ETOP 503689: Pre-optimized cell free lysates for rapid prototyping of genes and pathways

This ETOP project aimed to re-conceive how we engineer complex biological systems by linking pathway design, prospecting, and validation into an integrated framework. Specifically, our vision seeks to advance and interweave high-throughput cell-based systems and rapid cell-free technologies in a way suitable for automation and microdroplet manipulation in a DOE JGI user facility setting. The key technology to be investigated toward this vision is a cell-free platform for combinatorial assembly of pathways by mixing-and-matching crude cell lysates derived from a suite of flux-enhanced background strains, each enriched with pathway enzymes. Through this work, we have established rewired E. coli and S. cerevisiae cells suitable to generated rewired lysates. We have demonstrated that lysates derived from rewired cells can generate higher fluxes of products and can enable a more robust discovery method going from gene to function annotations. Finally, we demonstrate that these rewired lysates can be stored for up to a year and still retain function. Collectively, these technologies enable a more rapid screening approach for enzyme and pathway variants.

Prototyping and Manufacturing of Magnetic Gearbox Components using Innovations in Castings

In a previous collaboration with Emrgy (CRADA agreement NFE-17-06532), a housing component of Emrgy’s 10 kW gearbox was casted out of aluminum alloys using impression molds prepared using 3D printed techniques. (Henderson, 2018). After comparison with Emrgy’s existing hardware and material testing, the 3D printed impression molds were verified to represent a valid approach to advance technology readiness and commercialize Emrgy’s technology.

Prototype X-ray and Gamma Detection with Cyclotron Radiation Emission Spectroscopy

Cyclotron radiation emission spectroscopy, or CRES, is a novel approach to measuring the energy of an electron. By trapping a free electron in a high magnetic field, it undergoes cyclotron motion and emits radiofrequency (RF) waves. The frequency of this RF radiation is directly related to the energy of the electron. Because many cycles of the RF emission are recorded, the energy resolution of the CRES system is on the order of a single electron volt. To make a CRES system sensitive to photons, a target gas is used to induce a photoelectric effect, producing the electron that is subsequently trapped. By adding the binding energy of the target atom, the energy of the incident photon may be reconstructed. Using a xenon target gas, photoelectric interactions dominate up to approximately 300 keV, covering not only all atomic shell X-rays of the elements, but many low-lying nuclear states as well, including key transuranic elements related to nuclear security. CRES holds the potential of maintaining single-eV resolution up to this 300-keV range, thereby surpassing current state-of-the-art detectors by a factor of 10-100. The instrumental resolution of the system is limited by the uniformity of the applied magnetic field.

Bench testing of an early prototype pitch resonator WEC

This report describes a series of tests performed on a "pitch resonator'" concept for a wave energy converter. The overall testing campaign goals centered on risk reduction for the pitch resonator wave energy converter concept and model validation. Two modes of testing are captured in this report: one using a single degree of freedom test rig and one in which a six degree of freedom Stewart platform was employed.

Beam Optics Design of A Prototype 20 kW Conduction-Cooled SRF Accelerator for Medical Sterilization

Superconducting technology has significantly advanced the capabilities of particle accelerators, facilitating higher beam-power operations for fundamental research at a comparatively lower cost. However, the conventional implementation of superconducting technology introduces complexities in the form of cryogenic plants, cryogenic distribution systems and substantial construction and operational cost. In response to these challenges, recent research efforts at Fermilab have been dedicated to the development of a cryogen-free, conduction-cooled Nb3Sn-based superconducting technology. This paper outlines the beam optics design of a 20-kW conduction-cooled compact superconducting accelerator for medical sterilization. The paper reviews both the physics and practical constraints associated with high beam-power operation within the context of industrial applications. The focus is on providing insights into the potential of this innovative technology to overcome existing challenges and pave the way for more accessible and efficient industrial particle accelerators.

Specifications and a Prototype Software to Demonstrate a Data Catalog for Hanford Datasets

Environmental management activities at the Hanford Site produce extensive data about site conditions, contaminants, and cleanup activities. Managing, archiving, and accessing that data requires a high degree of collaboration among site contractors and a high level of awareness by project managers and staff. The Hanford Site has a range of databases (e.g., Hanford Environmental Information System [HEIS]) and their associated user interfaces (e.g., Environmental Dashboard Application [EDA], Virtual Library [VL]), as well as other document management systems (e.g., Integrated Document Management System [IDMS]). However, Hanford lacks a single unified resource to find data (which itself comes in multiple formats) amongst the multiple disparate systems, not to mention ad hoc data not contained in an official repository/database.

Design and Prototyping of Advanced Control Systems for Advanced Reactors Operating in the Future Electric Grid (Final Report)

Despite its significant advantages as a baseload, low-carbon energy source, the U.S. nuclear power industry has faced increasing difficulties in maintaining economic competitiveness in a rapidly evolving energy market. The economic conditions faced by the current fleet of nuclear power plants (NPPs) in the U.S. deregulated electricity market require a concerted effort to mitigate specific cost factors. Many units are struggling to stay competitive, and some premature shutdowns have occurred. Besides, in response to the large penetration of renewable energy sources, the role of nuclear power plants as pure baseload units needs to be reconsidered. Based on these experiences, operational flexibility is considered a fundamental requirement for the next generation of nuclear reactors to be competitive in the future energy market. The deployment of advanced reactors capable of operating within a new power grid paradigm, known as the Integrated Energy System (IES) was investigated. This approach combines new reactor designs with Thermal Energy Storage (TES) technologies, allowing the nuclear reactor to maintain a steady power output without the need for constant adjustments in response to load demand fluctuations. With this configuration, the reactor operates as a baseload unit, experiencing only very gradual power transients, while the energy storage facility within the power conversion cycle acts as a peaking unit.

Scalable Data Center Capacity for DOE's AI Prototype: A Rapidly Available Gigawatt Data Center for DOE

The multilaboratory Gigawatt Data Center working group was commissioned to identify approaches to rapidly establish federal data centers with scalable capacities up to 1,000 MW. These state-of-the-art facilities will serve as hubs for interdisciplinary collaboration, industry partnerships, and transformative applications of artificial intelligence. The proposed strategic shift includes facilitating multilaboratory collaboration, prioritizing operational efficiency, expanding public–private partnerships, optimizing investments, ensuring long-term contractual flexibility, supporting open science and secure data enclaves, and exploiting high-speed national networks. Owing to their extensive experience and best practices, the US Department of Energy national laboratories are uniquely positioned to lead this initiative. We recommend conducting a feasibility analysis to rapidly identify the optimal sites for this initiative, and the effort will likely involve private industry for design, construction, financing, and operational integration. We also propose establishing multiple geographically diverse sites to ensure energy resilience, high operational reliability, and a diverse user base, thereby effectively addressing the nation’s critical needs.
