Expression quantitative trait loci mapping identified PtrXB38 as a key hub gene in adventitious root development in Populus

Summary Plant establishment requires the formation and development of an extensive root system with architecture modulated by complex genetic networks. Here, we report the identification of the PtrXB38 gene as an expression quantitative trait loci (eQTL) hotspot, mapped using 390 leaf and 444 xylem Populus trichocarpa transcriptomes. Among predicted targets of this trans ‐eQTL were genes involved in plant hormone responses and root development. Overexpression of PtrXB38 in Populus led to significant increases in callusing and formation of both stem‐born roots and base‐born adventitious roots. Omics studies revealed that genes and proteins controlling auxin transport and signaling were involved in PtrXB38‐mediated adventitious root formation. Protein–protein interaction assays indicated that PtrXB38 interacts with components of endosomal sorting complexes required for transport machinery, implying that PtrXB38‐regulated root development may be mediated by regulating endocytosis pathway. Taken together, this work identified a crucial root development regulator and sheds light on the discovery of other plant developmental regulators through combining eQTL mapping and omics approaches.

Deep Lynx: Digital Engineering Integration Hub

The construction of megaprojects has consistently demonstrated challenges for project managers in regard to meeting cost, schedule, and performance requirements. Megaproject construction challenges are common place within megaprojects with many active projects in the United States failing to meet cost and schedule efforts by significant margins. Currently, engineering teams operate in siloed tools and disparate teams where connections across design, procurement, and construction systems are translated manually or over brittle point-to-point integrations. The manual nature of data exchange increases the risk of silent errors in the reactor design, with each silent error cascading across the design. These cascading errors lead to uncontrollable risk during construction, resulting in significant delays and cost overruns. Deep Lynx allows for an integrated platform during design and operations of mega projects.

Energy Material Network Data Hubs

In early 2015 the United States Department of Energy conceived of a consortium of collaborative bodies based on shared expertise, data, and resources that could be targeted towards the more difficult problems in energy materials research. The concept of virtual laboratories had been envisioned and discussed earlier in the decade in response to the advent of the Materials Genome Initiative and similar scientific thrusts. To be effective, any virtual laboratory needed a robust method for data management, communication, security, data sharing, dissemination, and demonstration to work efficiently and effectively for groups of remote researchers. With the accessibility of new, easily deployed cloud technology and software frameworks, such individual elements could be integrated, and the required collaboration architecture is now possible. The developers have leveraged open-source software frameworks, customized them, and merged them into a platform to enable collaborative energy materials science, regardless of the geographic dispersal of the people and resources. After five years in operations, the systems are demonstratively an effective platform for enabling research within the Energy Material Networks (EMN). This paper will show the design and development of a secured scientific data sharing platform, the ability to customize the system to support diverse workflows, and examples of the enabled research and results connected with some of the Energy Material Networks.

Mudstone Baffles and Barriers in Lower Cretaceous Strata at a Proposed CO 2 Storage Hub in Kemper County, Mississippi, United States

The Cretaceous and Tertiary deposits in Mississippi, Alabama, and the adjacent continental shelf constitute a widespread succession of sandstone, mudstone, and carbonate that has proven to be an important target for geologic CO 2 storage in the onshore Gulf of Mexico basin. Integrated analysis of stratigraphy, sedimentology, and reservoir properties based on cores and geophysical well logs indicates that the Paluxy Formation and Washita- Fredericksburg interval present gigatonne-class storage opportunities. The distribution, geometry, and composition of the area is a direct reflection of the original depositional environments, and understanding these factors is essential for understanding the geologic storage potential of the Paluxy Formation and Washita-Fredericksburg interval at the Kemper County energy facility in Mississippi. Geologic characterization of the Mississippi Embayment at the energy facility focused primarily on characterizing the confinement potential of the storage complex. Integration of core analyses and geophysical well logs has yielded a high-resolution stratigraphic analysis of storage reservoirs, baffles, barriers, and seals. Scanning electron microscopy (SEM) coupled with energy dispersive X-ray spectroscopy (EDS), and quantitative X-ray diffraction (XRD) was used to characterize microfabric, pore types, and mineralogy within mudstone of the east-central Mississippi Embayment at the Kemper County energy facility. Mudstone beds in the Paluxy Formation and Washita-Fredericksburg interval have variable thickness and continuity. High water saturation in the Cretaceous mudstone units influences swelling smectite clay and mudrock permeability based on pulse decay analysis is 1–96 nD. These low permeability values indicate that the mudstone units are effective baffles, barriers, and confining intervals that make significant migration of injected CO 2 out of the storage complex unlikely. The numerous baffles and barriers within the target reservoir intervals, moreover, favor the retention of multiple CO 2 in plumes within the abundant stacked sandstone bodies.
