Enantioselective ecotoxicity of promethazine in two freshwater organisms: daphnia (Daphnia magna) and zebrafish ( Danio rerio )

Abstract Chiral pharmaceuticals, racemic or enantiomerically pure forms and their metabolites, can reach aquatic ecosystems via wastewater effluents (inefficient treatment operations) or by direct human disposal. They may negatively affect nontarget organisms even at low environmental concentrations. To make an accurate risk evaluation, the (eco)toxicity of both enantiomers needs to be assessed. Promethazine (PMZ) is a chiral antihistamine that has been detected in aquatic ecosystems owing to its high consumption. Promethazine undergoes metabolism in the liver, producing chiral metabolites such as promethazine sulfoxide (PMZSO) and N-desmethylpromethazine (DMPMZ) that reach water bodies. However, knowledge regarding the enantioselective toxicity of PMZ and its metabolites on aquatic organisms is missing. This study aimed to explore the potential enantioselective toxicity of PMZ and its metabolites on two relevant freshwater organisms, daphniid and fish, representing different trophic levels. The half maximal effect concentrations (EC50s) in Daphnia magna of PMZ, DMPMZ, and PMZSO were 2.33, 2.31, > 4 mg L−1, respectively, > 4 and 2.50 mg L−1 for (R) and (S)-PMZ, respectively, and > 4 mg L−1 for the enantiomers of DMPMZ and PMZSO. In studies involving zebrafish, Danio rerio, (R, S)-PMZ showed a median lethal concentration (LC50) of .72 mg L−1, and specific assays revealed that (R)-PMZ exhibited more pronounced adverse effects on larvae at the embryonic, morphological, and biochemical level than the racemate and (S)-PMZ. Toxicity and potential bioaccumulation of these compounds in daphniids and fish were also conducted using in silico tests through proprietary software. The results revealed a concordance between the experimental and predicted EC50 and LC50 values in both species.

PQML: Enabling the Predictive Reproducibility on NISQ Machines for Quantum ML Applications

Quantum computing represents a groundbreaking approach to high-performance computing. In recent years, quantum computers have progressed from single-qubit processors to systems boasting over 400 qubits. The presence of such a large number of qubits offers significant advantages, including enhanced computational speed—a capability beyond classical computing methods. However, the current stage of quantum computing is referred to as the noisy intermediate-scale quantum (NISQ) era. The existence of noise in this era presents challenges in testing quantum computing applications, leading to considerable variance in application results. Furthermore, the diverse noise characteristics observed across different machines exacerbate this issue, complicating the selection of the appropriate machine for application execution. In response to these challenges, we introduce our Predictive Quantum Machine Learning (PQML) tool. This tool is designed to predict outcomes when executing identical quantum machine learning applications—specifically, a critical suite of variational quantum algorithms—across various quantum computers during the NISQ era. This effort relies on data collected over a 12-month period. To the best of our knowledge, this study represents the first attempt to ensure reproducibility across quantum computers for complex circuits. Additionally, we have developed a model capable of forecasting the accuracy of quantum computers for variational quantum algorithms, with a particular emphasis on quantum machine learning as a case study.

Atmosphere Explorer control system software (version 1.0)

The basic design is described of the Atmosphere Explorer Control System (AECS) software used in the testing, integration, and flight contol of the AE spacecraft and experiments. The software performs several vital functions, such as issuing commands to the spacecraft and experiments, receiving and processing telemetry data, and allowing for extensive data processing by experiment analysis programs. The major processing sections are: executive control section, telemetry decommutation section, command generation section, and utility section.

Proposal for hierarchical description of software systems

The programming of digital computers has developed into a new dimension full of diffculties, because the hardware of computers has become so powerful that more complex applications are entrusted to computers. The costs of software development, verification, and maintenance are outpacing those of the hardware and the trend is toward futher increase of sophistication of application of computers and consequently of sophistication of software. To obtain better visibility into software systems and to improve the structure of software systems for better tests, verification, and maintenance, a clear, but rigorous description and documentation of software is needed. The purpose of the report is to extend the present methods in order to obtain a documentation that better reflects the interplay between the various components and functions of a software system at different levels of detail without losing the precision in expression. This is done by the use of block diagrams, sequence diagrams, and cross-reference charts. In the appendices, examples from an actual large sofware system, i.e. the Marshall System for Aerospace Systems Simulation (MARSYAS), are presented. The proposed documentation structure is compatible to automation of updating significant portions of the documentation for better software change control.

Space Shuttle - The new baseline

Recent design alterations and refinements in the Space Shuttle program are considered with particular attention to the present baseline Shuttle configurations, performance characteristics, mission profiles, and flight and ground operations. Hardware and software development and testing, propulsion systems, entry trajectory events, and payload capability are covered. The refined Space Shuttle system design is characterized as one reflecting firm technical and economic considerations. Systems development is well under way, with all major contractors progressing on schedule.

Launch Vehicle Operations Simulator

The Saturn Launch Vehicle Operations Simulator (LVOS) was developed for NASA at Kennedy Space Center. LVOS simulates the Saturn launch vehicle and its ground support equipment. The simulator was intended primarily to be used as a launch crew trainer but it is also being used for test procedure and software validation. A NASA/contractor team of engineers and programmers implemented the simulator after the Apollo XI lunar landing during the low activity periods between launches.

Advanced application flight experiment breadboard pulse compression radar altimeter program

Design, development and performance of the pulse compression radar altimeter is described. The high resolution breadboard system is designed to operate from an aircraft at 10 Kft above the ocean and to accurately measure altitude, sea wave height and sea reflectivity. The minicomputer controlled Ku band system provides six basic variables and an extensive digital recording capability for experimentation purposes. Signal bandwidths of 360 MHz are obtained using a reflective array compression line. Stretch processing is used to achieve 1000:1 pulse compression. The system range command LSB is 0.62 ns or 9.25 cm. A second order altitude tracker, aided by accelerometer inputs is implemented in the system software. During flight tests the system demonstrated an altitude resolution capability of 2.1 cm and sea wave height estimation accuracy of 10%. The altitude measurement performance exceeds that of the Skylab and GEOS-C predecessors by approximately an order of magnitude.

Adaptation and optimization of a line-by-line radiative transfer program for the STAR-100 (STARSMART)

A program to calculate upwelling infrared radiation was modified to operate efficiently on the STAR-100. The modified software processes specific test cases significantly faster than the initial STAR-100 code. For example, a midlatitude summer atmospheric model is executed in less than 2% of the time originally required on the STAR-100. Furthermore, the optimized program performs extra operations to save the calculated absorption coefficients. Some of the advantages and pitfalls of virtual memory and vector processing are discussed along with strategies used to avoid loss of accuracy and computing power. Results from the vectorized code, in terms of speed, cost, and relative error with respect to serial code solutions are encouraging.

Shuttle orbiter Ku-band radar/communications system design evaluation

Tasks performed in an examination and critique of a Ku-band radar communications system for the shuttle orbiter are reported. Topics cover: (1) Ku-band high gain antenna/widebeam horn design evaluation; (2) evaluation of the Ku-band SPA and EA-1 LRU software; (3) system test evaluation; (4) critical design review and development test evaluation; (5) Ku-band bent pipe channel performance evaluation; (6) Ku-band LRU interchangeability analysis; and (7) deliverable test equipment evaluation. Where discrepancies were found, modifications and improvements to the Ku-band system and the associated test procedures are suggested.

Charge injection device evaluation hardware

A microprocessor based system which provides maximum testing flexibility via software reconfiguration is described. This system has the capability to evaluate a charge injection device (CID) and to characterize its performance. The construction and operation of the CID offer advantages over the various CCD configurations available. These advantages include a less sophisticated manufacturing process, no images smearing during readout, high transfer efficiencies, and high resistance to blooming due to optical overload.

Structure, composition and thermal state of the crust in Brazil

Aeromagnetic survey data of the Minas Gerais areas were analyzed and compared with MAGSAT anomaly data. Initially the data set was used to determine the thickness of the magnetized crust. The value obtained is in agreement with crustal models derived from deep seismic sounding and gravity data, indicating that the bottom of the magnetized layer occurs close to the Moho transition. An analysis of the long wavelength pattern indicates that the anomalies form a series of lineations along the magnetic E-W. These are shown to be the result of attenuation. On the MAGSAT total intensity map localized anomalies are superimposed onto an anomaly background formed by alternating positive and negative patches. Implementation and testing of analysis software for MAGSAT data are also discussed.

The P-POD Project

The programmable pilot oriented display (P-POD), a multiprocessor based flight information processing and display unit, is discussed. Communication protocols between the three Z-80 processors inside P-POD are reviewed. The interface between the video processor, P2, and the main processor, P1, was tested. Hardware and software were revised. Data flow between P1 and P2 is illustrated. While running diagnostic routines to exercise the P1/P2 interface at maximum speed an intermittent failure was observed. A check of possible noise sources or race conditions in the hardware did not reveal the source of this failure. Possible software timing problems were also fully investigated. To eliminate the effects of this type of failure a performance monitor routine was used to detect these occasional failures and correct them. The data transmission cycle between P1/P2 is either allowed to continue normal operation or is re-initialized and restarted when an error is detected.

Development of a preprototype times wastewater recovery subsystem, addendum

Six tasks are described reflecting subsystem hardware and software modifications and test evaluation of a TIMES wastewater recovery subsystem. The overall results are illustrated in a figure which shows the water production rate, the specific energy corrected to 26.5 VDC, and the product water conductivity at various points in the testing. Four tasks are described reflecting studies performed to develop a preliminary design concept for a next generation TIMES. The overall results of the study are the completion of major design analyses and preliminary configuration layout drawings.

A methodology for evaluation of an interactive multispectral image processing system

Because of the considerable cost of an interactive multispectral image processing system, an evaluation of a prospective system should be performed to ascertain if it will be acceptable to the anticipated users. Evaluation of a developmental system indicated that the important system elements include documentation, user friendliness, image processing capabilities, and system services. The criteria and evaluation procedures for these elements are described herein. The following factors contributed to the success of the evaluation of the developmental system: (1) careful review of documentation prior to program development, (2) construction and testing of macromodules representing typical processing scenarios, (3) availability of other image processing systems for referral and verification, and (4) use of testing personnel with an applications perspective and experience with other systems. This evaluation was done in addition to and independently of program testing by the software developers of the system.

Temporal and contextual knowledge in model-based expert systems

A basic paradigm that allows representation of physical systems with a focus on context and time is presented. Paragon provides the capability to quickly capture an expert's knowledge in a cognitively resonant manner. From that description, Paragon creates a simulation model in LISP, which when executed, verifies that the domain expert did not make any mistakes. The Achille's heel of rule-based systems has been the lack of a systematic methodology for testing, and Paragon's developers are certain that the model-based approach overcomes that problem. The reason this testing is now possible is that software, which is very difficult to test, has in essence been transformed into hardware.
