Production of Vanillin From Ferulic Acid by Pseudomonas putida KT2440 Using Metabolic Engineering and In Situ Product Recovery

Vanillin is the most in-demand flavouring compound in the world and because vanillin extracted from vanilla pods cannot meet the global demand, most vanillin on the market today is chemically synthesised. Increasing demands by consumers for natural ingredients have inspired efforts to develop vanillin derived from microbial sources. These efforts have been challenged by low titers, likely caused by the toxicity of vanillin to most microbial biocatalysts. In this study, we engineered a Pseudomonas putida KT2440-derived strain that accumulated vanillin from ferulic acid to 0.64 g/L. To increase the overall titre, we applied a hydrophobic polystyrene-based resin to vanillin-accumulating cultures, which enabled an increase in total vanillin recovery to an apparent titre of 3.35 g/L. This study demonstrates that P. putida can accumulate vanillin from ferulic acid to higher titers when vanillin is removed from the cultivation medium, mitigating its toxicity.

Production of drop-in biodiesel blendstocks via competitive acid-catalyzed dehydration reactions using ethanol oligomerization products

Ethanol can be converted into diesel fuel ethers using a three-step catalytic approach that involves ethanol oligomerization to larger alcohols, hydrogenolysis of the esters followed by dehydration of the C4 to C8 alcohols into ethers. In this paper we report results for the dehydration of a mixture of C4 – C8 alcohols using a zeolite Y catalyst in a continuous flow reactor. Mono-molecular dehydration of the alcohols produces olefins while bi-molecular dehydration of the alcohol produces ethers. Increasing the pressure increases the ether selectivity while decreasing the pressure produces more olefins. Linear alcohol feeds produce more C8+ ethers, while branched and secondary alcohols lead to more olefins. Olefin and coke selectivities increase with increasing carbon chain length of alcohols. Secondary alcohols lead to higher coke selectivities. Furthermore, ethanol/Butanol oligomerization experiments showed that the incoming dehydration feedstock can be grown to larger C6+ alcohol fractions, leading to higher yields of C10+ diesel-range ethers.

Data for An Orphan Gene BOOSTER Enhances Photosynthetic Efficiency and Plant Productivity

Seeds of Col-0 wild type, sig6 T-DNA mutants (CS877785, ABRC), PRL-1-OE, and sig6 T-DNA mutants transfected with PRL-1 (sig6::PRL-1) were planted in 1/2 MS media. Seedlings growth including chlorophyll development defects were investigated across the genotypes. Four-days-old-post-light exposure seedlings were harvested and performed RNAseq analysis with four biological replicates.

Production and analysis of output data products for Landsat-4 in the engineering check-out phase

The Landsat-D (now Landsat-4) program was initiated by NASA in the mid-1970's. The spacecraft was launched on July 16, 1982. Instruments on the satellite include the fourth Multispectral Scanner (MSS) and the new higher-resolution Thematic Mapper (TM). In order to achieve the planned engineering and scientific objectives, attention has to be given to the validation of spacecraft and instrument performance, the adjustment of ground processing procedures, and the definition of the measures of the TU utility. To satisfy these requirements, a Landsat Assessment System (LAS) was conceived, and the Applications Developmental Data System (ADDS) was instituted. The set of component systems collectively became known as the 'Scrounge' system. The present investigation is concerned with the Scrounge system and the results of radiometric and geometric data evaluations. Attention is given to the TM geometric correction, TM data resampling, a radiometric correction functional description, and the results of the TM geometric and radiometric correction.
