BBO-10203 inhibits tumor growth without inducing hyperglycemia by blocking RAS-PI3Kα interaction

BBO-10203 is an orally available drug that covalently and specifically binds to the rat sarcoma (RAS)–binding domain of phosphoinositide 3-kinase α (PI3Kα), preventing its activation by HRAS, NRAS, and KRAS. Here, it inhibited PI3Kα activation in tumors with oncogenic mutations in KRAS or PIK3CA and in tumors with human epidermal growth factor receptor 2 (HER2) amplification or overexpression. In preclinical models, BBO-10203 caused significant tumor growth inhibition across multiple tumor types and showed enhanced efficacy in combination with inhibitors of cyclin-dependent kinase 4/6 (CDK4/6), estrogen receptor (ER), HER2, and KRAS-G12C mutant, including in tumors harboring mutations in Kelch-like ECH-associated protein 1 (KEAP1) and serine/threonine kinase 11 (STK11). Notably, these antitumor effects occurred without inducing hyperglycemia, because insulin signaling does not depend on RAS-mediated PI3Kα activation to promote glucose uptake.

Multi-scale transport in the DIII-D ITER baseline scenario with direct electron heating and projection to ITER

Multi-scale fluctuations measured by turbulence diagnostics spanning long and short wavelength spatial scales impact energy confinement and the scale-lengths of plasma kinetic profiles in the DIII-D ITER baseline scenario with direct electron heating. Contrasting discharge phases with ECH + neutral beam injection (NBI) and NBI only at similar rotation reveal higher energy confinement and lower fluctuations when only NBI heating is used. Modeling of the core transport with TGYRO using the TGLF turbulent transport model and NEO neoclassical transport reproduces the experimental profile changes upon application of direct electron heating and indicates that multi-scale transport mechanisms are responsible for changes in the temperature and density profiles. Intermediate and high-k fluctuations appear responsible for the enhanced electron thermal flux, and intermediate-k electron modes produce an inward particle pinch that increases the inverse density scale length. Projection to ITER is performed with TGLF and indicates a density profile that has a finite scale length due to intermediate-k electron modes at low collisionality and increases the fusion gain. For a range of E×B shear, the dominant mechanism that increases fusion performance is suppression of outward low-k particle flux and increased density peaking.

Density wavenumber spectrum measurements, synthetic diagnostic development, and tests of quasilinear turbulence modeling in the core of electron-heated DIII-D H-mode plasmas

Abstract Measurements of the turbulent density wavenumber spectrum, δ n ˆ e ( k ⊥ ) , using the Doppler Back-Scattering (DBS) diagnostic are reported from DIII-D H-mode plasmas with electron cyclotron heating as the only auxiliary heating method. These electron-heated plasmas have low collisionality, ν e ∗ < 1 , T e / T i > 1 , and zero injected torque—a regime expected to be relevant for future fusion devices. We probe density fluctuations in the core ( ρ ≈ 0.7) over a broad wavenumber range, 0.5 ⩽ k ⊥ ⩽ 16 cm −1 ( 0.1 ⩽ k ⊥ ρ s ⩽ 5 ), to characterize plasma instabilities and compare with theoretical predictions. We present a novel synthetic DBS diagnostic to relate the back-scattered power spectrum, P s ( k ⊥ ) —which is directly measured by DBS—to the underlying electron density fluctuation spectrum, δ n ˆ e ( k ⊥ ) . The synthetic DBS P s ( k ⊥ ) spectrum is calculated by combining the SCOTTY beam-tracing code with a model δ n ˆ e ( k ⊥ ) predicted either analytically or numerically. In this work we use the quasi-linear code Trapped Gyro-Landau Fluid (TGLF) to approximate the δ n ˆ e ( k ⊥ ) spectrum. We find that TGLF, using the experimental profiles, is capable of closely reproducing the DBS measurements. Both the DBS measurements and the TGLF-DBS synthetic diagnostic show a wavenumber spectrum with variable decay. The measurements show weak decay ( k −0.6 ) for k < 3.5 cm −1 , with k −2.6 at intermediate- k ( 3.5 ⩽ k ⩽ 8.5 cm −1 ), and rapid decay ( k −9.4 ) for k > 8.5 cm −1 . Scans of physics parameters using TGLF suggest that the normalized ∇ T e scale-length, R / L T e , is an important factor for distinguishing microturbulence regimes in these plasmas. A combination of DBS observations and TGLF simulations indicate that fluctuations remain peaked at ITG-scales (low k ) while R / L T e -driven TEM/ETG-type modes (intermediate/high k ) are marginally sub-dominant.

Initial Results from High-Field-Side Transient CHI Start-Up on QUEST

Transient coaxial helicity injection (t-CHI) current start-up using a new design simple electrode configuration has been implemented on the QUEST. Discharges injected from the low field side (LFS) and from the high field side (HFS) were examined. Compared to the LFS injection case, the HFS injection has the advantages of providing access to a higher toroidal field and better controlling the location of the injector flux footprint location. Although the present PF coils on QUEST are not well positioned to form the injector flux on the HFS injector region and there has been a frequent occurrence of the spurious arcs, known as absorber arcs, HFS injection has shown flux evolution in a shape that is suitable for the formation of closed flux surfaces. The discharges were improved by installing an in-vessel-coil and adding a new cylindrical electrode to the existing CHI electrode. Overall, the results show that the new cylindrical electrode allowed the flux to evolve stably while allowing both the inner and the outer injector flux footprint to remain in the vicinity of the cylindrical electrode. This configuration which inherently generates a narrow injector flux footprint width resulted in discharges that strongly suggested the persistence of the CHI generated plasma after the injector current was reduced to zero. These studies have informed us of the need to improve the CHI gas injection system so that the absorber arcs could be better controlled in the HFS injection configuration.
