Effect of foot-and-mouth disease virus 3C protease B 2 β-strand proline mutagenesis on expression and processing of the P1 polypeptide using a plasmid expression vector

The production of experimental molecular vaccines against foot-and-mouth disease virus utilizes the viral encoded 3C protease for processing of the P1 polyprotein. Expression of wild type 3C protease is detrimental to host cells. The molecular vaccine constructs containing the 3C protease L127P mutant significantly reduce adverse effects associated with protease expression while retaining the ability to process and assemble virus-like particles. In published 3C protease crystal structures, the L127 residue is contained within the B 2 β-strand as part of the A 2 –B 2 β-sheet. To provide insight into the mechanism by which the L127P mutant alters the properties of the 3C protease, we performed scanning proline mutagenesis of residues 123–128 of the B 2 β-strand and monitored expression and P1 processing. Simultaneously, we utilized random mutagenesis of the full 3C sequence to identify additional mutations presenting a phenotype similar to the L127P mutation. Six of the tested mutants enhanced expression over wild type, and the I22P, T100P and V124P mutations surpassed the L127P mutation in certain cell lines. These data are interpreted in conjunction with published 3C protease crystal structures to provide insight into the mechanism by which these mutations enhance expression.

User's Guide for Monthly Vector Wind Profile Model

The background, theoretical concepts, and methodology for construction of vector wind profiles based on a statistical model are presented. The derived monthly vector wind profiles are to be applied by the launch vehicle design community for establishing realistic estimates of critical vehicle design parameter dispersions related to wind profile dispersions. During initial studies a number of months are used to establish the model profiles that produce the largest monthly dispersions of ascent vehicle aerodynamic load indicators. The largest monthly dispersions for wind, which occur during the winter high-wind months, are used for establishing the design reference dispersions for the aerodynamic load indicators. This document includes a description of the computational process for the vector wind model including specification of input data, parameter settings, and output data formats. Sample output data listings are provided to aid the user in the verification of test output.

Scalable Risk Assessment of Rare Events in Power Systems With Uncertain Wind Generation and Loads

Risk assessment of rare events has become increasingly important in power system planning and operation with the increasing integration of renewable energy and the presence of system uncertainties. However, quantifying the risk posed by rare events via the traditional method, i.e., Monte Carlo sampling (MCS), incurs substantial computational expense stemming from the vast ensemble of power flow simulations. To accelerate the assessment, this paper proposes a Deep Neural Network (DNN)-kernelized vector-valued Gaussian Process (VVGP) approach with excellent computational efficiency while maintaining high accuracy. Consequently, serving as a surrogate model for the power flow solver, the DNN-kernelized VVGP enables significantly faster but accurate risk assessment compared to the power flow solver. The developed surrogate model evaluates low-order N - k events that contain more than 90% instances by adeptly capturing the topological features while the high-order N - k events are assessed via a power flow solver, thereby striking a balance between computational efficiency and uncertainty quantification accuracy. Moreover, the model incorporates a Support Vector Machine (SVM) classifier to resample concerning low-probability tail events to counteract the biases potentially introduced during the DNN-kernelized VVGP evaluations. Simulations conducted on the modified IEEE 24-bus, 118-bus, and European 1354-bus systems demonstrate that the proposed method maintains the accuracy benchmark set by MCS while significantly reducing computational demands in large-scale power systems as compared to other state-of-the-art methods.

Algorithms for solving large sparse systems of simultaneous linear equations on vector processors

Very efficient algorithms for solving large sparse systems of simultaneous linear equations have been developed for serial processing computers. These involve a reordering of matrix rows and columns in order to obtain a near triangular pattern of nonzero elements. Then an LU factorization is developed to represent the matrix inverse in terms of a sequence of elementary Gaussian eliminations, or pivots. In this paper it is shown how these algorithms are adapted for efficient implementation on vector processors. Results obtained on the CYBER 200 Model 205 are presented for a series of large test problems which show the comparative advantages of the triangularization and vector processing algorithms.

Searches for electroweak production of supersymmetric particles with compressed mass spectra in s = 13 TeV p p collisions with the ATLAS detector

This paper presents results of searches for electroweak production of supersymmetric particles in models with compressed mass spectra. The searches use 139/fb of sqrt(s) = 13 TeV proton-proton collision data collected by the ATLAS experiment at the Large Hadron Collider. Events with missing transverse momentum and two same-flavor, oppositely charged, low transverse momentum leptons are selected, and are further categorized by the presence of hadronic activity from initial-state radiation or a topology compatible with vector-boson fusion processes. The data are found to be consistent with predictions from the Standard Model. The results are interpreted using simplified models of R-parity-conserving supersymmetry in which the lightest supersymmetric partner is a neutralino with a mass similar to the lightest chargino, the second-to-lightest neutralino or the slepton. Lower limits on the masses of charginos in different simplified models range from 193 GeV to 240 GeV for moderate mass splittings, and extend down to mass splittings of 1.5 GeV to 2.4 GeV at the LEP chargino bounds (92.4 GeV). Similar lower limits on degenerate light-flavor sleptons extend up to masses of 251 GeV and down to mass splittings of 550 MeV. Constraints on vector-boson fusion production of electroweak SUSY states are also presented.

Search for nonresonant Higgs boson pair production in final state with two bottom quarks and two tau leptons in proton-proton collisions at $\sqrt{s}$ = 13 TeV

A search for the nonresonant production of Higgs boson pairs (HH ) via gluon-gluon and vector boson fusion processes in final states with two bottom quarks and two tau leptons is presented. The search uses data from proton-proton collisions at a center-of-mass energy of $\sqrt{s}$ = 13 TeV recorded with the CMS detector at the LHC, corresponding to an integrated luminosity of 138 fb -1 . Events in which at least one tau lepton decays hadronically are considered and multiple machine learning techniques are used to identify and extract the signal. The data are found to be consistent, within uncertainties, with the standard model (SM) predictions. Upper limits on the HH production cross section are set to constrain the parameter space for anomalous Higgs boson couplings. The observed (expected) upper limit at 95% confidence level corresponds to 3.3 (5.2) times the SM prediction for the inclusive HH cross section and to 124 (154) times the SM prediction for the vector boson fusion HH cross section. At 95% confidence level, the Higgs field self-coupling is constrained to be within -1.7 and 8.7 times the SM expectation, and the coupling of two Higgs bosons to two vector bosons is constrained to be within -0.4 and 2.6 times the SM expectation.

Search for nonresonant Higgs boson pair production in final states with two bottom quarks and two photons in proton-proton collisions at $ \sqrt{s} $ = 13 TeV

A search for nonresonant production of Higgs boson pairs via gluon-gluon and vector boson fusion processes in final states with two bottom quarks and two photons is presented. The search uses data from proton-proton collisions at a center-of-mass energy of $ \sqrt{s} $ = 13 TeV recorded with the CMS detector at the LHC, corresponding to an integrated luminosity of 137 fb$^{−1}$. No significant deviation from the background-only hypothesis is observed. An upper limit at 95% confidence level is set on the product of the Higgs boson pair production cross section and branching fraction into $ \gamma \gamma \mathrm{b}\overline{\mathrm{b}} $. The observed (expected) upper limit is determined to be 0.67 (0.45) fb, which corresponds to 7.7 (5.2) times the standard model prediction. This search has the highest sensitivity to Higgs boson pair production to date. Assuming all other Higgs boson couplings are equal to their values in the standard model, the observed coupling modifiers of the trilinear Higgs boson self-coupling κ$_{λ}$ and the coupling between a pair of Higgs bosons and a pair of vector bosons c$_{2V}$ are constrained within the ranges −3.3 < κ$_{λ}$< 8.5 and −1.3 < c$_{2V}$< 3.5 at 95% confidence level. Constraints on κ$_{λ}$ are also set by combining this analysis with a search for single Higgs bosons decaying to two photons, produced in association with top quark-antiquark pairs, and by performing a simultaneous fit of κ$_{λ}$ and the top quark Yukawa coupling modifier κ$_{t}$.[graphic not available: see fulltext]

Optimizing cloud motion estimation on the edge with phase correlation and optical flow

Abstract. Phase correlation (PC) is a well-known method for estimating cloud motion vectors (CMVs) from infrared and visible spectrum images. Commonly, phase shift is computed in the small blocks of the images using the fast Fourier transform. In this study, we investigate the performance and the stability of the blockwise PC method by changing the block size, the frame interval, and combinations of red, green, and blue (RGB) channels from the total sky imager (TSI) at the United States Atmospheric Radiation Measurement user facility's Southern Great Plains site. We find that shorter frame intervals, followed by larger block sizes, are responsible for stable estimates of the CMV, as suggested by the higher autocorrelations. The choice of RGB channels has a limited effect on the quality of CMVs, and the red and the grayscale images are marginally more reliable than the other combinations during rapidly evolving low-level clouds. The stability of CMVs was tested at different image resolutions with an implementation of the optimized algorithm on the Sage cyberinfrastructure test bed. We find that doubling the frame rate outperforms quadrupling the image resolution in achieving CMV stability. The correlations of CMVs with the wind data are significant in the range of 0.38–0.59 with a 95 % confidence interval, despite the uncertainties and limitations of both datasets. A comparison of the PC method with constructed data and the optical flow method suggests that the post-processing of the vector field has a significant effect on the quality of the CMV. The raindrop-contaminated images can be identified by the rotation of the TSI mirror in the motion field. The results of this study are critical to optimizing algorithms for edge-computing sensor systems.

Measurement and interpretation of same-sign W boson pair production in association with two jets in $pp$ collisions at $\sqrt{s}$ = 13 TeV with the ATLAS detector

This paper presents the measurement of fiducial and differential cross sections for both the inclusive and electroweak production of a same-sign W-boson pair in association with two jets (W ± W ± jj) using 139 fb –1 of proton-proton collision data recorded at a centre-of-mass energy of $\sqrt{s}$ = 13 TeV by the ATLAS detector at the Large Hadron Collider. The analysis is performed by selecting two same-charge leptons, electron or muon, and at least two jets with large invariant mass and a large rapidity difference. The measured fiducial cross sections for electroweak and inclusive W ± W ± jj production are 2.92 ± 0.22 (stat.) ± 0.19 (syst.) fb and 3.38 ± 0.22 (stat.) ± 0.19 (syst.) fb, respectively, in agreement with Standard Model predictions. The measurements are used to constrain anomalous quartic gauge couplings by extracting 95% confidence level intervals on dimension-8 operators. A search for doubly charged Higgs bosons H ±± that are produced in vector-boson fusion processes and decay into a same-sign W boson pair is performed. The largest deviation from the Standard Model occurs for an H ±± mass near 450 GeV, with a global significance of 2.5 standard deviations.

Differential cross-section measurements of the production of four charged leptons in association with two jets using the ATLAS detector

Differential cross-sections are measured for the production of four charged leptons in association with two jets. These measurements are sensitive to final states in which the jets are produced via the strong interaction as well as to the purely-electroweak vector boson scattering process. The analysis is performed using proton-proton collision data collected by ATLAS at $\sqrt{s}$ = 13 TeV and with an integrated luminosity of 140 fb –1 . The data are corrected for the effects of detector inefficiency and resolution and are compared to state-of-the-art Monte Carlo event generator predictions. The differential cross-sections are used to search for anomalous weak-boson self-interactions that are induced by dimension-six and dimension-eight operators in Standard Model effective field theory.

Search for Higgs boson production in association with a high-energy photon via vector-boson fusion with decay into bottom quark pairs at $\sqrt{s}$ = 13 TeV with the ATLAS detector

A search is presented for the production of the Standard Model Higgs boson in association with a high-energy photon. With a focus on the vector-boson fusion process and the dominant Higgs boson decay into b-quark pairs, the search benefits from a large reduction of multijet background compared to more inclusive searches. Results are reported from the analysis of 132 fb –1 of pp collision data at $\sqrt{s}$ = 13 TeV collected with the ATLAS detector at the LHC. The measured Higgs boson signal yield in this final-state signature is 1.3 ± 1.0 times the Standard Model prediction. The observed significance of the Higgs boson signal above the background is 1.3 standard deviations, compared to an expected significance of 1.0 standard deviations.

Leptonic scalars at the LHC

We explore the collider prospects of neutrino non-standard interaction with a Standard Model (SM) gauge-singlet leptonic scalar ϕ carrying two units of lepton-number-charge. These leptonic scalars are forbidden from interacting with the SM fermions at the renormalizable level and, if one allows for higher-dimensional operators, couple predominantly to SM neutrinos. For masses at or below the electroweak scale, ϕ decays exclusively into neutrinos. Its characteristic production signature at hadron collider experiments like the LHC would be via the vector boson fusion process and leads to same-sign dileptons, two forward jets in opposite hemispheres, and missing transverse energy, i.e., $$ pp\to {\mathrm{\ell}}_{\alpha}^{\pm }{\mathrm{\ell}}_{\beta}^{\pm } jj+{E}_T^{\mathrm{miss}}\left(\alpha, \beta =e,\mu, \tau \right) $$ pp → ℓ α ± ℓ β ± jj + E T miss α β = e μ τ . Exploiting the final states of electrons and muons, we estimate, for the first time, the sensitivity of the LHC to these lepton-number-charged scalars. We show that the LHC sensitivity is largely complementary to that of low-energy precision measurements of the decays of charged leptons, charged mesons, W , Z and the SM Higgs boson, as well as the neutrino beam experiments like MINOS, and searches for neutrino self-interactions at IceCube and in cosmological observations. For ϕ mass larger than roughly 10 GeV, our projected LHC sensitivity would surpass all existing bounds.

${\Lambda}_c^{+}$ polarimetry using the dominant hadronic mode

The polarimeter vector field for multibody decays of a spin-half baryon is intro duced as a generalisation of the baryon asymmetry parameters. Using a recent amplitude analysis of the ${\Lambda}_c^{+}$ → $pK^-π^+$ decay performed at the LHCb experiment, we compute the distribution of the kinematic-dependent polarimeter vector for this process in the space of Mandelstam variables to express the polarised decay rate in a model-agnostic form. The obtained representation can facilitate polarisation measurements of the ${\Lambda}_c^{+}$ baryon and eases inclusion of the ${\Lambda}_c^{+}$ → $pK^-π^+$ decay mode in hadronic amplitude analyses.

Differential cross-sections for events with missing transverse momentum and jets measured with the ATLAS detector in 13 TeV proton-proton collisions

Measurements of inclusive, differential cross-sections for the production of events with missing transverse momentum in association with jets in proton-proton collisions at √s = 13 TeV are presented. The measurements are made with the ATLAS detector using an integrated luminosity of 140 fb –1 and include measurements of dijet distributions in a region in which vector-boson fusion processes are enhanced. They are unfolded to correct for detector resolution and efficiency within the fiducial acceptance, and are designed to allow robust comparisons with a wide range of theoretical predictions. A measurement of differential cross sections for the Z → νν process is made. The measurements are generally well-described by Standard Model predictions except for the dijet invariant mass distribution. Auxiliary measurements of the hadronic system recoiling against isolated leptons, and photons, are also made in the same phase space. Ratios between the measured distributions are then derived, to take advantage of cancellations in modelling effects and some of the major systematic uncertainties. These measurements are sensitive to new phenomena, and provide a mechanism to easily set constraints on phenomenological models. To illustrate the robustness of the approach, these ratios are compared with two common Dark Matter models, where the constraints derived from the measurement are comparable to those set by dedicated detector-level searches.

Sensitivity to longitudinal vector boson scattering and doubly charged Higgs boson production in $W^± W^± jj$ at future hadron colliders

We study the sensitivity to longitudinal vector boson scattering at a 27-, 50-, and 100-TeV pp collider using events containing two leptonically decaying same-electric-charge 𝑊 bosons produced in association with two jets. The baseline FCC-hh detector parametrization within the D ELPHES framework is used under the assumption of fully efficient pileup mitigation. A tightly constrained phase space with a dijet mass greater than 2 TeV is considered in order to suppress the impact of potential instrumental backgrounds. Based on this setup, the expected sensitivity to the production of longitudinally polarized same-sign 𝑊 boson pairs is evaluated. Additionally, expected limits are set on doubly charged Higgs bosons produced via vector boson fusion processes and decaying to same-sign 𝑊 boson pairs.

A Method of Ground-Based Cloud Motion Predict: CCLSTM + SR-Net

Ground-based cloud images can provide information on weather and cloud conditions, which play an important role in cloud cover monitoring and photovoltaic power generation forecasting. However, the cloud motion prediction of ground-based cloud images still lacks advanced and complete methods, and traditional technologies based on image processing and motion vector calculation are difficult to predict cloud morphological changes. In this paper, we propose a cloud motion prediction method based on Cascade Causal Long Short-Term Memory (CCLSTM) and Super-Resolution Network (SR-Net). Firstly, CCLSTM is used to estimate the shape and speed of cloud motion. Secondly, the Super-Resolution Network is built based on perceptual losses to reconstruct the result of CCLSTM and, finally, make it clearer. We tested our method on Atmospheric Radiation Measurement (ARM) Climate Research Facility TSI (total sky imager) images. The experiments showed that the method is able to predict the sky cloud changes in the next few steps.
