Adapting a Low-Cost and Open-Source Commercial Pipetting Robot for Nanoliter Liquid Handling

Nanoliter liquid handling capabilities in bioanalytical workflows can reduce reagent costs and improve sample processing efficiency. Here we report the successful modification of a low-cost commercially available pipetting robot, the Opentrons 1 (OT1), using inexpensive 3D-printed parts to accommodate low-nanoliter volumes. The pipetting accuracy and precision of the modified OT1 for 50 nL volumes of water was 0.2% and 2%, respectively. The accuracy and precision at 20 nL was 1% and 3.8%, respectively. The platform was successfully applied to the preparation of low-nanogram proteomic samples for liquid chromatography-mass spectrometry analysis. The modified OT1 is thus an effective platform for nanoliter liquid handling and, at a total cost of less than $6000 including the commercial liquid handler and all modifications, is far less expensive and more versatile than other platforms with similar capabilities, placing this technology within reach of a far broader scientific community.

Improving AHU Performance by Minimizing Approach Temperature, Reducing Air Maldistribution, and Efficiently Handling Sensible and Latent Loads (Phase 1 Interim Final Technical Report)

Residential air handling units (AHUs) have stayed the same in form and efficiency for the past 30+ years, with incremental improvements made to address safety, functionality, and energy-efficiency. The purpose of this research in Phase I, Topic 9a: Next Generation Residential Air Handlers, was to improve AHU performance by minimizing heat exchanger (HX) approach temperature, reducing air maldistribution, and developing alternative system configurations which more efficiently handle sensible and latent loads. In this research Optimized Thermal Systems (OTS) developed, modeled, and evaluated multiple alternative system concepts. A dual vapor compression system separate sensible and latent cooling (SSLC) concept was studied to inform work on alternative concepts and to show best-case performance benefit. System concepts included ejector enhanced vapor compression cycles, desiccant assisted dehumidification, dual evaporator SSLC, and alternative AHU HX configurations. A dual vapor compression system showed COP improvement of 20%, however, required additional components, increased unit size, and increased cost. Two types of ejector enhanced vapor compression cycles with dual evaporators improved system COP by 9 to 11%, and reduced AHU losses by as much as 18%, with design changes limited to the AHU, no unit physical size increase, and a moderate increase to system first cost. Desiccant assisted air-conditioning required increased air flow rate resulting in higher fan power and the desiccant wheel increased sensible heat load leading to increased compressor power and reduced system COP. Dual evaporator cycles were found to degrade performance due to increased expansion losses. Optimized single slab HX designs used in place of the traditional A-coil HX led to 44–49% reduction in aluminum, 47–60% less refrigerant charge, and improved HX velocity distribution.

Radial Internal Material Handling System (RIMS) for Circular Habitat Volumes

A Radial Internal Material Handling System (RIMS) has been developed to service a circular floor area in variable gravity. On planetary surfaces, pressurized human habitable volumes will require a means to carry heavy equipment between various locations within the volume of the habitat, regardless of the partial gravity (Earth, moon, Mars, etc). On the NASA Habitat Demonstration Unit (HDU), a vertical cylindrical volume, it was determined that a variety of heavy items would need to be carried back and forth from deployed locations to the General Maintenance Work Station (GMWS) when in need of repair, and other equipment may need to be carried inside for repairs, such as rover parts and other external equipment. The vertical cylindrical volume of the HDU lent itself to a circular overhead track and hoist system that allows lifting of heavy objects from anywhere in the habitat to any other point in the habitat interior. In addition, the system is able to hand off lifted items to other material handling systems through the side hatches, such as through an airlock. This paper describes the RIMS system which is scalable for application in a variety of circular habitat volumes.

Handling Qualities of Large Rotorcraft in Hover and Low Speed

According to a number of system studies, large capacity advanced rotorcraft with a capability of high cruise speeds (approx.350 mph) as well as vertical and/or short take-off and landing (V/STOL) flight could alleviate anticipated air transportation capacity issues by making use of non-primary runways, taxiways, and aprons. These advanced aircraft pose a number of design challenges, as well as unknown issues in the flight control and handling qualities domains. A series of piloted simulation experiments have been conducted on the NASA Ames Research Center Vertical Motion Simulator (VMS) in recent years to systematically investigate the fundamental flight control and handling qualities issues associated with the characteristics of large rotorcraft, including tiltrotors, in hover and low-speed maneuvering.

NASA's Center Innovation Fund (CIF) Internal Research and Development (IRAD) Handling Qualities Study on the Mikrolar Motion Platform Test Report

In order to support the National Aeronautics and Space Administration’s (NASA) Extravehicular Activity and Surface Mobility Program (EHP), system engineers, designers, and researchers embarked on a yearlong challenge to develop a new lunar rover-based motion table simulator to study handling qualities of lunar rovers in the lunar South Pole region, support development research for any lunar rover vendor, and provide a lunar training capability for future lunar/planetary astronauts. In 1989, the United Kingdom’s Royal Air Force Institute of Aviation Medicine (RAF-IAM) stated that motion platforms are the only simulation devices capable of fully stimulating the body motion sensors. They confirmed that motion platforms can impart accelerations to the whole body and therefore exercise the automatic motion feedback-loop that operators are used to. With both visual and motion cues handling the vehicle becomes more realistic. Strachan (2019) con-firms motion cueing from a well set-up motion platform has been found to be important especially in conditions such as night or reduced visibility where motion cues may be more relied upon

Electrification Analysis: Container Ports' Cargo Handling Equipment

This one-page highlight details the key takeaways from a project that utilized NREL's Fleet Research, Energy Data, and Insights (FleetREDI) data analysis pipeline, the Electrification Analysis of Container Ports' Cargo Handling Equipment project. This project created a scalable solution to model energy demand per shipping container moved (kWh/TEU) for an all-electric cargo handling equipment fleet located at a maritime port. The model allows stakeholders to understand energy demand at each electric vehicle (EV) equipment level and is easily scalable to container demand and EV adoption rate projections.

D2NO: Efficient handling of heterogeneous input function spaces with distributed deep neural operators

Neural operators have been applied in various scientific fields, such as solving parametric partial differential equations, dynamical systems with control, and inverse problems. However, challenges arise when dealing with input functions that exhibit heterogeneous properties, requiring multiple sensors to handle functions with minimal regularity. To address this issue, discretization-invariant neural operators have been used, allowing the sampling of diverse input functions with different sensor locations. However, existing frameworks still require an equal number of sensors for all functions. We propose a novel distributed approach to further relax the discretization requirements and solve the heterogeneous dataset challenges. Our method involves partitioning the input function space and processing individual input functions using independent and separate neural networks. A centralized neural network is used to handle shared information across all output functions. This distributed methodology reduces the number of gradient descent back-propagation steps, improving efficiency while maintaining accuracy. Here, we demonstrate that the corresponding neural network is a universal approximator of continuous nonlinear operators and present three numerical examples to validate its performance.

Search for CME in U+U and Au+Au collisions in STAR with different approaches of handling backgrounds

We report the chiral magnetic effect (CME) refers to charge separation along a strong magnetic field between left- and right-handed quarks, caused by interactions with topological gluon fields from QCD vacuum fluctuations. We present two approaches to handle the dominant elliptic flow (v 2 ) background in the three-particle correlator (Δγ 112 ), sensitive to CME. In the first approach, we present the Δγ 112 and Δγ 123 measurements in U+U and Au+Au collisions. While hydrodynamic simulations including resonance decays and local charge conservation predict that Δγ 112 scaled by N part /ν 2 will be similar in U+U and Au+Au collisions, the projected B-field exhibits a distinct difference between the two systems and with varying N part . Therefore, U+U and Au+Au collisions provide configurations with different expectations for both CME signal and background. Moreover, the three-particle observable Δγ 123 scaled by N part /ν 3 provide baseline measurement for only the background. In the second approach, we handle the ν 2 background by measuring Δγ 112 with respect to the planes of spectators measured by Zero Degree Calorimeters and participants measured by Time Projection Chamber. These measurements contain different amounts of contributions from CME signal (along B-field, due to spectators) and ν 2 background (determined by the participant geometry). With the two Δγ 112 measurements, the possible CME signal and the background contribution can be determined. We report such a measurement in Au+Au collisions at $\sqrt{s_{NN}}$ = 27GeV with the newly installed event plane detector, and report the new findings in U+U system where the spectator-participant plane correlations are expected to differ from those in Au+Au collisions.

General Strategy for Incorporation of Functional Group Handles into Covalent Organic Frameworks via the Ugi Reaction

The library of imine-linked covalent organic frameworks (COFs) has grown significantly over the last two decades, featuring a variety of morphologies, pore sizes, and applications. An array of synthetic methods has been developed to expand the scope of the COF functionalities; however, most of these methods were designed to introduce functional scaffolds tailored to a specific application. Having a general approach to diversify COFs via late-stage incorporation of functional group handles would greatly facilitate the transformation of these materials into platforms for a variety of useful applications. In this paper, we report a general strategy to introduce functional group handles in COFs via the Ugi multicomponent reaction. To demonstrate the versatility of this approach, we have synthesized two COFs with hexagonal and kagome morphologies. We then introduced azide, alkyne, and vinyl functional groups, which could be readily utilized for a variety of post-synthetic modifications. This facile approach enables the functionalization of any COFs containing imine linkages.

The Potential Benefits of Handling Mixture Statistics via a Bi-Gaussian EnKF: Tests With All-Sky Satellite Infrared Radiances

The meteorological characteristics of cloudy atmospheric columns can be very different from their clear counterparts. Thus, when a forecast ensemble is uncertain about the presence/absence of clouds at a specific atmospheric column (i.e., some members are clear while others are cloudy), that column's ensemble statistics will contain a mixture of clear and cloudy statistics. Such mixtures are inconsistent with the ensemble data assimilation algorithms currently used in numerical weather prediction. Hence, ensemble data assimilation algorithms that can handle such mixtures can potentially outperform currently used algorithms. In this study, we demonstrate the potential benefits of addressing such mixtures through a bi-Gaussian extension of the ensemble Kalman filter (BGEnKF). The BGEnKF is compared against the commonly used ensemble Kalman filter (EnKF) using perfect model observing system simulated experiments (OSSEs) with a realistic weather model (the Weather Research and Forecast model). Synthetic all-sky infrared radiance observations are assimilated in this study. In these OSSEs, the BGEnKF outperforms the EnKF in terms of the horizontal wind components, temperature, specific humidity, and simulated upper tropospheric water vapor channel infrared brightness temperatures. This study is one of the first to demonstrate the potential of a Gaussian mixture model EnKF with a realistic weather model. Our results thus motivate future research toward improving numerical Earth system predictions though explicitly handling mixture statistics.

Assessment of ITER radiation environment during the remote-handling operation of In-Vessel components with D1SUNED

During ITER operational life, a remote-handled cask will be used to transfer In-Vessel components to the Hot Cell for maintenance, storage and decommissioning purposes. Due to the distribution of penetrations for system allocation in the facility, the radiation field of each transfer operation presents a high spatial variability; all operations must be studied independently for workers and electronics protection. In this paper, we present a fully representative approach to describe the radiation environment during the complete remote-handling scenario of In-Vessel components in the ITER facility. The impact of all relevant radiation sources during different stages of the operation is addressed. As-built structures and 2020 baseline designs are considered to produce the most detailed neutronics model of the Tokamak Complex, the 400,000-tonne civil structure hosting the tokamak, up to date. Novel capabilities of the D1SUNED code have allowed to compute the integral dose, the dose rate and the photon-induced neutron flux of both moving and static radiation sources. Time bins are included in the simulations to compute the dose rate caused by In-Vessel components at all positions along the transfer. The time evolution of the dose rate is built in video format with a 1-m resolution, especially valuable for hot-spots identification.

A Year-Round Energy-Efficient Fresh Air Handling System with Two-Stage Heat Pumps and Lake Water Pre-Treatment

Supplying fresh air is important for indoor air quality, but the energy consumption to treat fresh air is significant. The main problems in existing fresh air system include: supplying single-temperature water to treat fresh air and not applying natural energy sufficiently restrict the efficiency improvement of cooling and heat sources; unused air handling devices under most operating conditions throughout the year increase the fan energy consumption. Thus, this study proposes an efficient fresh air system that uses lake water to pre-treat fresh air and two-stage heat pumps for further treatment, and the unused air handling devices in the system are bypassed. A fresh air system in northern China is selected as a case to show the annual energy performance of the proposed system, and a comparative analysis is conducted between the proposed system and a traditional system. The results show that the annual energy-savings of the proposed system compared to the traditional system come from fans, and compressors, which are 56.5%, and 31.1%, respectively, with an entire system energy saving of 32.2%. The proposed system can achieve an annual system coefficient of performance (COP) of 5.5. This study provides potential for further improving the energy efficiency in fresh air treatment.

A semi-analytical model of RF condensation that can handle localized power depositions

A nonlinear effect, RF (radio frequency) condensation, can be used to facilitate RF stabilization of magnetic islands. Previously studied semi-analytical models for RF condensation are suited mainly for broad deposition profiles and are unable to handle power depositions that are localized in the interior of a magnetic island. The power deposition profile of electron cyclotron waves, in particular, can be highly localized near a cyclotron resonance. Here, a model is developed that can handle both localized profiles and broad profiles. This allows a comparison of RF condensation for narrow vs broad deposition profiles, and it allows a study of the dependence of RF condensation of localized deposition profiles on key parameters.

RICH (Robotic Interface Control & Handling) System for VULCAN

High flux neutron beam and high efficiency detectors warrant quick turn arounds of neutron diffraction measurements at the engineering materials diffractometer VULCAN. Efficient change and alignment of samples and automatic measurements at VULCAN are desired for better use of neutron beam time by users. In this work, we aim to develop a proof-of-concept Robotic Interactive Control & Handling System (RICH) for sample handling at VULCAN that could assist high throughput experiments and reduce the overhead time significantly. This was realized by a six-axis desktop robot with trained AI models. The trained AI models can recognize and locate samples in various types of positions in a live video stream. In addition, we developed smart algorithms which used our models on multiple cameras to recognize where samples are with respect to multiple point of views, and then using user-inputted parameters, align them to perform complex measurements.
