Cryptic cycling by electroactive bacterioplankton in Trout Bog Lake

The potential for extracellular electron transfer (EET) is a prevailing genomic feature of humic lake bacterioplankton. However, there has been little evidence for the substantial ecological contribution predicted by genetics. We hypothesized that anoxygenic phototrophic electrotrophs and accompanying heterotrophic electrogens cycle dissolved organic matter (DOM) between oxidized and reduced states. We predicted that such bacterioplankton would exhibit diel-scale oscillations due to the light dependency of photosynthesis. Using Trout Bog Lake in Wisconsin, USA, as our model ecosystem, we profiled the water column with depth-discrete metagenomic, physiochemical, and electrochemical analyses. We observed variation in oxidation reduction potential (ORP) in response to sunlight, initiating at depths populated by anoxygenic phototrophs with EET genes. We developed an automated buoy to measure electric current flow between many pairs of electrodes simultaneously, observing correlation in electron consumption to sunlight. Our results, combined with published metatranscriptomic analysis, indicate the occurrence of electron cycling between phototrophic oxidation (electrotrophic metabolism) by Chlorobium and anaerobic respiration (electrogenic metabolism) by Geothrix, involving DOM. We also repeatedly observed gradual seasonal increases in hypolimnion ORP throughout summer. These diel and seasonal patterns imply that electroactive DOM mediates the ecology of electroactive bacteria in lakes, controlling humic lake methane emissions.IMPORTANCEWe investigated the physical, chemical, and redox characteristics of a bog lake and electrodes hung therein to test the hypothesis that dissolved organic matter is being cycled between oxidized and reduced states by electroactive bacterioplankton powered by phototrophy. To do so, we performed field-based analyses on multiple timescales using both established and novel instrumentation. We paired these analyses with recently developed bioinformatics pipelines for metagenomics data to investigate genes that enable electroactive metabolism and accompanying metabolisms. Our results are consistent with our hypothesis and yet upend some of our other expectations. Our findings have implications for understanding greenhouse gas emissions from lakes, including electroactivity as an integral part of lake metabolism throughout more of the anoxic parts of lakes and for a longer portion of the summer than expected. Our results also give a sense of what electroactivity occurs at given depths and provide a strong basis for future studies.

A Multiyear Dataset of SSM/I-Derived Global Ocean Surface Turbulent Fluxes

The surface turbulent fluxes of momentum, latent heat, and sensible heat over global oceans are essential to weather, climate and ocean problems. Evaporation is a key component of the hydrological cycle and the surface heat budget, while the wind stress is the major forcing for driving the oceanic circulation. The global air-sea fluxes of momentum, latent and sensible heat, radiation, and freshwater (precipitation-evaporation) are the forcing for driving oceanic circulation and, hence, are essential for understanding the general circulation of global oceans. The global air-sea fluxes are required for driving ocean models and validating coupled ocean-atmosphere global models. We have produced a 7.5-year (July 1987-December 1994) dataset of daily surface turbulent fluxes over the global oceans from the Special Sensor microwave/Imager (SSM/I) data. Daily turbulent fluxes were derived from daily data of SSM/I surface winds and specific humidity, National Centers for Environmental Prediction (NCEP) sea surface temperatures, and European Centre for Medium-Range Weather Forecasts (ECMWF) air-sea temperature differences, using a stability-dependent bulk scheme. The retrieved instantaneous surface air humidity (with a 25-km resolution) validated well with that of the collocated radiosonde observations over the global oceans. Furthermore, the retrieved daily wind stresses and latent heat fluxes were found to agree well with that of the in situ measurements (IMET buoy, RV Moana Wave, and RV Wecoma) in the western Pacific warm pool during the TOGA COARE intensive observing period (November 1992-February 1993). The global distributions of 1988-94 seasonal-mean turbulent fluxes will be presented. In addition, the global distributions of 1990-93 annual-means turbulent fluxes and input variables will be compared with those of UWM/COADS covering the same period. The latter is based on the COADS (comprehensive ocean-atmosphere data set) and is recognized to be one of the best climatological analyses of fluxes derived from ship observations.

Comparison of buoy-mounted 74-kHz acoustic Doppler current profilers with vector-measuring current meters

In December 1991, the National Data Buoy Center (NDBC) deployed two meteorological buoys in the Southern California Bight on a transect between San Diego and San Clemente Island. Each buoy consisted of a 10-m discus hull instrumented to measure a suite of meteorological parameters, and, for the first time in the NDBC buoy program, acoustic Doppler current profilers (ADCPs) were included to gather hourly current profiles beneath the two buoys. Moorings instrumented with seven vector-measuring current meters (VMCMs) were deployed adjacent to the NDBC buoys for several months and provided current observations for comparison with the ADCP measurements. When wave-induced buoy motion is not overly large, the observations of horizontal current made by the ADCP and the VMCM are highly correlated. Time series of differences between ADCP and VMCM measurements are characterized by a mean difference (bias error) of about 0.01 m/s and standard deviation of about 0.035 m/s for 1-h observations. Estimates of current spectra from ADCP and VMCM records suggest that the ADCP system can be characterized by a white noise level of 2 x 10(exp -3) sq m/sq s/cph. However, when the in situ environment is such that large surface waves are present (including breaking waves and whitecaps), erroneous current values are usually reported by the ADCP. Mean values of vertical velocities reported by the ADCP appear to be much larger than what could be physically expected and are therefore deemed unreliable.

Mid-Atlantic Bight Wave Hindcast To Support DOE Lidar Buoy Deployments: Model Validation

This study presents a shelf scale wave hindcast for the Mid-Atlantic Bight to provide accurate wave data to complement the measured data gathered by two DOE lidar buoy deployments off the coasts of Virginia and New Jersey. PNNL developed a ~2 km resolution model based on Wavewatch III and forced with analyzed winds and ocean currents; and executed a four-year hindcast for the period from January 2014 to December 2017. The model results are compared well against 16 wave-measuring buoys and data derived from six satellite-borne radar altimeters. In addition, the model results for two storm events that generated large waves, Hurricane Hermine and the January 2016 Blizzard, are discussed in detail.
