecular networks underlying FPBC and fungal ecological roles, offering novel insights that can guide future genetic engineering of fungi for valorization of agriculture waste into value‐added bioproducts.

Engineering uranyl-chelating peptides from NikR for electrochemical peptide-based sensing applications

We have, for the first time, designed three uranyl-chelating peptides by modeling after the uranyl binding pocket found in a mutated Ni(II)-dependent transcriptional repressor (NikR). All three thiolated and methylene blue (MB)-modified peptides, NikR-5, NikR-11, and NikR-15, contain the five core amino acids responsible for target recognition, but the two longer peptides have either one or two additional glycine residues in between the five amino acids. These three peptide probes were then used in the fabrication of electrochemical peptide-based (E-PB) uranyl ion (U(VI)) sensors, with the goal of elucidating the effects of the added glycine residues and probe flexibility on target recognition. The sensing mechanism is similar to other “signal-off” E-PB sensors, in which binding of the target rigidifies the probe, resulting in a decrease in the redox signal from the tethered MB label. Although all three sensors responded to U(VI), the NikR-15 sensor's behavior was irreproducible and thus precluded from the rest of the study. The NikR-11 sensor showed the largest response to U(VI), whereas the NikR-5 sensor showed higher specificity for U(VI). The limit of detection was 50 nM for both sensors, which is well below the U.S. Environmental Protection Agency maximum contaminant level for uranium. Both sensors were further tested and proven functional in a 50% synthetic aquifer sample. Overall, the NikR-11 sensor, fabricated with the 11-amino acid probe, is deemed the optimal design. Incorporating glycine residues is a strategic way to lengthen the peptide slightly to improve target binding. This simple yet versatile approach to recreating binding pockets on electrode surfaces can potentially be employed in the design of other E-PB and surface-based metal ion sensors.

Application and performance of the novel Elcomat 5000 autocollimator in slope-measuring profilometry of beamline optics

Autocollimators are used in profilometers for the precise form measurement of beam-shaping optics for synchrotron light sources and x-ray free-electron lasers. This application requires using an aperture stop, typically 2.5 mm in diameter, which limits the footprint of the autocollimator’s measuring beam on the optics to achieve a sufficiently high lateral resolution. Central to the development of profilometry has been the availability of commercial autocollimators, such as the Elcomat 3000 from Möller–Wedel Optical. Although autocollimators are generally not designed for use with small apertures, this device has proven capable and has become the de facto standard. Now that it has been replaced by the Elcomat 5000, we evaluate the performance of several autocollimators of this type at small apertures, including the characterization of instrument transfer functions with a chirped height profile. We demonstrate that the Elcomat 5000 is capable of repeatable angle measurements with a standard deviation of 0.014 arcsec at an aperture diameter of 1.6 mm, whereas the Elcomat 3000 achieves 0.03 arcsec at 2.5 mm, comparing models with modified reticle designs. Here, we also investigate the influence of optical aberrations of the autocollimator’s objective and their changes with the path length to the surface under test. We characterize the sensitivity of the angle measurement to changes in environmental parameters, in particular, barometric pressure. These efforts are aimed at approaching fundamental limits in the characterization of the shape of optical surfaces with autocollimator-based profilometers so that these are ready for characterizing the next generation of synchrotron and XFEL beamline optics.

Bioenergy sorghum maintains photosynthetic capacity in elevated ozone concentrations

Abstract Elevated tropospheric ozone concentration (O 3 ) significantly reduces photosynthesis and productivity in several C 4 crops including maize, switchgrass and sugarcane. However, it is unknown how O 3 affects plant growth, development and productivity in sorghum ( Sorghum bicolor L.), an emerging C 4 bioenergy crop. Here, we investigated the effects of elevated O 3 on photosynthesis, biomass and nutrient composition of a number of sorghum genotypes over two seasons in the field using free‐air concentration enrichment (FACE), and in growth chambers. We also tested if elevated O 3 altered the relationship between stomatal conductance and environmental conditions using two common stomatal conductance models. Sorghum genotypes showed significant variability in plant functional traits, including photosynthetic capacity, leaf N content and specific leaf area, but responded similarly to O 3 . At the FACE experiment, elevated O 3 did not alter net CO 2 assimilation ( A ), stomatal conductance ( g s ), stomatal sensitivity to the environment, chlorophyll fluorescence and plant biomass, but led to reductions in the maximum carboxylation capacity of phosphoenolpyruvate and increased stomatal limitation to A in both years. These findings suggest that bioenergy sorghum is tolerant to O 3 and could be used to enhance biomass productivity in O 3 polluted regions.

Evaluation of short-term initial performance of a “Molekule Air” portable air cleaner

The aim of this project was to evaluate and characterize the performance of a MOLEKULE AIR air cleaner unit. The testing protocol comprised the evaluation of an unused Molekule Air unit operating at maximum (“boost” setting) fan speed setting. The device was tested over initial short-term periods of 70-80 h of continuous operation. The tested air cleaner was challenged in separate experiments with a well-characterized mixture of volatile organic compounds (VOCs), and with ozone. All tests were performed at realistic pollutant levels in a 20-m 3 environmental stainless-steel chamber. Variables controlled and measured in this study included the chamber temperature, relative humidity, the composition and concentration of the challenge VOCs, and the concentrations of ozone. Potential formation of byproducts was also investigated to assess the overall performance.

The SMART Cables Initiative for Ocean and Geophysical Observing [Slides]

SMART Cables are becoming a reality, and have the potential to revolutionize submarine oceanographic and geophysical sensing. Pilot / demonstration projects are underway to test the technology and explore the feasibility of incorporating both in-repeater standardized sensor packets but also ancillary sensor types on external, branching nodes. The projects currently in planning will set the standard and influence the norms for the future of SMART systems. The interest of all potential stakeholders and users, and input as to needs and concerns, is welcomed as testing proceeds.

Corrective Action Decision Document/Corrective Action Plan for Corrective Action Units 101 and 102: Central and Western Pahute Mesa Nevada National Security Site, Nevada (Rev. 1)

This corrective action decision document (CADD)/corrective action plan (CAP) has been prepared for the Central and Western Pahute Mesa (PM) Corrective Action Units (CAUs) 101 and 102. Corrective action is needed because underground radioactive contamination exists in these areas resulting from historic underground nuclear testing. The PM CAUs are located in the northwestern portion of the Nevada National Security Site and comprise 82 corrective action sites. A total of 85 underground nuclear detonations took place within the area covered by the PM CAUs between 1965 and 1992 and resulted in the release of radionuclides (RNs) in the subsurface in the vicinity of the test cavities.

Acceptance criteria for in situ surveillance of MSR materials based on thermally-loaded mechanical test articles

This report describes practices and acceptance test procedures for designing, running, and maintaining a material surveillance program in a future operating molten salt reactor. The programs described here rely on test data from passively actuated mechanical test articles inserted into critical regions of the reactor and periodically removed for out-of-reactor testing. The report defines definite acceptance procedures, based on the results of these tests, to determine whether a component can continue to operate accounting for the accumulation of environmentally-assisted mechanical damage in the component materials to date, and extrapolated out through the next inspection period. Additionally, the report describes work on a software tool implementing many of the surveillance methods and procedures described here and progress on simplified methods for inferring damage accumulation in the test articles, based on out-of-reactor thermal cycling, that do not rely on sophisticated numerical analysis.

A physics-based ensemble machine-learning approach to identifying a relationship between lightning indices and binary lightning hazard

To convert lightning indices generated by numerical weather prediction experiments into binary lightning hazard, a machine-learning tool was developed. This tool, consisting of parallel multilayer perceptron classifiers, was trained on an ensemble of planetary boundary layer schemes and microphysics parameterizations that generated four different lightning indices over 1 week. In a subsequent week, the multi-physics ensemble was applied and the machine-learning tool was used to evaluate the accuracy. Unintuitively, the machine-learning tool performed better on the testing dataset than the training dataset. Much of the error may be attributed to mischaracterizing the convection. The combination of the machine learning model and simulations could not differentiate between cloud-to-cloud lightning and cloud-to-ground lightning, despite being trained on cloud-to-ground lightning. It was found that the simulation most representative of the local operational model was the most accurate simulation tested.

Rainbows and Climate Change: A tutorial on climate model diagnostics and parameterization

Earth System Models (ESMs) must represent processes below the grid scale of a model using representations (parameterizations) of physical and chemical processes. As a tutorial exercise to understand diagnostics and parameterization, this work presents a representation of rainbows for an ESM: the Community Earth System Model version 2 (CESM2). Using the 'state' of the model, basic physical laws, and some assumptions, we generate a representation of this unique optical phenomena as a diagnostic output. Rainbow occurrence and it's possible changes are related to cloud occurrence and rain formation which are critical uncertainties for climate change prediction. The work highlights issues which are typical of many diagnostics parameterizations such as assumptions, uncertain parameters and the difficulty of evaluation against uncertain observations. Results agree qualitatively with limited available global 'observations' of Rainbows. Rainbows are seen in expected locations in the sub-tropics over the ocean where broken clouds and frequent precipitation occurs. The diurnal peak is in the morning over ocean and in the evening over land. The representation of rainbows is found to be quantitatively sensitive to the assumed amount of cloudiness and the amount of stratiform rain. Rainbows are projected to have decreased, mostly in the Northern Hemisphere, due to aerosol pollution effects increasing cloud coverage since 1850. In the future, continued climate change is projected to decrease cloud cover, associated with a positive cloud feedback. As a result the rainbow diagnostic projects that rainbows will increase in the future, with the largest changes at mid-latitudes. The diagnostic may be useful for assessing cloud parameterizations, and is an exercise in how to build and test parameterizations of atmospheric phenomena.

Environmental Degradation in Advanced Reactor Environments

The Advanced Materials and Manufacturing Technologies (AMMT) program operating under the Department of Energy Office of Nuclear Energy is accelerating the qualification and deployment of additively manufactured materials in advanced reactor environments. Laser powder bed fusion (LPBF) has emerged as a promising additive manufacturing technique to fabricate complex components with the potential of tailored material properties. In the nuclear industry, metal additive manufacturing can offer numerous advantages, such as reduced lead times, streamlined quality assurance, and cost-effective low-volume production of new and replacement components with conventional or novel materials and geometries. The rapid and effective qualification of the effect of processing variability on the performance and degradation of additively manufactured materials is essential for the deployment of these components into advanced reactor environments. This talk presents an overview of the Environmental Effects area activities within AMMT. AMMT is performing irradiation and corrosion testing of several materials built by LPBF, including 316L and 316H stainless steels. Our irradiation efforts encompass neutron irradiation testing and ion irradiation testing to provide a robust technical basis for understanding the effect of process variability on materials degradation. We develop focused irradiation test plans that align well with risk-informed and technologically inclusive approaches to licensing. We will also discuss our plans and specific concerns for corrosion testing additively manufactured material, with a primary focus on molten salt and liquid sodium environments.

Recent Advances in Assessing Environmental Effects of Marine Renewable Energy Around the World

Marine renewable energy (MRE) is increasingly of interest to coastal nations as a source of renewable energy that can support climate change mitigation goals as well as provide secure locally-produced energy for coastal and island communities. MRE extracts power from tidal streams, waves, ocean currents, run of rivers, and gradients in the ocean, with specialized devices developed and tested for each energy resource. Alongside development of MRE technologies and systems, first in Europe and then in North America, Australia, Asia, and other regions, it has been universally recognized that there is also a need to examine potential effects on marine animals, habitats, ecosystem processes, local communities and other sea users, to ensure that the MRE industry can be developed in a responsible and sustainable manner. This paper looks at the status of assessment and monitoring for potential environmental effects associated with MRE projects around the world. Over 80 projects were identified worldwide as having been tested, demonstrated, or commercially deployed with associated environmental monitoring. Five of the projects that represent tidal stream, wave, and run of river projects are examined in more detail to determine the types of data and information collected for those projects, the outputs of the monitoring campaigns, and the actions taken as a result of the data collection and analysis. Recommendations are provided for standardization of the monitoring approaches, instruments, and analysis methods at MRE project sites worldwide.

Uncertainty Analysis and Software Verification

Uncertainty analyses are an important part of calibrations and testing. They allow researchers insight on how to reduce and mitigate errors in testing. Each device used in testing introduces error in a system, as well as other sources such as environmental conditions, electronics, analog to digital, and random errors. Each source is carefully examined to identify how much error it introduces to a system. These sources are then combined using various methods of uncertainty calculations. To verify and validate software, a manual calculation is required to ensure the software is performing as intended. Using Excel to verify the calculations, we can identify discrepancies within the software. The root of the sum of the squares uncertainty (RSS) is used to find the combined uncertainty of a device at one and two standard deviations of the mean. Calibrations on accelerometers are performed using a vibration system along with a back-toback reference accelerometer. The vibration system takes a reference point at 100Hz frequency at 10g amplitude. The sensitivities are collected at each dialed in frequency. The sensitivity of the device represents the electrical output of the UUT (mV, pC, etc.) per unit of acceleration (g). The full history of sensitivities of selected accelerometers are used to find the averaged, standard deviation, and uncertainty of the device at each frequency tested. The uncertainty calculations from excel and the software are then compared.

A streamlined approach to characterize microalgae strains for biomass productivity under dynamic climate simulation conditions

Screening microalgae strains under static light and temperature flask conditions cannot directly quantify the biomass productivities of algae growing in the dynamically fluctuating light and temperature conditions of outdoor ponds. In this effort, we describe a testing pipeline that screens for productivity under climate simulated conditions. A validated, miniaturized Laboratory Environmental Algae Pond Simulator (mini-LEAPS) photobioreactor was used to determine optimal medium salinities and biomass productivities of cold-tolerant microalgae collected from Arctic, subarctic, Antarctic, and subalpine habitats. Strains characterized in this effort include: Chlorella antarctica UTEX1959, Chlorella sp. UTEXSNO69, Chloromonas rosae UTEXSNO11, Phaeodactylum tricornutum UTEX646, and Stichococcus minutus CCALA727. Observed productivities of the characterized strains ranged from 1.74 ± 0.25 to 8.18 ± 0.81 g m –2 day –1 (as ash-free dry weight). For each strain, a temperature tolerance profile was generated to identify the most appropriate season (s) for cultivation. The two most promising strains, Chlorella sp. UTEX SNO69 and P. tricornutum UTEX646, were tested alongside the DISCOVR winter benchmark strain Monoraphidium minutum 26B-AM in outdoor open ponds at the PNNL Algae Testbed (PAT) in Arizona. Further, under cold-season outdoor pond conditions, Chlorella sp. UTEXSNO69 achieved average areal biomass productivities of 8.21 ± 0.50 g m –2 day –1 , not significantly different from that of the benchmark strain (8.52 ± 0.43 g m –2 day –1 , p > 0.05). Under spring outdoor pond conditions, P. tricornutum UTEX646 achieved average areal biomass productivities 10.34 ± 0.27 g m –2 day –1 , not significantly different from that of the benchmark (10.07 ± 0.50 g m –2 day –1 ) under the tested conditions (p > 0.05). The streamlined pipeline was thus demonstrated to successfully characterize productive microalgae strains for outdoor deployment by weeding out algae strains that were not productive under the tested dynamic light and temperature conditions.

The Coupled Influence of Thermal Physiology and Biotic Interactions on the Distribution and Density of Ant Species along an Elevational Gradient

A fundamental tenet of biogeography is that abiotic and biotic factors interact to shape the distributions of species and the organization of communities, with interactions being more important in benign environments, and environmental filtering more important in stressful environments. This pattern is often inferred using large databases or phylogenetic signal, but physiological mechanisms underlying such patterns are rarely examined. We focused on 18 ant species at 29 sites along an extensive elevational gradient, coupling experimental data on critical thermal limits, null model analyses, and observational data of density and abundance to elucidate factors governing species’ elevational range limits. Thermal tolerance data showed that environmental conditions were likely to be more important in colder, more stressful environments, where physiology was the most important constraint on the distribution and density of ant species. Conversely, the evidence for species interactions was strongest in warmer, more benign conditions, as indicated by our observational data and null model analyses. Our results provide a strong test that biotic interactions drive the distributions and density of species in warm climates, but that environmental filtering predominates at colder, high-elevation sites. Such a pattern suggests that the responses of species to climate change are likely to be context-dependent and more specifically, geographically-dependent.
