Three-dimensional structure of Schistosoma japonicum glutathione S-transferase fused with a six-amino acid conserved neutralizing epitope of gp41 from HIV

The 3-dimensional crystal structure of glutathione S-transferase (GST) of Schistosoma japonicum (Sj) fused with a conserved neutralizing epitope on gp41 (glycoprotein, 41 kDa) of human immunodeficiency virus type 1 (HIV-1) was determined at 2.5 A resolution. The structure of the 3-3 isozyme rat GST of the mu gene class was used as a molecular replacement model. The structure consists of a 4-stranded beta-sheet and 3 alpha-helices in domain 1 and 5 alpha-helices in domain 2. The space group of the Sj GST crystal is P4(sub 3)2(sub 1)2 with unit cell dimensions of a = b = 94.7 A, and c = 58.1 A. The crystal has 1 GST monomer per asymmetric unit, and 2 monomers that form an active dimer are related by crystallographic 2-fold symmetry. In the binding site, the ordered structure of reduced glutathione is observed. The gp41 peptide (Glu-Leu-Asp-Lys-Trp-Ala) fused to the C-terminus of Sj GST forms a loop stabilized by symmetry-related GSTs. The Sj GST structure is compared with previously determined GST structures of mammalian gene classes mu, alpha, and pi. Conserved amino acid residues among the 4 GSTs that are important for hydrophobic and hydrophilic interactions for dimer association and glutathione binding are discussed.

Amino Acids in the Asteroidal Water-Bearing Salt Crystals Hosted in the Zag Meteorite

Solid evidence of liquid water in primitive meteorites is given by the ordinary chondrites H5 Monahans (1998) and H3-6 Zag. Aqueous fluid inclusion-bearing halite (NaCl) crystals were shown to be common in Zag. These striking blue/purple crystals (Figure 1), which gained the coloration from electron-trapping in the Cl-vacancies through exposure to ionizing radiation, were determined to be over 4.0-4.7 billion years old by I-Xe dating. The halite grains are present as discrete grains within an H-chondrite matrix with no evidence for aqueous alteration that indicates a xenogenic source, possibly ancient cryovolcanism. They were proposed to be formed from the cryovolcanic plumes on icy C-type asteroids (possibly Ceres), and were transferred and incorporated into the H chondrite parent asteroid following the eruption event(s). A unique aspect of these halites is that they contain abundant solid inclusions hosted within the halites alongside the water inclusions. The solid inclusions were suggested to be entrained within the fluid erupted from the cryovolcanic event(s), and were shown to be comprised of abundant organics. Spectrofluorometric study and Raman imaging of the halites have identified macromolecular carbon and aliphatic carbon compounds. In order to investigate the type of organics present in Zag and in particular within the fluid-bearing halites, we studied for the first time the amino acid contents of a selected mineral (halite) phase in a meteorite sample.

Insulin effect on amino acid uptake by unloaded rat hindlimb muscles

The effect of insulin on the uptake of alpha-amino-isobutyric acid (AIB) by unloaded rat hindlimb muscles was investigated using soleus and extensor digitorum longus (EDL) muscles from intact and adrenalectomized (ADX) rats that were tail-casted for six days. It was found that, at insulin levels above 0.00001 units/ml, the in vitro rate of AIB uptake by muscles from intact animals was stimulated more in the weight bearing muscles than in unloaded ones. In ADX animals, this differential response to insulin was abolished.

Glutamate and aspartate are decreased in the skin in amyotrophic lateral sclerosis

We measured the levels of amino acids in biopsied skin from eight patients with amyotrophic lateral sclerosis (ALS) and seven controls. The most conspicuous changes in ALS patients were as follows. First, the contents of the acidic amino acids glutamate and aspartate were significantly decreased in ALS, and were negatively and significantly associated with the duration of illness. Second, the levels of the collagen-associated amino acids hydroxyproline, proline, glycine, alanine, and hydroxylysine were significantly decreased in ALS, and correlated inversely with the duration of illness. These results suggest that there are abnormalities of acidic amino acids and collagen-associated amino acids in the skin of patients with ALS. These changes may underlie the pathogenesis of ALS.

The radiolysis and radioracemization of amino acids on clays

The effects of the surfaces of kaolinite and bentonite clays on the radiolysis and radioracemization of L-leucine and its hydrochloride salt have been investigated experimentally. L-leucine and its hydrochloride salt were deposited on the clays and the amino acid/clay preparations were irradiated by a Co-60 gamma-ray source which induced 2-89 percent radiolysis. The efficiency of radiolysis and radioracemization were measured using gas chromatography. Results were obtained for leucine in 0.1 M aqueous solution for comparison with the clay-deposted leucine and leucine hydrochloride. It is found that radiolysis and radioracemization in the samples occurred according to a pseudo-first-order rate law. Comparison of the specific rate constants showed that leucine and its hydrochloride salt were the most resistant to both radiolysis and radioracemization, followed by leucine and its hydrochloride salt on kaolin. Leucine and its HCl salt on bentonite, and leucine in aqueous solution were found to be the least resistant to radiolysis and radioracemization. The experimental results are intepreted with respect to the Vester-Ulbricht mechanism for the origin of optical activity.
