Diffraction-quality, ultraflexible protein single crystals engineered with DNA

DNA-functionalized colloidal nanoparticles assemble through flexible, nanoscale DNA hybridization interactions that limit atomic-level structural order. Here, we report a valence-centric strategy that enables DNA-bonded, protein single crystals with unconventional mechanical properties. An octameric enzyme, glutarate L-2-hydroxylase, was site- and number-selectively conjugated with eight self-complementary single-stranded DNA, yielding octavalent molecular bonds. The resulting conjugate assembled into the designed body-centered tetragonal crystals that diffracted to 1.42- to 2.61-angstrom resolution, with contacts mediated by B-form DNA helices spanning 17 to 25 angstroms. Increasing oligonucleotide length induces anisotropic lattice expansion while preserving atomic periodicity, even with partial DNA occupancy. Mechanistic studies suggest that the dynamic motion of unhybridized DNA facilitates crystallization, analogous to fluctuating electron clouds in atomic bonding. Compared with native protein crystals, DNA-hybridized crystals are 23-fold softer. These results challenge the assumption that flexibility is incompatible with structural order and establish a programmable framework for biomolecular crystallization and nanomaterials engineering with atomic precision.

Ground-Based Microwave Measurements of ClO and O3 Over Scott Base, Antarctica

We present measurements from a new microwave instrument that has been deployed at Scott Base, Antarctica, primarily to measure ClO. The new instrument allows for the extension of the time series of lower stratospheric ClO measurements begun in 1996 at this site, while some of the technical improvements offer the possibility of making additional measurements. We show measurements of ClO and O 3 in both the upper and lower stratosphere near both the fall and spring equinox and document the diurnal variations of these measurements throughout the season. The ground-based measurements are compared with Aura Microwave Limb Sounder (MLS) measurements, which are available twice per day, and with a NASA Goddard 2D model (GSFC2D) simulation. We show that, in agreement with the GSFC2D model diurnal variations, upper stratospheric ClO mixing ratios gradually increase throughout the daylight hours, while in the lower stratospheric austral spring, when heterogeneous chemistry associated with polar stratospheric clouds occurs, daily maximum ClO mixing ratios of near 2 ppbv occur near noon. We also show measurements of stratospheric O 3 , primarily in the upper stratosphere, which, while exhibiting a low bias of ∼15%–20% relative to Aura MLS, show temporal variations in good agreement with MLS.

The 1996 Workshop was held on three consecutive days and was divided into five sessions. The first day consisted of a General Primary Battery Session and a Nickel-Hydrogen Battery On-Orbit Reconditioning Experience Focused Session. The second day consisted of a Nickel-Hydrogen Session and a Nickel-Cadmium Session. The third and final day was devoted to an Other Secondary Technologies Session which covered sodium-sulfur, nickel-zinc, nickel-metal hydride, and lithium ion technologies.

MSFC Thermal Testing Lessons Learned Course. Section 3: Lessons Learned from Protoflight StarBurst Instrument Thermal Vacuum Test

MSFC thermal engineers will be presenting lessons learned while preparing and conducting thermal tests. This course will cover practical real world examples of thermal chamber and vacuum chamber testing from engineering development units to system level testing on flight hardware. Special topics include large scale testing preparation, requirement tailoring from parent documents, and cryogenic development testing.

2005 NASA Seal/Secondary Air System Workshop

The 2005 NASA Seal/Secondary Air System workshop covered the following topics: (i) Overview of NASA’s new Exploration Initiative program aimed at exploring the Moon, Mars, and beyond; (ii) Overview of the NASA-sponsored Propulsion 21 Project; (iii) Overview of NASA Glenn’s seal project aimed at developing advanced seals for NASA’s turbomachinery, space, and reentry vehicle needs; (iv) Reviews of NASA prime contractor, vendor, and university advanced sealing concepts including tip clearance control, test results, experimental facilities, and numerical predictions; and (v) Reviews of material development programs relevant to advanced seals development. Turbine engine studies have shown that reducing high-pressure turbine (HPT) blade tip clearances will reduce fuel burn, lower emissions, retain exhaust gas temperature margin, and increase range. Several organizations presented development efforts aimed at developing faster clearance control systems and associated technology to meet future engine needs. The workshop also covered several programs NASA is funding to develop technologies for the Exploration Initiative and advanced reusable space vehicle technologies. NASA plans on developing an advanced docking and berthing system that would permit any vehicle to dock to any on-orbit station or vehicle. Seal technical challenges (including space environments, temperature variation, and seal-on-seal operation) as well as plans to develop the necessary “androgynous” seal technologies were reviewed. Researchers also reviewed tests completed for the shuttle main landing gear door seals.

Generalizable Web User Interface for Scalable and Streamlined Deployment of Building Energy Management Systems in Small and Medium-Sized Commercial Buildings

Small and medium-sized commercial buildings (SMCBs) comprise 94% of US commercial buildings yet face significant barriers to implementing building energy management systems despite advances in smart device technology. Existing solutions present critical limitations: cloud-based API solutions simplify deployment but create vendor lock-in constraints; commercial integrated software solutions ensure compatibility via standardized protocols but require substantial cost and technical expertise; open-source IoT platforms offer cost-effective vendor independence but provide insufficient standardized protocol support for commercial building automation. This research presents a generalizable web user interface framework that bridges the gap between evolving smart device capabilities and lagging software infrastructure for SMCBs. The proposed system integrates VOLTTRON open-source middleware with an automated configuration converter that transforms unified specifications written in YAML, a human-readable data-serialization format, into system-specific files, streamlining manual setup processes. The vendor-agnostic architecture supports industry-standard protocols (BACnet and Modbus) and semantic building models while providing adaptive web interfaces that dynamically adjust to various building configurations. Demonstrations through simulation-based testing and a field deployment show automatic interface adaptation across heterogeneous HVAC systems and multizone monitoring. The automated configuration converter also substantially reduces labor-intensive setup.

Update of the IUE battery in-flight performance

The in-flight performance data of two 17-cell, 6-ampere-hour nickel cadmium spacecraft batteries are presented covering 22 months of operation. Fluctuations in the battery voltage and the battery temperature are presented for spacecraft movement throughout a beta range of 0 to 130 deg. The battery discharge voltages during the peak eclipse seasons, daily seasons, and daily eclipse periods are noted. Finally, the spacecraft data are compared to data from a 6-ampere-hour test pack and test flight data.

HRMS Sky Survey Wideband Feed System Design for DSS 24 Beam Waveguide Antenna

The High-Resolution Microwave Survey (HRMS) Sky Survey project will be implemented on the DSS 24 beam waveguide (BWG) antenna over the frequency range of 2.86 to 10 GHz. Two wideband, ring-loaded, corrugated feed horns were designed to cover this range. The horns match the frequency-dependent gain requirements for the DSS 24 BWG system. The performance of the feed horns and the calculated system performance of DSS 24 are presented.
