A global dataset of terrestrial biological nitrogen fixation

Biological nitrogen fixation (BNF) is the main natural source of new nitrogen inputs in terrestrial ecosystems, supporting terrestrial productivity, carbon uptake, and other Earth system processes. We assembled a comprehensive global dataset of field measurements of BNF in all major N-fixing niches across natural terrestrial biomes derived from the analysis of 376 BNF studies. The dataset comprises 32 variables, including site location, biome type, N-fixing niche, sampling year, quantification method, BNF rate (kg N ha −1 y −1 ), the percentage of nitrogen derived from the atmosphere (%N dfa ), N fixer or N-fixing substrate abundance, BNF rate per unit of N fixer abundance, and species identity. Overall, the dataset combines 1,207 BNF rates for trees, shrubs, herbs, soil, leaf litter, woody litter, dead wood, mosses, lichens, and biocrusts, 152 herb %N dfa values, 1,005 measurements of N fixer or N-fixing substrate abundance, and 762 BNF rates per unit of N fixer abundance for a total of 424 species across 66 countries. This dataset facilitates synthesis, meta-analysis, upscaling, and model benchmarking of BNF fluxes at multiple spatial scales.

Phylogenetic and ecological drivers of the avian lung mycobiome and its potentially pathogenic component

Vertebrate lungs contain diverse microbial communities, but little is known about the drivers of community composition or consequences for health. Microbiome assembly by processes such as dispersal, coevolution, and host-switching can be probed with comparative surveys; however, few studies exist for lung microbiomes, particularly for the fungal component, the mycobiome. Distinguishing among fungal taxa that are generalist or specialist symbionts, potential pathogens, or incidentally inhaled spores is urgent because of potential for emerging diseases. Here, we characterize the avian lung mycobiome and test the relative influences of environment, phylogeny, and functional traits. We used metabarcoding and culturing from 195 lung samples representing 32 bird species across 20 families. We identified 526 fungal taxa as estimated by distinct sequence types (zOTUs) including many opportunistic pathogens. These were predominantly from the phylum Ascomycota (79%) followed by Basidiomycota (16%) and Mucoromycota (5%). Yeast and yeast-like taxa (Malassezia, Filobasidium, Saccharomyces, Meyerozyma, and Aureobasidium) and filamentous fungi (Cladosporium, Alternaria, Neurospora, Fusarium, and Aspergillus) were abundant. Lung mycobiomes were strongly shaped by environmental exposure, and further modulated by host identity, traits, and phylogenetic affinities. Our results implicate migratory bird species as potential vectors for long-distance dispersal of opportunistically pathogenic fungi.

The impact of spectroscopic incompleteness in direct calibration of redshift distributions for weak lensing surveys

ABSTRACT Obtaining accurate distributions of galaxy redshifts is a critical aspect of weak lensing cosmology experiments. One of the methods used to estimate and validate redshift distributions is to apply weights to a spectroscopic sample, so that their weighted photometry distribution matches the target sample. In this work, we estimate the selection bias in redshift that is introduced in this procedure. We do so by simulating the process of assembling a spectroscopic sample (including observer-assigned confidence flags) and highlight the impacts of spectroscopic target selection and redshift failures. We use the first year (Y1) weak lensing analysis in Dark Energy Survey (DES) as an example data set but the implications generalize to all similar weak lensing surveys. We find that using colour cuts that are not available to the weak lensing galaxies can introduce biases of up to Δz ∼ 0.04 in the weighted mean redshift of different redshift intervals (Δz ∼ 0.015 in the case most relevant to DES). To assess the impact of incompleteness in spectroscopic samples, we select only objects with high observer-defined confidence flags and compare the weighted mean redshift with the true mean. We find that the mean redshift of the DES Y1 weak lensing sample is typically biased at the Δz = 0.005−0.05 level after the weighting is applied. The bias we uncover can have either sign, depending on the samples and redshift interval considered. For the highest redshift bin, the bias is larger than the uncertainties in the other DES Y1 redshift calibration methods, justifying the decision of not using this method for the redshift estimations. We discuss several methods to mitigate this bias.

Mapping and Synthesis of International Biomass Supply Assessments

This report, Mapping and Synthesis of International Biomass Supply Assessments (or Global Biomass Resource Assessment) is the first step in a long-term process to assemble data from around the globe into a virtual repository that can be updated and provide user-friendly access to the data. The Clean Energy Ministerial (CEM) Biofuture Platform Initiative recommended that research be completed to “address the need for internationally accepted benchmarks quantifying sustainable biomass feedstock supplies.” To act upon the CEM Biofuture recommendation, in 2024, the U.S. Department of Energy (DOE) commissioned Oak Ridge National Laboratory (ORNL) to prepare this report as the primary deliverable for a one-year assignment to assemble data into a citable form that could help resolve the persistent question presented related to bioenergy policy, “Is there enough sustainable biomass?” In response to that query, this report includes information received by August 2024 from national CEM representatives, collaborators, and public sources on current and future sustainable biomass supplies in 62 nations, and subsequently documents (a) the approach used by ORNL to analyze and categorize the information received in a manner that enables aggregation and comparability; and (b) recommendations for next steps and guidelines to help others update and harmonize future assessments of global sustainable biomass supplies.

A stable 15-member bacterial SynCom promotes Brachypodium growth under drought stress

Introduction: Rhizosphere microbiomes are known to drive soil nutrient cycling and influence plant fitness during adverse environmental conditions. Field-derived robust Synthetic Communities (SynComs) of microbes mimicking the diversity of rhizosphere microbiomes can greatly advance a deeper understanding of such processes. However, assembling stable, genetically tractable, reproducible, and scalable SynComs remains challenging. Methods: Here, we present a systematic approach using a combination of network analysis and cultivation-guided methods to construct a 15-member SynCom from the rhizobiome of Brachypodium distachyon. This SynCom incorporates diverse strains from five bacterial phyla. Genomic analysis of the individual strains was performed to reveal encoded plant growth-promoting traits, including genes for the synthesis of osmoprotectants (trehalose and betaine) and Na+/K+ transporters, and some predicted traits were validated by laboratory phenotypic assays. Results: The SynCom demonstrates strong stability both in vitro and in planta. Most strains encoded multiple plant growth-promoting functions, and several of these were confirmed experimentally. The presence of osmoprotectant and ion transporter genes likely contributed to the observed resilience of Brachypodium to drought stress, where plants amended with the SynCom recovered better than those without. We further observed preferential colonization of SynCom strains around root tips under stress, likely due to active interactions between plant root metabolites and bacteria. Discussion: Our results demonstrate that trait-informed construction of synthetic communities can yield stable, functionally diverse consortia that enhance plant resilience under drought. Preferential colonization near root tips points to active, localized plant-microbe signaling as a component of stress-responsive recruitment. This stable SynCom provides a scalable platform for probing mechanisms of plant-microbe interaction and for developing microbiome-based strategies to improve soil and crop performance in variable environments.

Image animation for theme enhancement and change detection

Animated displays are useful in enhancing subtle temporally related changes in scenes viewed by satellites capable of providing repetitive coverage. The detectability of fixed features is also improved through the help of the powerful visual integration process. To expedite the process of assembling and displaying well-registered, time-lapse sequences and to provide means for making quantitative measurements of radiances, displacements, and areas, an electronic satellite image analysis console was constructed. During the LANDSAT-1 program, this equipment was applied to the needs of a number of earth resource investigators with interests principally related to dynamic hydrology. The measurement of the areal extent of snow cover within defined drainage basins is discussed as a representative applications example.

Advanced thermionic energy conversion

Basic analytical and experimental exploration was conducted on several types of advanced thermionic energy converters, and preliminary analysis was performed on systems utilizing advanced converter performance. The Pt--Nb cylindrical diode which exhibited a suppressed arc drop, as described in the preceding report, was reassembled and the existence of the postulated hydrid mode of operation was tentatively confirmed. Initial data obtained on ignited and unignited triode operation in the demountable cesium vapor system essentially confirmed the design principles developed in earlier work, with a few exceptions. Three specific advanced converter concepts were selected as candidates for concentrated basic study and for practical evaluation in fixed-configuration converters. Test vehicles and test stands for these converters and a unique controlled-atmosphere station for converter assembly and processing were designed, and procurement was initiated.

Extreme ultraviolet composite spectra of representative solar features

The spectral identifications and absolute intensities of lines in the EUV spectrum for various solar features, such as the quiet sun, active regions, coronal holes, and the off-limb corona, are presented and discussed. Composite spectra have been assembled and processed from the ATM ultraviolet spectrometer data with a spectral resolution of 1.6 A over the range 280-1350 A. These spectra represent a self-consistent set of observations with good photometric calibration, in which intercomparison of intensities from different regions is used to assist in line identifications and to draw general conclusions on average physical conditions in the solar atmosphere. The quiet-sun line intensities obtained near solar minimum are compared with OSO 6 results near solar maximum to provide an estimate for the changes over the solar cycle.

Development and fabrication of improved power transistor switches

A new class of high-voltage power transistors was achieved by adapting present interdigitated thyristor processing techniques to the fabrication of npn Si transistors. Present devices are 2.3 cm in diameter and have V sub CEO (sus) in the range of 400 to 600V. V sub CEO (sus) = 450V devices were made with an (h sub FE)(I sub C) product of 900A at V sub CE = 2.5V. The electrical performance obtained was consistent with the predictions of an optimum design theory specifically developed for power switching transistors. The device design, wafer processing, and assembly techniques are described. Experimental measurements of the dc characteristics, forward SOA, and switching times are included. A new method of characterizing the switching performance of power transistors is proposed.

The role of Space Station Freedom in the Human Exploration Initiative

Exploration accommodation requirements for Space Station Freedom (SSF) and mission-supporting capabilities have been studied. For supporting the Human Exploration Initiative (HEI), SSF will accommodate two functions with augmentations to the baseline Assembly Complete configuration. First, it will be an earth-orbiting transportation node providing facilities and resources (crew, power, communications) for space vehicle assembly, testing, processing and postflight servicing. Second, it will be an in-space laboratory for science research and technology development. The evolutionary design of SSF will allow the on-orbit addition of pressurized laboratory and habitation modules, power generation equipment, truss structure, and unpressurized vehicle processing platforms.
