Nutrient and moisture limitations reveal keystone metabolites linking rhizosphere metabolomes and microbiomes

Plants release a wealth of metabolites into the rhizosphere that can shape the composition and activity of microbial communities in response to environmental stress. The connection between rhizodeposition and rhizosphere microbiome succession has been suggested, particularly under environmental stress conditions, yet definitive evidence is scarce. In this study, we investigated the relationship between rhizosphere chemistry, microbiome dynamics, and abiotic stress in the bioenergy crop switchgrass grown in a marginal soil under nutrient-limited, moisture-limited, and nitrogen (N)-replete, phosphorus (P)-replete, and NP-replete conditions. We combined 16S rRNA amplicon sequencing and LC-MS/MS-based metabolomics to link rhizosphere microbial communities and metabolites. We identified significant changes in rhizosphere metabolite profiles in response to abiotic stress and linked them to changes in microbial communities using network analysis. N-limitation amplified the abundance of aromatic acids, pentoses, and their derivatives in the rhizosphere, and their enhanced availability was linked to the abundance of bacterial lineages from Acidobacteria, Verrucomicrobia, Planctomycetes, and Alphaproteobacteria. Conversely, N-amended conditions increased the availability of N-rich rhizosphere compounds, which coincided with proliferation of Actinobacteria. Treatments with contrasting N availability differed greatly in the abundance of potential keystone metabolites; serotonin and ectoine were particularly abundant in N-replete soils, while chlorogenic, cinnamic, and glucuronic acids were enriched in N-limited soils. Serotonin, the keystone metabolite we identified with the largest number of links to microbial taxa, significantly affected root architecture and growth of rhizosphere microorganisms, highlighting its potential to shape microbial community and mediate rhizosphere plant–microbe interactions.

Powdery mildew induces chloroplast storage lipid formation at the expense of host thylakoids to promote spore production

Powdery mildews are obligate biotrophic fungi that manipulate plant metabolism to supply lipids to the fungus, particularly during fungal asexual reproduction when lipid demand is high. We found levels of leaf storage lipids (triacylglycerols, TAGs) are 3.5-fold higher in whole Arabidopsis (Arabidopsis thaliana) leaves with a 15-fold increase in storage lipids at the infection site during fungal asexual reproduction. Lipid bodies, not observable in uninfected mature leaves, were found in and external to chloroplasts in mesophyll cells underlying the fungal feeding structure. Concomitantly, thylakoid disassembly occurred and thylakoid membrane lipid levels decreased. Genetic analyses showed that canonical endoplasmic reticulum TAG biosynthesis does not support powdery mildew spore production. Instead, Arabidopsis chloroplast-localized DIACYLGLYCEROL ACYLTRANSFERASE 3 (DGAT3) promoted fungal asexual reproduction. Consistent with the reported AtDGAT3 preference for 18:3 and 18:2 acyl substrates, which are dominant in thylakoid membrane lipids, dgat3 mutants exhibited a dramatic reduction in powdery mildew-induced chloroplast TAGs, attributable to decreases in TAG species largely comprised of 18:3 and 18:2 acyl substrates. This pathway for TAG biosynthesis in the chloroplast at the expense of thylakoids provides insights into obligate biotrophy and plant lipid metabolism, plasticity, and function. By understanding how photosynthetically active leaves can be converted into TAG producers, more sustainable and environmentally friendly plant oil production may be developed.

Direct evidence of ferromagnetism in MnSb 2 Te 4

We report the magnetic imaging of ferromagnetic domains in the van der Waals single crystal MnSb 2 Te 4 from two different sources using cryogenic magnetic force microscopy. The magnetic field dependence of the domains reveals very weak pinning of domain walls in MnSb 2 Te 4 , resulting in a negligibly small magnetic hysteresis loop. Furthermore, the temperature dependence of the domain contrast reveals a mean field like behavior, in good agreement with that of bulk magnetization measurements.

Robust A -Type Order and Spin-Flop Transition on the Surface of the Antiferromagnetic Topological Insulator MnBi 2 Te 4

Here, we present microscopic evidence of the persistence of uniaxial A-type antiferromagnetic order to the surface layers of MnBi 2 Te 4 single crystals using magnetic force microscopy. Our results reveal termination-dependent magnetic contrast across both surface step edges and domain walls, which can be screened by thin layers of soft magnetism. The robust surface A-type order is further corroborated by the observation of termination-dependent surface spin-flop transitions, which have been theoretically proposed decades ago. Our results not only provide key ingredients for understanding the electronic properties of the antiferromagnetic topological insulator MnBi 2 Te 4 , but also open a new paradigm for exploring intrinsic surface metamagnetic transitions in natural antiferromagnets.

Two-channel anomalous Hall effect in SrRuO 3

The Hall effect in SrRuO 3 thin films near the thickness limit for ferromagnetism shows an extra peak in addition to the ordinary and anomalous Hall effects. This extra peak has been attributed to a topological Hall effect due to two-dimensional skyrmions in the film around the coercive field; however, the sign of the anomalous Hall effect in SrRuO 3 can change as a function of saturation magnetization. Here we report Hall peaks in SrRuO 3 in which volumetric magnetometry measurements and magnetic force microscopy indicate that the peaks result from the superposition of two anomalous Hall channels with opposite sign. These channels likely form due to thickness variations in SrRuO 3 , creating two spatially separated magnetic regions with different saturation magnetizations and coercive fields. Overall, the results are central to the development of strongly correlated materials for spintronics.

The Arctic

Arctic observations in 2023 provided clear evidence of rapid and pronounced climate and environmental change, shaped by past and ongoing human activities that release greenhouse gases into the atmosphere and push the broader Earth system into uncharted territory. This chapter provides a snapshot of 2023 and summarizes decades-long trends observed across the Arctic, including warming surface air and sea-surface temperatures, decreasing snow cover, diminishing sea ice, thawing permafrost, and continued mass loss from the Greenland Ice Sheet and Arctic glaciers. These changes are driving a transition to a wetter, greener, and less frozen Arctic, with serious implications for Arctic peoples and ecosystems, as well as for low- and midlatitudes

CFD-Enhanced Calibration of a Multihole Probe for Small Uncrewed Aircraft Systems

A robust calibration method with a 9-hole probe (9HP) for inertial wind vector measurements from a small uncrewed aircraft system (sUAS) is presented. Calibration accuracy is improved by using computational fluid dynamics (CFD) to estimate corrections, such as those for test section blockage, that are generally applied in wind tunnel testing. A method for estimating experimental bias is presented to account for flow variations over the pressure taps as the 9HP is repositioned in the test section. Installation effects from flow over the airframe and upwash produced by the wings are estimated from CFD simulations of the entire airframe at the expected cruise speed. An initial CFD analysis has demonstrated a quantifiable linear relationship between the measured and actual angle of attack as measured from the relative wind frame. In conclusion, the objective of these additional steps is to increase the accuracy of 9HP calibration.

Overview of leakage scenarios in supervised machine learning

Machine learning (ML) provides powerful tools for predictive modeling. ML’s popularity stems from the promise of sample-level prediction with applications across a variety of fields from physics and marketing to healthcare. However, if not properly implemented and evaluated, ML pipelines may contain leakage typically resulting in overoptimistic performance estimates and failure to generalize to new data. This can have severe negative financial and societal implications. Our aim is to expand understanding associated with causes leading to leakage when designing, implementing, and evaluating ML pipelines. Illustrated by concrete examples, we provide a comprehensive overview and discussion of various types of leakage that may arise in ML pipelines.

NARUC grid data sharing playbook

In 2022, the National Association of Regulatory Utility Commissioners (NARUC) launched an initiative to support its members in addressing issues related to grid data sharing. The Grid Data Sharing Collaborative was funded by the DOE’s Office of Electricity and Office of Cybersecurity, Energy Security, and Emergency Response (CESER). NARUC invited programmatic, policy, technical, and cybersecurity subject matter experts from public utility commissions, utilities, non-governmental organizations, energy service companies, and DOE to join the two-year Grid Data Sharing Collaborative to help develop a flexible framework for states to use as a starting point when navigating complex decision-making inherent in grid data sharing. The framework took shape through a series of intensive workshops during which Collaborative participants explored illustrative use cases to identify data needs, articulate the benefits and risks of sharing such data, and assess the trade-offs. Along the way, participants offered suggestions for how the framework could be used in practice. The purpose of this playbook is to describe the elements of the Grid Data Sharing Framework and to begin supporting its implementation.

The S-PLUS Fifth Data Release: Over 4500 Square Degrees of the Southern Sky and a Multicolor View of the Hydra and Antlia Galaxy Clusters

Abstract We present the fifth data release (DR5) of the Southern Photometric Local Universe Survey (S-PLUS), covering 4592 deg 2 across 2491 fields. Observations were conducted with the T80-South, a Brazilian robotic telescope equipped with the Javalambre 12-filter system, containing five broadband and seven narrowband filters. Data products feature FITS images and extensive catalogs containing fluxes, magnitudes, and shape parameters for over 113 million detections. In addition, several value-added catalogs are provided, offering photometric redshifts (photo- zs ), object classifications, masks, and extinction coefficients. For the first time, this release includes coverage of 110 deg 2 along the Galactic disk, facilitating new research into Galactic structure and stellar populations. The release also provides full coverage of the Hydra Supercluster and numerous other nearby clusters with improved data reduction and calibration, enhancing photo- z accuracy, which is vital for large-scale structure studies. A preliminary analysis of the Hydra and Antlia galaxy clusters up to 5 × R 200 yields an updated catalog of 1706 cluster members based on both spectroscopic and high-quality photo- zs . Our photometric data shows that both clusters have a similar proportion of galaxies with an H α excess relative to their clustercentric distance, though Hydra has a higher fraction near its center. Additionally, the spatial distribution of all objects in our sample highlights a bridge connecting both clusters. We verify that S-PLUS DR5 provides a solid foundation for future scientific investigations, ranging from solar system studies to cosmology.
