Antimotion-sickness efficacy of scopolamine 12 and 72 hours after transdermal administration

The antimotion sickness remedy, transdermal therapeutic system-scopolamine, administered in this experiment was scheduled to deliver 1.0 mg of scopolamine over a period of 3 d, and this paper compares its efficacy 12 and 72 h after administration. In a double-blind study, six male college students were individually exposed to a standardized provocative test in a slow rotation room after six apparently identical treatments comprising four placebos and two medications. Efficacy was categorized as beneficial, inconsequential, or detrimental. None of the responses was detrimental. Following the first administration of the therapeutic system, there were four beneficial responses after 12 h but none was beneficial after 72 h. Following the second treatment regimen, there were four beneficial responses after 12 h and three beneficial responses after 72 h. Great individual differences were demonstrated, two subjects accounting for six beneficial responses and two accounting for only one beneficial response. The difference in efficacy after 12 and 72 h has practical and theoretical significance.

Administrative automation in a scientific environment

Although the scientific personnel at GSFC were advanced in the development and use of hardware and software for scientific applications, resistance to the use of automation or purchase of terminals, software and services, specifically for administrative functions was widespread. The approach used to address problems and constraints and plans for administrative automation within the Space and Earth Sciences Directorate are delineated. Accomplishments thus far include reduction of paperwork and manual efforts; improved communications through telemail and committees; additional support staff; increased awareness at all levels on ergonomic concerns and the need for training; better equipment; improved ADP skills through experience; management commitment; and an overall strategy for automating.

Impact of the 1985 Space World Administrative Radio Conference on frequency/orbit planning and use

The 1985 World Administrative Radio Conference (WARC-ORB-85) was held to determine which space radio services should be planned and which planning methods should be used. The second session of this Conference (WARC-ORB-88) will meet to develop the required plans. This paper presents the results of WARC-ORB-85, assesses the impact of those decisions, and identifies the intersessional work to be conducted by administrations and the CCIR (Consultative Committee on International Radio). The major decisions of WARC-ORB-85 were: (1) the restriction of additional planning to the fixed satellite service at identified frequencies; and (2) the selection of a planning method consisting of two parts (a) an allotment plan, and (b) improved procedures. The paper also discusses WARC-ORB-85 decisions relative to the Region 2 broadcast satellite service plans at 12 GHz, feederlink planning for Regions 1 and 3 broadcast satellites at 12 GHz, and sound broadcast satellite service.

Effects of Long-Term Daily Administration of Prostaglandin-E2 on Maintaining Elevated Proximal Tibial Metaphyseal Cancellous Bone Mass in Male Rats

The effects of long-term prostaglandin E(sub 2) (PGE(sub 2)) on cancellous bone in proximal tibial metaphysis were studied in 7 month old male Sprague-Dawley rats given daily subcutaneous injections of 0, 1, 3, and 6 mg PGE(sub 2)/kg/day and sacrificed after 60, 120, and 180 days. Histomorphometric analyses were performed on double fluorescent-labeled undecalcified bone specimens. After 60 days of treatment, PGE(sub 2) produced diffusely labeled trabecular bone area, increased trabecular bone area, eroded and labeled trabecular perimeter, mineral apposition rate, and bone formation rate at all dose levels when compared with age-matched controls. In rats given PGE(sub 2) for longer time periods (120 and 180 days), trabecular bone area, diffusely labeled trabecular bone area, labeled perimeter, mineral apposition, and bone formation rates were sustained at the elevated levels achieved earlier at 60-day treatment. The eroded perimeter continued to increase until 120 days, then plateau. The observation that continuous systemic PGE(sub 2) administration to adult male rats elevated metaphyseal cancellous bone mass to 3.5-fold of the control level within 60 days and maintained it for another 120 days indicates that the powerful skeletal anabolic effects of PGE2 can be sustained with continuous administration .

Effects of 2-deoxy-D-glucose administration on cytokine production in BDF1 mice

Physical exercise and diet changes have been shown to affect immune parameters, and similar effects are also induced by the administration of a nonmetabolizable glucose analog, 2-deoxy-D-glucose (2-DG). The present study was designed to characterize the effects of glucoprivation induced by 2-DG administration on concentrations of tumor necrosis factor-alpha (TNF-alpha), interleukin-1beta (IL-1beta), and IL-6 in the blood and interferon-gamma (IFN-gamma), IL-2, and IL-4 in vitro production by partially purified T splenocytes in BDF1 mice. Mice (n = 8 per group) were injected intraperitoneally one or three times with 0, 500, 750, or 1000 mg/kg of 2-DG, and blood and spleens were collected 2 h after the last injection. Partially purified T splenocytes were cultured 24 h in the presence of concanavalin A (ConA). A significant increase in the corticosterone levels with the amount of 2-DG injected was observed after one or three injections (p<0.05). The amount of 2-DG injected was associated with an increase in TNF-alpha, IL-1beta, and IL-6 concentrations in the blood of mice after one or three injections of 2-DG (p<0.05). A significant decrease in in vitro proliferation of partially purified splenocytes in the presence of ConA was associated with a decrease in IFN-gamma production in the culture supernatants and an increase in IL-1 receptor expression on the cell surface (p<0.05).

Effects of methiothepin on changes in brain serotonin release induced by repeated administration of high doses of anorectic serotoninergic drugs

We previously observed, using in vivo microdialysis, that the potassium-evoked release of frontocortical serotonin (5-HT) is suppressed after rats receive high doses (30 mg/kg, i.p., daily for 3 days) of fluoxetine, a selective blocker of 5-HT reuptake. We now describe similar impairments in 5-HT release after repeated administration of two other 5-HT uptake blockers, zimelidine and sertraline (both at 20 mg/kg, i.p. for 3 days) as well as after dexfenfluramine (7.5 mg/kg, i.p. daily for 3 days), a drug which both releases 5-HT and blocks its reuptake. Doses of these indirect serotonin agonists were about 4-6 times the drug's ED50 in producing anorexia, a serotonin-related behavior. In addition, methiothepin (20 microM), a non-selective receptor antagonist, locally perfused through the dialysis probe 24 h after the last drug injection, enhanced K(+)-evoked release of 5-HT at serotoninergic nerve terminals markedly in control rats and slightly in rats treated with high doses of dexfenfluramine or fluoxetine. On the other hand, pretreatment with methiothepin (10 mg/kg, i.p.) one hour before each of the daily doses of fluoxetine or dexfenfluramine given for 3 days, totally prevented the decrease in basal and K(+)-evoked release of 5-HT. Finally, when methiothepin was injected systemically the day before the first of 3 daily injections of dexfenfluramine, it partially attenuated the long-term depletion of brain 5-HT and 5-HIAA levels induced by repeated administration of high doses of dexfenfluramine. These data suggest that drugs which bring about the prolonged blockade of 5-HT reuptake - such as dexfenfluramine and fluoxetine - can, by causing prolonged increases in intrasynaptic 5-HT levels as measured by in vivo microdialysis, produce receptor-mediated long-term changes in the processes controlling serotonin levels and dynamics.

National Aeronautics and Space Administration Biological Specimen Repository

The National Aeronautics and Space Administration Biological Specimen Repository (Repository) is a storage bank that is used to maintain biological specimens over extended periods of time and under well-controlled conditions. Samples from the International Space Station (ISS), including blood and urine, will be collected, processed and archived during the preflight, inflight and postflight phases of ISS missions. This investigation has been developed to archive biosamples for use as a resource for future space flight related research. The International Space Station (ISS) provides a platform to investigate the effects of microgravity on human physiology prior to lunar and exploration class missions. The storage of crewmember samples from many different ISS flights in a single repository will be a valuable resource with which researchers can study space flight related changes and investigate physiological markers. The development of the National Aeronautics and Space Administration Biological Specimen Repository will allow for the collection, processing, storage, maintenance, and ethical distribution of biosamples to meet goals of scientific and programmatic relevance to the space program. Archiving of the biosamples will provide future research opportunities including investigating patterns of physiological changes, analysis of components unknown at this time or analyses performed by new methodologies.

Tools for Administration of a UNIX-Based Network

Several computer programs have been developed to enable efficient administration of a large, heterogeneous, UNIX-based computing and communication network that includes a variety of computers connected to a variety of subnetworks. One program provides secure software tools for administrators to create, modify, lock, and delete accounts of specific users. This program also provides tools for users to change their UNIX passwords and log-in shells. These tools check for errors. Another program comprises a client and a server component that, together, provide a secure mechanism to create, modify, and query quota levels on a network file system (NFS) mounted by use of the VERITAS File SystemJ software. The client software resides on an internal secure computer with a secure Web interface; one can gain access to the client software from any authorized computer capable of running web-browser software. The server software resides on a UNIX computer configured with the VERITAS software system. Directories where VERITAS quotas are applied are NFS-mounted. Another program is a Web-based, client/server Internet Protocol (IP) address tool that facilitates maintenance lookup of information about IP addresses for a network of computers.

Validation of a Best-Fit Pharmacokinetic Model for Scopolamine Disposition after Intranasal Administration

An intranasal gel formulation of scopolamine (INSCOP) was developed for the treatment of Motion Sickness. Bioavailability and pharmacokinetics (PK) were determined per Investigative New Drug (IND) evaluation guidance by the Food and Drug Administration. Earlier, we reported the development of a PK model that can predict the relationship between plasma, saliva and urinary scopolamine (SCOP) concentrations using data collected from an IND clinical trial with INSCOP. This data analysis project is designed to validate the reported best fit PK model for SCOP by comparing observed and model predicted SCOP concentration‐time profiles after administration of INSCOP.

Project Mercury: Man-In-Space Program of the National Aeronautics and Space Administration

The purpose of this staff study, made at the request of the chairman, is to serve members of the Committee on Aeronautical and Space Sciences as a source of basic information on Project Mercury, the man-in-space program of the National Aeronautics and Space Administration. The study is largely derived from unclassified information released by the National Aeronautics and Space Administration and testimony concerning Project Mercury given during hearings before this committee. The program descriptions are based upon current program planning. Since this is a highly advanced research and development program, the project is obviously subject to changes that may result from future developments and accomplishments characteristic of such research activities. Certain information with respect to revised schedules, obtained on a classified basis by the committee during inspection trips, is necessarily omitted. The appendixes to the study include information that may prove helpful on various aspects of space flight and exploration. Included are unofficial comments and observations relating to Russia's manned space flight activities and also a complete chronology of all satellites, lunar probes, and space probes up to the present.

Dose-Dependent Sensorimotor Impairment in Human Ocular Tracking After Acute Low-Dose Alcohol Administration

Changes in oculomotor behaviours are often used as metrics of sensorimotor disruption due to ethanol (EtOH); however, previous studies have focused on deficits at blood-alcohol concentrations (BACs) above about 0.04%.We investigated the dose dependence of the impairment in oculomotor and ocular behaviours caused by EtOH administration across a range of ultra-low BACs (≤0.035%).We took repeated measures of oculomotor and ocular performance from sixteen participants, both pre- and post-EtOH administration. To assess the neurological impacts across a wide range of brain areas and pathways, our protocol measured 21 largely independent performance metrics extracted from a range of behavioural responses ranging from ocular tracking of radial step-ramp stimuli, to eccentric gaze holding, to pupillary responses evoked by light flashes. Our results show significant impairment of pursuit and visual motion processing at 0.015% BAC, reflecting degraded neural processing within extrastriate cortical pathways. However, catch-up saccades largely compensate for the tracking displacement shortfall caused by low pursuit gain, although there still is significant residual retinal slip and thus degraded dynamic acuity. Furthermore, although saccades are more frequent, their dynamics are more sluggish (i.e. show lower peak velocities) starting at BAC levels as low as 0.035%. Small effects in eccentric gaze holding and no effect in pupillary response dynamics were observed at levels below 0.07%, showing the higher sensitivity of the pursuit response to very low levels of blood alcohol, under the conditions of our study.

From Here to There - Bird's Eye Perspective on Grant Lifecycle and Administration

“What gets measured gets done ”or “measure it to manage it”. The statements have become matter of fact. However, measurement (and even metrics) alone cannot fully communicate an organization’s story without the point of accountability. As a dynamic org, we must measure what is necessary, using the appropriate lenses to analyze the most relevant information that allows robust decision-making. A key function of Program Planning and Control (PP&C) is to assist HRP in the development of such strategy that ensures the proper execution of program research goals. With introspection of the Technical Officer role as defined by 2 CFR(i.e., Part 200, Part 1800)and subsequent governance under the NASA Grant and Cooperative Agreement Manual (GCAM), we’ve realized an obvious disconnect among Principal Investigator’s (PI),technical communication with the elements and some of the administrative/fiduciary requirements that should run parallel to the science work. This find has introduced new program risks as evidenced through incomplete deliverables or other less-than-successful requirements through the lifecycle for work we fund. One solution the Grants Technical Officer proposes is for PP&C to share in the communication with stakeholders of grants administration much earlier in the award process in attempt to reduce programmatic risks; particularly in data, reporting and performance management. As such, the planned IWS 2022 session will focus on(plenary) topics that include (1) definition of the grant lifecycle, (2) anatomy of a grant(NF1687 the award document), and (3)performance measurement/post-award monitoring. As HH&P pursues its strategic objectives to evolve the directorate into a data rich and knowledge rich organization, it has become increasingly important to develop the multiple levels of insight across the program to ensure data and other reporting(qualitative/quantitative), alongside proper administration. And the desired outcome is to make certain HRP meets/exceeds the accountability standard that our research information is received, maintained and accessible in posterity.

General Services Administration Reclamation Yard, SWMU 010, 2021 Groundwater Monitoring Report

This report presents a summary of the groundwater monitoring activities that occurred in November and December 2021 at General Services Administration Reclamation Yard, Solid Waste Management Unit 010, located at the John F. Kennedy Space Center (KSC), Florida. The site is monitored under KSC’s Resource Conservation and Recovery Act Corrective Action Program. This approach also meets the requirements of Chapter 62-780, Florida Administrative Code. For the purposes of this report, two separate plumes, known as the Polychlorinated Biphenyl (PCB)/Volatile Organic Aromatic (VOA) Plume and the Chlorinated Volatile Organic Compound (VOC) Plume, were identified for this site. The contaminants of concern (COCs) for the PCB/VOA Plume consists of PCBs, 1,2,4-trichlorobenzene, and breakdown products of 1,2,4-trichlorobenzene. The COCs for the Chlorinated VOC Plume are tetrachloroethene, trichloroethene, cis-1,2-dichloroethene, and vinyl chloride. The activities presented in this report include two field events: (1) November 2021 – PCB/VOA Plume groundwater sampling via Direct Push Technology – 70 groundwater samples were collected from 10 locations at varying intervals; and (2) December 2021 – Site-wide semi-annual water level measurements of 54 monitoring wells, redevelopment of 5 monitoring wells, and sampling of 35 monitoring wells. Results for the PCB/VOA plume demonstrated COCs have not rebounded since the 2018 source removal, with groundwater cleanup target levels (GCTLs) exceeded in the 10-30 ft. below ground surface interval only. In the Chlorinated VOC plume area, there were no detections of COCs above a GCTL for at least the second consecutive sampling event for the sampled wells. Cleanup objectives have been met, and it is recommended that VOC monitoring be discontinued. Underground injection control parameters will continue to be monitored in three wells until those analytes return to background concentrations.

General Services Administration Reclamation Yard Solid Waste Management Unit 010: 2022 Groundwater Monitoring Report Kennedy Space Center, Florida

This report presents a summary of the groundwater monitoring activities that occurred in September and October 2022 at General Services Administration Reclamation Yard, Solid Waste Management Unit 010, located at the John F. Kennedy Space Center (KSC), Florida. The site is monitored under KSC’s Resource Conservation and Recovery Act Corrective Action Program. This approach also meets the requirements of Chapter 62-780, Florida Administrative Code. The COCs for the Chlorinated VOC Plume were tetrachloroethene, trichloroethene, cis-1,2-dichloroethene, and vinyl chloride. Groundwater monitoring for VOCs in the Chlorinated VOC Plume area was discontinued following the December 2021 annual monitoring event. Annual groundwater monitoring for underground injection control (UIC) parameters continues for both the PCB/VOA Plume and the Chlorinated VOC Plume areas. The activities presented in this report include one field event: September and October 2022, which includes sitewide semi-annual water level measurements of 55 monitoring wells, redevelopment of all monitoring wells listed in the sampling plan, and groundwater sampling of 28 monitoring wells. The sampling and analysis demonstrated that the PCB/VOA plume is not expanding and is significantly reduced compared to before a 2018 source removal interim measure. Recommendations are made for the 2023 groundwater monitoring program.
