Bridging the gap: linking Torulaspora delbrueckii genotypes to fermentation phenotypes and wine aroma

Abstract Climate change and consumer preferences are driving innovation in winemaking, with a growing interest in non-Saccharomyces species. Among these, Torulaspora delbrueckii (Td) has gained recognition for its ability to reduce volatile acidity and enhance aromatic complexity in wine. However, knowledge regarding its phenotypic and genomic diversity impacting alcoholic fermentation remains limited. Aiming to elucidate the metabolic differences between Td and Saccharomyces cerevisiae (Sc) and the Td intraspecies diversity, we conducted a comprehensive metabolic characterization of 15 Td strains. This analysis delved beyond standard fermentation parameters (kinetics and major metabolites production) to explore non-conventional aromas and establish genotype-phenotype links. Our findings confirmed that most Td strains produce less acetic acid and more succinate and glycerol than Sc. The overall aromatic profiles of Td strains differed from Sc, exhibiting higher levels of monoterpenes and higher alcohols, while producing less acetate esters, fatty acids, their corresponding ethyl esters, and lactones. Moreover, we identified the absence of genes responsible for specific aroma profiles, such as decreased ethyl esters production, as well as the absence of cell wall genes, which might negatively affect Td performance when compared to Sc. This work highlights the significant diversity within Td and underscores potential links between its genotype and phenotype.

Quantum limits of superconducting-photonic links and their extension to millimeter waves

Photonic addressing of superconducting circuits has been proposed to overcome wiring complexity and heat-load challenges. However, such superconducting-photonic links suffer from an efficiency-noise trade-off that limits scalability. This trade-off arises because increasing power conversion efficiency entails reducing optical power, which makes the converted signal susceptible to shot noise. We analyze this trade-off and find that the laser-driven qubit gate infidelity scales inversely with the number of photons used. While methods such as nonlinear detection or squeezed light could mitigate this effect, we consider generating higher-frequency electrical signals, such as millimeter waves (100 GHz), using laser light. At these higher frequencies, circuits have higher operating temperatures and cooling power budgets and alleviate constraints posed by the trade-off. Therefore, we demonstrate an optically driven cryogenic millimeter-wave source with a maximum power efficiency of 8 ×10 −5 that can generate a maximum of 0.7 μ⁢W of 80 GHz power, with a 1200-thermal-photon equivalent of added noise at 4K. Using this source, we perform frequency-domain spectroscopy of superconducting NbTiN resonators at 80–90 GHz. Our results show a promising approach to lessen the efficiency-noise constraints on superconducting-photonic links, while leveraging the benefits of photonic signal delivery. Further optimization of power efficiency and noise at high frequencies could make scalable photonic control of superconducting qubits viable at temperatures exceeding 1K.

Freely jointed chain models with extensible links

We report analytical relations for the mechanical response of single polymer chains are valuable for modeling purposes, on both the molecular and the continuum scale. These relations can be obtained using statistical thermodynamics and an idealized single-chain model, such as the freely jointed chain model. To include bond stretching, the rigid links in the freely jointed chain model can be made extensible, but this almost always renders the model analytically intractable. Here, an asymptotically correct statistical thermodynamic theory is used to develop analytic approximations for the single-chain mechanical response of this model. The accuracy of these approximations is demonstrated using several link potential energy functions. This approach can be applied to other single-chain models, and to molecular stretching in general.

A Simple Space Vector Modulation of High-Frequency AC Linked Three-Phase-to-Single-Phase/DC Converter

High frequency (HF) ac linked converter functions well to interface between buses with different voltage types. Using a three-phase-to-single-phase matrix converter, a single-phase cyclo converter, and a HF transformer, we can compose a HF ac linked converter to interface between three-phase ac grids and single-phase ac or dc distributed generations (DGs). In this paper, a novel space vector modulation (SVM) method is proposed for such kind of converter. The proposed SVM method converts the mains three-phase ac voltage to a HF ac voltage at the primary side of transformer, and then unfolds the HF ac voltage to a single-phase ac or dc voltage at the secondary side of transformer. Without need to compute separate switching signals for the primary and secondary converters and then coordinate their operations elaborately, the proposed SVM method can be easily implemented by incorporating the HF chopping into the SVM of traditional indirect matrix converter. Therefore, no complicated duty cycle computation or elaborate switching states combination is required. Simulation and hardware-in-loop implementation demonstrate the validity of the proposed method.

Employing Weibull Analysis and Weakest Link Theory to Resolve Crystalline Silicon PV Cell Strength Between Bare Cells and Reduced- and Full-Sized Modules

Weibull analysis and weakest link theory are employed to resolve the probability of crystalline silicon PV cell fracture when measured as bare cells and when stressed in reduced- and full-sized modules. Experimental results indicated that the characteristic cell strength is reduced by ~20% once packaged into the laminate of a one-cell module and loaded in four-point flexure (4PF). This experimental observation was shown consistent with a weakest link theory prediction that the strength limiting flaws reside on the surface of the cell's edge. Here, the analysis is ultimately extended to present the equivalent loading of four-cell modules by uniform pressure and 4PF and a uniformly loaded full-sized module and demonstrates that smaller, representative, modules must be loaded to a much higher level than their parent full-sized modules to achieve an equivalent probability for cell fracture.

Coordinated Control of Natural and Sub-Synchronous Oscillations via HVDC Links in Great Britain Power System

Conventional power oscillation damping (POD) controllers have been designed to be effective in damping either inter-area low-frequency oscillations (LFO) or sub-synchronous oscillations (SSO). For those POD controls, model-based control design method has been widely implemented with wide-area feedback signals from phasor measurement units (PMU). However, the model accuracy of a large realistic power system can hardly be guaranteed, which will finally impact the POD damping performance. In this paper, a measurement-based method is proposed for POD design using High Voltage Direct Current links (HVDC) to realize the coordinated control of both LFO and SSO. For each type of oscillations, a band-pass filter is added to avoid the interactions between different modes. Case studies are carried out on the Great Britain 36-bus power system model with multiple HVDC links in DIgSILENT/PowerFactory. Simulation results demonstrate that the proposed PODs are effective in damping inter-area LFO and SSO simultaneously.

Reduced Common-Mode Voltage PWM Scheme for Full-SiC Three-Level Uninterruptible Power Supply With Small DC-Link Capacitors

In this article, a pulsewidth modulation (PWM) scheme for three-level full-SiC uninterruptible power supplies is developed to achieve a high power density. Two key passive components are selected for size reduction of the ac-ac stage: common-mode (CM) EMI filter, and dc-link capacitors. To reduce the CM noise, a new vector combination is proposed based on synchronous switching among three-phases. The proposed combinations align CM voltage (CMV) to be a single pulse per switching period. Owing to the simple shape, CMV cancellation between a three-level rectifier and inverter can be maximally utilized. A transition between the three combinations can control the drift of neutral point voltage. An equivalent carrier-based implementation is developed. Second, a simple algorithm to compensate neutral point voltage fluctuation is proposed both for differential mode (DM) and CM output voltage. Low-order harmonics on three-phase currents and an additional high-frequency CM noise by misaligned switching instant can be eliminated. The proposed compensation can be implemented by a simple correction on carrier slopes and injected zero-sequence voltage. The proposed PWM scheme is verified with 20-kW full-SiC UPS switching at 60 kHz with 140 μF dc-link capacitors.

A genetic link between leaf carbon isotope composition and whole-plant water use efficiency in the C 4 grass Setaria

Genetic selection for whole-plant water use efficiency (yield per transpiration; WUE plant ) in any crop-breeding programme requires high-throughput phenotyping of component traits of WUE plant such as intrinsic water use efficiency (WUE i ; CO 2 assimilation rate per stomatal conductance). Measuring WUE i by gas exchange measurements is laborious and time consuming and may not reflect an integrated WUE i over the life of the leaf. Alternatively, leaf carbon stable isotope composition (δ 13 C leaf ) has been suggested as a potential time-ntegrated proxy for WUE i that may provide a tool to screen for WUE plant . However, a genetic link between δ 13 C leaf and WUE plant in a C 4 species has not been well established. Thus, to confirm if there is a genetic relationship in a C 4 plant between δ 13 C leaf and WUE plant under well watered and water-limited growth conditions, a high-throughput phenotyping facility was used to measure WUE plant in a recombinant inbred line (RIL) population created between the C 4 grasses Setaria viridis and S. italica . Three quantitative trait loci (QTL) for δ 13 C leaf were found and co-localized with transpiration, biomass accumulation, and WUE plant . Additionally, WUE plant for each of the δ 13 C leaf QTL allele classes was negatively correlated with δ 13 C leaf , as would be predicted when WUE i influences WUE plant . These findings demonstrate that δ 13 C leaf is genetically linked to WUE plant , likely to be through their relationship with WUE i , and can be used as a high-throughput proxy to screen for WUE plant in these C 4 species.

A Micro-Mechanical Constitutive Model to Predict Hygrothermal Aging of Cross-Linked Polymers

Abstract A multi-physics material model is presented to describe the effects of temperature, oxygen, and humidity on the constitutive response of cross-linked polymers. The effect of hygrothermal damage on the mechanical integrity of the polymer matrix can be considered as the result of damage accumulation of two independent aging mechanism namely, i) thermo-oxidative, and ii) hydrolytic aging. In order to capture the mutual effects of thermo-oxidative and hydrolytic aging, an assumption has been made that each of the aging phenomenon can be superposed to each other. In fact, each of them works independently and as a result, they can compete with each other. Utilizing the theory of network decomposition, all phenomena and their correlation were modeled and thus, the strain energy function of the polymer matrix is written with respect to four independent mechanisms, i) the shrinking original matrix that has neither been attacked by water nor oxygen, ii) conversion of the first network to two new network due to the reduction and formation of cross-links, and iii) energy loss from network degradation due to attack of the water molecules to polymer active agents. Moreover, the proposed model is micro-mechanically based and is mainly relevant on thin samples due to our underlying assumption of homogeneous diffusion of oxygen and water throughout the matrix. The model has been validated against extensive data-sets obtained from experiments we specifically designed for concept validation.

Exploration of structure-activity relationships for the SARS-CoV-2 macrodomain from shape-based fragment linking and active learning

The macrodomain of severe acute respiratory syndrome coronavirus 2 nonstructural protein 3 is required for viral pathogenesis and is an emerging antiviral target. We previously performed an x-ray crystallography–based fragment screen and found submicromolar inhibitors by fragment linking. However, these compounds had poor membrane permeability and liabilities that complicated optimization. Here, we developed a shape-based virtual screening pipeline—FrankenROCS. We screened the Enamine high-throughput collection of 2.1 million compounds, selecting 39 compounds for testing, with the most potent binding with a 130 μM median inhibitory concentration (IC 50 ). We then paired FrankenROCS with an active learning algorithm (Thompson sampling) to efficiently search the Enamine REAL database of 22 billion molecules, testing 32 compounds with the most potent binding with a 220 μM IC 50 . Further optimization led to analogs with IC 50 values better than 10 μM. This lead series has improved membrane permeability and is poised for optimization. FrankenROCS is a scalable method for fragment linking to exploit synthesis-on-demand libraries.

Linking the Salmonella enterica 1,2-Propanediol Utilization Bacterial Microcompartment Shell to the Enzymatic Core via the Shell Protein PduB

Bacterial microcompartments (MCPs) are protein-based organelles that house the enzymatic machinery for metabolism of niche carbon sources, allowing enteric pathogens to outcompete native microbiota during host colonization. While much progress has been made toward understanding MCP biogenesis, questions still remain regarding the mechanism by which core MCP enzymes are enveloped within the MCP protein shell. Here, we explore the hypothesis that the shell protein PduB is responsible for linking the shell of the 1,2-propanediol utilization (Pdu) MCP from Salmonella enterica serovar Typhimurium LT2 to its enzymatic core. Using fluorescent reporters, we demonstrate that all members of the Pdu enzymatic core are encapsulated in Pdu MCPs. We also demonstrate that PduB is critical for linking the entire Pdu enzyme core to the MCP shell. Using MCP purifications, transmission electron microscopy, and fluorescence microscopy, we find that shell assembly can be decoupled from the enzymatic core, as apparently empty MCPs are formed in Salmonella strains lacking PduB. Mutagenesis studies reveal that PduB is incorporated into the Pdu MCP shell via a conserved, lysine-mediated hydrogen bonding mechanism. Finally, growth assays and system-level pathway modeling reveal that unencapsulated pathway performance is strongly impacted by enzyme concentration, highlighting the importance of minimizing polar effects when conducting these functional assays. Together, these results provide insight into the mechanism of enzyme encapsulation within Pdu MCPs and demonstrate that the process of enzyme encapsulation and shell assembly are separate processes in this system, a finding that will aid future efforts to understand MCP biogenesis.

Tracking the spatial and temporal links between late Cenozoic extension and magmatism in the Death Valley region (California, USA)

The relationship between late Cenozoic magmatism and extension in the central Basin and Range province (western United States) is complex, necessitating high-precision geochronology to understand its spatiotemporal connections. In the Death Valley region (California), the lack of high-precision U-Pb zircon ages has limited our understanding of the timing of pluton formation and its links to regional extension. We present new high-precision chemical abrasion-isotope dilution-thermal ionization mass spectrometry 206 Pb/ 238 U zircon ages and trace element analyses for eight Death Valley plutons. Our findings reveal three distinct phases of intrusive magmatism: (1) emplacement of shallow rapakivi granites at 13.2 Ma, (2) construction of the mid-crustal Black Mountains intrusive complex at 11.3 Ma, and (3) late emplacement of shallow, compositionally diverse intrusions at 8.2 Ma. A gap in zircon crystallization between 10 Ma and 8.2 Ma coincides with exhumation of the Black Mountains and a transition from sill to dike emplacement. The dominance of rapakivi granites in the Death Valley region, which is rare among Cenozoic granitoids, is likely a result of rapid crustal extension that induces adiabatic decompression. A comparison of the timing of volcanism, plutonism, and tectonic events in Death Valley reveals that intrusive magmatism closely tracks the locus of extension, underscoring the plutonic record as a vital link for understanding regional tectonics and changes in plate boundary dynamics during this period.

Deployed quantum link characterization via Bayesian ancilla-assisted process tomography

The development of large-scale quantum networks requires reliable quantum channels, the quality of which can be quantified by the framework of quantum process tomography. Here, in this work, we leverage ancilla-assisted process tomography (AAPT) and Bayesian inference to probe a 1.6 km deployed fiber-optic link. We send one of the two polarization-entangled photons at Alice in one building to Bob in another, exploiting the local qubit as an ancilla system to characterize the corresponding quantum channel. Monitoring over a 24 h period returns a steady process fidelity of 97.6(1)%, while controllable spectral filtering with passbands from 0.025 to 4.38 THz finds fidelities that first increase, and then level off with bandwidth, suggesting both stable operation with time and minimal polarization mode dispersion. To our knowledge, these results represent the first AAPT of a deployed quantum link, revealing a valuable tool for in situ analysis of entanglement-based quantum networks.

Topological Link Models of Multipartite Entanglement

We introduce a novel model of multipartite entanglement based on topological links, generalizing the graph/hypergraph entropy cone program. We demonstrate that there exist link representations of entropy vectors which provably cannot be represented by graphs or hypergraphs. Furthermore, we show that the contraction map proof method generalizes to the topological setting, though now requiring oracular solutions to well-known but difficult problems in knot theory.

Chemically Cross-Linked Cellulose Nanocrystal Aerogels for Effective Removal of Cation Dye

In this study, porous aerogels were prepared by directional freeze-drying via cross-linking cellulose nanocrystals (CNCs) with poly(methyl vinyl ether-co-maleic acid) (PMVEMA) and poly(ethylene glycol) (PEG). The thermal properties and physical adsorption performance toward cation methylene blue dye of the obtained CNC aerogels were investigated. The maximum degradation temperature was increased from 324°C of CNCs to 355°C of cross-linked CNC aerogels. The dye adsorption isotherm results showed that the maximum methylene blue adsorption capacity of CNC aerogels was 116.2 mg g −1 , according to the Langmuir model, which was mainly due to the electrostatic attractions between negatively charged carboxyl groups or sulfonate groups on the CNC aerogles and cation MB molecules. The reusability test showed that the CNC aerogels contained the same dye adsorption performance in five adsorption/desorption cycles. Overall, this study described an ideal alternative for water purification with high dye adsorption capacity and enhanced physical performance.
