Residuals-based distributionally robust optimization with covariate information

We consider data-driven approaches that integrate a machine learning prediction model within distributionally robust optimization (DRO) given limited joint observations of uncertain parameters and covariates. Our framework is flexible in the sense that it can accommodate a variety of regression setups and DRO ambiguity sets. We investigate asymptotic and finite sample properties of solutions obtained using Wasserstein, sample robust optimization, and phi-divergence-based ambiguity sets within our DRO formulations, and explore cross-validation approaches for sizing these ambiguity sets. Through numerical experiments, we validate our theoretical results, study the effectiveness of our approaches for sizing ambiguity sets, and illustrate the benefits of our DRO formulations in the limited data regime even when the prediction model is misspecified.

Reducing nitrous oxide emissions and optimizing nitrogen-use efficiency in dryland crop rotations with different nitrogen rates

Recent interests in improving agricultural production while minimizing environmental footprints emphasized the need for research on management strategies that reduce nitrous oxide (N 2 O) emissions and increase nitrogen-use efficiency (NUE) of cropping systems. This study aimed to evaluate N 2 O emissions, annualized crop grain yield, emission factor, and yield-scaled- and NUE-scaled N 2 O emissions under continuous spring wheat (Triticum aestivum L.) (CW) and spring wheat-pea (Pisum sativum L.) (WP) rotations with four N fertilization rates (0, 50, 100, and 150 kg N ha(-1)). The N 2 O fluxes peaked immediately after N fertilization, intense precipitation, and snowmelt, which accounted for 75-85% of the total annual flux. Cumulative N 2 O flux usually increased with increased N fertilization rate in all crop rotations and years. Annualized crop yield and NUE were greater in WP than CW for 0 kg N ha(-1) in all years, but the trend reversed with 100 kg N ha(-1) in 2013 and 2015. Crop yield maximized at 100 kg N ha(-1), but NUE declined linearly with increased N fertilization rate in all crop rotations and years. As N fertilization rate increased, N fertilizer-scaled N2O flux decreased, but NUE-scaled N 2 O flux increased non-linearly in all years, regardless of crop rotations. The yield-scaled N 2 O flux decreased from 0 to 50 kg N ha -1 and then increased with increased N fertilization rate. Because of non-significant difference of N 2 O fluxes between 50 and 100 kg N ha(-1), but increased crop yield, N 2 O emissions can be minimized while dryland crop yields and NUE can be optimized with 100 kg N ha -1 , regardless of crop rotations.

Capacitated p -hub approach for park-and-ride facility location problem under nested logit demand function: polyhedral approaches

By generalizing the unconstrained p-hub approach for the park-and-ride (P&R) facility location problem under the multinomial logit demand function, the capacitated p-hub approach for the problem under the nested logit demand function captures a broader range of real-world cases. To solve this problem optimally, we introduce a mixed-integer linear program and accelerate its solution by enhancing the branch-and-cut procedure. To address the problem at a large scale, we introduce two other polyhedral approaches: variable neighborhood search (VNS) and adaptive randomized rounding (ARR). Downtown areas in Seoul have a high modal share of public transportation and congested road traffic, yet P&R has not been widely implemented. Therefore, we apply the ARR procedure to solve a real-world problem using traffic and geographic data from the Seoul metropolitan area. ARR performs better than VNS and addresses real-world cases. The solutions obtained by ARR present a phased expansion plan that encourages policymakers to start installing a small number of P&Rs immediately.

Tailoring Microbial Fitness Through Computational Steering and CRISPRi-Driven Robustness Regulation

The widespread application of genetically modified microorganisms (GMMs) across diverse sectors underscores the pressing need for robust strategies to mitigate the risks associated with their potential uncontrolled escape. This study merges computational modeling with CRISPR interference (CRISPRi) to refine GMM metabolic robustness. Utilizing ensemble modeling, we achieved high-throughput in silico screening for enzymatic targets susceptible to expression alterations. Translating these insights, we developed functional CRISPRi, boosting fitness control via multiplexed gene knockdown. Our method, enhanced by an insulator-improved gRNA structure and an off-switch circuit controlling a compact Cas12m, resulted in rationally engineered strains with escape frequencies below National Institutes of Health standards. The effectiveness of this approach was confirmed under various conditions, showcasing its ability for secure GMM management. This research underscores the resilience of microbial metabolism, strategically modifying key nodes to halt growth without provoking significant resistance, thereby enabling more reliable and precise GMM control. A record of this paper's transparent peer review process is included in the supplemental information.
