Diversity in the Common Fold: Structural Insights into Class D β-Lactamases from Gram-Negative Pathogens

Class D β-lactamases (DBLs) represent a major threat to antibiotic efficacy by hydrolyzing β-lactam drugs, including last-resort carbapenems, thereby driving antimicrobial resistance in Gram-negative bacteria. The enzymes share a structurally conserved two-domain α/β architecture with seven active-site motifs and three flexible extended loops (the P-loop, Ω-loop, and newly designated B-loop) that surround the active site. While each of these loops is known to influence enzyme function, their coordinated roles have not been fully elucidated. To investigate the significance of their interplay, we compared the sequences and crystal structures of 40 DBLs from clinically relevant Gram-negative pathogens and performed molecular dynamics simulations on selected representatives. Combined structural and dynamical analyses revealed a strong correlation between B-loop architecture and carbapenemase activity in the pathogens Klebsiella and Acinetobacter, particularly regarding loop length and spatial organization. These findings emphasize the B-loop’s critical contribution, in concert with the P- and Ω-loops, in tuning active site versatility, substrate recognition, catalytic activity, and structural stability. A deeper understanding of how these motifs and loops govern DBL function may inform the development of novel antibiotics and inhibitors targeting this class of enzymes.

Metal tube can be folded for compact stowage, is self-erecting

Metal tube configuration reduces the section modulus to that of a thin plate, thus permitting the section to be bent into a coil for stowage in limited space without destructive yielding of the material. It is readily released to serve as a rigid fluid transportation conduit or structural member.

Folded stick module

Integrated circuit modules can be compactly assembled into short-run complex electronic assemblies by mounting them on a Stick module. This module provides a method of high-density packaging for industrial operations that require the fabrication of compact circuitry configurations.

Self-Alining, Latching Joint For Folding Structural Elements

Structural column elements assembled quickly and easily with aid of new center joint. Joint alines column elements automatically and fastens them together securely. Tapered half columns are stacked like paper cups, unfolded, and connected to other similar elements to form truss structures.

Dynamics and structure of the Alpine Fold Belt

The structure and present-day dynamics of the Alps interms of geodesy and gravimetry are discusssed. A strong correlation of precise leveling and isostatic gravity along the central Alpine chain, especially in Canton Graubunden, East Switzerland are shown. It is assumed that the uplift is partly controlled by isostatic rebound effects. Field observations indicate that these phenomena are still active in the Alps. The study of the uplift processes by applying a number of geodetic and gravimetric measuring techniques, such as the determination of nonperiodic secular variations of gravity, of the deflections of the vertical and tilt changes monitored by hydrostatic leveling is proposed.

Analysis of a single-fold deployable truss beam preloaded by extension of selected face diagonal members

A technique for preloading a deployable box truss beam by extension of one face diagonal per bay was studied to determine if it would result in uniform loading of truss joints without causing excessive truss deformations. Results indicate that it is possible to accomplish uniform loading in the beam region way from beam boundaries, whereas in the regions near boundaries the member loading becomes non-uniform with magnitudes greater than those in the uniform load region. Also, the type of deformation which results in the beam depends on the pattern of preloaded members.

Folding, articulated, square truss

A larger, stronger deployable boom was developed to handle the requirements of larger, heavier payloads in space. The main components of the boom and its deployer are described and their functions explained. Desirable features of the boom are identified and physical properties are reported.

Correlations between ozone and carbon monoxide in the lower stratosphere, folded tropopause, and maritime troposphere

The variations of ozone and carbon monoxide with respect to the atmospheric flow structure are investigated for two aircraft flights: one in the upper troposphere/lower stratosphere over the continental United States on April 20, 1984, and the other in the marine troposphere over the eastern Pacific on April 26, 1984. The high frequency (1 Hz) and precision of the measurements allow detailed analysis of the species distribution down to length scales of the order of 1 km. Correlations between the two species are made on large, medium, and small scales of motion, and the results are interpreted. In general, the correlations are negative with some regions of positive correlation. The interpretation of these results is discussed.

Folding Truss Structure

Concept for foldable and deployable truss offers advantages of strength, rigidity, and mechanical simplicity. Structure consists of series of boxlike bays with 9-ft sides. Each box has panels on top and bottom and two sides. Two remaining sides open. Panels hinged at connecting edges. Adapted to terrestrial transportable structures, scaffolds, cranes, and rows of cubicles.

Lighted, Folding Inspection Mirror

Compact, inexpensive tool used in place of expensive borescopes. Shortens inspection/photographing process. Includes two small metal or glass mirrors hinged together. Two 3-V light bulbs attached along edges of one mirror and connected to battery of two cells. Inserted into narrow opening of clevis or tand, and surface viewed and photographed in opposite mirror. Useful in assembly of segments of solid rocket motors as well as in postflight assessment, engineering evaluation, and refurbishment. Also applied in general to inspection and photographing of inner sealing surfaces to which access difficult.

Synchronously deployable double fold beam and planar truss structure

A deployable structure that synchronously deploys in both length and width is disclosed which is suitable for use as a structural component for orbiting space stations or large satellites. The structure is designed with maximum packing efficiency so that large structures may be collapsed and transported in the cargo bay of the Space Shuttle. The synchronous deployment feature allows the structure to be easily deployed in space by two astronauts, without a complex deployment mechanism. The structure is made up of interconnected structural units, each generally in the shape of a parallelepiped. The structural units are constructed of structural members connected with hinged and fixed connections at connection nodes in each corner of the parallelepiped. Diagonal members along each face of the parallelepiped provide structural rigidity and are equipped with mid-length, self-locking hinges to allow the structure to collapse. The structure is designed so that all hinged connections may be made with simple clevis-type hinges requiring only a single degree of freedom, and each hinge pin is located along the centerline of its structural member for increased strength and stiffness.

Observations of Convective and Dynamical Instabilities in Tropopause Folds and their Contribution to Stratosphere-Troposphere Exchange

SONEX has provided a number of opportunities to learn more about the fine-scale structure of the atmosphere. Coupled with the MOZAIC work on layers we can synthesize a new 3-D view of the fine scales that influence atmospheric chemistry. As a side issue we are also relating the features to clear air turbulence which would be, we think, a useful connection for commercial aviation.
