De Novo Design of Proteins That Bind Naphthalenediimides, Powerful Photooxidants with Tunable Photophysical Properties

De novo protein design provides a framework to test our understanding of protein function and build proteins with cofactors and functions not found in nature. Here, we report the design of proteins designed to bind powerful photooxidants and the evaluation of the use of these proteins to generate diffusible small-molecule reactive species. Because excited-state dynamics are influenced by the dynamics and hydration of a photooxidant’s environment, it was important to not only design a binding site but also to evaluate its dynamic properties. Thus, we used computational design in conjunction with molecular dynamics (MD) simulations to design a protein, designated NBP (NDI Binding Protein), that held a naphthalenediimide (NDI), a powerful photooxidant, in a programmable molecular environment. Solution NMR confirmed the structure of the complex. We evaluated two NDI cofactors in this de novo protein using ultrafast pump–probe spectroscopy to evaluate light-triggered intra- and intermolecular electron transfer function. Moreover, we demonstrated the utility of this platform to activate multiple molecular probes for protein labeling.

Optimal strategies for optical quantum memories using long-lived noble-gas spins

Nuclear spins of noble gases exhibit exceptionally long coherence times and can potentially serve as a long-lived storage medium for quantum information. We analyze and compare the performance of two mechanisms for mapping the quantum state of light onto the collective spin state of noble gases. The first mechanism utilizes collisional exchange with the electronic spin state of metastable noble-gas atoms, while the second relies on spin-exchange collisions with ground-state alkali-metal atoms. We describe the operation of an optical quantum memory relying on these two mechanisms using a compact model and study strategies that optimize the memory storage efficiency. Through numerical simulations, we identify optimal sequences for storing optical signals with different signal bandwidths and electronic spin relaxation rates. This work highlights the qualitative difference between the two approaches for using noble gases as long-lived quantum memories at noncryogenic conditions and outlines the regimes in which they are expected to be efficient.

A New Reduced Order Model For The Mechanistic Creep Behavior Of UO 2

This manuscript describes an ongoing NEAMS effort to better determine the performance of advanced nuclear fuels, in particular the creep behavior of doped UO$_2$ for light water reactors. In our previous work, we outlined a method to utilize data generated from lower length scale simulations and implement it into the engineering scale fuel performance analysis. This process has been further refined, and in addition, new data has been used to train the surrogate model which has also been substantially improved since the previous iteration. The new model is compared against the current empirical model used in BISON using both scoping calculations to define the performance over the parameter space and using integral instrumented fuel assessment cases to determine the impact of these models on the overall fuel performance. Suggestions and guidance for future improvements to this method are provided to ensure the model covers relevant parameter space and phenomena.

2x2 Demonstrator High-Coverage Light Readout System in a Novel LArTPC

DUNE is a long-baseline Liquid Argon Time Projection Chamber (LArTPC) neutrino oscillation experiment. Its near detectors, located at Fermi National Laboratory, will constrain systematic errors on far detector measurements. The 2x2 Demonstrator, a novel proof of concept prototype for the ND LArTPC, has a modular TPC design that mitigates event pileup in a high-occupancy nearline beam environment, further aided by a native 3D pixelated charge readout and a 30% coverage, low profile light readout. This will be the first large-scale LArTPC to utilize pixels in place of wire planes

Domesticating the green alga Scenedesmus obliquus

Microalgae cultivation processes exist only for a few species used in commercial applications. Their domestication is still in its infancy, with major advances required, specifically to overcome limiting factors in microalgae production. The goal is to develop superior strains without the need of genetic technologies that would require lengthy regulatory permits, and often limit consumer acceptance. Adaptive Laboratory Evolution techniques, alone or in conjunction with sexual recombination, can allow for rapid develop of improved strains and their industrial production. Approaches include increased photon utilization efficiency by cultures operating under full sunlight conditions due to higher light saturation levels, allowing for higher productivities under outdoor conditions. Decades of research yielded some promising results under controlled conditions with a few specific mutant strains. However, these failed to achieve the anticipated higher productivities in actual algal mass cultures, in part due to the inability of mutations to overcome photoinhibition, reactive oxygen species, and other pleiotropic impacts on photosynthesis.

Fast quantum ghost imaging with a single-photon-sensitive time-stamping camera

Quantum ghost imaging (QGI) leverages correlations between entangled photon pairs to reconstruct an image using light that has never physically interacted with an object. Despite extensive research interest, this technique has long been hindered by slow acquisition speeds, due to the use of raster-scanned detectors or the slow response of intensified cameras. Here, we utilize a single-photon-sensitive time-stamping camera to perform QGI at ultra-low-light levels with rapid data acquisition and processing times, achieving high-resolution and high-contrast images in under 1 min. Our work addresses the trade-off between image quality, optical power, data acquisition time, and data processing time in QGI, paving the way for practical applications in biomedical and quantum-secured imaging.

2019 California Vehicle Survey

The 2019 California Vehicle Survey of residential and commercial light-duty fleet owners in California assessed consumer preferences for vehicles and included a targeted sample of plug-in electric vehicle (PEV) owners. In addition to economic and demographic data, the survey integrated light-duty vehicle holding and use information with vehicle choice data, which was collected via a set of eight exercises on vehicle and fuel type choice. The PEV owner survey participants provided additional data on charging behavior, electricity rates, and their main motivations for purchasing PEVs.

ComStock Measure Documentation: Lighting Control for Load Shedding

This report describes the modeling methodology for a single end-use savings shape measure - lighting control for load shedding - and briefly introduces key results. The lighting control for load shedding measure applies lighting dimming control to reduce the lighting load during the building's electricity peak window every weekday. The measure takes daily peak load schedule inputs generated by the method "Dispatch Schedule Generation" described in the "Supplemental Documentation: Dispatch Schedule Generation for Demand Flexibility Measures" to determine the start and end times of the predicted peak window, and then adjusts the lighting dimming level by a percentage reduction from the original schedules during the peak window to reduce the peak demand. The measure is applicable to (large, medium and small) offices, warehouses, and primary and secondary schools, which correspond to approximately 68% of the stock floor area of commercial buildings in ComStock analysis. The measure demonstrates 2%-7% daily peak demand reduction performance for applicable buildings, and 0.43% total site energy savings (0 trillion British thermal units [TBtu]) for the U.S. commercial building stock modeled in ComStock.

Model-agnostic search for dijet resonances with anomalous jet substructure in proton–proton collisions at $\sqrt{s}$ = 13 TeV

This paper presents a model-agnostic search for narrow resonances in the dijet final state in the mass range 1.8-6 TeV. The signal is assumed to produce jets with substructure atypical of jets initiated by light quarks or gluons, with minimal additional assumptions. Search regions are obtained by utilizing multivariate machine-learning methods to select jets with anomalous substructure. A collection of complementary anomaly detection methods - based on unsupervised, weakly supervised, and semisupervised algorithms - are used in order to maximize the sensitivity to unknown new physics signatures. These algorithms are applied to data corresponding to an integrated luminosity of 138 fb -1 , recorded by the CMS experiment at the LHC, at a center-of-mass energy of 13 TeV. No significant excesses above background expectations are seen. Exclusion limits are derived on the production cross section of benchmark signal models varying in resonance mass, jet mass, and jet substructure. Many of these signatures have not been previously sought, making several of the limits reported on the corresponding benchmark models the first ever. When compared to benchmark inclusive and substructure-based search strategies, the anomaly detection methods are found to significantly enhance the sensitivity to a variety of models.
