The adsorption and reaction of adenine nucleotides on montmorillonite

The binding of AMP to Zn(2+)-montmorillonite is investigated in the presence of salts and Good's zwitterion buffers, PIPES and MES. The initial concentrations of nucleotide and the percent adsorbtion are used to calculate the adsorption isotherms, and the Langmuir adsorption equation is used for the analysis of data. The adsorption coefficient was found to be three times greater in the presence of 0.2 M PIPES than in its absence. In addition, basal spacings measured by X-ray diffraction were increased by the buffer. These results are interpreted in terms of a model in which the adsorption of AMP is mediated by a Zn(2+) complex of PIPES in different orientations in the interlamellar region of the montmorillonite. Mixed ligand complexes of this type are reminiscent of the complexes observed between metal ions and biological molecules in living systems.

Performance boundaries for prioritized multiplexing systems

Systems in which many data sources are multiplexed over a single communication channel are considered. Data from all the sources are generated in fixed-length packets and are stored in a common buffer with finite capacity. Packets that overflowed or were removed from the buffer prior to transmission are lost. The system performance measure is the set of packet loss probabilities associated with the sources. Queueing disciplines vary depending on the stringency of prioritization and the utilization of the system resources. The set of all possible performances is characterized as the set of all queueing disciplines is spanned. Whether a given performance is possible can be deduced. Strategies that achieve the minimum overall loss probability are identified. The extreme disciplines are specified, and their performances are calculable by means of a given algorithm.

Continuous flow electrophoresis system experiments on shuttle flights STS-6 and STS-7

A space continuous flow electrophoresis system (CFES) was developed that would incorporate specific modifications to laboratory instruments to take advantage of weightlessness. The specific objectives were to use a model sample material at a high concentration to evaluate the continuous flow electrophoresis process in the CFES instrument and compare its separation resolution and sample throughput with related devices on Earth and to expand the basic knowledge of the limitations imposed by fluid flows and particle concentration effects on the electrophoresis process by careful design and evaluation of the space experiment. Hemoglobin and polysaccharide were selected as primary samples. The results from space show a large band spread of the high concentration of the single species of hemoglobin that was due to the mismatch of electrical conductivity between the sample and the buffer. On STS-7 the major objective was to evaluate the influence of the electrical properties of the sample constituents on the resolution of the CFES. As expected, the polystyrene latex microspheres dispersed in a solution with 3 times the electrical conductivity of the curtain buffer separated with a larger band spread than in the 2nd experiment.

A modular BLSS simulation model

A bioregenerative life support system (BLSS) for extraterrestrial use will be faced with coordination problems more acute than those in any ecosystem found on Earth. A related problem in BLSS design is providing an interface between the various life support processors, one that will allow for their coordination while still allowing for system expansion. A modular model is presented of a BLSS that interfaces system processors only with the material storage reservoirs, allowing those reservoirs to act as the principal buffers in the system and thus minimizing difficulties with processor coordination. The modular nature of the model allows independent development of the detailed submodels that exist within the model framework. Using this model, BLSS dynamics were investigated under normal conditions and under various failure modes. Partial and complete failures of various components, such as the waste processors or the plants themselves, drive transient responses in the model system, allowing the examination of the effectiveness of the system reservoirs as buffers. The results from simulations help to determine control strategies and BLSS design requirements. An evolved version could be used as an interactive control aid in a future BLSS.

Seal Technology for Liquid Oxygen (LOX) Turbopumps

Two types of advanced seals for liquid oxygen (LOX) turbopumps were investigated. One was a spiral-groove face seal whose function is to seal high-pressure LOX at the impeller end of the turbopump. The other was a floating-ring, Rayleigh-step, helium buffered seal used to prevent LOX ingress to the turbine side of the unit. For each seal type, two sizes were investigated (50 and 20 mm). A turbine-driven test rig was designed and manufactured, and a test program was completed on the 50 mm floating-ring, Rayleigh-step, helium buffered seal. Significant results were: vaporization in the flow path could cause failure by overheating; therefore, the spiral-groove pumping portion of the seal that provides the fluid film must circulate fluid without disruption if vaporization occurs in the sealing dam. This is successfully accomplished by a pressure-balanced spiral-groove concept that is described. The spiral-groove configuration is affected by turbulence in the fluid film and pressure drops due to fluid inertia at sudden contractions. The net results of these effects are deep grooves, large operating films, and high power loss when compared against seals operating with laminar films. Turbulence and inertia are induced by the high-density and low-viscosity characteristics of LOX. The program clearly pointed out the need to consider system environmental factors such as thermal and centrifugal distortions and rotor vibrations in the seal design.

AVIRIS onboard data handling and control

The timing and flow of detector and ancillary data for the Airborne Visible/Infrared imaging spectrometer (AVIRIS) are controlled within the instrument by its digital electronics assembly. In addition to providing detector and signal chain timing, the digital electronics receives, formats, and rate-buffers digitized science data; collects and formats ancillary (calibration and engineering) data; and merges both into a single tape record. Overall AVIRIS data handling is effected by a combination of dedicated digital electronics to control instrument timing, image data flow, and data rate buffering and a microcomputer programmed to handle real-time control of instrument mechanisms and the coordinated preparation of ancillary data.

Mathematical models of continuous flow electrophoresis: Electrophoresis technology

Two aspects of continuous flow electrophoresis were studied: (1) the structure of the flow field in continuous flow devices; and (2) the electrokinetic properties of suspended particles relevant to electrophoretic separations. Mathematical models were developed to describe flow structure and stability, with particular emphasis on effects due to buoyancy. To describe the fractionation of an arbitrary particulate sample by continuous flow electrophoresis, a general mathematical model was constructed. In this model, chamber dimensions, field strength, buffer composition, and other design variables can be altered at will to study their effects on resolution and throughput. All these mathematical models were implemented on a digital computer and the codes are available for general use. Experimental and theoretical work with particulate samples probed how particle mobility is related to buffer composition. It was found that ions on the surface of small particles are mobile, contrary to the widely accepted view. This influences particle mobility and suspension conductivity. A novel technique was used to measure the mobility of particles in concentrated suspensions.

Solar-powered alkali metal vapor lasers

The emission spectrum of the A(1 Sigma u +) - X(1 Sigma g +) band of Na2 has been recorded following excitation by monochromatic radiation in the region of X-A and X-B absorption. The spectral profile has been investigated as a function of excitation wavelength, sodium vapor temperature and buffer gas pressure. Additionally, gain measurements were made for the satellite of the A-X band as a function of the sodium vapor temperature and buffer gas pressure.

Sulfur speciation in hydrous experimental glasses of varying oxidation state - Results from measured wavelength shifts of sulfur X-rays

The focusing geometry of an electron microprobe has been used to measure the wavelength shifts of sulfur X-rays from hydrous experimental melts synthesized at oxygen fugacities that range from near the iron-wustite buffer to the magnetite-hermatite buffer. It is found that the proportion of dissolved sulfur which is present as sulfate increases with increasing oxygen fugacity. It is noted that in natural melts that have equilibrated at or below fayalite-magnetite-quartz values of +1, sulfur is probably present mainly as S(2-).

Nucleation and growth of CrSi2 on Si(111)

The nucleation and growth of CrSi2 on Si(111) by MBE and solid-phase epitaxy (SPE), was investigated using SEM and TEM observations of 2-mm-thick layers grown under a variety of conditions, including the use of a CoSi2 buffer. During growth, the wafers were monitored in situ using reflection high-energy electron diffraction. Island growth was observed, with islands found to nucleate with three epitaxial orientations. The morphology of CrSi2 islands was found to be affected by the degree and direction of substrate misorientation, the growth technique, and the use of CoSi2 buffer. However, the reconstruction of the Si surface does not appear to be an important factor in controlling SPE growth.

Electrohydrodynamic distortion of sample streams in continuous flow electrophoresis

Continuous flow electrophoresis experiments were carried out, using an electrolyte and a sample both made of aqueous solutions of phosphate buffer (with polystyrene latex added for visibility), to investigate causes of the sample spreading in this procedure. It is shown theoretically that an electric field perpendicular to a circular filament of conducting fluid surrounded by a fluid of different conductivity produces an electrohydrodynamic flow, which distorts the filament into an ellipse. Experimental results were found to be fully consistent with theretical predictions. It was found that the rate of distortion of the sample stream into a ribbon was proportional to the square of the applied voltage gradient. Furthermore, the orientation of the ribbon depends on the ratios of dielectric constant and electrical conductivity between the buffer and the sample.

Carrying Synchronous Voice Data On Asynchronous Networks

Buffers restore synchronism for internal use and permit asynchronism in external transmission. Proposed asynchronous local-area digital communication network (LAN) carries synchronous voice, data, or video signals, or non-real-time asynchronous data signals. Network uses double buffering scheme that reestablishes phase and frequency references at each node in network. Concept demonstrated in token-ring network operating at 80 Mb/s, pending development of equipment operating at planned data rate of 200 Mb/s. Technique generic and used with any LAN as long as protocol offers deterministic (or bonded) access delays and sufficient capacity.

Scan line graphics generation on the massively parallel processor

Described here is how researchers implemented a scan line graphics generation algorithm on the Massively Parallel Processor (MPP). Pixels are computed in parallel and their results are applied to the Z buffer in large groups. To perform pixel value calculations, facilitate load balancing across the processors and apply the results to the Z buffer efficiently in parallel requires special virtual routing (sort computation) techniques developed by the author especially for use on single-instruction multiple-data (SIMD) architectures.

Data acquisition for a real time fault monitoring and diagnosis knowledge-based system for space power system

The design and implementation of the real-time data acquisition and processing system employed in the AMPERES project is described, including effective data structures for efficient storage and flexible manipulation of the data by the knowledge-based system (KBS), the interprocess communication mechanism required between the data acquisition system and the KBS, and the appropriate data acquisition protocols for collecting data from the sensors. Sensor data are categorized as critical or noncritical data on the basis of the inherent frequencies of the signals and the diagnostic requirements reflected in their values. The critical data set contains 30 analog values and 42 digital values and is collected every 10 ms. The noncritical data set contains 240 analog values and is collected every second. The collected critical and noncritical data are stored in separate circular buffers. Buffers are created in shared memory to enable other processes, i.e., the fault monitoring and diagnosis process and the user interface process, to freely access the data sets.

Input/output behavior of supercomputing applications

The collection and analysis of supercomputer I/O traces and their use in a collection of buffering and caching simulations are described. This serves two purposes. First, it gives a model of how individual applications running on supercomputers request file system I/O, allowing system designer to optimize I/O hardware and file system algorithms to that model. Second, the buffering simulations show what resources are needed to maximize the CPU utilization of a supercomputer given a very bursty I/O request rate. By using read-ahead and write-behind in a large solid stated disk, one or two applications were sufficient to fully utilize a Cray Y-MP CPU.

Actively controlled shaft seals for aerospace applications

The main objective is to determine the feasibility of utilizing controllable mechanical seals for aerospace applications. A potential application was selected as a demonstration case: the buffer gas seal in a LOX (liquid oxygen) turbopump. Currently, floating ring seals are used in this application. Their replacement with controllable mechanical seals would result in substantially reduced leakage rates. This would reduce the required amount of stored buffer gas, and therefore increase the vehicle payload. For such an application, a suitable controllable mechanical seal was designed and analyzed.
