Sostdc1 Regulates NK Cell Maturation and Cytotoxicity [Sostdc1 regulates natural killer cell maturation and cytotoxicity]

NK cells are innate-like lymphocytes that eliminate virally infected and cancerous cells, but the mechanisms that control NK cell development and cytotoxicity are incompletely understood. We identified roles for sclerostin domain–containing-1 ( Sostdc1 ) in NK cell development and function. Sostdc1 -knockout ( Sostdc1 –/– ) mice display a progressive accumulation of transitional NK cells (tNKs) (CD27 + CD11b + ) with age, indicating a partial developmental block. The NK cell Ly49 repertoire in Sostdc1 –/– mice is also changed. Lower frequencies of Sostdc1 –/– splenic tNKs express inhibitory Ly49G2 receptors, but higher frequencies express activating Ly49H and Ly49D receptors. Yet, the frequencies of Ly49I + , G2 + , H + , and D + populations were universally decreased at the most mature (CD27 – CD11b + ) stage. We theorized that the Ly49 repertoire in Sostdc1 –/– mice would correlate with NK killing ability and observed that Sostdc1 –/– NK cells are hyporesponsive against MHC class I–deficient cell targets in vitro and in vivo, despite higher CD107a surface levels and similar IFN-γ expression to controls. Consistent with Sostdc1’s known role in Wnt signaling regulation, Tcf7 and Lef1 levels were higher in Sostdc1 –/– NK cells. Expression of the NK development gene Id2 was decreased in Sostdc1 –/– immature NK and tNK cells, but Eomes and Tbx21 expression was unaffected. Reciprocal bone marrow transplant experiments showed that Sostdc1 regulates NK cell maturation and expression of Ly49 receptors in a cell-extrinsic fashion from both nonhematopoietic and hematopoietic sources. Taken together, these data support a role for Sostdc1 in the regulation of NK cell maturation and cytotoxicity, and identify potential NK cell niches.

The diurnal cycle of the smoky marine boundary layer observed during August in the remote southeast Atlantic

Ascension Island (8° S, 14.5° W) is located at the northwestern edge of the south Atlantic stratocumulus deck, with most clouds in August characterized by surface observers as “stratocumulus and cumulus with bases at different levels”, and secondarily as “cumulus of limited vertical extent” and occurring within a typically decoupled boundary layer. Field measurements have previously shown that the highest amounts of sunlight-absorbing smoke occur annually within the marine boundary layer during August. On more smoke-free days, the diurnal cycle in cloudiness includes a nighttime maximum in cloud liquid water path and rain, an afternoon cloud minimum, and a secondary late-afternoon increase in cumulus and rain. The afternoon low-cloud minimum is more pronounced on days with a smokier boundary layer. The cloud liquid water paths are also reduced throughout most of the diurnal cycle when more smoke is present, with the difference from cleaner conditions most pronounced at night. Precipitation is infrequent. An exception is the mid-morning, when the boundary layer deepens and liquid water paths increase. The data support a view that a radiatively enhanced decoupling persisting throughout the night is key to understanding the changes in the cloud diurnal cycle when the boundary layer is smokier. Under these conditions, the nighttime stratiform cloud layer does not always recouple to the sub-cloud layer, and the decoupling maintains more moisture within the sub-cloud layer. After the sun rises, enhanced shortwave absorption in a smokier boundary layer can drive a vertical ascent that momentarily couples the sub-cloud layer to the cloud layer, deepening the boundary layer and ventilating moisture throughout, a process that may also be aided by a shift to smaller droplets. After noon, shortwave absorption within smokier boundary layers again reduces the upper-level stratiform cloud and the sub-cloud relative humidity, discouraging further cumulus development and again strengthening a decoupling that lasts longer into the night. The novel diurnal mechanism provides a new challenge for cloud models to emulate. The lower free troposphere above cloud is more likely to be cooler, when boundary layer smoke is present, and lower free-tropospheric winds are stronger and more northeasterly, with both (meteorological) influences supporting further smoke entrainment into the boundary layer from above.

An attribution of the low single-scattering albedo of biomass burning aerosol over the southeastern Atlantic

Abstract. Aerosol over the remote southeastern Atlantic is some of the most sunlight-absorbing aerosol on the planet: the in situ free-tropospheric single-scattering albedo at the 530 nm wavelength (SSA530 nm) ranges from 0.83 to 0.89 within ORACLES (ObseRvations of Aerosols above CLouds and their intEractionS) aircraft flights from late August–September. Here we seek to explain the low SSA. The SSA depends strongly on the black carbon (BC) number fraction, which ranges from 0.15 to 0.4. Low organic aerosol (OA)-to-BC mass ratios of 8–14 and modified combustion efficiency values >0.975 point indirectly to the dry, flame-efficient combustion of primarily grass fuels, with back trajectories ending in the miombo woodlands of Angola. The youngest aerosol, aged 4–5 d since emission, occupied the top half of a 5 km thick plume sampled directly west of Angola with a vertically consistent BC:ΔCO (carbon monoxide) ratio, indicating a homogenization of the source emissions. The younger aerosol, transported more quickly off of the continent by stronger winds, overlaid older, slower-moving aerosol with a larger mean particle size and fraction of BC-containing particles. This is consistent with ongoing gas condensation and the coagulation of smaller non-BC particles upon the BC-containing particles. The particle volumes and OA:BC mass ratios of the older aerosol were smaller, attributed primarily to evaporation following fragmentation, instead of dilution or thermodynamics. The CLARIFY (CLoud–Aerosol–Radiation Interaction and Forcing: Year 2017) aircraft campaign sampled aerosols that had traveled further to reach the more remote Ascension Island. CLARIFY reported higher BC number fractions, lower OA:BC mass ratios, and lower SSA yet larger mass absorption coefficients compared to this study's. Values from one ORACLES 2017 flight, held midway to Ascension Island, are intermediate, confirming the long-range changes. Overall the data are most consistent with continuing oxidation through fragmentation releasing aerosols that subsequently enter the gas phase, reducing the OA mass, rather than evaporation through dilution or thermodynamics. The data support the following best fit: SSA530nm=0.801+0055⋅(OA:BC) (r=0.84). The fires of southern Africa emit approximately one-third of the world's carbon; the emitted aerosols are distinct from other regional smoke emissions, and their composition needs to be represented appropriately to realistically depict regional aerosol radiative effects.

Seawater pH reconstruction using boron isotopes in multiple planktonic foraminifera species with different depth habitats and their potential to constrain pH and <i>p</i>CO2 gradients

Boron isotope systematics of planktonic foraminifera from core-top sediments and culture experiments have been studied to investigate the sensitivity ofδ11B of calcite tests to seawater pH. However, our knowledge of the relationship between δ11B and pH remains incomplete for many taxa. Thus, to expand the potential scope of application of this proxy, we report δ11B data for seven different species of planktonic foraminifera from sediment core tops. We utilize a method for the measurement of small samples of foraminifera and calculate the δ11B-calcite sensitivity to pH for Globigerinoides ruber, Trilobus sacculifer (sacc or without sacc), Orbulina universa, Pulleniatina obliquiloculata, Neogloboquadrina dutertrei, Globorotalia menardii, and Globorotalia tumida, including for unstudied core tops and species. These taxa have diverse ecological preferences and are from sites that span a range of oceanographic regimes, including some that are in regions of air–sea equilibrium and others that are out of equilibrium with the atmosphere. The sensitivity of δ11Bcarbonate to δ11Bborate (e.g., Δδ11Bcarbonate/Δδ11Bborate) in core tops is consistent with previous studies for T. sacculifer and G. ruber and close to unity for N. dutertrei, O. universa, and combined deep-dwelling species. Deep-dwelling species closely follow the core-top calibration fo rO. universa, which is attributed to respiration-driven microenvironments likely caused by light limitation and/or symbiont–host interactions. Our data support the premise that utilizing boron isotope measurements of multiple species within a sediment core can be utilized to constrain vertical profiles of pH andpCO2 at sites spanning different oceanic regimes, thereby constraining changes in vertical pH gradients and yielding insights into the past behavior of the oceanic carbon pumps.

Structure analyses reveal a regulated oligomerization mechanism of the PlexinD1/GIPC/myosin VI complex

The GIPC family adaptor proteins mediate endocytosis by tethering cargo proteins to the myosin VI motor. The structural mechanisms for the GIPC/cargo and GIPC/myosin VI interactions remained unclear. PlexinD1, a transmembrane receptor that regulates neuronal and cardiovascular development, is a cargo of GIPCs. GIPC-mediated endocytic trafficking regulates PlexinD1 signaling. Here, we unravel the mechanisms of the interactions among PlexinD1, GIPCs and myosin VI by a series of crystal structures of these proteins in apo or bound states. GIPC1 forms a domain-swapped dimer in an autoinhibited conformation that hinders binding of both PlexinD1 and myosin VI. PlexinD1 binding to GIPC1 releases the autoinhibition, promoting its interaction with myosin VI. GIPCs and myosin VI interact through two distinct interfaces and form an open-ended alternating array. Our data support that this alternating array underlies the oligomerization of the GIPC/Myosin VI complexes in solution and cells.

Structure-based nuclear import mechanism of histones H3 and H4 mediated by Kap123

Kap123, a major karyopherin protein of budding yeast, recognizes the nuclear localization signals (NLSs) of cytoplasmic histones H3 and H4 and translocates them into the nucleus during DNA replication. Mechanistic questions include H3- and H4-NLS redundancy toward Kap123 and the role of the conserved diacetylation of cytoplasmic H4 (K5ac and K12ac) in Kap123-mediated histone nuclear translocation. Here, we report crystal structures of full-length Kluyveromyces lactis Kap123 alone and in complex with H3- and H4-NLSs. Structures reveal the unique feature of Kap123 that possesses two discrete lysine-binding pockets for NLS recognition. Structural comparison illustrates that H3- and H4-NLSs share at least one of two lysine-binding pockets, suggesting that H3- and H4-NLSs are mutually exclusive. Additionally, acetylation of key lysine residues at NLS, particularly H4-NLS diacetylation, weakens the interaction with Kap123. These data support that cytoplasmic histone H4 diacetylation weakens the Kap123-H4-NLS interaction thereby facilitating histone Kap123-H3-dependent H3:H4/Asf1 complex nuclear translocation.

The Missing Correlation Between the Potential Rate Impacts of Rooftop Solar and the Timing of State Net Metering Policy Revisions

Data supporting the article “The Missing Correlation Between the Potential Rate Impacts of Rooftop Solar and the Timing of State Net Metering Policy Revisions” (https://www.nlr.gov/docs/fy25osti/93543.pdf). Residential solar photovoltaic (PV) output in most states is credited at the retail electricity rate, a policy commonly known as net metering. Twelve states have replaced net metering with alternative rate structures that reduce PV adopter bill savings. Proponents of these revisions argue that net metering increases the electricity rates of customers without PV. Here, we analyze the degree to which the timelines of net metering revisions have correlated with potential electricity rate impacts. We estimate that potential rate impacts at the end of 2023 were less than 1% of typical customer bills in 37 of 44 states that have offered net metering. There are no statistically significant differences in average or median estimated rate impacts between states that have and have not revised net metering. Nine of the states that had revised net metering did so when estimated impacts were less than 1% of typical customer bills. Many states have retained net metering into higher PV deployment levels with increased risk of potential rate impacts. Only two states-California and Hawaii-retained net metering beyond estimated rate impacts of 5%, and both have revised net metering. These findings do not suggest a clear, consistent link between net metering revision timelines and potential rate impacts. The timing and nature of net metering revisions are ultimately policy decisions based on state-level priorities and considerations.

Impurity screening behavior of the high-field side scrape-off layer in near-double-null configurations: prospect for mitigating plasma-material interactions on RF actuators and first-wall components

The impurity screening response of the high-field side (HFS) scrape-off layer (SOL) to localized nitrogen injection is investigated on Alcator C-Mod for magnetic equilibria spanning lower-single null, double-null and upper single null configurations under otherwise identical plasma conditions. L-mode, EDA H-mode and I-mode discharges are investigated. HFS impurity screening is found to depend on magnetic flux balance and the direction of B×?B relative to the most active divertor. Impurity ‘plume’ emission patterns indicate that both parallel and perpendicular (E×B) flows in the SOL contribute to the ‘flushing’ of impurities towards the active divertor, thereby affecting the overall impurity screening behavior. Despite the fact that the HFS scrape-off layer is extremely narrow in near-double-null configurations, this SOL is able to screen locally injected nitrogen at least as effectively as the low-field side (LFS) SOL – up to a factor of 10 more effective, depending on specific plasma conditions and whether the magnetic geometry produces parallel flows that work with or against E×B flows. For situations in which the E×B drift of the impurity ions opposes parallel flow toward the primary divertor, HFS impurity screening is found to be least effective. When E×B drifts assist parallel flow toward the primary divertor, HFS impurity screening is found to be very effective. These data support the idea of placing RF actuators and close-fitting wall components on the high-field side of the tokamak. With this configuration, near-double-null magnetic topologies may be used for active control of plasma parameters at the antenna/plasma interface for optimal RF coupling, mitigate the generation of local impurities from plasma-material interactions and, taking advantage of favorable plasma flows and good screening properties of the HFS SOL, further minimize the impact of wall-born impurity sources on the plasma core.

High-field side scrape-off layer investigation: plasma profiles and impurity screening behavior in near-double-null configurations

New experiments on Alcator C-Mod reveal that the favorable impurity screening characteristics of the high-field side (HFS) scrape-off layer (SOL), previously reported for single null geometries, is retained in double null configurations, despite the formation of an extremely thin SOL. In balanced double-null, nitrogen injected locally into the HFS SOL is better screened by a factor of 2.5 compared to the same injection into the low field side (LFS) SOL. This result is insensitive to plasma current and Greenwald fraction. Nitrogen injected into the HFS SOL is not as well screened (only a factor of 1.5 improvement over LFS) in unbalanced double-null discharges, when the primary divertor is in the direction of B×?B. In this configuration, impurity ‘plume’ emission patterns indicate that an opposing E X B drift competes with the parallel impurity flow to the divertor. In balanced double-null plasmas, the dispersal pattern exhibits a dominant E X B motion. Unbalanced discharges with the primary divertor opposite the direction of B×?B exhibit excellent HFS screening characteristics – a factor of 5 enhancement compared to LFS. These data support the idea that future tokamaks should locate all RF actuators and close-fitting wall structures on the HFS and employ near-double-null magnetic topologies, both to precisely control plasma conditions at the antenna/plasma interface and to maximally mitigate the impact of local impurity sources arising from plasma-material interactions.
