Hot-melt extrudability of amorphous solid dispersions of flubendazole-copovidone: An exploratory study of the effect of drug loading and the balance of adjuvants on extrudability and dissolution

The FDA-approved anthelmintic flubendazole has shown potential to be repositioned to treat cancer and dry macular degeneration; however, its poor water solubility limits its use. Amorphous solid dispersions may overcome this challenge, but the balance of excipients may impact the preparation method and drug release. Here, we evaluate the influence of adjuvants and drug loading on the development of an amorphous solid dispersion of flubendazole-copovidone by hot-melt extrusion. The drug, copovidone, and adjuvants (magnesium stearate and hydroxypropyl cellulose) mixtures were statistically designed, and the process was performed in a twin-screw extruder. The study showed that flubendazole and copovidone mixtures were highly extrudable, except when drug loading was high (>40%). Furthermore, magnesium stearate positively impacted the extrusion and was more effective than hydroxypropyl cellulose. The extruded materials were evaluated by modulated differential scanning calorimetry and X-ray powder diffraction, obtaining positive amorphization and physical stability results. Pair distribution function analysis indicated the presence of drug rich domains with medium-range order structure and no evidence of polymer-drug interaction. All extrudates presented faster dissolution (HCl, pH 1.2) than pure flubendazole, and both adjuvants had a notable influence on the dissolution rate. In conclusion, hot-melt extrusion may be a viable option to obtain stable flubendazole: copovidone amorphous dispersions.

An exploratory study on helium mobility in amorphous and crystallized bulk metallic glasses

In this study, Zr 52.5 Cu 17.9 Ni 14.6 Al 10 Ti 5 and Cu 60 Zr 20 Hf 10 Ti 10 amorphous and crystallized bulk metallic glasses (BMGs) were implanted at room temperature by 150 keV 3 He + ions to fluences of 2 × 10 15 cm -2 and 5 × 10 15 cm -2 , producing peak implanted He concentrations of ~1,100 and 3,500 appm, respectively. Nuclear reaction analysis (NRA) performed on post-implantation annealed samples at various temperatures between 250 and 600 °C (~0.30 to 0.75 of the absolute melting temperature T m of the BMGs) revealed no appreciable He migration. Complementary thermal desorption spectroscopy (TDS) on as-implanted samples, which involved in situ annealing, showed appreciable He release only at temperatures higher than ~700 °C (~0.76-0.90 T m ) in all the samples. Half or more of the implanted He did not desorb up to the maximum investigated TDS annealing temperature of 770 °C (~0.85-0.97 T m ). The lack of pronounced He diffusion up to 600 °C as observed from the NRA and TDS data was attributed to He trapping in free volume sites present in the BMGs, which seems to play a similar role to vacancies in crystalline materials. It was also observed that in both the amorphous and crystallized forms of the Zr BMG, the sample implanted to the lower fluence released more He as compared to the sample implanted with a higher fluence. The crystalline forms of both the Cu and Zr BMGs released more He as compared to their amorphous counterparts.

Investigating the influence of environmental information on perceived indoor environmental quality: An exploratory study

Under the assumption that information can impact perception, most research on human sensation and satisfaction with indoor environmental quality (IEQ) parameters has been conducted with respondents uninformed about the test conditions. Therefore, researchers know little about the impact of information on perception. These potential effects are increasingly relevant as quantitative information about indoor environments becomes accessible via low-cost, wirelessly connected sensors. In this experimental study, 48 subjects were exposed to varied indoor environmental conditions and provided with different types of environmental information. The subjects' sensation and satisfaction were compared when they were blinded or provided with quantitative information about and/or qualitative ratings of specific parameters. The results indicate that accurate information on parameter values influenced how the subjects perceived the indoor air quality (IAQ) but not how they perceived the thermal, acoustic, or visual quality. The subjects rated the IAQ more positively when they were informed that there were nonzero ventilation rates. The qualitative ratings influenced the subjects' perceptions of all four environmental factors, but in different directions. The subjects generally had more positive sensation and higher satisfaction when they were told that the parameter values and qualitative ratings were more favorable than the test conditions. However, the improved sensation and satisfaction were often not as good as when the environmental conditions were actually improved and the subjects were provided with accurate information. Here, these findings affirm the critical need for more research on the impacts of information on perceptions of the indoor environment.

Exploratory study reveals far reaching systemic and cellular effects of verapamil treatment in subjects with type 1 diabetes

Currently, no oral medications are available for type 1 diabetes (T1D). While our recent randomized placebo-controlled T1D trial revealed that oral verapamil had short-term beneficial effects, their duration and underlying mechanisms remained elusive. Now, our global T1D serum proteomics analysis identified chromogranin A (CHGA), a T1D-autoantigen, as the top protein altered by verapamil and as a potential therapeutic marker and revealed that verapamil normalizes serum CHGA levels and reverses T1D-induced elevations in circulating proinflammatory T-follicular-helper cell markers. RNA-sequencing further confirmed that verapamil regulates the thioredoxin system and promotes an anti-oxidative, anti-apoptotic and immunomodulatory gene expression profile in human islets. Moreover, continuous use of oral verapamil delayed T1D progression, promoted endogenous beta-cell function and lowered insulin requirements and serum CHGA levels for at least 2 years and these benefits were lost upon discontinuation. Thus, the current studies provide crucial mechanistic and clinical insight into the beneficial effects of verapamil in T1D.

Preventing leaching from lead water pipes with electrochemistry: an exploratory study

Toxic levels of lead leaching from ageing water distribution infrastructure affect over 5000 public drinking water systems in the US. Pipe replacement, the most effective solution to this problem, is prohibitively expensive. Additionally, chemical conditioning of drinking water using orthophosphates, although cost-effective, does not quickly stop lead leaching once it has started. We propose a novel approach to stop lead leaching: to rapidly form an insoluble scale within lead pipes using an external power supply. We report on the feasibility of this approach by first anodizing lead coupons and lead pipes reclaimed from a local water distribution using a phosphate electrolyte and different potentials, pH values, and phosphate concentrations. We subsequently exposed these anodized lead coupons and pipes to synthetic tap water to evaluate their lead leaching rates. We found that polarizing lead coupons in the presence of a 0.05 M phosphate solution decreased lead leaching by up to a 100-fold, relative to leaching from polished bare lead. Similarly, polarizing the reclaimed lead pipes (with a preexisting scale) decreased lead leaching from an average of 36 ppb to 7 ppb. These results were observed when applying potentials that favor Pb(IV) formation, which resulted in the buildup of PbO 2 and Pb 5 (PO 4 ) 3 OH in both lead coupons and pipes. Our findings indicate that this novel technology has the potential to rapidly decrease equilibrium lead levels in tap water below the EPA action limit of 15 ppb, and thus deserves further exploration.

Exploratory investigation on the use of low-cost alternative media for ultrafine grinding of coal

In this study, the energy efficiency of three alternative grinding media including, coarse silica beads, fine silica beads, and granular sand, were evaluated and compared against standard stainless steel. Stirred mill test results show that both silica beads and granular sand were more efficient than stainless steel in producing ultrafine particles following the order of coarse silica beads>granular sand>fine silica beads>stainless steel. The finest product size corresponding to a P 80 value of 2.33 μm was generated by coarse silica beads with a nominal size of 420×1190 μm, which also consumed the least amount of grinding energy. The specific energy input was reduced by almost 50% as opposed to that of stainless steel, while the operating work index was reduced by 87%. Overall, the test data indicate that grinding media density, media particle size and shape all play a role in determining the optimized grinding behavior of the coal material.

Exploratory Investigation of Disengagements and Crashes in Autonomous Vehicles Under Mixed Traffic: An Endogenous Switching Regime Framework

Autonomous Vehicles (AVs) have a large potential to improve traffic safety but also pose some critical challenges. While AVs may help reduce crashes caused by human error, they still may experience failures of technologies and sensing, as well as decision-making errors in a mixed traffic environment. A disengagement refers to an AV transitioning control from autonomous systems to the trained test driver. The safety critical nature of disengagements makes it imperative to analyze disengagements and crashes together. In this study, we analyze both crashes and disengagements from real-world AV driving in California to fill the knowledge gap regarding the relationship between disengagements and crashes in a mixed traffic environment. A nested logit model was calibrated using three different outcomes: (1) disengagement with a crash, (2) disengagement with no crash, and (3) no disengagement with a crash. Furthermore, endogenous switching regime models were also calibrated to draw distinctions between the relation of disengagements and crashes while accounting for endogeneity effects. The results show that factors related to AV systems (such as software failures) and other roadway participants increase the propensity of a disengagement without a crash. Furthermore, AVs were observed to disengage less often as the technology matured over time. Marginal effects revealed an 8% decrease. The results thus suggest that disengagements are a part of AVs' safe performance and disengagement alerts may need to be triggered in order to avoid certain failures with current technology.

On a High-Mirror Stellarator Reactor Exploratory Concept With Neutrons Concentrated on Centrifuge Liquids

In the framework of fusion energy research based on magnetic confinement, stellarators allow numerous degrees of freedom for the design of the magnetic trap and plasma shape. Taking advantage of these features, some plasma shapes might benefit several of the many integrated elements involved in commercial fusion reactors, like, e.g., decreasing the number, mass and complexity of replaced activated in-vessel components (IVC) (i.e., by using liquids), extraction of large power, tritium generation, and remote maintenance. Certainly, free-surface liquid materials were proposed for tokamaks and field-reversed configuration (FRC) to try to improve some of such elements, i.e., in advanced power extraction (APEX) studies. Some reactor-relevant quasi-isodynamic (QI) magnetic configurations exhibit a relatively straight sector of plasma and high magnetic mirror. The combination of those elements and possibilities in a single stellarator reactor concept might have some advantages, in spite of the uncertainties due to the current low technological readiness level (TRL). The proposed and studied reactor concept is based on a vacuum vessel having short curved sectors and longer wide cylindrical sectors, which encloses a high-mirror low-vertical-excursion magnetic configuration, and swirling liquids or rotating cylinders, which centrifuge molten Li salts located at the low field region. Thus, the molten salts (if possible covered by a thin layer of liquid lithium) would be located on the internal perimeter of the cylinder, to act as particle exhaust (except for helium), neutron power extractor, and tritium breeder. The high-mirror feature tries to concentrate the neutron power at the cylindrical sectors, which might avoid using breeding materials at the curved sectors. The different elements of the concept are exploratorily studied and defined, and the difficulties assessed.

An exploratory steady–state redox model of photosynthetic linear electron transport for use in complete modelling of photosynthesis for broad applications

A photochemical model of photosynthetic electron transport (PET) is needed to integrate photophysics, photochemistry, and biochemistry to determine redox conditions of electron carriers and enzymes for plant stress assessment and mechanistically link sun-induced chlorophyll fluorescence to carbon assimilation for remotely sensing photosynthesis. Towards this goal, we derived photochemical equations governing the states and redox reactions of complexes and electron carriers along the PET chain. These equations allow the redox conditions of the mobile plastoquinone pool and the cytochrome b 6 f complex (Cyt) to be inferred with typical fluorometry. The equations agreed well with fluorometry measurements from diverse C 3 /C 4 species across environments in the relationship between the PET rate and fraction of open photosystem II reaction centres. We found the oxidation of plastoquinol by Cyt is the bottleneck of PET, and genetically improving the oxidation of plastoquinol by Cyt may enhance the efficiency of PET and photosynthesis across species. Redox reactions and photochemical and biochemical interactions are highly redundant in their complex controls of PET. Although individual reaction rate constants cannot be resolved, they appear in parameter groups which can be collectively inferred with fluorometry measurements for broad applications. The new photochemical model developed enables advances in different fronts of photosynthesis research.

ftmsRanalysis: An R package for exploratory data analysis and interactive visualization of FT-MS data

The high-resolution and mass accuracy of Fourier transform mass spectrometry (FT-MS) has made it an increasingly popular technique for discerning the composition of soil, plant and aquatic samples containing complex mixtures of proteins, carbohydrates, lipids, lignins, hydrocarbons, phytochemicals and other compounds. Thus, there is a growing demand for informatics tools to analyze FT-MS data that will aid investigators seeking to understand the availability of carbon compounds to biotic and abiotic oxidation and to compare fundamental chemical properties of complex samples across groups. We present ftmsRanalysis, an R package which provides an extensive collection of data formatting and processing, filtering, visualization, and sample and group comparison functionalities. The package provides a suite of plotting methods and enables expedient, flexible and interactive visualization of complex datasets through functions which link to a powerful and interactive visualization user interface, Trelliscope. Example analysis using FT-MS data from a soil microbiology study demonstrates the core functionality of the package and highlights the capabilities for producing interactive visualizations.

Exploratory splitter bend system designs for FFA@CEBAF

An upgrade to the Continuous Electron Beam Accelerator Facility (CEBAF) at the Thomas Jefferson National Accelerator Facility (JLAB) is anticipated to provide an electron beam of over 20 GeV using the existing superconducting-RF linear accelerator and new fixed-field alternating (FFA) gradient recirculation arcs made up of Halbach-style permanent magnets. In the current design, the FFA arcs will carry six beams with energies of approximately 11, 13, 16, 18, 20, and 22 GeV which will require horizontal splitter lines to match the beam from the preceding linac. In this paper, we describe two alternative splitter beamline designs that are tuned to match the beam's Twiss parameters, R56, time-of-flight, bend-plane offset, and dispersion into the FFA cells.

Exploratory Efforts to Constrain Geologic Material Properties from Remote Sensing Data: Joint Study (FY2020 Final Report)

Identification and characterization of underground events from surface or remote data requires a thorough understanding of the rock material properties. However, material properties usually come from borehole data, which is expensive and not always available. A potential alternative is to use topographic characteristics to approximate the strength, but this has never been done before quantitatively. Here we present the results from the first steps towards this goal. We have found that there are strong correlations between compressive and tensile strengths and slopes, but these correlations vary depending on data analysis details. Rugosity may be better correlated to strength than slope values. More comprehensive analyses are needed to fully understand the best method of predicting strength from topography for this area. We also found that misalignment of multiple GIS datasets can have a large influence on the ability to make interpretations. Lastly, these results will require further study in a variety of climatic conditions before being applicable to other sites.
