Building a custom high-throughput platform at the Joint Genome Institute for DNA construct design and assembly—present and future challenges

Abstract The rapid design and assembly of synthetic DNA constructs have become a crucial component of biological engineering projects via iterative design–build–test–learn cycles. In this perspective, we provide an overview of the workflows used to generate the thousands of constructs and libraries produced each year at the U.S. Department of Energy Joint Genome Institute. Particular attention is paid to describing pipelines, tools used, types of scientific projects enabled by the platform and challenges faced in further scaling output.

Thermal-vacuum testing and in-situ optical alignment measurements of the HALOE telescope/sun sensor assembly

Design features, test data and projected performance levels of the telescope/sun sensor assembly for the Halogen Occultation Experiment (HALOE) on board the Upper Atmosphere Research Satellite are described. HALOE will gather data on stratospheric chemical species with an IR telescope pointed through the atmosphere at the sun during occultation periods. The pointing accuracy will need to be 1 arcmin elevation and 1.6 arcmin azimuthal. Adjustments of the gimbaled instrument are guided by a sun sensor and its associated electronics. The components were subjected to thermal-vacuum, optical boresight stability tests and the data generated were compared with a finite element model of the telescope and sun sensor. The tests consisted of exposure to various thermal gradients while the telescope and sun sensor alignments were tracked by optical deflectometry. The thermal behavior was compared with predictions made with a finite difference model. Alignments were within tolerable ranges and the thermal behavior model was concluded valid for predicting the thermal behavior of orbiting instruments.

Estimation of bearing contact angle in-situ by X-ray kinematography

The mounted, preloaded contact angle of the structural bearings in the assembled design mechanical assembly was measured. A modification of the Turns method is presented, based upon the clarity and definition of moving parts achieved with X-ray technique and cinematic display. Contact angle is estimated by counting the number of bearings passing a given point as a function of number of turns of the shaft. Ball and pitch diameter variations are discussed. Ball train and shaft angle uncertainties are also discussed.

Design and assembly sequence analysis of option 3 for CETF reference space station

A design and assembly sequence was conducted on one option of the Dual Keel Space Station examined by a NASA Critical Evaluation Task Force to establish viability of several variations of that option. A goal of the study was to produce and analyze technical data to support Task Force decisions to either examine particular Option 3 variations in more depth or eliminate them from further consideration. An analysis of the phasing assembly showed that use of an Expendable Launch Vehicle in conjunction with the Space Transportation System (STS) can accelerate the buildup of the Station and ease the STS launch rate constraints. The study also showed that use of an Orbital Maneuvering Vehicle on the first flight can significantly benefit Station assembly and, by performing Station subsystem functions, can alleviate the need for operational control and reboost systems during the early flights. In addition to launch and assembly sequencing, the study assessed stability and control, and analyzed node-packaging options and the effects of keel removal on the structural dynamics of the Station. Results of these analyses are presented and discussed.

Design and Assembly Process Implementation for Embedding Passive and Active Components Introduction to IPC-7092

"This presentation describes the design and assembly challenges for implementing passive and active components, in either formed or inserted methodology, into a printed board. The completed structure including internal electronic components is ready for surface mount and/or through-hole component attachment. The multilayered structure becomes a complete product ready for further processing in an assembly process and be made from organic, inorganic (ceramic) or both types of material."

Fuel cell design and assembly

The present invention is directed to a novel bipolar cooling plate, fuel cell design and method of assembly of fuel cells. The bipolar cooling plate used in the fuel cell design and method of assembly has discrete opposite edge and means carried by the plate defining a plurality of channels extending along the surface of the plate toward the opposite edges. At least one edge of the channels terminates short of the edge of the plate defining a recess for receiving a fastener.

Predictability of Localized Plasmonic Responses in Nanoparticle Assemblies

Design of nanoscale structures with desired optical properties is a key task for nanophotonics. In this work, the correlative relationship between local nanoparticle geometries and their plasmonic responses is established using encoder-decoder neural networks. In the im2spec network, the relationship between local particle geometries and local spectra is established via encoding the observed geometries to a small number of latent variables and subsequently decoding into plasmonic spectra; in the spec2im network, the relationship is reversed. Surprisingly, these reduced descriptions allow high-veracity predictions of local responses based on geometries for fixed compositions and surface chemical states. Analysis of the latent space distributions and the corresponding decoded and closest (in latent space) encoded images yields insight into the generative mechanisms of plasmonic interactions in the nanoparticle arrays. Ultimately, this approach creates a path toward determining configurations that yield the spectrum closest to the desired one, paving the way for stochastic design of nanoplasmonic structures.

Design and assembly of a catalyst bed gas generator for the catalytic decomposition of high concentration hydrogen peroxide propellants and the catalytic combustion of hydrocarbon/air mixtures

A method for designing and assembling a high performance catalyst bed gas generator for use in decomposing propellants, particularly hydrogen peroxide propellants, for use in target, space, and on-orbit propulsion systems and low-emission terrestrial power and gas generation. The gas generator utilizes a sectioned catalyst bed system, and incorporates a robust, high temperature mixed metal oxide catalyst. The gas generator requires no special preheat apparatus or special sequencing to meet start-up requirements, enabling a fast overall response time. The high performance catalyst bed gas generator system has consistently demonstrated high decomposition efficiency, extremely low decomposition roughness, and long operating life on multiple test articles.
