Elucidating Phosphate and Cadmium Cosorption Mechanisms on Mineral Surfaces with Direct Spectroscopic and Modeling Evidence

The simultaneous sorption of cations and anions at the mineral–water interface can substantially alter their individual sorption characteristics; however, this phenomenon lacks a mechanistic understanding. Our study provides direct spectroscopic and modeling evidence of the molecular cosorption mechanisms of the cadmium ion (Cd 2+ ) and phosphate (P) on goethite and layered manganese (Mn) oxide of birnessite, through in situ attenuated total reflection Fourier-transform infrared (ATR-FTIR), P K-edge X-ray absorption near-edge structure (XANES) spectroscopy, and surface complexation modeling. Phosphate synergistically cosorbed with Cd on goethite predominantly through P-bridged ternary complexes (≡Fe–P–Cd) and electrostatic interactions at wide pH conditions. Likewise, P and Cd exhibited synergistic cosorption on birnessite by forming P-bridged ternary complexes (≡Mn–P–Cd) and weak competitive sorption at the layer edge sites. As pH and Cd loading increased, the surface P species transitioned from a binary complex to a ternary complex and/or Cd 3 (PO 4 ) 2 precipitate for both goethite and birnessite. Compared to that in solution at pH 8, the formation of Cd 3 (PO 4 ) 2 was inhibited by the presence of goethite and birnessite, ascribed to the specific adsorption of P and Cd, more pronounced in birnessite due to the stronger sorption of Cd at its vacant sites. Finally, the discovered cosorption mechanisms of P and Cd have important implications for understanding and predicting their mobility and availability in Cd-contaminated settings.

Charge density waves and pinning by lattice anisotropy in 214 cuprates

The detection of static charge density waves (CDWs) in La 2-x ⁢Sr x ⁢CuO 4 (LSCO) with x ~ 0.12 at relatively high temperatures has raised the question of what lattice feature pins the CDWs. Some recent structural studies have concluded that some form of monoclinic distortion, indicated by the appearance of certain weak Bragg peaks (type M peaks) at otherwise forbidden positions, are responsible for CDW pinning. Here, as a test of this idea, we present neutron diffraction results for a single crystal of La 2-x ⁢Ba x CuO 4 (LBCO) with x = 1/8, which is known to undergo two structural transitions on cooling, from high-temperature tetragonal (HTT) to low-temperature orthorhombic (LTO) near 240 K, involving a collective tilt pattern of the corner-sharing CuO 6 octahedra, and from LTO to low-temperature tetragonal (LTT) near 56 K, involving a new tilt pattern and the appearance of intensity at peaks of type T. We observe both type M and type T peaks in the LTT phase, while the type M peaks (but not type T) are still present in the LTO phase. Given that CDW order is observed only in the LTT phase of LBCO, it is apparent that the in-plane Cu-O bond anisotropy associated with the octahedral tilt pattern is responsible for charge pinning. We point out that evidence for a similar, but weaker, bond anisotropy has been observed previously in LSCO and should be responsible for CDW pinning there. In the case of LBCO, the monoclinic distortion may help to explain previously reported magneto-optical evidence for gyrotropic order.

Fuzzy spheres in stringy matrix models: quantifying chaos in a mixed phase space

We consider a truncation of the BMN matrix model to a configuration of two fuzzy spheres, described by two coupled non-linear oscillators dependent on the mass parameter μ. The classical phase diagram of the system generically (μ ≠ 0) contains three equilibrium points: two centers and a center-saddle; as μ → 0 the system exhibits a pitchfork bifurcation. We demonstrate that the system is exactly integrable in quadratures for μ = 0, while for very large values of μ, it approaches another integrable point characterized by two harmonic oscillators. The classical phase space is mixed, containing both integrable islands and chaotic regions, as evidenced by the classical Lyapunov spectrum. At the quantum level, we explore indicators of early and late time chaos. The eigenvalue spacing is best described by a Brody distribution, which interpolates between Poisson and Wigner distributions; it dovetails, at the quantum level, the classical results and reemphasizes the notion that the quantum system is mixed. We also study the spectral form factor and the quantum Lyapunov exponent, as defined by out-of-time-ordered correlators. These two indicators of quantum chaos exhibit weak correlations with the Brody distribution. We speculate that the behavior of the system as μ → 0 dominates the spectral form factor and the quantum Lyapunov exponent, making these indicators of quantum chaos less effective in the context of a mixed phase space.

What is the Role of Relative Humidity on Conductivity in Polymer Electrolytes?

Ion transport properties in polymer electrolytes have been widely studied under rigorously dry and highly water-swollen conditions. However, the transition between these extremes, i.e., the low hydration regime, is still poorly understood at the molecular level and relevant to applications ranging from battery electrolytes to electrophoretic separations. Here, in this study, we apply atomistic molecular dynamics simulations to probe the role of hydration on ion mobilities and conductivities in LiTFSI-doped polyethers at low water content (less than 10% water by volume), which corresponds to 0−80% relative humidity conditions. With increasing water content, Li + ions exhibit two distinct regimes in their mobilities. At low water content, the majority of the Li + ions are weakly hydrated (two or fewer water molecules per Li + ion), maintaining strong interactions with their polymer solvation cage, and therefore exhibit only a weak increase in mobility relative to dry conditions. As water content increases, Li + ions form complete hydration shells, residing within water-rich domains that promote faster mobilities. Further, Li + mobilities increase more significantly in polymer electrolytes composed of more hydrophilic polymers and higher salt concentrations due to the formation of larger water-rich domains. In contrast, TFSI − has comparatively weak interactions with both the polymer and water, exhibiting a monotonic increase in mobility as a function of water content. Overall, these results help clarify the molecular mechanisms underlying ionic mobilities and conductivities in polymer electrolytes at low water content.

Conversion of real-world aluminum scrap streams into high-performance Al–Mg–Si–Cu automotive alloys using shear assisted processing and extrusion

The conversion of post-consumer aluminum (Al) scrap into usable Al alloys without adding primary Al is challenging because of excess impurities. In this work, >99% post-consumer Twitch, used beverage cans (UBCs), and remelt scrap ingots (RSIs) were used as feedstock materials. As-cast and solution heat-treated feedstock billets were extruded using Shear Assisted Processing and Extrusion (ShAPE) at ~510°C, followed by press quenching. To explore the development of the microstructure, texture, and underlying mechanisms and how they contribute to the overall strengthening in as-extruded and artificially aged samples, scanning electron microscopy (SEM) and electron backscatter diffraction (EBSD) were used to collect microstructure and texture data. The enhanced strength and ductility were corroborated with the microstructural features and crystallographic texture. Simple shear $\textrm{A}/\bar{\textrm{A}}$, $\textrm{A}_1^*/\textrm{A} _2^*$ texture components along with weak $\textrm{C}$ and $\textrm{B}/\bar{\textrm{B}}$ texture components were formed during extrusion; the texture was strengthened after heat treatment. The refined second-phase particles helped to retain the deformed microstructure and texture. The contributions of dislocation and precipitate strengthening were maximized when billets were solution-heat-treated prior to extrusion. This is attributed to the formation of effective supersaturated solid solutions during the ShAPE process, which precipitate out during the peak age treatment. Overall, the highest yield strength of 305 MPa, ultimate tensile strength of 350 MPa, and elongation of 12% were achieved in artificially aged samples, which are comparable to those of Al 6082-T6.

Primordial black hole dark matter: A quantitative parameter sensitivity comparison across formation mechanisms and particle candidates

Primordial black holes (PBHs) in the asteroid-mass window ( 10 17 – 10 22 g ) can account for all of the dark matter without violating any observational constraint, yet are routinely dismissed as fine-tuned. I put that dismissal to the test by applying three complementary sensitivity measures uniformly across a broad landscape: three noninflationary PBH production mechanisms, six classes of inflationary PBH models, and seven particle dark matter benchmarks, all evaluated against the same observable target. Three distinct naturalness universality classes emerge, determined entirely by the analytic structure of the abundance map rather than by the nature of the dark matter candidate. Biased-domain-wall PBHs, in their least model-dependent (free- V b ) form, have the same low sensitivity, Δ = 4.5 , as off-resonance weakly interacting massive particles and freeze-in particles ( Δ = 2 ), a sensitivity that, because it is constant over the entire parameter space of the construction, also coincides trivially with its own Wilson-normalized average within that parameter space (Section Definition and conventions), an equivalence that concerns only the space over which Δ is computed and is not a naturalness statement about the construction as a whole; a further reduction to Δ = 2 is possible only under the additional, independently motivated but not required, assumption that the domain-wall bias is generated by Planck-suppressed operators; early matter-domination PBHs occupy an intermediate tier alongside coannihilating weakly interacting massive particles (WIMPs), unified by a structural identity in which the sensitivity measure equals the logarithm of the ratio of the formation scale to the matter–radiation equality scale; first-order phase transition PBHs, once the more accurate super-exponential collapse probability is used in place of the single-exponential approximation, instead belong to the same highly sensitive tier as resonant WIMP annihilation and single-field inflationary collapse, for a structurally distinct reason; single-field ultraslow-roll inflationary collapse is severely tuned for a distinct reason: a double exponential in which the power spectrum amplitude is itself exponentially sensitive to the inflaton potential coefficients, on top of the exponential collapse sensitivity of the abundance map. My main conclusion is that the claim that PBH dark matter is generically fine-tuned conflates the worst case with a landscape spanning every naturalness tier. The Barbieri-Giudice sensitivity computed here and the Wilson-normalized measure of Iovino and Riotto answer distinct and mutually consistent questions about the same construction, a distinction I clarify within the two-layer decomposition.

Synthetic tensor gauge fields

Synthetic gauge fields have provided physicists with a unique tool to explore a wide range of fundamentally important phenomena. However, most experiments have focused on synthetic vector gauge fields. The very rich physics brought about by coupling tensor gauge fields to fracton phases of matter remains unexplored in laboratories. Here, we propose schemes to realize synthetic tensor gauge fields that address dipoles instead of single particles. A lattice tilted by a strong linear potential and a weak quadratic potential yields a rank-2 electric field for a dipole formed by a particle-hole pair. Such a rank-2 electric field leads to a new type of Bloch oscillations, which modulates the quadrupole moment and preserves the dipole moment of the system. In higher dimensions, the interplay between interactions and vector gauge potentials imprints a phase to the ring-exchange interaction and thus generates synthetic tensor gauge fields. Such tensor gauge fields make it possible to realize a dipolar Harper-Hofstadter model in laboratories. The resultant dipolar Chern insulators feature chiral edge currents of dipoles in the absence of net charge currents. Published by the American Physical Society 2025

Constraining the Stellar-to-Halo Mass Relation with Galaxy Clustering and Weak Lensing from DES Year 3 Data

We develop a framework to study the relation between the stellar mass of a galaxy and the total mass of its host dark matter halo using galaxy clustering and galaxy-galaxy lensing measurements. We model a wide range of scales, roughly from $\sim 100 \; {\rm kpc}$ to $\sim 100 \; {\rm Mpc}$, using a theoretical framework based on the Halo Occupation Distribution and data from Year 3 of the Dark Energy Survey (DES) dataset. The new advances of this work include: 1) the generation and validation of a new stellar mass-selected galaxy sample in the range of $\log M_\star/M_\odot \sim 9.6$ to $\sim 11.5$; 2) the joint-modeling framework of galaxy clustering and galaxy-galaxy lensing that is able to describe our stellar mass-selected sample deep into the 1-halo regime; and 3) stellar-to-halo mass relation (SHMR) constraints from this dataset. In general, our SHMR constraints agree well with existing literature with various weak lensing measurements. We constrain the free parameters in the SHMR functional form $\log M_\star (M_h) = \log(εM_1) + f\left[ \log\left( M_h / M_1 \right) \right] - f(0)$, with $f(x) \equiv -\log(10^{αx}+1) + δ[\log(1+\exp(x))]^γ/ [1+\exp(10^{-x})]$, to be $\log M_1 = 11.506^{+0.325}_{-0.404}$, $\log ε= -1.632^{+0.306}_{-0.181}$, $α= -1.638^{+0.108}_{-0.099}$, $γ= 0.596^{+0.251}_{-0.210}$ and $δ= 3.810^{+2.045}_{-1.811}$. The inferred average satellite fraction is within $\sim 5-35\%$ for our fiducial results and we do not see any clear trends with redshift or stellar mass. Furthermore, we find that the inferred average galaxy bias values follow the generally expected trends with stellar mass and redshift. Our study is the first SHMR in DES in this mass range, and we expect the stellar mass sample to be of general interest for other science cases.

Fast Growth of Ga2O3 on Highly Offcut (100)-Oriented Substrates

Beta-Ga2O3 has emerged as a leading candidate for next-generation power electronics, radio frequency (RF) switches, and extreme environment electronics due to a wide band gap (4.6 - 4.9 eV), high dopability (approximately 40 meV activation energy for an isolated silicon donor), and melt growth characteristics resulting in commercially available 4-inch substrates and commercial demonstrations of 6-inch substrates by multiple techniques. The (100) surface of Ga2O3 is highly desirable from a device and epitaxy standpoint - bulk growth of (100) material is more scalable than (010), the surface is nearly lattice-matched to p-type partner NiO, and Al2O3 incorporates at higher concentrations without phase separation. More importantly, the impact ionization coefficients along the [100] direction are low, leading to the highest possible critical fields. This is advantageous compared to the current state of the art, (001), due to reduced surface defects and increased possible breakdown voltage. However, the epitaxial growth rate on (100) surfaces is less than 10% of other faces due to weak bonding and favorable desorption, and on-axis (100) growth easily forms twin domains. Recent demonstrations have shown growth rate improvements from 0.4 nm/min to 1.5 nm/min by growing on (100) wafers that are offcut 6 degrees in the -c direction. These films show step-flow growth from (20-1) step-edges and high electron mobility due to suppressed twins. Despite these exciting results, offcuts greater than 6 degrees have not been explored due to the waste associated with grinding and polishing large offcuts. In this talk we will discuss the molecular beam epitaxy (MBE) growth and properties of Beta-Ga2O3 grown on (100) substrates offcut in the -c direction up to 13.4 degrees. These large offcuts are enabled by edge-fed film-defined growth (EFG) where the offcut is grown into the surface by pulling the crystal through the EFG die with the seed crystal rotated by the desired offcut angle. We will demonstrate that 13.4 degrees offcut substrates still exhibit a terraced (100) surface, and that a >10x increase (>5 nm/min) in growth rate is achieved. As previously reported on lower offcuts, we observe reversal of substrate twin domains around the (001) direction at the substrate-epilayer interface. We will discuss electrical properties including record-low (by MBE) unintentional doping densities of < 5E15 cm-3 and critical breakdown field in Schottky barrier diodes comparable with state-of-the-art (001) Ga2O3 without edge termination.

Reassessment of Dose Contributions from the Paducah Cask Special Waste Form Buried in the E-Area Low-Level Waste Facility

During a recent revision to the groundwater and inadvertent human intruder screening analysis report (Aleman and Hamm, 2023), weaknesses within the screening logic were observed resulting in the addition of U-235 being brought back into the list of radionuclides requiring inventory limits within the Paducah Cask special waste form (SWF). In addition, while reviewing the earlier vadose zone transport analysis efforts, an inconsistency was observed in the initial U-235P inventory that was supplied to the Consolidated Waste Tracking System (CWTS) back in 2009.

Fractional AC Josephson effect in a topological insulator proximitized by a self-formed superconductor

A lateral Josephson junction in which the surface of a 3D topological insulator serves as the weak link should support topologically protected excitations related to Majorana fermions. The resulting 4π-periodic current-phase relationship could be detected under high-frequency excitation by the suppression of odd Shapiro steps. Here, in this study, we demonstrate such devices through the self-formation of a Pd-Te superconducting layer from a telluride topological insulator, and observe suppressed first and third Shapiro steps. Other devices, including those where the Pd-Te layer is bolstered by an additional Al layer, show no suppression of Shapiro steps, a difference supported by simulations. Though we rule out the known trivial causes of suppressed Shapiro steps in our devices, we nevertheless argue that corroborating measurements and disorder-aware theoretical descriptions of these systems are needed before confidently claiming the observation of Majorana states.

Crystal structure and polymorphic forms of auranofin revisited

Auranofin, initially developed as a treatment for rheumatoid arthritis, is currently under extensive investigation as a potential drug for various conditions, including cancer, bacterial infections, and parasitic infections. The compound is a known inhibitor of thioredoxin reductase (TXNRD1) and related selenoproteins. Although preliminary studies on the auranofin crystal polymorphism exist, and a low-quality crystal structure has been reported, a comprehensive crystallographic characterization remains unexplored. Utilizing X-ray crystallography techniques, we conducted detailed structural analysis of auranofin and compared our findings with related organogold compounds. Implementation of Hirshfeld atom refinement (HAR) enabled a more accurate hydrogen atom positioning in the structure. The crystal packing reveals a layered arrangement stabilised by numerous weak hydrogen bonds and dispersive interactions. Notably, our attempts to reproduce the previously reported polymorphic form of auranofin, purportedly more water-soluble, were unsuccessful despite following published protocols. To our knowledge, this is the first study describing a “disappearing polymorph” phenomenon of any pharmaceutically relevant transition metal coordination compound. Our findings may have significant implications for medicinal chemistry and pharmacology of coordination complexes, suggesting the need for systematic revision of historical crystallographic data in this field.

Supernova Axions Convert to Gamma Rays in Magnetic Fields of Progenitor Stars

It has long been established that axions could have been produced within the nascent proto-neutron star formed following the type II supernova SN1987A, escaped the star due to their weak interactions, and then converted to gamma rays in the Galactic magnetic fields; the nonobservation of a gamma-ray flash coincident with the neutrino burst leads to strong constraints on the axion-photon coupling for axion masses m a ≲10 –10 eV. In this Letter, we use SN1987A to constrain higher mass axions, all the way to m a ~10 –3 eV, by accounting for axion production from the Primakoff process, nucleon bremsstrahlung, and pion conversion along with axion-photon conversion on the still-intact magnetic fields of the progenitor star. Moreover, we show that gamma-ray observations of the next Galactic supernova, leveraging the magnetic fields of the progenitor star, could detect quantum chromodynamics axions for masses above roughly 50 μ⁢eV, depending on the supernova. We propose a new full-sky gamma-ray satellite constellation that we call the GALactic AXion Instrument for Supernova (GALAXIS) to search for such future signals along with related signals from extragalactic neutron star mergers.

Impact of Newly Measured 𝛽-Delayed Neutron Emitters around 78 Ni on Light Element Nucleosynthesis in the Neutrino Wind Following a Neutron Star Merger

Neutron emission probabilities and half-lives of 37 𝛽-delayed neutron emitters from 75 Ni to 92 Br were measured at the RIKEN Nishina Center in Japan, including 11 one-neutron and 13 two-neutron emission probabilities and six half-lives for the first time that supersede theoretical estimates. These nuclei lie in the path of the weak 𝑟 process occurring in neutrino-driven winds from the accretion disk formed after the merger of two neutron stars synthesizing elements in the 𝐴∼80 abundance peak. The presence of such elements dominates the accompanying kilonova emission over the first few days and have been identified in the AT2017gfo event, associated to the gravitational wave detection GW170817. Abundance calculations based on over 17,000 simulated trajectories describing the evolution of matter properties in the merger outflows show that the new data lead to an increase of 50%–70% in the abundance of Y, Zr, Nb, and Mo. This enhancement is large compared to the scatter of relative abundances observed in old very metal poor stars and thus is significant in the comparison with other possible astrophysical processes contributing to the light-element production. These results underline the importance of including experimental decay data for very neutron-rich 𝛽 -delayed neutron emitters into 𝑟 -process models.

The Two Arctic Wintertime Boundary Layer States: Disentangling the Role of Cloud and Wind Regimes in Reanalysis and Observations During MOSAiC

The wintertime central Arctic atmosphere comprises a radiatively clear and a radiatively opaque state, which are linked to synoptic forcing and mixed-phase clouds. Weather and climate models often lack process representations surrounding these states, but prior work mostly treated the problem as an aggregate of synoptic conditions, resulting in partially overlapping biases. Here, we disaggregate the Arctic states and confront ERA5 reanalysis with observations from the MOSAiC campaign over the central Arctic sea ice during winter 2019/2020. Low-level winds and liquid water path (LWP) are combined to derive different synoptic classes. Results show that the clear state is primarily formed by weak/moderate winds and the absence of liquid-bearing clouds, while strong winds and enhanced LWP primarily form the radiatively opaque state. ERA5 struggles to reproduce these basic statistics, shows too weak sensitivity of thermal radiation to synoptic forcing, and overestimates thermal radiation for similar LWP amounts. The latter is caused by a warm bias, which has a pronounced inversion structure and is largest in clear and calm conditions. Under strong synoptic forcing, the warm bias is constant with height and discrepancies in mixed-phase cloud altitude appear. Separating synoptic conditions is regarded as useful for process-oriented evaluation of the Arctic troposphere in models.

Adsorption of Ag, Au, Cu, and Ni on MoS 2 : theory and experiment

Abstract Here, we present results of a computational and experimental study of adsorption of various metals on MoS2. In particular, we analyzed the binding mechanism of four metallic elements (Ag, Au, Cu, Ni) on MoS2. Among these elements, Ni exhibits the strongest binding and lowest mobility on the surface of MoS2. On the other hand, Au and Ag bond very weakly to the surface and have very high mobilities. Our calculations for Cu show that its bonding and surface mobility are between these two groups. Experimentally, Ni films exhibit a composition characterized by randomly oriented nanoscale clusters. This is consistent with the larger cohesive energy of Ni atoms as compared with their binding energy with MoS2, which is expected to result in 3D clusters. In contrast, Au and Ag tend to form atomically flat plateaued structures on MoS2, which is contrary to their larger cohesive energy as compared to their weak binding with MoS2. Cu displays a surface morphology somewhat similar to Ni, featuring larger nanoscale clusters. However, unlike Ni, in many cases Cu exhibits small plateaued surfaces on these clusters. This suggests that Cu likely has two competing mechanisms that cause it to span the behaviors seen in the Ni and Au/Ag film morphologies. These results indicate that calculations of the initial binding conditions could be useful for predicting film morphologies. In addition, out calculations show that the adsorption of adatoms with odd electron number like Ag, Au, and Cu results in 100% spin-polarization and integer magnetic moment of the system. Adsorption of Ni adatoms, with even electron number, does not induce a magnetic transition.

Co-, Ni- and Fe-rich grain-boundary phases enhance creep resistance in θ'-strengthened Al-Cu alloys

Here, microstructural evolution and creep response were investigated in the cast Al-5.0Cu-0.3Mn-0.2Zr (wt.%) alloy with and without addition of slow-diffusing, intermetallic-forming elements Fe, Ni, or Co. Baseline Al-5.0Cu-0.3Mn-0.2Zr alloy exhibits high creep resistance at 300 °C, up to ~75 MPa, which is attributed to high-aspect-ratio, intragranular θ'-Al 2 Cu precipitates that effectively suppress dislocation climb. However, θ'-Al 2 Cu precipitate-free zones form along grain boundaries upon heat treatment, whose extent is amplified during subsequent creep. Such weak regions experience dislocation creep, leading to strain localization and acceleration of grain-boundary sliding. Adding Ni and Co, individually or in combination, leads to the formation of grain-boundary precipitates (Al 9 Co 2 , Al 3 Ni 2 ) which are resistant to coarsening, thus suppressing the formation of θ'-Al 2 Cu precipitate-free zones. This microstructure provides high creep resistance at stresses up to 75–80 MPa, with strain rates much lower than in unmodified Al-5.0Cu-0.3Mn-0.2Zr. Adding Fe, which results in extensive decoration of grain boundaries with coarsening-resistant Al 7 Cu 2 Fe, and then increasing the Cu content to compensate for the Cu loss to this new phase, leads to a new Al-7.4Cu-1.6Fe-0.3Mn-0.2Zr alloy with creep resistance at 300 °C that surpasses known cast aluminum alloys. Adding Fe to improve the creep resistance of Al-Cu alloys is both cost-effective and sustainable. Our findings offer guidelines applicable to various alloy systems on controlling the evolution of precipitate-free zones and its ensuing effects on creep deformation.

CT584 Is Not a Protective Vaccine Antigen against Respiratory Chlamydial Challenge in Mice

Background: Chlamydia trachomatis is the most prevalent bacterial sexually transmitted pathogen in humans worldwide. Since chlamydial infection is largely asymptomatic with the potential for serious complications, a preventative vaccine is likely the most viable long-term answer to this public health threat. Cell-free protein synthesis (CFPS) utilizes the cellular protein manufacturing machinery decoupled from the requirement for maintaining cellular viability, offering the potential for flexible, rapid, and decentralized production of recombinant protein vaccine antigens. Methods: Here, we use CFPS to produce the full-length putative chlamydial type three secretion system (T3SS) needle-tip protein, CT584, for evaluation as a vaccine antigen in mouse models. High-speed atomic force microscopy (HS-AFM) (RIBM, Tsukuba, Japan) imaging and computer simulations confirm that CFPS-produced CT584 retains a native-like structure prior to immunization. Female mice were primed with CT584 adjuvanted with CpG-1826 intranasally (i.n.) or CpG-1826 + Montanide ISA 720 intramuscularly (i.m.), followed four weeks later by an i.m. boost before respiratory challenge with 10 4 inclusion forming units (IFU) of Chlamydia muridarum. Results: Immunization with CT584 generated robust antibody responses but weak cell-mediated immunity and failed to protect against i.n. challenge as demonstrated by body weight loss, increased lung weights, and the presence of high numbers of IFUs in the lungs. Conclusion: While CT584 was not a protective vaccine candidate, the speed and flexibility with which CFPS can be used to produce other potential chlamydial antigens make it an attractive technique for antigen production.
