Design synthesis of a boron/epoxy reinforced metal shear web.

An advanced composite shear web design concept has been developed for the Space Shuttle Orbiter main engine thrust beam structure. Various web concepts were synthesized by a computer-aided adaptive random search procedure. A practical concept is identified having a titanium-clad, boron/epoxy plate with vertical boron/epoxy reinforced stiffeners. Baseline composite and titanium shear resistant designs are compared; the composite concept is 28% lighter than the titanium web. Element test results show the metal cladding effectively reinforces critical composite load transfer and fastener hole areas making the composite web concept practical for other shear structure applications.-

General aviation design synthesis utilizing interactive computer graphics

Interactive computer graphics is a fast growing area of computer application, due to such factors as substantial cost reductions in hardware, general availability of software, and expanded data communication networks. In addition to allowing faster and more meaningful input/output, computer graphics permits the use of data in graphic form to carry out parametric studies for configuration selection and for assessing the impact of advanced technologies on general aviation designs. The incorporation of interactive computer graphics into a NASA developed general aviation synthesis program is described, and the potential uses of the synthesis program in preliminary design are demonstrated.

New frontiers in design synthesis

The Intelligent Synthesis Environment (ISE), which is one of the major strategic technologies under development at NASA centers and the University of Virginia, is described. One of the major objectives of ISE is to significantly enhance the rapid creation of innovative affordable products and missions. ISE uses a synergistic combination of leading-edge technologies, including high performance computing, high capacity communications and networking, human-centered computing, knowledge-based engineering, computational intelligence, virtual product development, and product information management. The environment will link scientists, design teams, manufacturers, suppliers, and consultants who participate in the mission synthesis as well as in the creation and operation of the aerospace system. It will radically advance the process by which complex science missions are synthesized, and high-tech engineering Systems are designed, manufactured and operated. The five major components critical to ISE are human-centered computing, infrastructure for distributed collaboration, rapid synthesis and simulation tools, life cycle integration and validation, and cultural change in both the engineering and science creative process. The five components and their subelements are described. Related U.S. government programs are outlined and the future impact of ISE on engineering research and education is discussed.

Design synthesis and optimization of joined-wing transports

A computer program for aircraft synthesis using a numerical optimizer was developed to study the application of the joined-wing configuration to transport aircraft. The structural design algorithm included the effects of secondary bending moments to investigate the possibility of tail buckling and to design joined wings resistant to buckling. The structural weight computed using this method was combined with a statistically-based method to obtain realistic estimates of total lifting surface weight and aircraft empty weight. A variety of 'optimum' joined-wing and conventional aircraft designs were compared on the basis of direct operating cost, gross weight, and cruise drag. The most promising joined-wing designs were found to have a joint location at about 70 percent of the wing semispan. The optimum joined-wing transport is shown to save 1.7 percent in direct operating cost and 11 percent in drag for a 2000 nautical mile transport mission.

A Review of Polymeric Nanoparticle Design, Synthesis, and Biomedical Applications

Polymeric nanoparticles (PNPs) are solid particles that contain macromolecular polymers ranging in size from 1-1000 nm. They have a wide variety of structures that are determined by their identity as either natural biopolymers, synthetic polymers or a combination of both. In the design of PNPs, the polymeric system, size, internal structure, additional molecules, and synthetic pathway can be engineered, allowing for a multitude of properties including differing responses to physiological environments and the incorporation of a drug. The easily accessible fine tuning of PNPs allows for a multitude of applications in medicine. PNPs allow for increased pharmacokinetics, efficacy, safety, and targeting strength when utilized as carriers for drugs. Currently there are a wide range of PNPs approved by the Food and Drug Administration for use in clinical settings and many more being investigated in clinical studies. There is also research that is being conducted for the usage of PNPs in controlled drug delivery systems and as carriers through the blood brain barrier to aid in treating neurological diseases.

Design, synthesis, and biological testing of biphenylmethyloxazole inhibitors targeting HIV-1 reverse transcriptase

In this article, we report non-nucleoside inhibitors of HIV-1 reverse transcriptase (NNRTIs) using a biphenylmethyloxazole pharmacophore. A crystal structure of benzyloxazole 1 was obtained and suggested the potential viability of biphenyl analogues. In particular, 6a, 6b, and 7 turned out to be potent NNRTIs with low-nanomolar activity in enzyme inhibition and infected T-cell assays, and with low cytotoxicity. Though modeling further suggested that analogues with fluorosulfate and epoxide warheads might provide covalent modification of Tyr188, synthesis and testing did not find evidence for this outcome.

A robust Feasible Directions algorithm for design synthesis

A nonlinear optimization algorithm is developed which combines the best features of the Method of Feasible Directions and the Generalized Reduced Gradient Method. This algorithm utilizes the direction-finding sub-problem from the Method of Feasible Directions to find a search direction which is equivalent to that of the Generalized Reduced Gradient Method, but does not require the addition of a large number of slack variables associated with inequality constraints. This method provides a core-efficient algorithm for the solution of optimization problems with a large number of inequality constraints. Further optimization efficiency is derived by introducing the concept of infrequent gradient calculations. In addition, it is found that the sensitivity of the optimum design to changes in the problem parameters can be obtained using this method without the need for second derivatives or Lagrange multipliers. A numerical example is given in order to demonstrate the efficiency of the algorithm and the sensitivity analysis.

Oxadiazole-based Heterocycles as Building Block for Material Property Control: Design, Synthesis, and Characterization of 3,4-Bis(3-(4-nitro-1,2,5-oxadiazol-3-yl)- 1,2,4-oxadiazol-5-yl)-1,2,5-oxadiazole (LLM-210)

This paper presents an approach to novel oxadiazole-based melt-castable energetic material, LLM-210. Used in pour-in process and 3D printing, melt-castable energetic materials (MCEM) must possess required physical and safety properties of industry-standard energetic materials, also a narrow range of melting point between 80-100 °C. Melting point is difficult to accurately predict, therefore, searching for MCEM is challenging. 3,4-Bis(5-(4-nitro-1,2,5-oxadiazol-3-yl)-1,2,4-oxadiazol3-yl)-1,2,5-oxadiazole (LLM-205), composed of five-oxadiazole rings allied via carbon-carbon bonds, was selected as new MCEM candidate. The target molecule was synthesized and characterized, showing a density of 1.807g/cm3 ; thermal decomposing temperature at 299 °C; and insensitive to external stimuli, but the melting point of 104 °C was out of the rang. Based on the molecular properties and ab initio calculations, 3,4-bis(3-(4-nitro-1,2,5-oxadiazol-3-yl)-1,2,4-oxadiazol-5-yl)- 1,2,5-oxadiazole (LLM-210), an isomer of LLM-205, was designed and synthesized from a different way. LLM-210 was characterized as MCEM, possessing melting point of 87 °C; density of 1.812 g/cm3 ; and exothermally decomposing temperature at 314 °C; being insensitive to external stimuli. In this study, the relationship of molecular structure to properties of LLM-205 and LLM-210 based on the results of Density Functional Theory (DFT) and X-ray crystallographic analysis is also briefly discussed.

Design, synthesis, molecular docking, antimicrobial, and antioxidant activities of new phenylsulfamoyl carboxylic acids of pharmacological interest

Abstracts: The research explores the facile synthesis of some new phenylsulfamoyl carboxylic acids, their molecular docking, antimicrobial, and antioxidant activities. The procedure involved the mild reaction of amino acids with benzenesulfonyl chloride in a medium of aqueous base. The compounds were characterized using FTIR, {sup 1}H-NMR, {sup 13}C-NMR, and an elemental analysis. They were tested for their antimicrobial activities against Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa, Salmonella typhi, Candida albicans, and Aspergillus niger microorganisms. The antioxidant activity of the compounds were measured in vitro by the inhibition of generated stable 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical. The molecular docking was carried out properly and five different disease conditions were studied, namely: trypanosomiasis, malaria, bacterial, fungal infections, and oxidative stress. From the results, compounds 4c, 4d, 4e, and 4g possess more excellent in vitro antibacterial and antifungal activities than the standard drug Ofloxacin used. Compound 4e displayed the most excellent antioxidant activity. Compound 4g showed significant 2D interaction with amino acid residue of urate oxidase from Aspergillus flavus complexed with uracil. Interestingly, compounds 4a, 4c, 4d, 4e, and 4g exhibited excellent antibacterial, antifungal, antioxidant, antitrypanosome, and antimalaria activities comparable to the corresponding standard drugs such as Penicillin, Ketoconazole; α-Tocopherol, Melarsoprol, and Chloroquine respectively. All the compounds were confirmed drug-like according to “Lipinski’s rule of five”. The compounds were found to be promising antibacterial, antifungal, antioxidant, and antitrypanosome agents.
