Interdependent iron and phosphorus availability controls photosynthesis through retrograde signaling

Abstract Iron deficiency hampers photosynthesis and is associated with chlorosis. We recently showed that iron deficiency-induced chlorosis depends on phosphorus availability. How plants integrate these cues to control chlorophyll accumulation is unknown. Here, we show that iron limitation downregulates photosynthesis genes in a phosphorus-dependent manner. Using transcriptomics and genome-wide association analysis, we identify two genes, PHT4;4 encoding a chloroplastic ascorbate transporter and bZIP58 , encoding a nuclear transcription factor, which prevent the downregulation of photosynthesis genes leading to the stay-green phenotype under iron-phosphorus deficiency. Joint limitation of these nutrients induces ascorbate accumulation by activating expression of an ascorbate biosynthesis gene, VTC4 , which requires bZIP58. Furthermore, we demonstrate that chloroplastic ascorbate transport prevents the downregulation of photosynthesis genes under iron-phosphorus combined deficiency through modulation of ROS homeostasis. Our study uncovers a ROS-mediated chloroplastic retrograde signaling pathway to adapt photosynthesis to nutrient availability.

Vitamin interdependencies predicted by metagenomics-informed network analyses and validated in microbial community microcosms

Abstract Metagenomic or metabarcoding data are often used to predict microbial interactions in complex communities, but these predictions are rarely explored experimentally. Here, we use an organism abundance correlation network to investigate factors that control community organization in mine tailings-derived laboratory microbial consortia grown under dozens of conditions. The network is overlaid with metagenomic information about functional capacities to generate testable hypotheses. We develop a metric to predict the importance of each node within its local network environments relative to correlated vitamin auxotrophs, and predict that a Variovorax species is a hub as an important source of thiamine. Quantification of thiamine during the growth of Variovorax in minimal media show high levels of thiamine production, up to 100 mg/L. A few of the correlated thiamine auxotrophs are predicted to produce pantothenate, which we show is required for growth of Variovorax , supporting that a subset of vitamin-dependent interactions are mutualistic. A Cryptococcus yeast produces the B-vitamin pantothenate, and co-culturing with Variovorax leads to a 90-130-fold fitness increase for both organisms. Our study demonstrates the predictive power of metagenome-informed, microbial consortia-based network analyses for identifying microbial interactions that underpin the structure and functioning of microbial communities.

Hy-DAT: A Tool to Address Hydropower Modeling Gaps Using Interdependency, Efficiency Curves, and Unit Dispatch Models

As the power system continues to be flooded with intermittent resources, it becomes more important to accurately assess the role of hydro and its impact on the power grid. While hydropower generation has been studied for decades, dependency of power generation on water availability and constraints in hydro operation are not well represented in power system models used in the planning and operation of large-scale interconnection studies. There are still multiple modeling gaps that need to be addressed; if not, they can lead to inaccurate operation and planning reliability studies, and consequently to unintentional load shedding or even blackouts. As a result, it is very important thathydropower is represented correctly in both steady-state and dynamic power system studies. In this paper, we discuss the development and use of the Hydrological Dispatch and Analysis Tool (Hy-DAT) as an interactive graphical user interface (GUI), that uses a novel methodology to address the hydropower modeling gaps like water availability and inter-dependency using a database, and algorithms, to generate accurate representative models for power system simulation.

Interdependent Water And Power Infrastructure Model

The approach used is the Multi-Agent System (MAS) paradigms, where systems components are represented as agents, interacting both with each other, and with the environment in which they evolved. Agents behaviors correspond to components in the real Integrated Water-Power System. The model simulate actions and interactions of these (autonomous) agents to analyze their effects on the overall system. Agents in the water system capture components of water collection, treatment, transportation, distribution and use (e.g. pipe, canal, pump, water demands for agriculture, etc.). Agents in the power system capture components of power generation, transportation, distribution and use (electricity demands, sources, etc.).

Gas-Electric Interdependence

Presentation to February 2024 DOE Electricity Advisory Committee (EAC) Meeting regarding interrelationship between gas and electric. The EAC is a Federal Advisory Committee Act (FACA) committee. The purpose of the EAC is to provide advice to the U.S. Department of Energy in implementing the Energy Policy Act of 2005, executing the Energy Independence and Security Act of 2007, and modernizing the nation's electricity delivery infrastructure.

Using stable isotopes in water vapor to study the interdependence of clouds, atmospheric aerosols, and precipitation processes

This project deployed water vapor isotope measurements at the La Porte, Texas TRACER site during June-September 2022 to quantify mixing processes in sea breeze circulations. Using a Picarro L2130 analyzer, we collected high-frequency measurements of δD and δ¹⁸O that revealed how marine and terrestrial air masses mix during 46 identified sea breeze events. The isotopic tracers showed systematic changes during frontal passages, with composite analyses demonstrating vertical mixing between cool marine air and warmer air aloft. We developed a transilient matrix model that explicitly treats non-local mixing and isotopic fractionation to interpret the observations. The work directly supports TRACER's core objective of understanding convective initiation by constraining the vertical transport of water vapor and its interaction with Houston's aerosol-laden urban plume during sea breeze-convection coupling.

The interdependence of lake ice and climate in central North America

There are no author-identified significant results in this report. This investigation is to identify any correlations between the freeze/ thaw cycles of lakes and regional weather variations. ERTS-1 imagery of central Canada and north central United States is examined on a seasonal basis. The ice conditions of certain major study lakes are noted and recorded on magnetic tape, from which the movement of a freeze/thaw transition zone may be deduced. Weather maps and tables are used to establish any obvious correlations. The process of selecting major study lakes is discussed, and a complete lake directory is presented. Various routines of the software support library are described, accompanied by output samples. Procedures used for ERTS imagery processing are presented along with the data analysis plan. Application of these procedures to selected ERTS imagery has demonstrated their utility. Preliminary results show that the freeze/thaw transition zone can be monitored from ERTS.

The Interdependence of Lake Ice and Climate in Central North America

The author has identified the following significant results. A comparison of lake freeze transition zone migration with the movement of large pressure centers reveals the following consistencies: (1) polar continental cyclones originate within and/or travel along the trend of the transition zone; (2) polar continental anticyclones fail to cross the transition zone; (3) polar outbreak anticyclones pass through the transition zone, apparently unaffected. In addition, storm centers associated with the transition zone undergo significant intensification manifest by a deepening of the pressure through and increased precipitation outside the zone.
